This study was performed to investigate the correlations between steroids and TGF-${\beta}_1$ levels and delayed parturition in SCNT clone calving. The recipients pregnant by AI were used as control (AI-R). All AI-R were labored by natural delivery (n=5, day $284{\pm}0.71$ of pregnancy). The recipients pregnant by SCNT embryo (SCNT-R) showing no signs of delivery about 10 days after expected date were operated by Caesarean section (n=5, day 292). The blood and placentome samples were obtained and weighed at parturition. The concentrations of plasma progesterone (P4) and Estradiol-$17{\beta}$ (E2) were measured by radioimmunoassay (RIA). The levels of plasma and placental TGF-${\beta}_1$ levels were examined by ELISA. The placentomes from SCNT-R were overweight (p<0.05) compared to those of AI-R. The plasma P4 (p<0.01) level in SCNT-R at parturition was significantly higher compared to that of AI-R. In contrast, the plasma E2 level in the SCNT-R was significantly lower compared to that of AI-R (p<0.05). The plasma and placental TGF-${\beta}_1$ protein levels in the SCNT-R were significantly higher than those of AI-R at parturition, respectively (p<0.01). Based on these results, aberrant expressions of steroid hormones and high levels of plasma and placental TGF-${\beta}_1$ protein at parturition may be one of the key indicators on delayed parturition of SCNT clone calving.

This study Investigated the effects of $estradiol-l7{\beta} (E_2)$, $17{\alpha}-methyltestosterone$ (MT) and high temperature (WT) on gonadal sex differentiation in black rockfish, Sebastes srhlegeli. fish were reared to oral adminstration of E, at nominal concentrations of $20, 40 and 60 {\mu}g/g diet$, and MT at nominal concentration of 20 and $50 {\mu}g/g$ diet from 56 days to 77 days after parturition. In the treatment of WT, water temperature of the breeding tanks ranged $27.0{\pm}0.5^{\circ}C$ more than $10^{\circ}C$ approximately, in comparison to the control and other experimental group, In the process of sex differentiation until 56 days after parturition, gonads were composed of mostly gonia cells, sexually undifferentiated. In contrast, 128 dal's after parturition, the ovaries were composed of ovarian cavity and lamellae, and oogonia and perinucleolus oocytes were distributed in the lamellae of ovary, and the testes were composed of a number of seminiferous tubule, and spermatogonia were distributed in the seminiferous tubule, and also melanophore scattered in the matrix layer of testis. In the sex ratio, more females than male were observed from $E_2$. treatment groups when compared to the control, but more males than females were observed from MT and WT treatment groups when compared to the control. In the results of the present study, the concentration and kinds of the sex steroid hormones, and also the rearing high temperature caused to the factor of sex determination in the process of sex differentiation of black rockfish.
Most progesterone enzyme immunoassays(EIA) are used liquid-phase double-antibody or single-antibody seperation. These methods consume considerable time and reagents because of the requirements for several washing and centrifugation steps involving the reactants. Because of these several problems, we were prompted to develop an effective enzyme-linked immunosorbent assay(ELISA) system that would be equal or superior to RIA for assay of progesterone. The results were obtained as follows. 1. Cross reaction of the progesterone antiserum with other steroids determined was shown with progesterone(100%), $11{\alpha}$-deoxycorti-costerone(2.271%), but the other steroids were shown below 0.9%. 2. Standard curve for progesterone ELISA was shown available difference according to progesterone concentration from 0 to 1,000pg/ml. 3. The lower limit of sensitivity was 0.2pg/well 4. Progesterone concentration was 1.6ng/ml for before parturition, and that was below 0.5ng/ml for after parturition. This development enzyme-linked immunosorbent assay for progesterone can be detected pregnancy diagnosis in cattle, and also applicable 10 research on physiological function including such as reproductive disorders.
This review focuses on modulation of growth hormone (GH) and its downstream actions on periparturient dairy cows undergoing physiological and metabolic adaptations. During the periparturient period, cows experience a negative energy balance implicating that the feed intake does not meet the total energy demand for the onset of lactation. To regulate this metabolic condition, key hormones of somatotropic axis such as GH, IGF-I and insulin must coordinate adaptations required for the preservation of metabolic homeostasis. The hepatic GHR1A transcript and GHR protein are reduced at parturition, but recovers on postpartum. However, plasma IGF-I concentration remains low even though hepatic abundance of the GHR and IGF-I mRNA return to pre-calving value. This might be caused by alternation in IGFBPs and ALS genes, which consequently affect the plasma IGF-I stability. Plasma insulin level declines in a parallel manner with the decrease in plasma IGF-I after parturition. Increased GH stimulates the lipolytic effects and hepatic glucose synthesis to meet the energy requirement for mammary lactose synthesis, suggesting that GH antagonizes insulin-dependent glucose uptake and attenuates insulin action to decrease gluconeogenesis.
Data from 5,311 ewes and 13,076 lambing from 1977 through 1994 were used to analyse both annual and cumulative outputs in terms on total number of lambs born, total lamb weight weaned and total wool produced per ewe for ewe longevity 1 to 8 depending on their productive life in the flock. Ewes at Khushab produced 0.08 more lambs per parturition than ewes at Khizerabad; however, 0.39 less lambs were weaned at Khushab than at Khizerabad. Similarly, cumulative number of lambs born was more at Khushab flock than Khizerabad flock (p < .01). However, total weight of lambs weaned was greater at Khizerabad than Khushab flock (p < .01) for each ewe longevity. Most ewes (35%) were sold/replaced just after their first parturition (i. e. ewe longevity 1). The overall mean for annual sale/replacement was 32 and 23% at Khushab and Khizerabad, respectively. Distribution of growth and reproductive traits from 1977-94 did not show upward or downward trend inspite of heavy sale/replacement except yearly variation. Lack of any genetic progress over the year suggested that random breeding was employed without any scientific selection programme. Annual means for lambs born, lambs weaned and weight of lambs weaned per ewe present in the flock were the highest for ewe longevity 2 compared with other ewe longevity groups. Relative efficiency in terms of net income was highest for ewe longevity 5 followed by ewe longevity 4 and 6 in both flocks.
The objectives of this study were to characterize the changes occurring in the levels of insulin-like growth factors (IGF-I and IGF-II) in bovine milk during a one-year lactation period, and to determine the parameters affecting IGF content in bovine milk. Milk was collected individually from lactating Holstein cows (n=70), and IGF-I and -II levels were determined via radioimmunoassay, using 125I after acid-ethanol treatment. The proximate compositions of the milk samples were determined using a near-infrared milk analyzer. The data were analyzed by the GLM and CORR procedures using SAS software to determine significant differences (p<0.05) occurring within groups (dairy farms, lactation periods, season, and parity). We noted an approximately six-fold reduction in the IGF-I concentration (from 2,462.7 to 353.0 ng/ml) and a three-fold drop in the IGF-II concentration (from 929.1 to 365.7 ng/ml) in the bovine colostrum, between 6 h after parturition and 18 h after parturition. IGF-I and -II content, measured at the early, middle, and late stages of lactation did not change significantly throughout the entirety of the lactation period. Interestingly, parity did not significantly affect IGF-I content, but did significantly affect IGF-II content between the primiparous and multiparous cows. We also found there were no significant relationships between IGF-I and total protein content or somatic cell counts (p<0.05).
A 4-year-old female reticulated giraffe (Giraffa camelopardalis reticulata), at Seoul Zoo, Gwacheon, Korea had a male calf with no help of proper restraint devices. The mother giraffe was in a danger of dystocia more than 7 hours in labor after showing the calf's toe of the foreleg which protruded from her vulva. After tugging with a snare of rope on the metacarpal bone of the calf and pulling it, the other toe emerged. Finally, with two snares around each of metacarpal bones, the calf was completely pulled out by zoo staff. After parturition, the dam was in normal condition for taking care of the calf and her progesterone hormone had also dropped down to a normal pre-pregnancy.
This study was conducted to find out the changes of progesterone and estrone sulphate concentration in serum of Korean native goats throughout gestation, and to apply the serum levels of the hormones to pregnancy diagnosis. Serum concentration of progesterone and estrone sulphate were assayed by radioimmunoassay. Serum progesterone concentration was similar to its luteal phase values during early pregnancy and remained at the high level continually at 20∼140days and decreased rapidly at the day of parturition. Serum estrone sulphate concentration showed to increase markedly at 40∼50dyas gestation and steadily increased to the maximum of 7.13ng/ml at 140days, but declined sharply at the day of parturition. The accuracy of pregnancy diagnosis by the measurement of serum progesterone at 20∼24days after mating was 85.7∼92.3% and that of non-pregnancy diagnosis was 100%, when the serum progesterone levels higher and lower than 3.0ng/ml were supposed to indicate pregnancy and non-pregnancy, respectively. The accuracy of pregnancy diagnosis by the mearsurement of serum estrone sulphate was found to be nearly 100% since 50days after mating, when the serum levels of estrone sulphate higher then 0.5ng/ml were diagnosed to be pregnant. The optimal sampling time for pregnancy diagnosis was considered to be at 50 days after mating or to be later. It appears that estrone sulphate values above 7.0ng/ml at any time in gestation are highly indicative of twin. But there was found no significant difference(P<0.05) in serum estrone sulphate concentration and number of kids between does with single and twin kids.
In order to estimate energy budget of the rockfish, Sebastes schlegeli juvenile, during the period from parturition to juvenile, of rockfish were reared at constant condition of water temperature($16^{\circ}C$ and $20^{\circ}C$). Energy used by the reared juveniles were calculated from estimates of data on growth, oxygen consumption, nitrogen excretion, feeding and energy content. After parturition, total length of juvenile was 4.56~5.49mm(average 4.97mm, n=20), and the average dally growth rates were 0.50mm at $20^{\circ}C$. The weight-specific oxygen consumption and nitrogen excretion rates decreased with increasing body weight. Temperature significantly affected oxygen consumption and nitrogen execretion rates, with the higher rates at $20^{\circ}C$ than $16^{\circ}C$. During the 25 days from parturition to jllveniles, feeding rates were 65.8cal at $16^{\circ}C$, and 89.2cal at $20^{\circ}C$. The assimilation effeciency estimated by nitrogen content of food and egested feces were 85.21% at $16^{\circ}C$ and $20^{\circ}C$. During the period from parturition to juvenile, energy was used the higher in body growth than in oxygen consumption. The gross growth efficiencies($K_1$)and net growth efficiencies($K_2$) of the rockfish, Sebastes schlegeli juvenile ranged from 43% to 47% and from 50% to 55%, respectively.
The objective of the present study was to investigate the effects of Fe-soy proteinate chelate (Fe-SP) on sows milk, piglet blood parameters and performance. A total of 15 sows of 3 wk before parturition and pigs after births to 3 wk were assigned to three dietary treatments: control (sow-basal diet, piglets with Fe injection); Fe-SP 100 (Fe 100 ppm as Fe-SP in sow and piglet diet); Fe-SP 200 (Fe 200 ppm as Fe-SP in sow and piglet diet). Each treatment had 5 replicates (sows) of six piglets per sow randomly selected from the same offspring. For this experiment, Fe-SP was manufactured. There were no significant differences among treatments in number of pigs born in total or alive per litter, birth weight, number of pigs weaned per litter and weaning weight. However, weight gain, feed intake and feed conversion ratio significantly (p<0.05) decreased as the supplementation level of Fe-SP increased. There were no significant differences among treatments in Fe content at 3 wk before parturition in sow blood. However, Fe content at 2 wk before parturition in sow blood significantly (p<0.05) increased as the supplementation of Fe-SP. While there were no significant differences among treatments in Fe content at 1 wk before parturition in sow blood, it tended to increase as the supplementation level of Fe-SP increased. There were no significant differences among treatments in Fe content of sow milk. However, it tended to increase as the supplementation level of Fe-SP increased. Iron content in the blood of piglets was significantly (p<0.05) higher in control (Fe injected) than Fe-SP 100 and Fe-SP 200 treatments at $1^{st}$ and $2^{nd}$ wk but it was significantly higher in Fe-SP 200 than others in $3^{rd}$ wk. Zinc content in the blood also significantly (p<0.05) increased as the Fe-SP supplementation level increased in $3^{rd}$ wk. In conclusion, Fe-SP supplementation significantly affected Fe content in the blood of piglets. Iron injection was more effective at $1^{st}$ and $2^{nd}$ wk, while Fe-SP 200 supplementation was effective at $3^{rd}$ wk in improving blood Fe level in piglets.
