The Effects of NOG-YONG Aqua-Aqupuncture Solution on Cytotoxicities in Primary Culture of Adult Rat Hepatocytes

  • This experimental study was carried out to investigate on the safety of Nog-yong aqua-acupuncture solution in primary culture of adult rat hepatocytes. Primary culture of adult rat hepatocytes has been considered as a ideal model for toxicological studies because cultured hepatocytes maintained many liver-specific functions. In this research, we investigated the effects of Nog-yong aqua-aqupunture(1-10$\mu\textrm{g}$/ml) on cytotoxicites in primary culture of adult rat hepatocytes using LDH release assays. Hepatic glutathione level. glutathione-S-transferase activity, and albumin synthesis were not affected by treatment with Nog-yong aqua-aqupuntur alone. Nog-yong aqua-aqupuncoure solution(0.5-$10{\mu}\textrm{g}$/ml) on cytotoxicites in primary culture of adult rat hepatocytes using LDH release not significantly affected normal functional charaterists.

Ultrastructural Study on the Morphogenesis of Rat Spleen (흰쥐 비장의 형태형성에 관한 미세구조적 연구)

  • Morphogenesis of rat spleen was studied by light and electron microscope from the fetal stage till the newborn or adult stages. The results indicate as follows: at the 14th day of gestation rat spleen, as an early form, consists of intercellular spaces and mesenchymal cells. And at this stage the spleen is in a premature state, then it appears its adult condition in structures after the 7th postnatal day. Erythropoiesis is shown to be an active process in rat spleen beginning about the 18th day of gestation, and once established the process continues at least till the 7th postnatal day. At the 20th day of gestation, there are splenic nodules, trabeculae, venous sinus, and granular leucocytes such as neutrophils and basophils in rat spleen. Lymphocytes appeared to be well differentiated at the 7th postnatal day and were present till the adult stage. While degenerating erythrocytes are phagocytosed by macrophages. In conclusion, rat spleen started to be appeared from the 14th day of gestation and erythropoiesis in rat spleen was carried out for about 10 days between prenatal and postnatal stage. Erythrophagocytosis was accomplished by macrophages and it is suggested that the proper functions of rat spleen set off from the 7 th postnatal day when its structures are similar to the adult's.

The Effects of Dietary Protein and Caffeine Consumption Levels on Calcium Phosphorus, Sodium and Potassium Metabolism in the Rats of Different Ages (단백질과 카페인의 섭취수준이 나이가 다른 흰쥐의 칼슘, 인 , 나트륨 및 칼륨 대사에 미치는 영향)

  • This study was designed to investigate the effects of dietary protein and caffeine consumption levels on Ca, P, Na and K metabolism. One hundred twenty rats were divided into twelve groups according to age, protein level and caffeine consumption such as group(120-130g young rat, 250-300g adult rat) , dietary protein group (20% normal protein , 85 low protein) , caffeine consumption group(0, 3.5mg, 7.0mg). Low protein diet containing high caffeine levels caffeine levels increased calcium, phosphorus, sodium and potassium contents of urine and fecal in rats. Young rat has higher level of calcium, phosphorus , sodium and potassium than adult rat. In the serum, calcium contents were not affected by age, dietary protein levels and caffeine consumption . However, phosphorus, sodium and potassium contents of serum in adult rat were higher than young rat. In the liver, potassium decreased with decreasing dietary protein levels. In the kidney, calcium , phosphorus and potassium contents were not different by age group, dietary protein levels and caffeine consumption , but sodium content was significantly reduced in the adult rat.

Differentiation and Distrbution of the Choline Acetyltransferase-immunoreactive Nerve Cells in the Magnocellular Preoptic Nucleus of the Rat Forebrains during the Postnatal Development

  • This study was performed to investigate the differentiation and distribution of choline acetyltransferase (ChAT}-immunoreactive cells in the magnacellular preoptic nucleus (MCPO) of the postnatal and adult rat forebrains, utilizing techniques of immunocytochemistry. According to the cell shape and the ratio of long axis versus short axis of cell soma, the ChATimmunoreactive nerve cells in the MCPO were classified into six types: 1) round, 2) oval, 3) elongated, 4) fusiform, 5) triangular, and 6) polygonal types. Frequency distributions of the oval and round nerve cells on the postnatal day (PND) 0 were observed to be high. But in the adult, frequency distributions of the same cells were shown to decrease. Compared to those of the postnatal rats, frequency distributions of elongated, fusiform, triangular, and polygonal nerve cells in the adult were increased. The total mean volumes of ChAT-immunoreactive cell somata in the MCPO of PND 0 rat were the lowest, while those in the PND 17 rat were shown to be the highest and decreased in the adult. The soma volumes of the immunoreactive cells at the PND 17 were evenly distributed, but those in the other developmental stages (e.g. PND 7 and adult) appeared to exhibit unimodal distributions. On the electron micrography, the free ribosomes, polysomes, and rough endoplasmic reticula (RER) of the nerve cells in the MCPO of PND 21 rat forebrains were immunoreactive to ChAT in the tissues untreated with triton X-100. According to the observations in the present study, it is considered that the ChAT-immunoreactive nerve cells in the MCPO of the rat forebrains are differentiated throughout the following processes during the postnatal development: 1) increase in cell soma volumes, 2) development of neurites, 3) increase in the frequency of differentiated cell types, and 4) decrease in cell soma volumes. The ribosomes, polysomes, and RER are considered to be closely related to the intracellular localization and biosynthesis of the ChAT but not Golgi complex.

  • The synthesis of steroid hormone starts from cholesterol. Steroidogenic acute regulatory protein (StAR) acutely transfers cholesterol from the outer mitochondrial membrane to the inner in the early step of steroidogenesis. Many kinds of steroid hormone are mainly synthesized in adrenal grand, ovary, and testis. Among the steroid hormone, testosterone is synthesized in Leydig cells of the testis, the production of testosterone significantly increases in adult testis after puberty onset. Therefore, we think that the expression of StAR mRNA in testis will change according to the testicular development. The aim of this study is to determine the distribution of StAR mRNA in immature and adult rat testes and to confirm the functions of StAR in these testes. Thus, in situ hybridization was used in rat testes of the 2, 4, and 10 weeks of age. StAR mRNA was expressed in Leydig cells. Positive signals of StAR mRNA were weakly detected in Leydig cells of the 2 weeks of age. But, StAR mRNA was strongly expressed in Leydig cells of the 4 and 10 weeks of age, where steroidogenesis actively occur. In our results, the pattern of StAR mRNA expression was similar to the pattern of testosterone production in immature and adult rat testes. In conclusion, we can suggest that StAR acts as an important factor to regulate the synthesis of testosterone in Leydig cells of the rat testis.

The immunohistochemical study on the morphological changes of oxytocin secreting neurons in the age-related rat hypothalamus (고령에 따른 흰쥐 시상하부 oxytocin 분비세포의 형태적 변화에 대한 면역조직화학적 연구)

  • This study was carried out to investigate the morphological changes of the oxytocin secreting neurons in age-related rat hypothalamus by means of immunohistochemistry. The experimental group was 20 aged female rats (Sprague-Dawley, 25~30 months), and the control group was 10 adult female rats (10 months). All animals were perfused transcardially with 4% paraformaldehyde-lysine-periocdate(PLP), and serial transverse brain sections($30{\mu}m$) were prepared by means of cryotome, and were stained with rabbit anti-oxytocin antisera immunohistochemically, using the free floating method and avidin-viotin peroxidase complex. The results were as followings. 1. The immunostained oxytocin secreting neurons were located at the paraventricular nucleus and supraoptic nucleus of age-related rat hypothalamus chiefly. 2. The numbers of oxytocin secreting neurons decreased at the paraventricular nucleus and supraoptic nucleus of age-related rat hypothalamus(p<0.01) 3. The oxytocin secreting neurons of age-related rat hypothalamus immunostained less than those of the adult rat hypothalamus, and the paraventricular nucleus immunostained greater than supraoptic nucleus in age-related rat. 4. The numbers and expansion of dendrite and axonal varicosities decreased in the age-related rat hypothalamus greatly.

  • Spherical multicellular aggregates of adult rat hepatocytes (spheroid) which have tissue like structure, were formed and immobilized in the pores of polyurethane foam (PUF) which was used as a culture substratum. These hepatocyte/spheroids, about 100 $\mu\textrm{m}$ in diameter, have maintained higher differentiated functions than those of hepatocyte/monolayer for about 3 weeks in serum-free medium. Then, we designed a prototype module of an artificial liver support system using a PUF/spheroid packed-bed, in which hepatocyte/spheroids were immobilized at high density. The urea synthesis activity of the artificial liver was maintained at least 10 days in 100% rat blood plasma. We start examining the performance of hybrid artificial liver in an ex vivo extracorporeal experiment with an acute hepatic failure rat.

  • Pituitary adenyl ate cyclase activating polypeptide (PACAP) was originally isolated from the ovine hypothalamus and stimulated cAMP production in anterior pituitary cells. It is known that PACAP stimulates cAMP accumulation and contributes to the spermatogenesis and steroidogenesis in rat testis. The principal aim of this study is to determinate the distribution of PACAP mRNA and protein in adult rat testis. For this study, we used in situ hybridization and immunohistochemistry techniques in adult rat testis. PACAP mRNA was stage specifically expressed in seminiferous tubules. Positive signals of PACAP mRNA were detected in the developing germ cells at stages HI-VII of the epithelial cycle. The strongest signals of PACAP mRNA and protein were detected in round spermatids at stages V to early VII of the cycle. These results demonstrate that PACAP which is synthesised in the developing germ cells contributes to the spermatogenesis in rat testis. Thus, we suggest that PACAP plays a critical role in the function of testis.

Distribution and Differentiation of the Choline Acetyltransferase-immunoreactive Nerve Cells in the Basal Nucleus of Meynert of the Rat Forebrains During the Postnatal Development (흰쥐 출생후 발생에 따른 전뇌 기저부 Meynert기저핵에서 Choline Acetyltranseferase 면역반응 신경세포의 분포 및 분화)

  • This study was performed to investigate the distribution and differentiation of choline acetyltransferase (ChAT)-immunoreactive cells in the basal nucleus of Meynert of the postnatal and adult rat forebrains, utilizing techniques of immunocytochemistry. According to the cell shape and the ratio of long axis vs short axis of cell soma, the ChAT-immunoreactive nerve cells in the basal nucleus of Meynert of the adult rat were classified into six types. In the adult rat, the frequency distributions (FD) of round, oval, elongated, fusiform, triangular and polygonal cells were 9.4%, 35.5%, 32.1%, 5.9%, 9.1% and 8.0%, respectively. The FD of oval and round nerve cells on the postnatal day (PND) 14 were observed to be 18.7% and 51.5%, respectively. Those were shown to be progressively decreased during developmental process to the adult. Also, those of elongated and triangular nerve cells on the PND 21 were observed to be 30.4% and 10.1%, respectively. Those were shown to be same phenomenon a,1 those in the round and oval cells. Meanwhile, those of the triangular and polygonal nerve cells were progressively increased from the early postnatal stage to the adult. The total mean volumes of ChAT-immunoreactive cell somata in the PND 7 rat were the lowest $(1,083{\mu}m^3)$ and those in the PND 21 rat were shown to be the highest $(5,045{\mu}m^3)$. But in the adult, those were decreased to $(2,731{\mu}m^3)$. Those in the PND 21 rat were shown to be about 84.7% larger than those in the adult. On the electron micrography, the cell organelles such as ribosomes, polysomes, rough endoplasmic reticula (RER) and mitochondria were well developed in the PND 21 rat forebrains, but Golgi complexes were shown to be proliferating phase. Especially, ribosomes, polysomes and RER were immunoreactive in the tissues treated with 0.05% triton X-100. According to the observations in the present study, it is considered that the ChAT-immunoreactive nerve cells in the basal nucleus of Meynert of the rat forebrains are differentiated throughout the following processes of changes during the postnatal development: 1) increase of cell soma volumes with the differentiation of tell organelles and neurites, 2) increase in the FD of differentiated tell types and 3) cell schrinkage without cell loss. The ribosomes, polysomes and RER are considered to be closely related to the intracellular localization and biosynthesis of the ChAT but not Colgi complex.

Expression of Peroxiredoxin I and II in Neonatal and Adult Rat Lung Exposed to Hyperoxia (고산소에 노출된 신생 백서와 성숙 백서에 있어서Peroxiredoxin I과 II의 발현)

  • Background : In mammals, the activity of antioxidant enzymes is increased in adult lung to adapt to hyperoxia. The increase of these activities is augmented in neonates and is known as an important mechanism of tolerance to high oxygen levels. Peroxiredoxin(Prx) is an abundant and ubiquitous intracellular antioxidant enzyme. Prx I and II are major cytosolic subtypes. The aim of this study was to examine th Prx I and II mRNA and protein expression levels in adult rat lungs and to compare then with those of neonatal rat lungs exposed to hyperoxia. Materials and Methods : Adult Sprague-Dawley rats and neonates that were delivered from timed pregnant Sprague-Dawley rat were randomly exposed to normoxia or hyperoxia. After exposure to high oxygen level for a set time, the bronchoalveolar lavage fluid and lung tissue were obtained. The Prx I and II protein expression levels were measured by western blot analysis using polyclonal rabbit anti-Prx I or anti-Prx II antibodies and the relative expression of the Prx I and Prx II per Actin protein were obtained as an internal standard. The Prx I and II mRNA expression levels were measured by northernblot analysis using Prx I and Prx II-specific cDNA prepared from pCRPrx I and pCRPrx II, and the relative Prx I and Prx II expression levels per Actin mRNA were obtained as an internal standard. Results : Hyperoxia induced some peak increase in the Prx I mRNA levels after 24 hour in adult rats. Interestingly, hyperoxia induced a marked increase of Prx I mRNA 24 hour in neonatal rats. However, hyperoxia did not induce an alteration in the expression of Prx II mRNA in both the adult and neonatal rat lungs. Hyperoxia did not induce an alteration in the expression of the Prx I and Prx II protein in both the adult and neonatal rat lungs. Hyperoxia did not induce an alteration in the amount of Prx I and Prx II protein all the times in the bronchoalveolar fluid of adult rats. Conclusion : Prx I and II is differently regulated by hyperoxia in adult and neonatal rat lung at the transcriptional level. The prominent upregulation of Prx I mRNA in neonates compared to those in adults by hyperoxia may be another mechanism of resistance to high oxygen levels in neonate.
