분화된 3T3-L1 세포에서 수세미오이 메탄올 추출물의 지방분해 효과 (Lipolytic Effect of Methanol Extracts from Luffa cylindrica in Mature 3T3-L1 Adipocytes)

  • 분화된 3T3-L1 지방세포에서 수세미오이의 메탄올 추출물이 지방의 축적 및 분해에 미치는 영향을 확인하였다. 수세미오이의 메탄올 추출물 안전범위(0~1000 ${\mu}g$/mL)에서 지방대사 관련한 여러 과정을 확인한 결과 $600\;{\mu}g$/mL 이상의 농도에서 세포 내 지방축적이 억제되었고 특히 지방구내의 triglyceride 함량이 억제되는 것으로 나타났다. 이러한 결과에 대한 기전을 위해 포도당의 유입 억제로 인한 세포내 triglyceride의 합성감소와 지방분해효소의 활성화로 인한 세포 내 triglyceride의 분해 증가의 두 가지로 확인하였다. 우선적으로 수세미오이의 메탄올 추출물의 포도당 유입 억제 효과를 실험하였지만 아무런 영향이 나타나지 않았다. 하지만 지방분해효소 중 LPL을 제외한 HSL과 ATGL의 유전자 발현이 증가됨에 따라 세포 내 triglyceride이 지방산과 glycerol로 분해되었을 것으로 생각되며 이러한 결과는 세포외로 유리된 glycerol의 함량이 수세미오이의 메탄올 추출물 $1000\;{\mu}g$/mL의 농도에서 증가한 것을 통해 재확인할 수 있었다. 하지만 $600\;{\mu}g$/mL의 농도에서 triglyceride 분해는 촉진되었으나 glycerol의 유리가 유의적으로 나타나지 않은 것은 세포 내에서 분해된 glycerol의 일부가 세포 내 생합성에 재사용되었을 것으로 생각되며 실제 세포 외로 유리된 양을 확인하기 위해서는 보다 높은 농도가 필요한 것으로 여겨진다. 전반적으로 본 실험에서는 수세미오이의 메탄올 추출물이 분화된 3T3-L1 지방세포에서 지방구의 생성을 억제하였으나 효과 농도가 다른 많은 천연물에 비해 다소 높게 나타남으로써 제약으로서의 생산으로는 제한점이 있을 것으로 생각되는 반면 식품으로서 꾸준히 사용될 경우 항비만 소재로서의 가능성이 있을 것으로 생각된다.

Korean Curcuma longa L. induces lipolysis and regulates leptin in adipocyte cells and rats

  • BACKGROUND/OBJECTIVES: Turmeric (Curcuma longa L.) has been reported to have many biological functions including anti-obesity. Leptin, peptide hormone produced by adipocytes and its concentration is increased in proportion to the amount of the adipocytes. In the present study, we examined the effects of Korean turmeric on the regulation of adiposity and leptin levels in 3T3-L1 adipocytes and rats fed a high-fat and high-cholesterol diet. MATERIALS/METHODS: Leptin secretion, free fatty acid and glycerol contents in 3T3-L1 adipocytes were measured after incubation of cells with turmeric for 24 hours. Rats were divided into four experimental groups: a normal diet group (N), a high-fat and high-cholesterol diet group (HF), a high-fat and high-cholesterol diet group supplemented with 2.5% turmeric extracts (TPA group) and a high-fat and high-cholesterol diet group supplemented with 5% turmeric extracts (TPB group). Serum samples were used for the measurement of leptin concentration. RESULTS: Contents of free fatty acid and glycerol showed concentration dependent increase in response to turmeric extracts. Effects of turmeric extracts on reduction of lipid accumulation in 3T3-L1 cells were examined by Oil Red O staining. Treatment with turmeric extracts resulted in increased expression levels of adipose triglyceride lipase and hormone-sensitive lipase mRNA. The concentration of leptin from 3T3-L1 adipocytes was significantly decreased by turmeric. Proportional abdominal and epididymal fats weights of the turmeric 5% supplemented group, TPB has significantly decreased compared to the HF group. The serum levels of leptin in the TPA and TPB groups were significantly lower than those of the HF group. CONCLUSIONS: Based on these results, we suggested that Korean turmeric may contribute to the decreasing of body fat and regulating leptin secretion.

Effects of Pueraria lobata Root Ethanol Extract on Adipogenesis and Lipogenesis During 3T3-L1 Differentiation into Adipocytes

  • We evaluated the inhibitory effect of Pueraria lobata root ethanol extract (PLREE) on lipid accumulation during 3T3-L1 differentiation to adipocytes by measuring the intracellular expression of adipogenic, lipogenic, and lipolytic markers and lipid accumulation. The total polyphenol and flavonoid content of PLREE were 47 and 29 mg/g, respectively. The electron donating capacity of PLREE at $1,000{\mu}g/mL$ was 48.8%. Treatment of 3T3-L1 preadipocytes with 100, 250, or $500{\mu}g/mL$ PLREE for 8 days dose-dependently promoted the differentiation of 3T3-L1 cells. In contrast, the lipid content of PLREE-treated cells was significantly reduced by 7.8% (p < 0.05), 35.6% (p < 0.001), and 42.2% (p < 0.001) following treatment with 100, 250, and $500{\mu}g/mL$ PLREE, respectively, as compared to differentiated control cells. PLREE upregulated peroxisome proliferator-activated receptor ${\gamma}$ mRNA and protein, and sterol regulator element-binding protein-1c mRNA levels, but did not affect CCAAT/enhancer binding-protein ${\beta}$ and ${\alpha}$ mRNA levels. PLREE also downregulated acetyl-CoA carboxylase mRNA and protein, fatty acid synthase (FAS) protein, and leptin mRNA levels, but did not affect FAS mRNA expression. PLREE upregulated adipose triglyceride lipase mRNA and protein expression, and hormone-sensitive lipase (HSL) protein expression, but did not affect HSL mRNA expression. In conclusion, we found that PLREE enhanced adipogenesis, but reduced lipogenesis, resulting in decreased lipid accumulation in 3T3-L1 cells.

실비산의 항산화 및 지방축적 억제 효과 (Anti-oxidation and Fat Accumulation Inhibitory Effects of Silbi-san)

  • Objectives The purpose of this study was to investigate the effects of Silbi-san on the antioxidant and fat accumulation inhibition and to analyze the anti-obesity effect by analyzing the changes in serum lipid composition in obese mice. Methods We compared contents of phytochemicals like total polyphenols and total flavonoid and antioxidant activities such as 2,2-dipheny-1-picrylhydrazyl and 2.2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) radical scavenging activity. After Silbi-san in 3T3-L1 cells in vitro and mouse adipose tissue ex vivo, we quantified intracellular triglyceride accumulation and lipolysis. Moreover, the anti-obesity activity though inhibiting pancreatic lipase were analyzed. In 3T3-L1 cells, morphological changes showed that control cells had many lipid while cells treated with Silbi-san had less lipid accumulation. 30% EtOH Silbi-san treatment also suppressed the fat absorption by inhibiting the activity of pancreatic lipase and led to high lipolysis through promoting glycerol release. The experimental group was divided into four groups: Normal group fed normal feed, Control group fed 60% high fat diet (HFD) and distilled water, drug group fed 60% high fat diet and 200 mg/kg of Silbi-san water extract, drug group fed 60% HFD and 200 mg/kg of Silbi-san 30% ethanol extract. Results Serum total cholesterol content and serum low density lipoprotein-cholesterol content were significantly decreased in the Silbi-san extract group compared to the control group, serum high density lipoprotein-cholesterol content was significantly increased in Silbi-san extract group. Conclusions In this study, the antioxidant and fat accumulation inhibitory effects of Silbi-san were confirmed.

The effect of eleutherococcus senticosus on metabolism-associated protein expression in 3T3-L1 and C2C12 cells

  • [Purpose] In vivo studies have demonstrated the ergogenic benefits of eleutherococcus senticosus (ES) supplementation. ES has been observed to enhance endurance capacity, improve cardiovascular function, and alter metabolic functions (e.g., increased fat utilization); however, the exact mechanisms involved remain unknown. We aimed to determine whether ES could effectively induce fat loss and improve muscle metabolic profiles through increases in lipolysis- and lipid metabolism-associated protein expression in 3T3-L1 adipocytes and C2C12 skeletal muscle cells, respectively, to uncover the direct effects of ES on adipocytes and skeletal muscle cells. [Methods] Different doses of ES extracts (0.2, 0.5, and 1.0 mg/mL) were added to cells (0.2 ES, 0.5 ES, and 1.0 ES, respectively) for 72 h and compared to the vehicle control (control). [Results] The intracellular triacylglycerol (TG) content significantly decreased (p < 0.05 for 0.2 ES, p < 0.01 for 0.5 ES and 1.0 ES) in 3T3-L1 cells. Adipose triglyceride lipase, which is involved in active lipolysis, was significantly higher in the 1.0 ES group than in the control group (p < 0.01) of 3T3-L1 adipocytes. In C2C12 cells, the mitochondrial protein voltage-dependent anion channel (VDAC) was significantly increased in the 1.0 ES group (p < 0.01). Furthermore, we found that 1.0 ES activated both 5' AMP-activated protein kinase (AMPK) and acetyl-CoA carboxylase (ACC) in skeletal muscle cells (p < 0.01). [Conclusion] These findings suggest that ES extracts decreased TG content, presumably by increasing lipase in adipocytes and metabolism-associated protein expression as well as mitochondrial biogenesis in muscle cells. These effects may corroborate previous in vivo findings regarding the ergogenic effects of ES supplementation.

"Dietary supplementation of L-tryptophan" increases muscle development, adipose tissue catabolism and fatty acid transportation in the muscles of Hanwoo steers

  • This study investigated the effects of dietary rumen-protected L-tryptophan (TRP) supplementation (43.4 mg of L-tryptophan kg-1 body weigt [BW]) for 65 days in Hanwoo steers on muscle development related to gene expressions and adipose tissue catabolism and fatty acid transportation in longissimus dorsi muscles. Eight Hanwoo steers (initial BW = 424.6 kg [SD 42.3]; 477 days old [SD 4.8]) were randomly allocated to two groups (n = 4) of control and treatment and were supplied with total mixed ration (TMR). The treatment group was fed with 15 g of rumen-protected TRP (0.1% of TMR as-fed basis equal to 43.4 mg of TRP kg-1 BW) once a day at 0800 h as top-dressed to TMR. Blood samples were collected 3 times, at 0, 5, and 10 weeks of the experiment, for assessment of hematological and biochemical parameters. For gene study, the longissimus dorsi muscle samples (12 to 13 ribs, approximately 2 g) were collected from each individual by biopsy at end of the study (10 weeks). Growth performance parameters including final BW, average daily gain, and gain to feed ratio, were not different (p > 0.05) between the two groups. Hematological parameters including granulocyte, lymphocyte, monocyte, platelet, red blood cell, hematocrit, and white blood cell showed no difference (p > 0.05) between the two groups except for hemoglobin (p = 0.025), which was higher in the treatment than in the control group. Serum biochemical parameters including total protein, albumin, globulin, blood urea nitrogen, creatinine phosphokinase, glucose, nonesterified fatty acids, and triglyceride also showed no differences between the two groups (p > 0.05). Gene expression related to muscle development (Myogenic factor 6 [MYF6], myogenine [MyoG]), adipose tissue catabolism (lipoprotein lipase [LPL]), and fatty acid transformation indicator (fatty acid binding protein 4 [FABP4]) were increased in the treatment group compared to the control group (p < 0.05). Collectively, supplementation of TRP (65 days in this study) promotes muscle development and increases the ability of the animals to catabolize and transport fat in muscles due to an increase in expressions of MYF6, MyoG, FABP4, and LPL gene.

차전초 잎의 물 추출물이 고지방식이로 비만 유도된 C57BL/6 생쥐의 체지방 감소에 미치는 영향 (Effect of Plantaginis asiaticae Folium water extract on body fat loss in high fat-induced obese C57BL/6 mice)

  • Objective : Plantago asiatica L. has been used for many years in Korea, China, and Japan for the treatment of many diseases such as anti-inflammatory, anti-asthma and diuretic. But the anti-obesity effect of Plantaginis asiaticae Folium has not been studied yet. Thus, this study aimed to demonstrate the anti-obesity efficacy of Plantaginis asiaticae Folium water extract (PAF) in high fat diet-induced obesity animal model. Method : Male, five-weeks old C57BL/6 mice were divided into 5 groups; ND (normal diet + 0.9% saline), HFD (high-fat diet + 0.9% saline), PC (high-fat diet + Garcinia cambogia 500 mg/kg), PAF 100, 300 (high-fat diet + PAF 100, 300 mg/kg. Treatments were performed daily for 6 weeks per os. We evaluated the changes of body weight, white adipose tissues weight, hepatic and fecal lipids level, pancreatic lipase activity and lipid profiles in plasma. Result : Body weights, adipose tissue weights and FER were significantly lower in mice fed the high-fat diet with PAF treatment than in mice fed the high-fat diet alone. In plasma, TG, TC, LDL-C, ATS and ALT levels significantly decreased in PAF groups compared with HFD group. Also, hepatic triglyceride (TG) levels decreased and fecal TG increased in PAF groups compared to HFD group. In addition, the activity of pancreatic lipase was decreased in the PAF groups compared to that of HFD group. Conclusion: These results suggest PAF extract is effective in preventing and improvement obesity, so it might be a potential and safe material for anti-obesity agent of weight control or functional supplements.

Effects of dietary n-6:n-3 polyunsaturated fatty acid ratio on growth performance, blood indexes, tissue fatty acid composition and the expression of peroxisome proliferator-activated receptor gamma signaling related genes in finishing pigs

  • Objective: This study investigated the effects of dietary n-6:n-3 polyunsaturated fatty acid (PUFA) ratio on growth performance, blood indexes, tissue fatty acid composition and the gene expression in finishing pigs. Methods: Seventy-two crossbred ([Duroc×Landrace]×Yorkshire) barrows (68.5±1.8 kg) were fed one of four isoenergetic and isonitrogenous diets with n-6:n-3 PUFA ratios of 2:1, 3:1, 5:1, and 8:1. Results: Average daily gain, average daily feed intake and gain-to-feed ratio had quadratic responses but the measurements were increased and then decreased (quadratic, p<0.05). The concentrations of serum triglyceride, total cholesterol and interleukin 6 were linearly increased (p<0.05) with increasing of dietary n-6:n-3 PUFA ratio, while that of high-density lipoprotein cholesterol tended to decrease (p = 0.062), and high-density lipoprotein cholesterol:low-density lipoprotein cholesterol ratio and leptin concentration were linearly decreased (p<0.05). The concentration of serum adiponectin had a quadratic response but the measurement was decreased and then increased (quadratic, p<0.05). The proportion of C18:3n-3 was linearly decreased (p<0.05) in the longissimus thoracis (LT) and subcutaneous adipose tissue (SCAT) as dietary n-6:n-3 PUFA ratio increasing, while the proportion of C18:2n-6 and n-6:n-3 PUFA ratio were linearly increased (p<0.05). In addition, the expression levels of peroxisome proliferator-activated receptor gamma (PPARγ) and lipoprotein lipase in the LT and SCAT, and adipocyte fatty acid binding protein and hormone-sensitive lipase (HSL) in the SCAT had quadratic responses but the measurements were increased and then decreased (quadratic, p<0.05). The expression of HSL in the LT was linearly decreased (p<0.05) with increasing of dietary n-6:n-3 PUFA ratio. Conclusion: Dietary n-6:n-3 PUFA ratio could regulate lipid and fatty acid metabolism in blood and tissue. Reducing dietary n-6:n-3 PUFA ratio (3:1) could appropriately suppress expression of related genes in PPARγ signaling, and result in improved growth performance and n-3 PUFA deposition in muscle and adipose tissue in finishing pigs.

브로콜리 싹 에탄올 추출물이 고지방식이를 급여한 흰쥐의 콜레스테롤 저하 및 비만 억제효과에 미치는 영향 (Effect of Broccoli Sprouts on Cholesterol-lowering and Anti-obesity Effects in Rats Fed High Fat Diet)

  • 고지방식이로 고지혈 및 비만이 유도된 흰쥐에서 브로콜리 싹 에탄올 추출물 투여로 인한 혈 중 지질대사 개선 및 지방조직의 항비만효과에 미치는 영향에 대하여 조사하였다. 5주령 된 흰쥐 수컷 36마리를 1주일간 적응시킨 후 정상 식이군(ND), 고지방식이군(HFD), 정상식이와 브로콜리 싹 에탄올 추출물 저용량 병합 투여군(ND-BSL), 정상식이와 브로콜리 싹 에탄올 추출물 고용량 병합 투여군(ND-BSH), 고지방식이와 브로콜리 싹 에탄올 추출물 저용량 병합 투여군(HFD-BSL) 및 고지방식이와 브로콜리 싹 에탄올 추출물 고용량 병합 투여군(HFD-BSH)으로 나누어 4주간 사육하였다. 고지방식이로 증가되어진 체중증가량은 브로콜리 싹 에탄올 추출물 고용량 병합 투여로 유의하게 감소되었으며, 정상식이군(ND)과는 비슷한 수준으로 저하되었다. 고지방 식이군들(HFD, HFD-BSL 및 HFD-BSH)이 정상식이군들 (ND, ND-BSL 및 ND-BSH)에 비하여 식이섭취량은 낮았으나, 식이효율은 높게 나타났다. 간조직의 무게는 실험 군간에 유의차가 없었으나, 부고환 및 장간막지방조직의 무게는 고지방식이를 급여한 흰쥐에게 브로콜리 싹 에탄올 추출물을 고용량 병합 투여한 경우에는 고지방식이에 비하여 유의하게 저하되었다. 고지방식이로 증가되어진 혈청 중의 ALT 및 AST 활성, 중성지방, 총콜레스테롤 및 LDL-콜레스테롤 함량, 심혈관위험지수와 동맥경화지수도 브로콜리 싹 에탄올 추출물 고용량 병합 투여로 감소되었다. 혈청 중의 ALP 활성은 실험 군 간에 유의차가 없었다. 혈청 중의 HDL-콜레스테롤 함량은 정상식이군(ND)에 비하여 고지방식이군(HFD)이 유의하게 감소하였으나, 정상식이 혹은 고지방식이를 급여한 흰쥐에게 브로콜리 싹 에탄올 추출물을 투여하였을 경우에는 영향을 미치지 않았다. 고지방식이로 증가되어진 간조직과 장간막지방조직 중의 총콜레스테롤 함량은 브로콜리 싹 에탄올 추출물 병합 투여로 저하되었다. 지방조직 중의 HR-LPL과 TE-LPL 활성 모두 정상식이군 (ND)에 비하여 고지방식이군(HFD)이 증가하였고, 브로콜리 싹 에탄올 추출물 고용량 병합 투여한 모든 군은 고지방 식이군(HFD)에 비하여 유의하게 감소되었으나 정상식이군 (ND)과는 비슷한 활성을 나타내었다. 특히 브로콜리 싹 에탄올 추출물의 효과는 고용량 병합 투여군이 저용량 병합투여군에 비하여 더 좋은 효과를 나타내었다. 혈청 및 조직 중의 지질 함량 변화는 정상식이군들(ND, ND-BSL 및 ND-BSH) 간에는 유의차가 없었다. 따라서 고지방식이를 급여하면서 브로콜리 싹 에탄올 추출물을 병합 투여 시에만 효과가 있는 것으로 여겨진다. 이상의 결과 브로콜리 싹 에탄올 추출물이 체중 및 지방조직의 무게 감소와 더불어 혈청지질 개선 효과와 LPL 활성을 저하시켜 고지혈증 예방효과와 지방축적 억제 효과가 있는 것으로 사료되나 브로콜리 싹 추출물의 어떤 성분이 콜레스테롤 저하효과 및 비만을 억제시킬 수 있는지는 보다 자세한 연구가 요구된다.

식이급여형태가 고지방식이를 급여한 흰쥐 지방조직의 지방합성 및 지방합성 효소활성에 미치는 영향 (Effect of Meal Pattern on Lipogenesis and Lipogenic Enzyme Activity in Rat Adipose Tissue Fed High Fat Diet)

  • 본 연구는 식이급여형태를 달리하면서 고지방식이를 급여한 흰쥐의 부고환지방조직과 장간막지방조직의 지방조직 부위별 지방합성량, 지방합성에 관여하는 주된 효소인 ME, G6PDH 및 6PGDH 활성 및 지방축적의 주된 효소인 LPL 활성에 미치는 영향을 구명하였다. Sprague-Dawley 계통 수컷 흰쥐 16마리를 체중에 따라 2군으로 나누어 4주간 실시하였고, ad libitum군은 실험 식이를 무제한 급여하였으며, meal-fed군은 1일 1회 3시간씩 제한 급여하였다. 지방조직의 지방합성량은 포도당이 총지방으로 변하는 양으로 측정하였고, 지방조직의 G6PDH, 6PGDH 및 ME 활성은 NADPH 생성량으로 측정하였다. 또한 지방조직의 LPL 활성과 혈청 지단백질, 중성지방 및 insulin 함량도 측정하였다. Meal-fed군이 ad libitum군에 비해 식이섭취량 저하로 인한 증체량 및 부고환지방조직과 장간막지방조직의 무게가 유의하게 저하되었다. 혈청 총콜레스테롤, HDL-콜레스테롤 및 LDL-콜레스테롤 함량은 식이급여형태에 따른 유의차가 없었으나, 혈청 중성지방 함량은 meal-fed군이 ad libitum군에 비해 유의하게 증가하였고, insulin 함량은 저하되었다. 지방합성량 및 G6PBH과 6PGDH 활성은 부고환지방조직과 장간막지방조직 모두 meal-fed군이 ad libitum군에 비해 증가하였으나, ME 활성은 식이급여형태에 따른 유의차가 없었다. 지방조직의 HR-LPL과 TE-LPL 활성은 meal-fed군이 ad libitum군에 비해 유의하게 높았다. 지방조직 부위에 따른 차이는 지방함량, 지방합성량 및 G6PDH와 LPL 활성은 부고환지방조직에 비해 장간막지방조직에서 낮은 경향이었다. 이와 같이 meal feeding시 섭취한 에너지를 체지방 형태로 더 많이 보유하도록 지방조직내 지방합성량과 지방합성 관련 효소들의 활성을 증가시켜 체내 대사에 영향을 미치는 것으로 사료된다. 또한 지방조직 부위에 따라 지방대사에 미치는 영향이 다르다는 것을 알 수 있었다.
