박테리오파아지 표면 발현 시스템을 이용한 Maackia amurensis Hemagglutinin (MAH)의 기능적 발현 (Functional Display of Maackia amurensis Hemagglutinin (MAH) on Bacteriophage)

  • A library of unlimited number of novel lectins with diverse specificities has been previously generated by randomly mutating the carbohydrate-recognition domain of Maackia amurensis hemagglutinin (MAH). To establish the experimental environment capable of selecting high affinity mutant lectins in E. coli, phage display system was adapted. Carbohydrate binding capacity of two phagemid vectors, pComb3 and pComb8 displaying wild-type MAH lectin was assessed. Specific bindings of pComb3 and pComb8 phages expressing w.t. MAH to affinity-purified polyclonal anti-MAH antibody and to glycophorin was demonstrated. Both phages also showed strong hemagglutinating activity to intact but not sialidase-treated human erythrocytes, which is consistent to the specificity of native MAH. Taken together, two different phage display vectors successfully allowed the expression of active MAH as a fusion protein on the surface of bacteriophage, which will lead to preparation of unique plant lectins with high affinity toward a variety of carbohydrate chains.

  • The photoelectrochemical conversion in SnO2 films deposited by spray pyrolysis using SnCl4-alcohol solution and N2 gas has been studied. The photocurrent increases with increasing deposition temperature up to 40$0^{\circ}C$ and then decreases, and the electron affinity decreases as the deposition temperature increases to 40$0^{\circ}C$. As the concentration of the spray solution increases, the photocurrent reaches a maximum value at the concentration of 0.05M, and the electron affinity is consistent. As the thickness of the film increases, the photocurrent increases with a maximum value at the thickness of 4600$\AA$, and electron affinity does not change.

금속소재 부품의 고장분석 사례 (Failure Analysis of Metallic Components)

  • Failure analyses were conducted on a crank shaft and a chock liner by using X-ray diffraction, optical microscopy and SEM/EDS techniques. In the crank shaft, a crack developed where a maximum tensile stress coincided with band structure formed by hot forging. The maximum tensile stress was observed to originate from volume expansion during high frequency induction heat treatment and the band structure to develop between upper and lower dies during hot forging. In the chock liner, the wear mechanism varied with the chemical affinity and hardness of liner material relative to friction pair of housing liner. Brass of low chemical affinity and hardness compared to housing liner showed uniform adhesive wear. STS 304 and STS 420J2 of high chemical affinity showed galling and scoring respectively.

칠성장어 (Lampetra japonica)젖 산수소이 탈효소 동위효소의 정제 및 면역화학

  • 칠성장어(Lampetro juponico) 골격근의 젖산수소이탈효소(EC 1. 1. 1.27, Lactate dehvdrogenase LDH) 동위효소 꺽지(Coreoperco herzi)의 LDH Aa 동위효소 및 소(Bos taurus)의 LDH B4 동위효소를 affinity chromatography로 정제하였고, 대구(Gadus macrocephcfus)의 LDH Ca 동위효소를 affinity chromatography와 OEAE-Sephacel chromatography로 정제하였다. 칠성장어 골격근 LDH 동위효소는 affinity chromatography에서 buffer를 유입한 후 용출되는 분획에서 모두 확인되었고, 정제한 칠성장어 골격근의 LDH에 대한 항체는 꺽지 LDH A4, C4 및 54 동위효소, 대구 LDH A4 및 B4 동위효소 그리고 생쥐(MUS musculus) LDH A4 등위효소와 복합체를 형성하였다. 칠성장어 골격근조직에서는 비각우 및 Ldh절가 발현되고 하부단위체 A의 구조는 하부단위체 B의 구조와 유사하며, 하부단위체 A는 진화상 보존적 이지만 하부단위체 C는 진화속도가 상당히 빠른 것으로 사료된다.

  • The utilization of resonance should be considered to get the maximum effect of ultrasonic to atomize liquid. The ultrasonic generator, transducer, horn, and all attached parts are used to produce the resonance, and specially the characteristics of liquids such as liquid load, property, and etc., for the liquid atomization affinity are considered. In this study, the variable device of liquid load was made and distilled water and city water selected as experimental liquids were sprayed by a twin-fluid spray method and their diameters, distributions, and spray quantum of spray droplets were measured by the light scattering system. And all data were observed, compared and considered relatively. In results, a lot of phenomena of liquid atomization affinity by ultrasonic appeared in accordance with liquid loads, namely head h.

  • Octadetkanubc으로 수식된 아미노산을 DPPC 혹은 DSPC와 혼합하여 리포솜-아미노산 결합체를 제조하였다. 리포솜의 크기는 100nm이고 구형이었다. DPPC와 글루탐산을 혼합하여 제조한 리포솜-아미노산 결합체가 글루타민이나 아스파라긴을 사용했을 때보다 포도당과의 친화성이 컸다. 제조한 리포솜의 안정성 면에서도 DPPC와 glucamic acid으로 구성된 리포솜-아미노산 결합체의 안정성이 높았다. 결국 포도당 친화성과 안정성을 갖춘 리포솜은 DPPC 와 글루탐산의 비가 7 : 3 으로 제조된 리포솜이었다.

Immunological Recognition by Artificial Neural Networks

  • The binding affinity between the T-cell receptors (TCRs) and antigenic peptides mainly determines immunological recognition. It is not a trivial task that T cells identify the digital sequences of peptide amino acids by simply relying on the integrated binding affinity between TCRs and antigenic peptides. To address this problem, we examine whether the affinity-based discrimination of peptide sequences is learnable and generalizable by artificial neural networks (ANNs) that process the digital experimental amino acid sequence information of receptors and peptides. A pair of TCR and peptide sequences correspond to the input for ANNs, while the success or failure of the immunological recognition correspond to the output. The output is obtained by both theoretical model and experimental data. In either case, we confirmed that ANNs could learn the immunological recognition. We also found that a homogenized encoding of amino acid sequence was more effective for the supervised learning task.

Purification of the Candida utilis Extracellular Invertase using Affinity Chromatography

  • The extracellular invertase (EC 3.2.1.26) (Candida utilis) preparation was obtained from the liquid medium after desalting and freeze drying. This prepared enzyme was used for the comparative purification on 4 activated matrices by liquid column affinity chromatography method. In this method there were used controlled porous glass (CPG) silanized covalently activated by keratin, silanized silica gel and silica gel covalently covered by keratin. It was found that the invertase purification process was better using both CPG matrices (silanized CPG and keratin activated CPG) than these with two silica gel supports. Also the elution coefficient of the invertase from the two CPG columns was about 93 to 94%. Two silica gel supports found to be superior in terms of purification efficiency. The invertase purification process was confirmed by PAGE electrophoresis.

대두품종(大豆品種)과 선발(選拔) Rhizobium japonicum간의 숙주(宿主) 친화성(親和性) (Host Specificities Between Soybean Varieties and Rhizobium japonicum Strains)

  • 우수(優秀) 대두접종제(大豆接種劑)로 이용할 균주(菌株)의 선발을 목적으로 국내(國內)에서 선발한 R. japonicum 균주(菌株)들과 현재 대두장려품종으로 보급되고 있는 대두품종들과의 상호(相互) 접종실험(接種實驗)을 통하여 숙주친화성(宿主親和性) 정도를 조사하였다. 얻어진 결과는 다음과 같다. 1. 공시(供試) 근류균주별(根瘤菌株別) 대두품종(大豆品種)과의 친화성(親和性) 정도를 보면 R. japonicum R-214, R-138 균주(菌株)가 근류형성능(根瘤形成能), 질소고정활성(窒素固定活性) 및 총(總) 질소흡수량(窒素吸收量)도 높아 대두품종(大豆品種)과 친화성(親和性) 범위가 비교적 넓었으며 R. japoncum R-224 균주(菌株)는 아주 낮은 것으로 판단되었다. 2. 대두품종별(大豆品種別) 근류균(根瘤菌)과의 친화성(親和性)은 황금(黃金)콩, 장백(長白)콩 등이 높았으며 그 다음이 장엽(長葉)콩, 단엽(短葉)콩, 백운(白雲)콩, 동북태(東北太) 등의 순이었고 방사(放射)콩이 제일 낮았다. 3. 일반적으로 질소고정활성(窒素固定活性)이 우수(優秀)한 균주(菌株)가 열등한 균주(菌株)보다 숙주친화성(宿主親和性) 범위가 넓은 것으로 판단되었다.

해명(海冥) 회종편(回腫片)에서 Morphine과 Naloxone 작용(作用)에 미치는 전해질(電解質)의 영향(影響) (Influences of Electrolytes on the Action of Morphine and Naloxone in Guinea-pig Ileum)

  • Guinea-Pig의 myenteric plexus-longitudinal muscle preparation에서 morphine과 naloxone 효과(效果)에 미치는 전해질(電解質)의 영향(影響)을 관찰(觀察)하였다. 표본(標本)은 Krebs-Henseleit bicarbonate tufter solution 으로 채운 organ bath에 현수(懸垂)하고 0.2Hz로 전기자극(電氣刺戟)하였다. Morphine은 전기자극(電氣刺戟)에 의(依)한 근편수축(筋片收縮)을 억제(抑制)하였으며, 이때 $ID_{30}$은 약(約) 190nM이었다. 이와같은 morphine의 억제작용(抑制作用)은 bath내(內) $Na^+$ 또는 $K+$ 농도(濃度)를 감소(減少)시키거나 $Mg^{2+}$을 가(加)하면 강화(强化)되었으며, $Ca^{2+}$ 농도(濃度)를 증가(增加)시키거나 $Mg^{2+}$를 농도(濃度)를 감소(減少)시키면 약화(弱化)되었다. Naloxone은 morphine의 작용(作用)을 억제(抑制)하였으며, 이때 naloxone에 대(對)한 affinity index인 $pA_2$ value는 약(約) 8.8이었고 that내(內) 전해질(電解質) 농도(濃度)를 변동(變動)시켜도 영향(影響)받지 않았다. 이 성적(成積)은 전해질(電解質) 변동(變動)으로 인(因)한 morphine의 작용변동(作用變動)은 전해질(電解質)이 opiate receptor의 affinity를 변동(變動)시킨다는 opiate-receptor binding실험(實驗)에서와는 달리 전해질(電解質) 변동(變動)에 의(依)한 functional opiate receptor의 affinity 변동(變動)에 의(依)한 것이 아님을 시사(示唆)한다.
