제주도 넙치 양식현장에서 배합사료 성장 평가 (Evaluation of extruded pellet for growth performance of olive flounder Paralichthys olivaceus in Jeju farm field)

  • This study was conducted to evaluate extruded pellet (EP) diet compared with a raw fish moist pellet (MP) diet for flounder, Paralichthys olivaceus on field feeding experiments in Jeju area. The experimental EP diet (juvenile, growing and adult) was prepared based on the former studies on nutrient requirements and feed ingredient of flounder. Fish were distributed randomly to each aquarium as a group of 102,300 fish (initial mean weight 97 g) in field experiment I and reared randomly to each aquarium as a group of 40,000 fish (initial mean weight 120 g) in field experiment II. In field feeding experiment I conducted in commercial flounder farm, survival, wt. gain (94-1,090 g), feed efficiency and condition factor of fish fed experimental EP were comparable to those of fish fed MP, but considerably higher protein efficiency ratio were observed in fish fed experimental EP. In field feeding experiment II, no significant difference was observed in final mean weight of fish fed the experimental EP (1,234 g) and MP (1,286 g), and any noticeable problem caused by feeding the experimental EP was not found during the whole period of feeding trial. Survival of fish fed the experimental EP was lower than that of MP, and there was no significant difference in feed efficiency between the fish fed experimental EP and MP. Therefore, these results strongly suggest that diet EP could be developed to replace MP diet for the marketable size (1.3 kg) of production for flounder without adverse effects on growth performance. In particular, abdominal dropsy and reduction of condition factor, which are frequently occurred in commercial EP feeding farms, were not observed in flounder fed experimental EP.

Expression Pattern of Early Growth Response Gene 1 during Olive Flounder (Paralichthys olivaceus) Embryonic Development

  • The early growth response protein 1 (Egr-1) is a widely reported zinc finger protein and a well known transcription factor encoded by the Egr-1 gene, which plays key roles in many aspects of vertebrate embryogenesis and in adult vertebrates. The Egr-1 expression is important in the formation of the gill vascular system in flounders, which develops during the post-hatching phase and is essential for survival during the juvenile period. However, the complete details of Egr-1 expression during embryo development in olive flounder are not available. We assessed the expression patterns of Egr-1 during the early development of olive flounders by using reverse transcription polymerase chain reaction (RT-PCR) analysis. Microscopic observations showed that gill filament formation corresponded with the Egr-1 expression. Thus, we showed that Egr-1 plays a vital role in angiogenesis in the gill filaments during embryogenesis. Further, Egr-1 expression was found to be strong at 5 days after hatching (DAH), in the development of the gill vascular system, and this strong expression level was maintained throughout all the development stages. Our findings have important implications with respect to the biological role of Egr-1 and evolution of the first respiratory blood vessels in the gills of olive flounder. Further studies are required to elucidate the Egr-1-mediated stress response and to decipher the functional role of Egr-1 in developmental stages.

Cloning, expression, and activity of type IV antifreeze protein from cultured subtropical olive flounder (Paralichthys olivaceus)

  • Antifreeze proteins (AFPs) lower the freezing point but not the melting point of aqueous solutions by inhibiting the growth of ice crystals via an adsorption-inhibition mechanism. However, the function of type IV AFP (AFP IV) is questionable, as its antifreeze activity is on the verge of detectable limits, its physiological concentration in adult fish blood is too low to function as a biological antifreeze, and its homologues are present even in fish from tropic oceans as well as freshwater. Therefore, we speculated that AFP IV may have gained antifreeze activity not by selective pressure but by chance. To test this hypothesis, we cloned, expressed, and assayed AFP IV from cultured subtropical olive flounder (Paralichthys olivaceus), which do not require antifreeze protein for survival. Among the identified expressed sequence tags of the flounder liver sample, a 5'-deleted complementary DNA (cDNA) sequence similar to the afp4 gene of the longhorn sculpin was identified, and its full-length cDNA and genome structure were examined. The deduced amino acid sequence of flounder AFP IV shared 55, 53, 52, and 49 % identity with those of Pleuragramma antarcticum, Myoxocephalus octodecemspinosus, Myoxocephalus scorpius, and Notothenia coriiceps, respectively. Furthermore, the genomic structure of this gene was conserved with those of other known AFP IVs. Notably, the recombinant AFP IV showed a weak but distinct thermal hysteresis of $0.07{\pm}0.01^{\circ}C$ at the concentration of 0.5 mg/mL, and ice crystals in an AFP IV solution grew star-shaped, which are very similar to those obtained from other polar AFP IVs. Taken together, our results do not support the hypothesis of evolution of AFP IV by selective pressure, suggesting that the antifreeze activity of AFP IV may have been gained by chance.

부유물질증가에 따른 저서성 해양생물의 독성평가에 관한 연구 (Ecotoxicological Effects of the Increased Suspended Solids on Marine Benthic Organisms)

  • Environmental impacts of suspended solids (SS) released in coastal area by dredging, reclamation and construction can cause serious damages to coastal habitats and benthic organisms. Acute toxicity tests (4-7 days) were conducted to identify the relationship between SS concentration and mortality of three marine benthic species; benthic copepod (Tigriopus japonicus) adult, Pacific abalone (Haliotis discus hannai) spat, and olive flounder (Paralichthys olivaceus) fry. Benthic copepod was the most sensitive to SS followed by olive flounder fry and Pacific abalone spat, with an $LC_{50}$ (lethal concentration of 50% mortality) value of 61.0 mg/L and LOEC (lowest observed effective concentration) value of 31.3 mg/L for benthic copepod. LOEC and 7 day-$LC_{50}$ for Pacific abalone spat were 500.0 mg/L and 1887.7 mg/L, and those for olive flounder fry were 125.0 mg/L and 156.9 mg/L, respectively. The tolerance limits of the test species to SS revealed the various concentration ranges of SS, which reflects the physiology and ecology of the test species. These results are very valuable for the determination of SS concentration of effluents released into the coastal area by dredging, reclamation and construction etc. Also, sharp increase of SS can cause long-term damages to the benthic and sessile fauna by blanketing of benthic substratum. These experimental procedures for marine bioassay and acute toxicity results can be a useful guideline for practical management planning of SS discharge into coastal area.

  • This study was conducted to investigate the effect of extruded pellet (EP) size on the growth performance of olive flounder Paralichthys olivaceus at the juvenile (Exp-I; $62.2{\pm}0.05$), growing (Exp-II; $150{\pm}1$) and sub-adult (Exp-III; $299{\pm}1$) stages. In Exp-I (3, 4, 5 and 6 mm diets), Exp-II (4, 9, 11 and 13 mm diets) and Exp-III (9, 11, 13 and 16 mm diets), triplicate groups of fish were fed four EP diets of different pellet sizes. In Exp-I, larger pellet sizes were significantly better than the smallest pellet size (3 mm). In Exp-II, significantly improved feed and protein efficiency were observed with increasing pellet size. The feed conversion ratios of the fish fed the 4 and 9 mm diets were significantly lower than those of fish fed 11 and 13 mm diets. The dietary digestibility of protein and energy was higher in smaller EPs than in larger sizes. In Exp-III, the growth performance of fish was not affected by the pellet size. However, feed intake was significantly affected, increasing with increasing pellet size. Therefore, the optimum pellet sizes for olive flounder under the conditions in this study were 5 mm for 60-150 g fish and 9 mm for 150-500 g fish.

  • We investigated the effects of feeding rate on the growth, blood components, and histology of sub-adult olive flounder Paralichthys olivaceus. Optimum feeding rate (initial fish mean weight : $384.2{\pm}5.91g$) was determined under the optimum water temperature. Two replicated groups of fish were fed a commercial diet at rates of 0%, 0.3%, 0.5%, and 0.7% body weight (BW) per day, and to satiation. The feeding trial was conducted using a flow-through system with ten 1.2-metric ton aquaria receiving filtered seawater at $20-24.5^{\circ}C$ for 3 weeks. After the feeding trial, the weight gain (WG) and specific growth rate (SGR) were significantly higher in fish fed at 0.7% BW/day and those fed to satiation (0.9% BW/day) than in fish fed at other feeding rates or in the unfed fish. These parameters were negative and significantly lower in the unfed fish than in those fed the experimental diet at all feeding rates. There were no significant differences in WG and SGR among fish fed at 0.3 and 0.5% BW/day and among those fed at 0.7% BW/day and to satiation. The histological changes in the hepatopancreas, kidney, and anterior intestine of fish fed at 0, 0.5, and 0.9 % BW/day did not differ much. Broken-line regression analysis of weight gain showed that the optimum feeding rate of olive flounder weighing 384 g was 0.74% BW per day at the optimum water temperatures.

Two replicated groups of fish were fed a commercial diet at rates of 0%, 0.3%, 0.4%, 0.5%, and 0.6% body weight (BW) per day, and to satiation (0.75%). Feeding trial was conducted under a flow-through system with 12 1.2-metric ton aquaria receiving filtered seawater at $12-14^{\circ}C$ for 4 weeks. Weight gain (WG) and specific growth rate (SGR) for fish fed at 0.6% BW per day was significantly higher than that of unfed fish (0%) and fish fed at 0.3% and 0.4% BW per day. There were no significant differences in WG and SGR among fish fed at 0.5%, 0.6%, and 0.75%. These parameters were negative and significantly lower in the starved fish than in fish fed the experimental diet at all feeding rates. Survival for unfed fish (0%) was significantly lower than that of fish fed at 0.4%, 0.5%, and 0.6%. Hematocrit and hemoglobin content of fish fed at 0% and 0.75% (satiation) were significantly lower than that of fish fed at 0.4% BW per day. Total protein content in unfed fish was significantly lower than those in other treatments. Broken-line regression analysis of weight gain showed that the optimum feeding rate of olive flounder weighing 370 g was 0.51% BW per day at the low water temperature.

제주지역 양식넙치의 연쇄구균증 발생동향 및 원인균에 대한 분자적 동정 (Characterization of Streptococcosis Occurrence and Molecular Identification of the Pathogens of Cultured Flounder in Jeju Island)

  • 제주지역 양식넙치의 주요 세균성 질병의 일종인 연쇄구균증 발병동향과 특성을 분석하였다. 제주지역 양식넙치의 연쇄구균증은 연중 발생하는 양상을 보였으나 상대적으로 고수온기에 발생율이 높았으며, 30cm 이상의 개체에서 주로 관찰되었다. 2003년 6월부터 2005년 5월 사이에 분리된 연쇄구균 균주를 multiplex PCR assay 기법을 이용하여 종 조성을 분리한 결과 Streptococcus iniae와 S. parauberis로 동정되었으며 그 비율은 각각 46%와 54%이었다. S. iniae는 상대적으로 9월과 10월에 높은 분리율을 나타내었으나 S. parauberissms 3월과 4월에 분리율이 높게 나타났다. S. iniae와 S. parauberis 감염개체의 병리학적 중상은 일부분 유사하였으나 S. iniae 분리개체는 복부팽만, 탈장, 복수저류증상이 주로 관찰되는 반면, S. parauberis 분리개체는 체색흑화, 무안측 발적 중상이 두드러지게 관찰되었다. 이 두 종의 병원체 모두 ampicillin과 amoxicillin에 감수성이 높았으며, S. iniae 균주들은 S. parauberis 균주들보다 상대적으로 doxycycline, erythromycin, oxytetracycline에 감수성이 높았다.

  • We examined the cause of a disease outbreak in adult olive flounder Paralichthys olivaceus, which occurred at a Korean aquaculture farm in Korea in 2011. The principal signs included an expanded abdomen and congested liver, with persistent mortality (a little over two months). At the beginning of the outbreak, farm administrators misjudged the disease as bacterial in origin, because of the aforementioned signs, persistent mortality, and the detection of bacterial species, including Vibrio spp. and Streptococcus spp. Moreover, the detection of viral hemorrhagic septicemia virus (VHSV) by reverse trasnscription-PCR analysis was complicated by use of the VHS-VN primer set, which has been in general use recently, because it produced weak bands in some samples. Therefore, we recommend the use of at least two different primer sets in the diagnosis of VHSV. Our histopathological findings indicate that necrotizing myocarditis could be considered a pathogenic sign of VHSV infection.
