운동훈련이 흰쥐 노화심근에 미치는 영향 I. 장기간 지구력 운동 훈련 (Effects of the Exercise Training on Aging Heart in Rat I. Long Term Endurance Exercise)

  • 장기간 반복 주행운동이 횐 쥐의 심근에 미치는 효과를 규명하기 위하여 생후 3개월,10개 월 및 20개월 된 횐 쥐를 운동군과 대조군으로 대별하여 motor driven treadmill을 이용하여 Park등이 사용한 방법에 준해서 5개월간, 주 5일 20분간 운동을 시킨 후 심근의 조직 및 세포학적 변화를 관찰하고 심근 세포 내 미세구조 변화를 입체해석학적으로 비교 분석하였다. 연령 증가에 따라 장기간 반복운동이 휜쥐 심근미세구조에 미치는 영향은 8, 15개월의 운동군과 대조군 사이에 뚜렷한 차이를 인정할 수 없으며, 15개월의 운동군에서 대조군에 비하여 변성된 사립체, 리소조옴, 지방적, 공포, 노화색소 등이 증가하는 경향이 있었다. 25개월 운동군은 같은 연령 대조군에 비하여 근원섬유 수축대, 근원섬유 소실, 윤반분리,세포간질 증식, 핵의 변성,교원섬유 근섬유내 침입 등 매우 심한 변화를 보였다. 조직상에 나타나는 early lipofusin과 미세구조상에 나타나는 노화색소는 8, 15개월의 운동군과 대조군 사이에 유의한 차이는 없었으며, 25개월 운동군은 같은 연령 대조군에 비하여 유의한 차이로 증가하였다. Glucose-6-phosphatase 활성도 8, 15개월군에서 운동군과 대조군에서 모두 활성이 높았으며 25개월의 대조군과 운동군에서는 모두 활성이 거의 나타나지 않았다. 미세구조 변화를 입체해석학적으로 분석한 결과 8, 15개월의 대조군과 운동군에서 체적 밀도의 모든 항은 양군사이에 유의한 차이가 없었다. 25개월에서는 세포간질이 대조군에 비하여 운동군이 유의한 증가를 나타내었고, 근원섬유는 유의한 차이는 없지만 증가하는 경향을 보였으며, 사립체는 대조군에 비하여 유의 한 감소를 나타내었고 근형질세망의 체적밀도는 감소하는 경향을 보였다. 사립체와 근원섬유 비는 8개월 운동군에서 유의한 차이는 없지만 대조군에 비하여 높게 나타났으며, 15개월의 운동군과 대조군 사이에서는 차이를 인정할 수 없었다. 그러나 25개월 운동군은 대조군에 비하여 유의한 차이로 감소하였다. 연령증가에 따른 사립체 내막 표면밀도와 사립체수는 대조군과 운동군 사이에는 유의한 변화는 없었다. 본 연구의 성적을 검토한 결과 젊은층(3개월군)과 중령층(10개월군)의 횐 쥐에서는 반복된 지구력운동이 심장에 미치는 역효과를 인정할 수 없었으며 젊은 층의 횐 쥐에서는 오히려 심장기능 강화를 보이는 경향이 나타났으며, 노화층(20개월군)에서 운동군에서는 스트레스로 작용하여 심장기능의 저하를 초래하였다고 생각된다.

주요 식이섬유질원이 첨가된 식이가 노화 흰쥐의 장내효소 및 유해산물에 미치는 영향 (Effects of Dietary Fiber on the Bacterial Enzymes and Putrefactive Metabolite in Aged Rats)

  • This study was performed to investigate the influcence of various dietary fiber sources in Korea for activities of bacterial enzymes (${\beta}$-glucosidase, ${\beta}$-glucuronidase) and amounts of putrefactive product (indole) in aged rats. ${\beta}$-Glucosidase activity in the intestinal content was significantly lower in the seamustard 15% group than in other groups whereas the activity of ${\beta}$-glucuronidase was higher in the mugwort 15% group than other experimental groups. The amount of indole and pH in the intestinal content of aged rats were significantly lower in mugwort groups than in other groups.

Effect of Glutinous Barley Intake on Lipid Metabolism in Middle-Aged Rats Fed a High-Fat Diet

  • This study was designed to determine whether dietary glutinous barley (GB) affects lipid metabolism in middle-aged rats previously fed a high-fat diet. To induce obesity, 20 male 9-month-old Sprague Dawley rats were raised for 1 month on a diet containing 20%(w/w) lipid. The rats were allocated to 1 of 2 groups of 10 rats each and for the subsequent 2 months were fed an 8%(w/w) lipid diet containing well-milled rice (WMR) or GB powder. Rats fed the GB diet had significantly lower concentrations of plasma triglyceride, plasma total cholesterol, and liver cholesterol than rats fed the WMR diet. Fecal excretions of triglyceride and bile acids were significantly greater for the GB group than for the WMR group. In conclusion, dietary GB has positive effects on lipid metabolism: it decreases plasma cholesterol concentration by increasing fecal excretion of bile acids.

환소단(還少丹) 뇌조직(腦組織)의 산화작용(酸化作用)에 미치는 영향(影響) (The Antioxidant Effects of HWANSODAN on the Brain Tissue of aged Rat)

  • The effect of HWANSODAN(HSD), on the level of brain antioxidants was examined in aged rat. The experimental groups were divided into three groups and treated as follows ; normal group(negative control), Vt.E administrated Group(HSD). The purified microsome from brain tissue, those were measired the amounts of oxidant materials like Malonfialdehyde(MDA) and H2O2, then activities of antioxidants enzymes like superoxide dismutase, catalase, NADPH-cytochrome P-450 reductase.The results were as follows;1. In TBA reaction to measure the amount of MDA, HSD group and Vt.E group did not showed signigicant decrease.2. In the formation of Hydrogen peroxide, HSD group and Vt.E group showed a little increase.3. The activity of Superoxide dismutase was increased significantly in HSD group and Vt.E group.4. In the activity of Catalase, Vt.E group was increased significantly and HSD group a little increased. 5. The activity of NADPH-cytochrome P-450 reductase in the HSD group and Vt.E group showed significantly increase.According to the above results, it is suggested that HWANSODAN(HSD) has some antioxidant effects on the tissue of brain.

Morus Nigra Extract Attenuates Cognition Impairment and GABAergic Interneuron Degeneration in Aged Rat Brain

  • Purpose: Aging process comes with cognitive impairment due to decreased neuronal cell number, activity, and neuronal circuit. Alteration of inhibitory neurons contributes to cognitive impairment in normal aging and is responsible for disrupting the excitation/inhibition balance by reducing the synthesis of gamma-aminobutyric acid (GABA). Morus nigra (Mulberry) is a natural physiologically active substance that has been proven to have anti-oxidant, anti-diabetic, and anti-inflammatory effects through many studies. This study aimed to evaluate the effects of the mulberry extract (ME) on cognitive function through anti-oxidant enzyme and GABAergic neuronal activity in aged rat brain. Methods: Sprague Dawley rats were randomly assigned as the young group (8 weeks, n= 8), aging group (67 weeks, n= 8), and aging+ mulberry extract group (67 weeks, n= 8). The aging+ mulberry extract group was orally administered 500 mg/kg/d mulberry extract for 6 weeks. Results: The aging+ mulberry extract group improved spatial and short-term memory. The antioxidant potential of ME increased the expression of superoxide dismutase-1 (SOD-1) and decreased inducible nitric oxide synthase (iNOS). Also, the aging+ mulberry extract group significantly increased the expression of GABAergic interneuron in hippocampus cornu ammonis1 (CA1) compared to the aging group. Conclusion: The number of GABAergic inhibitory interneurons was deceased and memory functions in the aging process, but those symptoms were improved and restored by mulberry extract administration.

카드뮴이 신겨중 인지질 대사에 미치는 영향 (Effect of Cadmium on Phospholipied Metabolism in Nervous System)

  • The effect of acute cadmium-neuropathy on phospholipid metabolism in rat sciatic nerve was investigated. An animal model of cadmium neuropathy was induced by feeding diet containing cadmium to Sprague-Dawley rat for two weeks. Four weeks aged Sprague-Dawley rats were divided into four groups : normal control group, 10ppm-cadmium treated group, 100ppm-cadmium treated group, 1000ppm-cadmium treated group, reference drug, myo-inositol-treated group. All rats were sacrificed at the end of two weeks. The rate of incorporation of 2-[3H]myo-inositol into polyphosphinositide was significantly decreased while the rates of incorporation into phospholipid of titratedserine, ethanolamine and choline were unchanged in sciatic nerve obtained from cadmium-treated rat. Continuously the activities of three enzymes concerned with inositol phospholiped metabolism were measured in homogenates of rat sciatic nerves. Cystidine diglyceride transferase and phophatidylinositol kinase showed significantly decreased activities while phosphatidylinositol-4-phosphate kinase did not show any significant change in activity by cadmium treatment. However these deficits of inositol phospholipid metabolism were ameliorated by myo-inositol administration and these effectiveness were more potent in lower dose cadmiumtreated rats than higher dose cadmium-treated rats. These results suggest that cadmium intoxicated peripheral nerve with perturbation of the ployphosphoinositide metabolism and alteration of the enzyme activity which concerned with myo-inositol metabolism.

납 중독 랫드의 말초신경내 myo-inositol 수송 체계에 관한 연구 (A study on myo-inositol transport system in peripheral nerve isolated from lead-intoxicated rat.)

  • In our previous studies, we reported that lead intoxicated nerve cell by inhibition of the Na$^{+}$-K$^{+}$ ATPase activity and reduction of myo-inositol in nerve cell. As the second series of experiments, in order to understand toxic mechanism of lead for nerve cell, the characteristics of myo-inositol transport system and the effect of lead on its system have been studied in the sciatic nerves of control and lead-treated rats. A lead intoxicated animal model was induced by feeding diet containing lead to Sprague-Dawley rat for two weeks. Four weeks aged Sprague-Dawley rats were divided into three group : normal control group, 10ppm-lead treated group, 100ppm-lead treated group. All rats were sacrified at the end of two weeks. The rate o myo-inositol transport by sciatic nerve isolated from lead-treated rat was significantly decreased compared with that of control rat. This deficit results from that myo-inositol transport system which is carrier mediated and sodium-potassium dependent was inhibited by the lead treatment (both 10ppm and 100ppm) due to increase of the Km value without affecting Vmax value for myo-inositol carrier. These observations suggest that the toxic mechanism of lead on nerve myo-inositol transport system might be a change of affinity without change of maximum transport velocity for carrier.

Minoxidil Regulates Aging-Like Phenotypes in Rat Cortical Astrocytes In Vitro

  • Mainly due to the slanted focus on the mechanism and regulation of neuronal aging, research on astrocyte aging and its modulation during brain aging is scarce. In this study, we established aged astrocyte culture model by long-term culturing. Cellular senescence was confirmed through SA-β-gal staining as well as through the examination of morphological, molecular, and functional markers. RNA sequencing and functional analysis of astrocytes were performed to further investigate the detailed characteristics of the aged astrocyte model. Along with aged phenotypes, decreased astrocytic proliferation, migration, mitochondrial energetic function and support for neuronal survival and differentiation has been observed in aged astrocytes. In addition, increased expression of cytokines and chemokine-related factors including plasminogen activator inhibitor -1 (PAI-1) was observed in aged astrocytes. Using the RNA sequencing results, we searched potential drugs that can normalize the dysregulated gene expression pattern observed in long-term cultured aged astrocytes. Among several candidates, minoxidil, a pyrimidine-derived anti-hypertensive and anti-pattern hair loss drug, normalized the increased number of SA-β-gal positive cells and nuclear size in aged astrocytes. In addition, minoxidil restored up-regulated activity of PAI-1 and increased mitochondrial superoxide production in aged astrocytes. We concluded that long term culture of astrocytes can be used as a reliable model for the study of astrocyte senescence and minoxidil can be a plausible candidate for the regulation of brain aging.

정상 및 당뇨 흰쥐에서 주령과 절식에 의한 양파의 혈당조절작용 변화 (Influence of Age and Fasting on the Anti-hyperglycemic Effect of Onion in Normal and Diabetic rat)

  • Onion (Allium cepa Linn) has been reported to have hypoglycemic activity in human and several animal models. In the present study, we performed intraperitoneal glucose tolerance test (IPGTT) in young (1.5mo) and aged (5 mo) rats treated with onion in order to determine whether aging can influence on the anti-hy-perglycemic effect of onion. In addition, we investigated the hypoglycemic effect of onion in streptozotocin- induced diabetic rats treated with aqueous extracts of onion (500 mg/kg, i.p., daily) for 4 weeks. Blood glucose level was determined in fasted and fed rats by using a glucometer (Johnson & Johnson). In glucose tolerance test, blood glucose level was maximally increased 15 min after glucose load (2 g/kg, i.p.), and recovered to the basal level 3 hr after glucose challenge in young and old rats. The maximum blood glucose levels of young and aged rat were 184$\pm$7.49 and 225.2$\pm$ 12.55 mg/dl, respectively. A single i.p. injection of aqueous extract of onion (1 g/kg) 30 min before glucose challenge significantly decreased blood glucose levels at 15, 30, 60, 90 min after glucose load in aged rats, while the administration of onion did not show any significant effect in young rats. In onion-treated diabetic rats, significant hypoglycemic effect (p<0.05) was observed, and the effect was greater in fasted rats than in fed. In conclusion, these results suggest that anti-hyperlycemic effect of onion can be changed by age and fasting.
