Specific Recognition of Unusual DNA Structures by Small Molecules: An Equilibrium Binding Study

  • The binding interaction of ethidium to a series of synthetic deoxyoligonucleotides containing a B-Z junction between left-handed Z-DNA and right-handed B-DNA, was studied. The series of deoxyoligonucleotides was designed so as to vary a dinucleotide step immediately adjacent to a B-Z junction region. Ethidium binds to the right-handed DNA forms and hybrid B-Z forms which contain a B-Z junction, in a highly cooperative manner. In a series of deoxyoligonucleotides, the binding affinity of ethidium with DNA forms which were initially hybrid B-Z forms shows over an order of magnitude higher than that with any other DNA forms, which were entirely in B-form DNA The cooperativity of binding isotherms were described by an allosteric binding model and by a neighbor exclusion model. The binding data were statistically compared for two models. The conformation of allosterically converted DNA forms under binding with ethidium is found to be different from that of the initial B-form DNA as examined by CD spectra. The ratio of the binding constant was interestingly correlated to the free energy of base unstacking and the conformational conversion of the dinucleotide. The more the base stacking of the dinucleotide is unstable, or the harder the conversion of B to A conformation, the higher the ratio of the binding constant of ethidium with the allosterically converted DNA forms and with the initial B-Z hybrid forms. DNA sequence around a B-Z junction region affects the binding affinity of ethidium. The results in this study demonstrate that ethidium could preferentially interact with unusual DNA structures.

녹두(Vigna radiata L.) Trypsin Inhibitor의 정제 및 약물학적 특성 (Characterization and Pharmacological Effect of Mung Bean Trypsin Inhibitor)

  • 우리나라에서 식용으로 뿐만아니라 한방재료로 널리 사용되고 있는 녹두(vigna radiata L. wilczek) 로부터 trypsin inhibitor (Mung bean trypsin inhibitor, MBTI)를 분리정제하여 그 특성을 조사하였다 또한 병태동물모델 즉, septic shock induced guinea pig model을 이용하여 MBTI의 약물학적 효과를 평가하였다. MBTI의 분리 및 정제과정은 Sephadex C-50 chromatography, DEAE-celluloseion exchange chromatography 및 trypsin affinity column 을 차례로 이용하였다. 정제한 MBTI는 전기영동 및 아미노산 서열분석결과 분자량 약 8,000 Da 의 BBI-type (Bowman-birk inhibitor type)임을 알 수 있었으며 이들의 생화학적 특성을 구명하였다. 또한 pseudomonal elastase로 유도된 septic shock guinea pig model에서 MBTI 10 mg/kg를 전처치한 결과 hypotention shock 유발이 억제됨을 알 수 있었다.

  • 생물학적 자극통제 수단으로 활용하기 위한 돼지 웅성 페르몬성 분자를 탐색하고자 일련의 냄새 분자로서 2-(cyclo-hexyloxy) tetrahydrofurane 유도체들의 정량적인 구조와 수용체인 porcine odorant binding protein (pOBP)간의 결합 친화력 상수$(p(Od)_{50})$에 대한 비교 분자장 분석(CoMFA)을 실행하였다. 가장 양호한 CoMFA 모델 AIV $(r^2_{cv}.(q^2)=0.886$$r^2_{ncv}.=0.984$)은 기질 분자 내 입체 중심(chiral center)의 절대 배열이 $C_1(R),C_2(S)$인 분자를 atom based fit 방법으로 배열하였을 경우의 standard field와 indicator field가 조합된 CoMFA장의 조건에서 유도되었다. 이 CoMFA 모델은 입체장 40.8% 정전기장 14.6%및 소수성장 44.6%가 결합 친화력 상수에 영향을 미치는 요소임을 나타내었다. 등고도의 분석 결과로부터 효과적인 결합 친화력 냄새 분자를 수식하는 데 몇 가지 가치 있는 정보를 얻을 수 있었다.

PET 초극세사 염색에서 분산염료의 친화력과 온도 의존성 (Influence of Temperature and Affinity of Disperse Dye on Dyeing of PET(Polyethylene Terephthalate) Microfiber)

  • 본 연구에서는 PET 초극세사의 분산염료 등온흡착염색에서 염료의 분배계수와 표준친화력의 시료 굵기에 따른 영향성과 확산계수의 온도 의존성을 알아보았다. 염색은 해도형 초극세사 0.2, 0.06, 0.01 dpf 급과 일반 극세사 0.5 dpf 급을 anthraquinone계 염료 C.I. Disperse Blue 56으로 무한염욕조건 100, 110, 120, $130^{\circ}C$에서 등온염색을 진행하였다. 그 결과 Nernst type과 잘 일치하는 분산염료의 시료 굵기별 등온흡착곡선을 얻었다. 해도형 PET 초극세사의 해성분 용출은 NaOH 수용액을 이용한 PET 알칼리 가공을 통하여 이루어졌다. 염색에 사용된 분산염료는 흡광도 측정을 토대로 고온에서 장시간 염색 진행시 안정성이 확보됨을 확인하였다. 초극세사의 염색은 결론적으로 동일 온도에서 굵기가 감소함에 따라 시료 표면적의 증가로 인해 분배계수 및 표준친화력은 증가하였다. 모든 굵기에서 염색현상은 발열반응이기 때문에 염색 온도가 증가할수록 분배계수가 감소하였고 이에 따라 표준친화력은 동일 굵기에서 대략 9% 감소하였다. 또한 굵기가 감소할수록 동일 온도에서 최대로 염착되는 염료 농도가 증가하였고, 염색 과정에서 발생되는 염색열은 0.01 dpf 초극세사에서 -10.64 kcal/mol로 가장 낮게 나타났다. 시료 굵기가 감소할수록 염액과 접촉 가능한 표면적은 증가하였지만, 그에 반해 초극세사 사이의 공간이 조밀하게 되어 염액이 원활히 이동하기 어렵게 된다. 그 때문에 확산계수는 시료의 굵기가 극세화될수록 감소하였다. 이러한 경향성은 고온으로 갈수록 더 뚜렷하게 나타났으며. 시료의 굵기가 증가할수록 확산계수의 온도 의존성은 $130^{\circ}C$에서 40%까지 증가하였다.

지능형 빌딩 시스템(IBS)의 등급 결정을 위한 ANP 모형 -IBS 구현 이득을 기준으로- (An Analytic Network Process Model for Ranking Decision of Intelligent Building System)

  • In this paper, the conceptual framework of IBS(Intelligent Building Systems) is redefined, and we propose two ANP(Analytic Network Process) models for ranking IBS. Traditional models have ranked IBS according to technical features or the number of elements in IBS. But, we consider relative functional importance among IBS elements for efficient building operation. According to the structure of interactive-relationship among IBS elements, we present two types of model. The one is Model A that is composed of both hierarchical and network structures. It has $12{\times}12$ supermatrix consists of interdependent relationship between 6 benefit elements(Productivity, Saving, Safety, Convenience, Pleasantness, Environment Affinity) and 6 IBS elements(Building Operation, Security, Safety, Telecommunication(TC), Office Automation(OA), other elements). Each of 6 IBS elements has subelements in hierarchical structure. The other is Model B that has $25{\times}25$ supermatrix consists of interdependent relationship between 6 benefit elements and 19 IBS sub elements. Merits and demerits of each model are discussed in detail.

굴패각 모르타르에의 휘발성 유기화합물 흡착에 대한 연구 (A Study on Isothermal Adsorption of VOCs onto Gypsum Mortar Incorporating Oyster Shell)

  • 굴패각과 석고를 대상으로 고형화시킨 후 포름알데히드 및 벤젠으로 대표되는 휘발성 유기화합물(volatile organic compounds, VOCs)를 흡착챔버 내에서 노출시켜 흡착특성을 관찰하였다. VOC를 챔버에 노출 후 굴패각과 같은 친환경재료를 혼입한 모르타르를 각각 개별적으로 주입한 후 180분간 흡착특성을 고찰하였다. 굴 패각분말을 0, 10, 30, 50%로 혼입한 시험체를 대상으로 포름알데히드와 VOC(벤젠, 톨루엔, 자일렌)를 $27.7{\sim}28.5mg/m^3$의 고농도 조건과 $175{\sim}150{\mu}g/m^3$의 저농도 조건에서 각각 노출시켜 흡착성 실험을 수행한 결과, 전반적으로 노출된 조건과 무관하게 굴패각의 혼입 함량이 많을수록 흡착성이 증가함을 관찰할 수 있었다. 또한 대류-확산-흡착이 고려된 모델을 사용하여 실험과 비교하였다. 확산계수 및 확산 길이(diffusion length)로 대변되는 흡착재의 친화력(affinity)을 고려한 모델을 시험한 결과, 피흡착제의 종류와 흡착재의 친화성에 따른 차이를 잘 표현하였으며 전반적으로 실험데이터와 일치하는 경향을 보였다.

고객요구속성을 반영한 중.노년층 니트정장 품질특성 (The Quality Characteristics Based on Customer-required Attributes of Middle and Elderly Women's Formal Knitwear)

  • An important issue for the improvement of product quality is to identify and understand the customer-required attributes of a product. This paper identifies and analyzes customer-required attributes of formal knitwear for middle age and elderly women through the use of the Kano model in order to develop competitive products. Various research methods were used in this study. Customer voices for formal knitwear were collected by means of brainstorming and in-depth interviews. The collected customer voices were transformed into customer-required attributes with the use of an affinity diagram. In addition, the data was collected by means of a questionnaire and was statistically analyzed to examine the dimensions of customer-required attributes on the basis of the Kano model. The results are as follows. Five categories of customer-required attributes were identified (symbolism, aesthetic, fitness, usefulness, and maintenance). The analysis based on the Kano model identifies a strong one-dimensional quality in all of the categories of customer-required attributes and the attractive quality that includes a one-dimensional quality in the categories of fitness and usefulness. The findings of this study demonstrate the applicability of the affinity diagram for apparel that educates formal knitwear developers of the customer-required attributes that they should strategically focus on in order to develop competitive products.

금속 친화성 액 이상분계 시스템에서 Phsphoprotein분배 및 분배예측 (Phosphoprotein Partitioning in Metal-Affinity Aqueous Two-Phase Systems and Prediction of Partitioning Behavior)

  • Fe(III) IDA -PEG/ dextran 액 이상 분계 시스템에서 phosphoprotein의 분배 예측을 위한 모델식이 유도되었다. 본 모델식에는 단백칠 분배에 수소이온과 hydroxide 이온의 저해효과가 포함되어 었다. 표변에 각각 1,27~의 phosphoryl 그 룹을 갖고 있는 ovalbumin의 AI, A2, 성분을 정제한 후 단백질 분배 실험을 수행하였으며 실 험결과와 모텔식에의한 예측이 잘 얼치하는 것 을 확인하였다. Fe(III)IDA-PEG와 단백질 표면 p phosphoryl group과의 결합상수는 PEG 상과 dextran상에 셔 각각 $6.1{\times}, 10^3M^{-1} and 2.3{\times}10^4M^{-1}$이었으며,이것은 Cu(II)IDA-PEG와 단백질 표면 histidine과의 결합상수 보다 3-5배 높은 값이 였 다.

도시민의 라이프스타일에 따른 농촌어메니티자원 선호도 분석 (An Affinity analysis for Rural Amenity Resources according to the Life-Styles of Urbanites)

  • The demand of rural tourism industry has increased among the urbanites in South Korea, in due to the increase of leisure activity and the emergence of ageing society. Rural amenity resources are gaining various interests, in the value creation and promotion of tourism. In this study, the propensities of city dwellers were separated by life-style classification, and each affinity to the rural amenity resources was examined in accordance with the separation. A questionnaire survey of urbanites in the southern area of Gyeonggi-do, the most populous province in South Korea, was conducted to analyze the preference of city dwellers about rural amenity resource and life-style of themselves. For statistical verification, $IBM^{(R)}$ $SPSS^{(R)}$ Statistics 20 software was used for frequency, reliability, factor and multiple regression analysis of this research. The results of the statistical analyses found a noticeable characteristic in life-style classification. The affinities of urbanites can be classified into four congregations of life-style factors in this statistical model. Each congregation of the factors was named as 'Self-development-oriented', 'Leisure-oriented', 'Achievement-oriented', and 'Culture-oriented' life-style, to represent the characteristics for convenience' sake. Among these styles, only 'Self-development-oriented' and 'Achievement-oriented' showed the positive correlation with rural amenity resources in the multiple regression analysis. In addition, the rural amenity resources were also analyzed in accordance with the life-styles classification of urbanites. City dwellers showed the highest interest to the 'natural resource management facility resource' in natural resources, the 'traditional heritage resource' in cultural resources, and the 'community resource' in social resources. Meanwhile, they showed less interest to 'agricultural and scenery resources' in natural resources, 'specialty production resource' in cultural resources, and 'cooperative farming' in social resources. These characteristics can be constructed as meaning that the urbanites who concern self-development and achievement of their lives have high interest in rural amenity resources, and the main interest of them is not 'return-to-the-farm'(歸農) but 'return-to-the-home'(歸村).

Species Differences in Affinity and Efficacy of Carbachol for Ileal Muscarinic Receptors

  • The muscarinic effects of carbachol were compared on the isolated ileums of guinea-pig, rat and rabbit to elucidate the underlying mechanism of species differences in sensitivity for carbachol. The ED$\_$50/ value estimated on the guinea-pig ileum was 4 to 6-fold lower than those obtained on the rat and rabbit ileums, but the K$\_$A/ values of carbachol determined by functional assays were almost identical with 12-l7 ${\mu}$M in all of three ileums. The competition data of carbachol for [$^3$H]QNB binding were best described by a two-site model yielding the Ki values of 0.4-0.6${\mu}$M and 12-16${\mu}$M for high(K$\_$H/) and low(K$\_$L/) affinity sites, respectively. The low affinity dissociation constants(K$\_$L/) of carbachol determined from receptor binding studies thus were not significantly different from the K$\_$A/ values estimated from functional studies. The percentage of receptor occupation that carbachol requires for half-maximal response was approximately 3 to 5-fold lower in guinea-pig compared to rat and rabbit whereas the density of muscarinic binding sites per gram of ileum measured by [$^3$H]QNB saturation isotherms was two-fold higher in guinea-pig than that in rat and rabbit. Therefore, the numbers of muscarinic receptors occupied at ED$\_$50/ values of carbachol were about two-fold lower in guinea-pig, suggesting two-fold greater intrinsic efficacy. These results indicate that the guinea-pig ileum has higher muscarinic receptor density and greater intrinsic efficacy for carbachol than the rat and rabbit ileums.
