We investigated the absorption characteristics of protein-bound polysaccharide powders of various molecular weights isolated from the mushroom Agaricus blazei Murill. The monolayer moisture content calculated using the GAB equation showed a higher level of significance than did the BET equation. The higher the water activity, the lower the isosteric heat of sorption. The fitness of the isotherm curve was shown to be in the order of the Khun, Oswin, Caurie and Henderson models. The prediction model equations for moisture content were established by use of ln(time), water activity, and temperature.
In this study, we first prepared 3 kinds of powders with different molecular masses from the crude protein-bound polysaccharide extract of Agraricus blazei Murill through ultrafiltration, followed by spray-drying. Then, the antitumor activities of the powders were analyzed. Size exclusion chromatography coupled with a multi-angle laser-light-scattering system showed the 3 powders had the following molecular ranges: below 10 kDa (SD-1), 10 to 150 kDa (SD-2), and above 150 kDa (SD-3), representing peak molecular weights of $8.26{\times}10^3,\;9.65{\times}10^4$, and $5.94{\times}10^6\;g/mol$, respectively. All the powders stimulated macrophage RAW264.7 cells to produce nitric oxide, of which SD-2 and SD-3 were superior to the crude extract powder (CP-SD), while SD-1 showed the lowest activity. Similar results were found for their cytotoxicities against human cancer cell lines (A549, MCF-7, and AGS), where the highest activity was obtained with the SD-2 treatment for 72 hr at $1,000\;{\mu}g/mL$. The MCF-7 cell line was less sensitive to the powders than the other cells. From this research we found that ultrafiltration, in combination with spray-drying, is applicable for preparing protein-bound polysaccharide powders with higher antitumor activities.
The purpose of this study was to prepare the granule using protein-bound polysaccharide isolated from Agaricus blazei Murill. Moisture content was the highest in granule formed with dextrin(DE=9). Sugar content of granule in relation to the forming agent was the highest in granule formed with ${\beta}$-cyclodextrin. pH and protein content were not affected by the forming agent. L and b values were high in granules formed with dextrin(DE=9) and ${\beta}$-cyclodextrin, respectively. Solubility of granule formed with dextrin(DE=23) and ${\beta}$-cyclodextrin was higher than that of formed with dextrin(DE=9), while there was no significant difference between dextrin(DE=23) and ${\beta}$-cyclodextrin. Rate of water absorption was the highest in granule formed with ${\beta}$-cyclodextrin, while the lowest in granule formed with dextrin(DE=9). Overall acceptance of three granules were acceptable in granule formed with ${\beta}$-cyclodextrin.
The purpose of this study is to observe the effect of protein-bound polysaccharide (PBP) on proliferation of Th1 cells and cytotoxicity of cancer cell. Mushrooms (Ganoderma lucidum, Agaricus blazei, Lentinus edodes, Coriolus versicolor and Phellinus linteus) were fractionated by 100$^{\circ}C$ hot water for 3hr. PBP was stimulated and proliferated Th1 cells most at 10 mg/$m\ell$ concentration and the percentage of cell proliferation was 40%. It was estimated cytotoxicity of PBP against 7 kinds of cancer cell lines. Antitumor activities of Agaricus blazei against P388D1 and L1210 (tumor cell lines) were 2.4% and 39.7% survival rate, and Lentinus edodes was 48.4% and 52.5% survival rate, respectively. PBP mixtures of Agaricus and Lentinus edodes prolonged (27∼40%) significantly the survival rate of mice intraperitoneally implanted with sarcoma 180.
The basidomycetes fungus, Agaricus blazei Murill has been well known as the biologically active polysaccharides. They do not attack cancer cells directly, but produce their antitumor effects by activating various immune responses in the host. This present study was designed to investigate the immune activity of polysaccharides extracted from fruiting body, culture broth, and mycelia. In order to examine the optimum conditions of polysaccharide production, medium and C/N ratio were examined in flask culture. The fine polysaccharides were isolated and purified by DEAE cellulose and Sephadex CL-6B, respectively. Each fraction was spplied to recognize NO synthesis and TNF-${\alpha}$ for immune activity test.
Objective: This study aimed to investigate the effects of dietary supplementation with Agaricus blazei polysaccharide (ABP) at varying concentrations on the performance, egg quality, blood biochemistry, intestinal morphology, and microflora of quail. Methods: The study involved a total of 2,700 Korean quails, which were randomly divided into three groups. The measured variables encompassed productive performance, egg parameters, carcass parameters, serum metabolites, immune response parameters, antioxidative properties, and gut microbiome. Results: The addition of ABP did not have a significant effect on average daily feed intake. However, it was found to increase the average daily egg weight and egg production rate, reduce the feed-egg ratio. There were no significant impacts on egg quality measures such as egg shape index, egg yolk index and color, egg yolk and protein content. However, ABP supplementation significantly increased the Hough unit (p<0.01) and decreased the rate of unqualified eggs (p<0.01). Regarding serum parameters, the inclusion led to an increase in total protein concentration (p<0.05) and a reduction in low-density lipoprotein cholesterol (p<0.05). There were no significant effects observed on immune indicators such as immunoglobulin A (IgA) and IgM. ABP supplementation increased the levels of serum antioxidant indicators, including glutathione peroxidase, total superoxide dismutase (p<0.05), and total antioxidant capacity colorimeter (p<0.05). Furthermore, ABP supplementation significantly elevated the intramuscular fatty acid content in quail meat. Additionally, ABP supplementation demonstrated a significant improvement in the diversity of gut microbiota and induced alterations in the composition of the gut microbiota. Conclusion: The findings of this study indicate that dietary supplementation of ABP enhanced production performance and antioxidant capacity while increasing the levels of polyunsaturated fatty acids in quail muscle.
Cytoprotective ability of polysaccharides isolated from different edible mushrooms was investigated on the 7-ketocholesterol-induced damaged cell line. Polysaccharide extracts from six different edible mushrooms-Flammulina velutipes, Peurotus ostreatus, Lentinus edodes, Agrocybe aegerita, Agaricus blazei, and Cordyceps militaris- were prepared by hot water extraction and alcohol precipitation. Cytoprotective ability was evaluated by measuring the viable cells of the normal embryonic liver cell line (BNL CL. 2) in the presence of 7-ketocholesterol. At $80\;{\mu}g/mL$ of 7-ketocholesterol, cytotoxicity was very high with a loss of 98% of viable cells after 20 h of incubation. With the addition of $200\;{\mu}g/mL$ of each polysaccharide isolate to the cell line containing $80\;{\mu}g/mL$ of 7-ketocholesterol, polysaccharide isolates from both Flammulina velutipes and Peurotus ostreatus could significantly inhibit the 7-ketochoelsterol-induced cytotoxicity in the cells. But other polysaccharide isolates were not effective in inhibiting cell damage caused by the oxLDL-induced cytotoxicity.
This study was conducted to investigate the rheological properties of protein-bound polysaccharide powders (SD-1, 2, 3) using ultrafiltration (UF) and spray drying (SD) process from Agaricus blazei Murill. The calculated weight-average molar mass (Mw) in the positions at 29.7 mL (for SD-1), and at 27.8 mL (for SD-2), and at 18.7 mL (for SD-3) was $8.2{\times}10^3,\;9.6{\times}10^4$, and $5.9{\times}10^6g/mol$, respectively. As concentration increased the solution showed higher pseudoplasticity where the pseudoplasticity decreased as temperature increased. The flow behaviors of spray dried powder solutions were more fitted to Herschel-Bulkley equation than Power law equation. Apparent viscosity of SD-2 was more temperature-dependent than that of SD-1 and 3. However, the SD-3 tended to be more concentration-dependent than SD-1 and 2 as temperature increasing.
This research was performed to investigate the immnomodulative effects of ploysaccharides extracted from the fruiting body of Agarcus blazei cultivated with the media which are fermented with sugar cane bagasse containing Pueraria thunbergiana in open-air storage. In MTT test, methanol extracts from the fruiting body of A. blazei cultivated with P. thunbergiana media showed in colon carcinoma line(HT29) by 1.5∼3.5 fold and human heptoma cell line (HepG2) by 1.3 ∼2.4 fold antitumor activites compared to two types media (rice straw plus sugar cane bagasse, rice straw only) often used in the fauns. To clarify the antimutagenic principles, three extracts, Ab-l, Ab-2 and Ab-3, were separated by the solvent fractionations such as hot water, cold & hot sodium hydroxide respectively, and their antimutagenic effects was determined against N-methyl-N'-nitro-N-cnitrso-guanidine(MNNG) using Salmonella typhymurium. There was no significant differencies of inhibition levels among the used media, but Ab-3 tractions still showed a high antimutagenicity in the Ames test regardless of cultivating areas or media. To prove the cell immunofunction, nitric oxide (NO) produced from Raw 264.7 matrophage cultured with three fractions (Ab-l, Ab-2, Ab-3) was measured, and showed generally increase about 45 ∼58 percent compared to another two media (rice straw plus sugar cane bagasse, rice straw only), in the fraction of hot alklai extracts of the fruiting body cultivated with P. thunbergiana, which means that the media selection could be very important factors for improving medicinal effects in agaricus blazei fruiting body.
