Microbiology and Antibiotic Selection for Diabetic Foot Infections (감염된 당뇨병성 족부 병변의 균주 조사 및 항생제 선택)

  • Purpose: To help the empirical antibiotics selection in diabetic foot infection patients, we investigated prevalence of microorganisms and their antibiotics sensitivity results. Materials and Methods: Patients who came to our clinics to treat diabetic foot infections with deep ulceration and were followed up more than 6 months until complete recovery were adopted. From March 2006 to June 2009, there were 140 patients who corresponded with such a inclusion criteria. Wound cultures were done by deep tissue or bone debris at first visit to our clinics. Microorganisms which was documented by wound culture and most susceptible antibiotics by minimum inhibitory concentrations were surveyed retrospectively. Results: Microorganisms were confirmed in 113 cases (80.7%). In the other 27 cases (19.3%), there were no cultured microorganisms. In bacterial growth group, there were 72 cases (63.7%) of gram-positive bacteria and 41 cases (36.3%) of gram-negative bacteria. All of them were aerobic microorganisms and there were no anaerobic microorganisms. Methicillin-sensitive staphylococcus aureus was the most common pathogen and accounted for 35 cases (31.0%). As other common pathogens, there were Enterobacter cloacae (11 cases, 9.7%), pseudomonas aeruginosa (10 cases, 8.8%), Methicillin-resistant staphylococcus aureus (10 cases, 8.8%) and enterococcus faecalis (6 cases, 5.3%), and so on. Common susceptible antibiotics in gram positive microorganism were vancomycin (60 cases, 83.3%), teicoplanin (60 cases, 83.3%), nitrofurantoin (60 cases, 83.3%) and ciprofloxacin (53 cases, 73.6%). In gram negative ones, common susceptible antibiotics were imipenem (35 cases, 85.3%), piperacillin/tazobactam (33 cases, 80.5%) and gentamicin (31 cases, 75.6%). Conclusion: Methicillin-sensitive Staphylococcus aureus in gram positive and enterobacter cloacae in gram negative was the most common pathogen in each group. Ciprofloxacin and gentamicin might be adaptable as a first-line empirical antibiotics in infected diabetic foot patients.

Screening and Characterization of Psychrotrophic, Lipolytic Bacteria from Deep-Sea Sediments

  • Of 23 psychrotrophic bacteria isolated from the west Pacific deep-sea sediments, 19 were assigned to the $\gamma$-Proteobacteria, 3 to the <$\beta$-Proteobacteria, and 1 to the Gram-positive bacteria, as determined by their 16S rDNA sequences. Ten psychrotrophs, affiliated to the Psychrobacter, Pseudoalteromonas, and Pseudomonas genera in the $\gamma$-Proteobacteria group, were screened for lipolytic bacteria. The majority of the lipolytic isolates had growth temperatures between 4-$30^\circ{C}$, and all of them were neutrophilic, aerobic, or facultatively anaerobic, and some were able to produce multiple kinds of ectohydrolytic enzymes. The deep-sea strains Psychrobacter sp. wp37 and Pseudoalteromonas sp. wp27 were chosen for further lipase production analysis. Both strains had the highest lipase production when grown at 10 to $20^\circ{C}$; their highest lipase production occurred at the late-exponential growth stage; and the majority of the enzymes were excreted to the outside of the cells. Lipases from both strains had the same optimal reaction temperature and pH (20-$30^\circ{C}$, pH 7-8) and could retain about 60% of their highest activity at $4^\circ{C}$. Furthermore, SDS-PAGE and an in-gel activity test showed that they had the same high molecular mass of about 85 kDa.

  • This experiment was conducted to find out the changes in microflora of submerged soil uncultivated rice plant by application of CNP herbicide (2, 4, 6 - Trichlorophenyl-4-Nitrophenyl ether) under conditions applied with compost, rice straw, glucose or without organic material. The soil, sandy loam textured was incubated in green house for 66 days. Sampling and analysis of microorganisms were carried out during submergence periods. The results were summarilized as follows. 1. Number of aerobic total bacteria was increased by application of CNP herbicide during submerbed 50 days, afterthen, could not seen the difference. The application of rice straw increased number of aerobic bacteria regardless of CNP herbicide application or not, but glucose tended to decrease. 2. Number of Fungi was constantly maintained at $8-20{\times}10^3$ levels during period of submergence regardless of application of CNP herbicide and organic materials or not. 3. The CNP herbicide application tended to decrease the number of actinomycetes, particularly, in the treatments without organic substances and rice straw were remarkably decreased. 4. Anaerobic-and gram-negative bacteria populations were not showed any difference by application of herbicide and organic materials. 5. The ratios of aerobic bacteria to fungi and aerobic bacteria to actinomycetes appeared high values by application of herbicide and of organic substances. 6. At 66 days after submergence, the ratio of chromogenic actinomycetes to the total number of actinomycetes was lowered in application of herbicide. On the other hand, the percentage of both pretense-positive and cellulase-positive actinomycetes to the total isolates were higher in the treatment with herbicide than An without herbicide, particularly. The ratios of pretense-positive actinomycetes were high in the rice straw application regardless of herbicide application or not, but cellulase-positive actinomycetes was not remarkably difference.

Isolation and Characteristics of Composting-promoting-bacteria (부숙촉진 미생물 분리 및 분리균의 특성)

  • This experiment was conducted to determine the safety and feasibility of using compost-promoting-bacteria. Compost-promoting-bacteria was isolated from livestock compost containing sawdust. The isolated bacteria was identified as Bacillus subtilis LYH201 by the method of the composition of the fatty acid with MIDI system and Bergey's manual. This Bacillus subtilis LYH201 had the following characteristics : Gram-positive, straight rod ($0.5{\sim}0.7{\mu}m$ width, $2.5{\sim}3.0{\mu}m$ length), facultatively aerobic and product of xylanase, CMCase, catalase, oxidase, protease and $0.5{\sim}0.7{\mu}m$-amylase. Growth of Bacillus subtilis LYH201 at saccharose as carbon source(0.5%) was faster than other carbon source. Activity of cellulase. $0.5{\sim}0.7{\mu}m$-amylase and protease from Bacillus subtilis LYH201 after 24 hours at $50^{\circ}C$ by agar diffusion method was higher than that of low temperature. Optimum growth condition of Bacillus subtilis LYH201 was $50^{\circ}C$ and pH 6.

Microbial Community Changes in the Soil of Plastic Film House as Affected by Anaerobic Fermentation of Rice Bran or Wheat Bran (쌀겨와 밀기울의 토양 혐기발효 처리가 시설 재배지 토양의 미생물상에 미치는 영향)

  • Soil microbial community has been changed after the treatment of anaerobic fermentation using wheat bran or rice bran was applied to the soil. In the dilution plate technique, the number of anaerobic bacteria and fungi was higher in rice bran-treated soil than in non and wheat bran-treated soil, but of yeast was higher in wheat bran-treated soil than in non and rice bran-treated soil. Specially, the fungi were not detected in the wheat bran-treated soil. Identified by 16S rDNA sequencing, the number of aerobic bacteria was similar in all treatments, the dominant bacteria was the genus Bacillus. In the phospholipid fatty acid (PLFA) technique, both Gram-positive and Gram-negative bacteria change slightly in all treatments for 20 days of fermentation process but, after 20day, increased rapidly in wheat or rice bran-treated soil. In conclusion, the microbial communities structure was dramatically changed after the treatment of wheat or rice bran to soil.

In vitro and in vivo Antibacterial Activities of the New Quinolone, DWQ-013 (새로운 퀴놀론계 항균제 DWQ-013의 항균작용)

  • ln vitro and in vivo antibacterial activities of DWQ-013(1-cyclopropyl-6,8-difluoro-7-(3-methylthiomethylpyrrolidinyl)-1,4-dihydro-4-oxo-3-quinolinecarboxylic acid), a new fluoroquinolone antibacterial agent, were compared with those of ciprofloxacin, sparfloxacin and ofloxacin against aerobic and anaerobic standard strains and clinical isolates. DWQ-013 had a broad spectrum and potent antibacterial activity against Gram-positive and Gram-negative bacteria. The antibacterial activity of DWQ-013 against Staphylococcus aureus was equal to that of sparfloxacin(SPFX) and superior to those of ciprofloxacin(CPFX). The antibacterial activity against Gram-negative bacteria was slightly lower than those of ciprofloxacin and sparfloxacin. MIC of DWQ-013 against Pseudomonas aeruginosa$(0.781{\sim}1.563\;{\mu}g/ml)$ was usually equal to that of sparfloxacin$(0.781\;{\mu}g/ml)$ and was inferior to that of ciprofloxacin$(0.098\;{\mu}g/ml)$. The number of viable cells was decreased rapidly after addition of DWQ-013 at concentration of $1{\sim}2$ folds of MIC.

Antimicrobial Activity of Black Garlic Pomace Extract and Its Application to Cleansing of Fresh Spinach Leaves for Microbial Control (흑마늘박 추출물의 항균 활성과 미생물 제어를 위한 시금치 세척에의 이용)

  • In this study, the antimicrobial activity of black garlic pomace extract (BGPE) was examined, and its washing applicability to spinach was investigated. BGPE had antimicrobial activity against both Gram-positive (Listeria monocytogenes and Staphylococcus aureus) and Gram-negative (Escherichia coli O157:H7 and Salmonella Typhimurium) food-borne pathogens. In particular, antimicrobial activities of BGPE against Gram-positive bacteria were higher than those against Gram-negative bacteria. Spinach samples were treated with 0.5% BGPE to determine the effect of BGPE on reducing naturally existing microorganisms on the surface of spinach leaves. BGPE treatment reduced populations of total aerobic bacteria and yeast/molds in spinach by 1.23~1.35 log CFU/g and 0.82~1.12 log CFU/g during 9 days of storage, respectively, compared with those of control samples. After treatment, there were no significant differences in color quality such as Hunter L, a, and b values and total color difference (${\Delta}E$). These results clearly indicate that BGPE treatment can be useful for improving microbiological safety and maintaining color quality of spinach during storage.

Isolation of Bacteria from Clinical Specimens in Veterinary Medical Teaching Hospital and Trend of Antimicrobial Susceptibility (대학 동물병원 임상 검체로부터 분리된 호기성 세균과 항생제 감수성 양상)

  • Bacteria that are resistant to several different groups of antibiotics have increased during the past few years. The importance of surveillance of antimicrobial resistance is now widely recognized. Unfortunately, this development has not been documented continuously in veterinary medicine in Korea. Therefore, the clinical prevalence and trend of antimicrobial susceptibility of aerobic isolates were investigated in this study. Total 121 isolates of aerobic bacteria were isolated from clinical specimens of dogs and cats at Veterinary Medical Teaching Hospital of Seoul National University from May 2001 to October 2002. Among them, the most common isolated species was Staphylococcus spp. (48 isolates), followed by E.coli (26 isolates), Enterococcus spp. (21 isolates), Klebsiella pneumoniae (9 isolates), Streptococcus spp. (6 isolates), Enterobacter cloacae (3 isolates), Pseudomonas aeruginosa (3 isolates), Corynebacterium xerosis (2 isolates), Chryseomonas spp. (2 isolates), and Providencia stuartii (1 isolate). The susceptibility of isolates to antibiotics was determined by the disk diffusion method. Gram-positive bacterial isolates were showed high susceptibilities to amikacin, amoxacillin/clavulanate, ceftazidime, and oxacillin, while Gram-negative bacterial isolates were showed high susceptibilities to amikacin and ceftazidime. Staphylococcus spp. were showed high susceptibilities to amikacin, amoxicillin/clavulanate, ceftazidime, cephalothin, and oxacillin. Streptococcus spp. and E.coli were showed high susceptibilities to amikacin and ceftazidime. Of the 48 staphylococci, seven Methicillin Resistant staphylococci were observed (14.6%), distributed among S. auricularis (1), S. hemolyticus (2), S. sciuri (1), S. saprophyticus (1), S. warneri (2) isolates. One strain of E.coli and one strain of Corynebacterium xerosis were resistant to all antibiotics tested. And, resistance trends between the you 2000 (from July 1999 to September 2000) and 2002 (from May 2001 to October 2002) were compared. Resistance to antibiotics was increased in both Gram-positive and Gram-negative bacterial isolates (p< 0.05). The resistance rates of Staphylococcus spp., E.coli and Klebsiella pneumoniae to all antibiotics tested were also increased (p<0.05). This study investigated increasing resistance between the year 2000 and 2002 in Veterinary Medical Teaching Hospital of Seoul National University. Surveillance resistance is helpful to alert to veterinarian and select of appropriate therapy. Antimicrobial susceptibility surveillance of isolates should urgently be continued in veterinary medicine.

Changes of Soil Microbe communities in Plastic Film House by Green Manure Crops Cultivation

  • To improve the soil condition for no-tillage organic pepper cultivation, four different green manure crops were cultivated. Fertilizer supply was depended on the biomass of the cultivated green manure crops, nitrogen supplies were 314kg in Vicia villosa and 341kg $ha^{-1}$ in Vicia angustifolia. In the microbial community analyzed by phospholipid fatty acid (PLFA) method, soil microbe populations were different among the green manure crops and fungi group was increased at Vicia angustifloia and Vicia villosa. The biological ratio indexes of fatty acids in the soils, the ratio of Gram-negative to Gram-positive bacterial PLFA and Ratio of aerobes to anaerobes were high at Vicia hirsute and Vicia tetrasperma suggesting the enrich of the aerobic conditions. The ratio of saturated to unsaturated fatty acids increased at Vicia angustifloia and Vicia villosa suggesting anaerobic conditions. Abundant biomass and uncomposted organic matter, the ratio of fungi to bacteria was increased at Vicia angustifloia and Vicia villosa.

Isolation and Identification of a Bacterium Producing Antifungal Antibiotic (항곰팡이성 항생물질의 생산하는 균주의 분리 및 동정)

  • A bactirium prohibiting the growth of fungus Botrytis cineria KT 433 was isolate and identified from soil. The isolated strain was gram positive, aerobic bacteria with cream color, round and mucoid. It showed ord form of 0.45$\times$1.1 ${\mu}{\textrm}{m}$ at the cultivated for 24 hrs and the ellipsoided endospore wer observed after culting for 72 hrs. The optimum growth temperature and pH wer 35$^{\circ}C$ and pH 5.0~8.0, respectively. It could assimilate daxtrin, maltose, glucose, mannose, ribose and collobiose as a sole carbon source. The isolated was confirmed to be a Bacillus sp. strain from the findings. The antibiotic from the isolated strain was stable up to 121$^{\circ}C$. The strain, especially, showed specific activity for mold and yeast. However, there was not significant antibacterial activity.
