The effect of docosahexaenoic acid (DHA) on maze-learning ability in aged mice was investigated. Aged mice fed a diet deficient in n-3 fatty acid received a semi-purified diet of 0.5, 1 and 2% DHA-ethyl ester (DHA-EE) for 4 months. Maze-learning ability was assessed at 3 months after the start of the experiment. The time required to reach the maze exit and the number of times that a mouse strayed into blind alleys in the maze were measured in 3 trials conducted at 4-day intervals. The time was significantly less in the DHA-EE 0.5% and DHA-EE 2% groups than in the control group (p<0.05). The number of times mice strayed into blind alleys in the maze was significantly lower in the DHA-EE 2% group than in the control group (p<0.05). Mice fed DHA showed increased DHA and decreased level of arachidonic acid (AA) in the brain. These results suggest that the intake of a 2% DHA diet improves learning ability in aged mice, which is associated with a higher DHA content in the brain.

The aim of this study was to discover whether Yukmijihwangtang which was added Plygalae Radix, Acori graminei Rhizoma, has antioxidant effects on aged senescence-accelerated mouse (SAM) P8. The control group used 12 months old SAM P8 and the experimental group was treated with Yukmijihwangtang extracts from 8 to 12 months old. In the liver, the MnSOD of the control group were approximately 11% higher than that of the experimental group treated with herbal extracts, but CuZnSOD activities of the experimental group were approximately 20% higher than that of the control group. The catalase activities of the experimental group was approximately 67% higher than that of the control group. The hepatocytes, in the control group compared with the experimental group, contained the dilated cisternae of rough endoplasmic reticulum and the abnormal mitochondira which had electron-dense matrix and indistinguishable cristae. The inner cavity of the smooth endoplasmic reticulum were slightly dilated, and a number of vacuoles and transitional vesicles occured in of the hepatocytes of the control group. These results suggest that the herbal extract has an antioxidant effects on the liver of SAM P8.
Cardiovascular disease is the prime cause of morbidity and mortality and the population ages that may contribute to increase in the occurrence of cardiovascular disease. Arginase upregulation is associated with impaired endothelial function in aged vascular system and thus may contribute to cardiovascular disease. According to recent research, Korean Red Ginseng water extract (KRGE) may reduce cardiovascular disease risk by improving vascular system health. The purpose of this study was to examine mechanisms contributing to age-related vascular endothelial dysfunction and to determine whether KRGE improves these functions in aged mice. Young ($10{\pm}3$ weeks) and aged ($55{\pm}5$ weeks) male mice (C57BL/6J) were orally administered 0, 10, or 20 mg/mouse/day of KRGE for 4 weeks. Animals were sacrificed and the aortas were removed. Endothelial arginase activity, nitric oxide (NO) generation and reactive oxygen species (ROS) production, endothelial nitric oxide synthase (eNOS) coupling, vascular tension, and plasma peroxynitrite production were measured. KRGE attenuated arginase activity, restored nitric oxide (NO) generation, reduced ROS production, and enhanced eNOS coupling in aged mice. KRGE also improved vascular tension in aged vessels, as indicated by increased acetylcholine-induced vasorelaxation and improved phenylephrine-stimulated vasoconstriction. Furthermore, KRGE prevented plasma peroxynitrite formation in aged mice, indicating reduced lipid peroxidation. These results suggest KRGE exerts vasoprotective effects by inhibiting arginase activity and augmenting NO signaling and may be a useful treatment for age-dependent vascular diseases.
The fat pad defined as the epididymal fat is located at the head part of the epididymis. However, another fat mass is present near the caudal epididymis, named tail epididymal fat. The present research was focused to determine the expression of adipocyte-associated molecules in the mouse tail epididymal fat at different postnatal ages, including 2, 5, 8, and 12 months of age. The quantitative real-time PCR analysis showed continuous increases of expression levels of delta like non-canonical Notch ligand 1, leptin, and resistin as postnatally aged. The transcript level of peroxisome proliferator-activated receptor gamma was the highest at 5 months of age, remaining at a steady level until 12 months of age. Expression levels of fatty acid binding protein 4, leptin, and adiponectin were significantly increased until 8 months of age, persisting the level at 12 months of age. The transcript level of fatty acid synthase was significantly increased at 8 months of age, without a further change of the level at 12 months of age. These findings exhibited the expression of adipocyte-associated genes which were also detected at the ordinary epididymal fat pad. However, expression patterns of these genes in the tail epididymal fat are different with those in the distal and proximal epididymal fat, suggesting distinct characteristics and/or functions of the tail epididymal fat.

Aging is a major risk factor for common neurodegenerative diseases. Although multiple molecular, cellular, structural, and functional changes occur in the brain during aging, the involvement of caveolin-2 (Cav-2) in brain ageing remains unknown. We investigated Cav-2 expression in brains of aged mice and its effects on endothelial cells. The human umbilical vein endothelial cells (HUVECs) showed decreased THP-1 adhesion and infiltration when treated with Cav-2 siRNA compared to control siRNA. In contrast, Cav-2 overexpression increased THP-1 adhesion and infiltration in HUVECs. Increased expression of Cav-2 and iba-1 was observed in brains of old mice. Moreover, there were fewer iba-1-positive cells in the brains of aged Cav-2 knockout (KO) mice than of wild-type aged mice. The levels of several chemokines were higher in brains of aged wild-type mice than in young wild-type mice; moreover, chemokine levels were significantly lower in brains of young mice as well as aged Cav-2 KO mice than in their wild-type counterparts. Expression of PECAM1 and VE-cadherin proteins increased in brains of old wild-type mice but was barely detected in brains of young wild-type and Cav-2 KO mice. Collectively, our results suggest that Cav-2 expression increases in the endothelial cells of aged brain, and promotes leukocyte infiltration and age-associated neuroinflammation.
Background: Sarcopenia is a new and emerging risk factor aggravating the quality of life of elderly population. Because Korean Red Ginseng (RG) is known to have a great effect on relieving fatigue and enhancing physical performance, it is invaluable to examine its potential as an anti-sarcopenic drug. Methods: Anti-sarcopenic effect of non-saponin fraction of Korean Red Ginseng (RGNS) was evaluated in C2C12 myoblasts treated with C2-ceramide to induce senescence phenotypes, and 22-month-old mice fed with chow diet containing 2% RGNS (w/w) for 4 further months. Results: The RGNS treatment significantly alleviated cellular senescence indicated by intracellular lipid accumulation, increased amount of lysosomal β-galactosidase, and reduced proliferative capacity in C2C12 myoblasts. This effect was not observed with saponin fraction. In an aged mouse, the 4-month-RGNS diet significantly improved aging-associated loss of muscle mass and strength, assessed by the weights of hindlimb skeletal muscles such as tibialis anterior (TA), extensor digitorum longus (EDL), gastrocnemius (GN) and soleus (SOL), and the cross-sectional area (CSA) of SOL muscle, and the behaviors in grip strength and hanging wire tests, respectively. During the same period, an aging-associated shift of fast-to slow-twitch muscle in SOL muscle was also retarded by the RGNS treatment. Conclusions: These findings suggested that the long-term diet of RGNS significantly prevented aging-associated muscle atrophy and reduced physical performance, and thus RGNS has a strong potential to be developed as a drug that prevents or improves sarcopenia.

Gonadotropin-releasing hormone (GnRH), a critical hormone produced in the hypothalamus, is essential for regulating reproductive processes. It has also been demonstrated the presence of GnRH and its receptors (GnRHR) in ovarian and uterine tissues, but little was known about the regulation mechanism of their expression in these organs and ovarian aging. Therefore, the aim of this study was to investigate the expression of GnRHR in the ovary and uterus of mice, particularly after high-dose gonadotropin treatments and in relation to aging. Quantitative real-time-PCR (qRT-PCR) revealed that pituitary gland had the highest GnRHR expression in both young and aged mice. In addition, liver expression was higher in young mice, whereas thymus expression was higher in aged mice. GnRHR mRNA was present in the ovaries of both young and aged mice but nearly undetectable in the uterus of aged mice. We next examined the expression of GnRHR in the ovary and uterus in response to high-dose administration of pregnant mare serum gonadotropin (PMSG). After PMSG administration, GnRH mRNA levels were significantly decreased in the ovary but increased in the uterus. The expression of GnRH mRNA in these organs showed opposite trends to that of GnRHR expression. These results suggest the involvement of GnRH in age-related reproductive decline and the potential effects of high-dose gonadotropin treatments on reproductive organ function.
This experiment was to investigate the antioxidant effect of Sesimtang(SST) on brain tissues of mouse. The experimental groups were divided into three groups and treated ad follows for 15 days ; Normal group(NC), Vt.E admistrated group(PC), SST administrated Group(SST). After the extracting microsome from brain of mouse, those were measured the amounts of oxidant materials like MDA(malonaldehyde) and $H_2O_2$, then activities of antioxidant enzymes like SOD(superoxide dismutase), catalase, NADPH-cytochrome P-450 reductase. The results were as follows; 1. In TBA reaction to measure the amount of MDA, oxidant material of brain tissue of aged rat, both treated groups showed significant decrease. 2. Hydrogen peroxide formation was showed significant decrease in both treated groups than normal group. 3. Superoxide dismutase activity was increased in both treated groups than normal group, and showed little change in SST administrated group than normal group. 4. Catalase activity was increased in both treated groups than normal group. 5. NADPH-cytochrome P-450 reductase activity was increased in both treated groups than normal group.

As smartphones came into wide use recently, it has become increasingly popular not only among young people, but middle-aged people as well. Most smartphones use capacitive full touch screen, so touch commands are made by fingers unlike the PDAs in the past that use touch pens. In this case, a significant portion of the smartphone's screen is blocked by the finger so it is impossible to see the screens around the finger touching the screen, and difficulty occurs in precise control used for small buttons such as qwerty keyboard. To solve this problem, this research proposes a method of using simple AR markers to improve the interface of smartphones. Sticker-form marker is attached to fingernails and placed in front of the smartphone camera Then, the camera image of the marker is analyzed to determine the orientation of the marker to perceive as onRelease() or onPress() of the mouse depending on the marker's angle of rotation, and use its position as the position of the mouse cursor. This method can enable click, double-click, drag-and-drop used in PCs as well as touch, slide, long-touch-input in smartphones. Through this research, smartphone inputs can be made more precise and simple, and show the possibility of the application of a new concept of smartphone interface.
The Present study was examined the response of mice of 5 strains to superovulation treatment. Female mice of CF#1, C57BL/6, ICR, dd and CHN strains, aged 60 days were used. Superovulation was induced by the subcutaneous injection of 5 IU PMS and followed 48 hours later by the subcutaneous injection of 5 IU HeG. All females were killed 24 hours after RCG and then ova in the oviduct were counted under a microscope. The average number of ova ovulated in CF#1, C57BL/6, ICR, dd and CHN were 21.3, 17.9, 20.7, 21.0 and 20.5, respectively.
