Micropropagation and Mass Production of Adventitious Roots of Polygonatum odoratum via the Culture of Seedling Explnnts

  • Micropropagation and adventitious root production via the culture of Polygonatum odoratum were performed. Stem segments of seedlings of Polygonatum odoratum were the most efficient explants for adventitious shoot formation compared to leaf and root segments. Exogenous cytokinin treatment was required for adventitious shoot formation. Among the cytokinin (BA, Kinetin and Zeatin) tested, BA was most effective for shoot formation from stem segments. Auxin (NAA or IBA) in combination with cytokinin significantly enhanced adventitious shoot formation. Twenty five percent of explants produced adventitious shoots on medium with 2.0 mg/L BAP alone, while 83% of explants produced adventitious shoots on medium with the combination of 2.0 mg/L BAP and 0.1 mg/L IBA. Rooting of adventitious shoots was achieved after transferring to 112 MS medium supplemented with 0.1 mg/L IBA and 0.5 mg/L zeatin. When stem segments were cultured on MS medium with various kinds of auxin (IBA, NAA and 2,4-D), adventitious roots were formed from callus. frequency of adventitious root formation was highest in 2,4-D than NAA and IBA. When roots were in clusters together with parental stem segments, growth of roots actively occurred in hormone-free MS liquid medium. The above results represent that possible application for the mass production of roots and plantlets through in vitro culture system of Polygonatum odoratum.

  • When the leaves, roots and stem segments of seedling of Polygonatum odoratum were cultured on Murashige and Skoog medium with 2.0mg/l BAP, stem segments were the most efficient explants for adventitious shoot inductino. To observe the efficient combination of growth regulators on the adventitious shoot formation , stem segments were cultured on MS medium with various kinds of cytokinins (BAP, kinetin, zeatin). From this experiment, cytokinin treatement was prerequisite for theadventitious shoot formatino,especially BAP was the most effective. Auxin (NAA or IBA) in combination with cyotokinin highly enhanced the adventitious shoot formation. Twenty five percents of explants produced the adventitious shoots on medium with 2.0mg/l BAP solely, while 83% of explants produced the adventitious shoots on medium with 2.0mg/l BAP and 0.1mg/l IBA. Root formationform adventitious shoot was promoted after transfer to 1/2 MS medium supplemented with 0.1mg/l IBA and 0.5mg/l zeatin, thereafter the plantlets with shoots and roots were cultured on 1/2MS medium lacking growth regulators. When the stem segments were cultured to MS medium with 1.0mg/l 2,4 NAA and IBA , yellow and nodulous cali were formed from the stem segments which were developed into adventitious roots. These roots were actively grew after transferred to MS liquid medium lacking growth regulators.

포플라 잎절편의 부정아 분화와 Polyamine 함량의 연관성 (Correlations Between Polyamine and Adventitious Shoot Regeneration from Populus Leaf Segments)

  • Polyamine contents and the activities of their main biosynthetic enzymes, arginine decarboxylase (ADC) and ornithine decarboxylase(ODC), were investigated in Populus leaf segments during adventitious shoot regeneration with the addition of 1 millimolar polyamine synthesis inhibitors. From the study of polyamine synthesis inhibitors, ODC was found to be the principal route of polyamine biosynthesis in adventitious shoot regeneration of Populus leaf segments. The ADC inhibitor difluoromethyl arginine(DFMA) and ODC inhibitor difluoromethyl ornithine(DFMO) strongly reduced the putrescine content. On the contarary, DCHA, an inhibitor of spermidine synthase, increased it. Spermidine content was decreased with the treatment of each polyamine synthesis inhibitor, but the inhibitory effect of DCHA was stronger than any other polyamine inhibitor. The decreased polyamine level by polyamine synthesis inhibitors was restored with the exogenously applied polyamine. Comparing the polyamine contents with the adventitious shoot regeneration rate, were observed a close correlation between spermidine content and adventitious shoot regeneration of Populus leaf segments.

강원지역 패랭이꽃속의 캘러스로부터 식물체 재분화와 돌연변이체 유발 (Plant Regeneration and Mutagenesis from Organogenic Callus of Dianthus Distributed in Gangwon Province)

  • 패랭이꽃속의 분포와 개발을 위한 기초자료로 강원도에서 자생 2종과 수집가능한 7 외래종에 대한 기내배양체의 특성을 조사하였다. 캘러스의 유도를 위해 조직부위별 다양한 절편체들을 2,4-D. NAA, BA가 농도별로 처리된 MS 배지에 치상한 후, 27$^{\circ}C$ 광조건과 2.0mg/L 2,4-D와 0.5 mg/L BA의 조합배지에서 배양했을 때 3주 후 절단면에서 callus가 형성되기 시작하였으며, 패랭이꽃 (Dianthus chinensis)의 잎절편체에서 가장 양호하였다. Organogenic 캘러스의 선발은 기내에서 증식된 adventitious shoot의 절편체 유래 캘러스로부터 가능했으며 이들 캘러스를 1.0 mg/L BA와 0.1 mg/L NAA가 조합된 배지에 치상하였을 때 약 30% 정도의 shoot 형성률을 보였고 multiple shoot의 분화를 관찰할 수 있었다. In vitro에서 증식된 adventitious shoot 절편체를 1.0 mg/L BA와 0.1 mg/L NAA가 포함된 N6 배지에 치상했을 때, 캘러스 단계를 거치지 않고 직접 shoot이 형성되었고 D. gratianopol 유래 adventitious shoot 절편체는 52%의 높은 shoot 형성률을 보였다. 이들 multiple shoot로부터 재분화된 소식물체는 0.1 mg/L NAA가 포함된 MS 배지에서 뿌리가 유도되고 vermiculite가 담긴 pot에 이식 후 85% 습도하에서 활착되어 정상적인 개화된 개체를 획득할 수 있었다. 0.03 M EMS 처리된 패랭이꽃의 adventitious, shoot 절편체 유래 캘러스로부터 힌꽃의 패랭이꽃의 도연변체식물 (M23)이 분화되었으며 종자 형성이 가능하였다.

Medium compositions reveal potential organogenesis in the diploid and tetrploid Codonopsis lanceolata

  • Medium composition plays a key role on influencing organogenesis in plant tissue culture. This study was carried out to examine the effects of medium composition on organogenesis in diploid and tetraploid Codonopsis lanceolata and obtain in-vitro mass propagation of superior species of C. lanceolata. Diploid C. lanceolata was found to be declined regarding MS medium composition for each concentration. However, shoot and adventitious root formation were suppressed with higher mineral salt concentration, and active growth of shoot and adventitious root was exhibited as 4.9 cm and 3.2 cm respectively in 1/2 MS medium. While in tetraploid C. lanceolata, it showed 2.9 cm and 3.2 cm respectively in 1/4 MS medium. In the case of sucrose concentration, no consistent decrease was observed for growth of shoot and the adventitious root of diploid both at high and low concentration. The growth of shoot (at 3% concentration) and adventitious root (at 7% concentration) was 2.3 cm and 2.0 cm respectively. Although there was no difference in shoot formation of tetraploid C. lanceolata in all concentrations with the range of 1.7~1.8, there was a slight decrease in shoot growth at high concentration. Results revealed that the adventitious root formation was suppressed at high concentration. The concentration of agar exhibited no significant difference in shoot formation of diploid C. lanceolata at all concentrations. The maximum result of adventitious growth (4.1 cm) was observed at 0.8% concentration. Slight inhibition of shoot formation and root formation of tetraploid C. lanceolata was observed at higher concentration. Shoot formation of diploid C. lanceolata also exhibited inhibition at higher concentration. Shoot formation of diploid C. lanceolata was increased at lower pH and shoot growth was the highest (2.3 cm) at pH 3.8. Adventitious root formation was higher at lower pH. However, both the adventitious root formation and growth exhibited comparatively higher result at pH 5.8. Taken together, the levels of pH had an effect on shoot and root formation in diploid and tetraploid of C. lanceolata

홍경천 (Rhodiola sachalinensis A. Bor) 잎절편으로부터 식물체 재분화에 미치는 Cytokinin과 Putresine의 영향 (Effect of Cytokinin and Putrescine on Plant Regeneration from Leaf Explant of Rhodiola sachalinesis A. Bor)

  • 홍경천(Rhodilola sachalinensis A. Bor)의 잎절편로부터 부정아를 유도하고자 BA와 kinetin을 처리한 결과, BA의 처리가 kinetin의 처리보다 부정아 유도와 신초의 생장에 양호한 것으로 나타났으며, 특히 BA 1 mg/L처리에서 부정아 유도빈도와 신초길이는 각각 71%와 3.0 mm로 가장 높게 나타났다. 또한 고빈도의 부정아를 유도하고자 다양한 농도의 putrescine를 처리한 결과, putrescine 100 mM 처리에서 부정아 유도빈도가 BA 1 mg/L 단용처리보다 약 20%가 증가되었고 신초길이 역시 약 10 mm가 증가된 것으로 나타났다. 이상의 결과에서 유도된 부정아는 생장조절물질이 포함되지 않은 1/2MS배지에서 발근과 신장이 정상적으로 이루어졌으며 약 7 cm 정도 생장한 유식물체는 peatmoss:sand가 1:1로 혼합된 상토에서 약 95%이상 정상적으로 순화되었다.

갯방풍의 기내부정아 형성에 미치는 식물생정조절물질의 영향 (Influence of plant growth regulators on adventitious shoot formation of Glehnia littoralis Fr. Schmid)

  • Petiole explant of Glehnia littoralis Fr. Schmidt was in vitro cultured MS plant medium(DUCHEFA co.) supplemented with various plant growth regulators and examined to find out their optimum combination and concentration for plantlet regeneration. We investigated optimal condition for efficient plant regeneration through adventitious shoot formation on MS plant medium with various kinds of plant growth regulators. Embryogenic calli and adventitious shoot formation were greatly influenced by plant growth regulators. Embryogenic calli induction showed a good response on MS medium supplemented with NAA and BA than others. Especially, combination of 1.0 mg/L NAA and 0.5 mg/L BA on MS medium led to the greatest frequency in adventitious shoot. The results suggest that plant regulator selection be important factor to achieve an efficient regeneration.

Comparison of Adventitious Shoot Formation in Petiole Explant Cultures of 20 Cultivars of Catharanthus roseus

  • Petiole explants from 20 cultivars of Catharanthus roseus were cultured on various shoot-inducing media to assess their competence for adventitious shoot formation. After eight weeks of culture on Murashige and Skoog' s medium supplemented with 4.4 $\mu\textrm{m}$6-benzyladenine and 0.5 $\mu\textrm{m}$ $\alpha$-naphthaleneacetic acid, petiole explants from 'Cooler Icy Pink' exhibited the greatest frequency of adventitious shoot formation at 40%, which was followed by 'Little Bright Eye'. By comparing with a previous study on assessment of competence for adventitious shoot formation in hypocotyl explant cultures of various cultures, it is indicated that the relative degree of their competence among cultivars varies to the organ used for the source of explant. Excised adventitious shoots were readily rooted on half-strength MS basal medium. Regenerated plantlets were successfully transplanted to potting soil and grown to maturity in a greenhouse.

할미꽃 기내발근에 미치는 식물생장조절제의 영향 (Effects of Plant Growth Regulation on Adventitious Root Formation of Pulsatilla koreana Nakai)

  • 식물생장조절제의 종류와 농도를 모색하여 할미꽃의 발근 최적 조건을 조사하여 정상적인 재분화체를 획득하고자 하였다. 지금까지의 선행연구를 토대로 최적조건에서 shoot를 유도시킨 후 발근 유도 생장조절제에 바로 옮기면 shoot가 너무 작아 재분화체가 성장하지 못하고 고사되었다. 1/2 MS에서 키운 shoot를 3.0 mg/L NAA를 첨가한 배지에서 3일간 생장시킨 후, 1/3 MS로 옮겨 치상 한 경우, 약 10일 전후에 뿌리의 유도가 관찰되었다. 이렇게 뿌리가 유도된 재분화체는 염의 농도와 설탕의 농도를 동시에 순차적으로 낮추어 순화처리되었다.
