Effects of Dietary Levels of Glycine, Threonine and Protein on Threonine Efficiency and Threonine Dehydrogenase Activity in Hepatic Mitochondria of Chicks

  • This study was carried out to evaluate the relationship between threonine (Thr) efficiency and Thr dehydrogenase (TDG) activity as an indicator of Thr oxidation on chicks fed with levels of diets (CP [17.5% and 21.5%] and Thr [3.8 and 4.7 g/100 g CP]; glycine [Gly][0.64% and 0.98%] and true digestible Thr [dThr] [0.45% and 0.60%]). Calculation of the Thr efficiency was based on N-balance data and an exponential N-utilization model, and TDG activity was determined as accumulation of aminoacetone and Gly during incubation of hepatic mitochondria. This study found that in the liver of chicks who received a diet containing up to 0.79% Thr (4.7 g Thr/100 g of CP) in the 17.5% CP diet, no significant (p>0.05) effect on TDG activity was observed. However, significantly (p = 0.014) increased TDG activity was observed with a diet containing 21.5% CP (4.7 g Thr/100 g of CP) and the efficiency of Thr utilization showed a significant (p = 0.001) decrease, indicating the end of the Thr limiting range. No significant (p>0.05) effect on the total TDG activity and accumulation of Gly was observed with addition of Gly to a diet containing 0.45% dThr. In addition, addition of Gly to a diet containing 0.60% dThr also did not result in a change in accumulation of Gly. Due to an increase in accumulation of aminoacetone, an elevated effect on total TDG activity was also observed. No significant (p>0.05) reduction in the efficiency of Thr utilization was observed after addition of Gly at the level of 0.45% dThr. However, significantly (p<0.001) reduced efficiency of Thr utilization was observed after addition of Gly at the level of 0.60% dThr. Collectively, we found that TDG was stimulated not only by addition of Thr and protein to the diet, but also by addition of Gly, and efficiency of Thr utilization was favorably affected by addition of Gly at the level near to the optimal Thr concentration. In addition, no metabolic requirement of Gly through the TDG pathway was observed with almost the same accumulation of Gly and a slight increase in TDG activity by addition of Gly. Thus, our findings suggest that determination of TDG activity and parameter of efficiency of Thr utilization may be useful for evaluation of dietary Thr level.

  • The results, which was analytically surveyed the free amino acids by the automatic amino acid analyzer adding the enzyme on the Korean cow's fore shank muscles, are as follows: 1. The content of free amino acids in the fore shank muscles, without addition of the enzyme orderly contains alanine, glutamic acid, lysine, glycine, histidine, leucine, threonine, arginine, cystine, serine, proline, isoleucine, phenylalanine, tyrosine, methionine, aspartic acid and valine. 2. In accordance with the addition of the enzyme, by 0.01%, 0.05% and 0.1% the nine free amino acids of glutamic acid, glycine, alanine, cystine, valine, isoleucine, leucine, lysine and arginine were continuosly increased. 3. Proline and histidine were decreased at the enzyme addition of 0.01% after showing the high content at the control, but the quantity of free amino acids was increased according to the increase of the quantity of the enzyme. 4. Aspartic acid, threonine, serine, methionine, tyrosine and phenylalanine were increased till the enzyme addition of 0.05% and remarkably decreased from 0.1%. 5. At cooking the meat, the quantity of the enzyme addition was most effective at 0.05% of meat weight.

  • In addition to known cucurbitacins, a glucosphingosine type cerebroside and amino acids were isolated from the roots of Trichosanthes kirilowii. The structure of cerebroside was determined as soya-cerebroside I by means of spectroscopic methods. Fifteen amino acids were identified as aspartic acid, glutamic acid, serine, glycine, histidine, citrulline, threonine, alanine, proline, tyrosine, valine, isoleucine, leucine, phenylalanine and tryptophan, among which the major components such as citrulline, phenylalanine, leucine/isoleucine and valine were isolated.

  • In an attempt to develop natural seasoning materials by use of shellfishes and crustaceans, contents of taste components such as amino acids, nucleotide and its derivatives, the extractability of oyster (Crassostrea gigas), sea mussel(Mytilus coruscus) and crab(Portanus tribuerculata) were investigated. As a result of chemical analysis and sensory evaluation, the optimum condition of extraction could be concluded as extracting fresh or frozen raw materials for about 40 min. at $95^{\circ}C$ with 1.5 to 2 times of water by volume. The contents of free amino acids in the extractions were much in sequence as crab(1,886 mg%), mussel(765 mg%) and oyster(554 mg%), and the dominant amino acids in each extracts were identified as glutamic acid, alanine, glycine, proline and arginine in oyster, threonine, alanine, arginine, glycine and glutamic acid in mussel, arginine, proline, lysine, alanine and threonine in crab respectively. In addition, the major nucleotides affecting as taste enhancer of each extracts were estimated as inosine in oyster and crab, and inosine monophosphate in mussel respectively.

  • For the purpose of clarifying the changes of free amino acid content in seeds of Korean mung bean during the ripening process, samples ranging in five stages-10, 15, 20, 25 and 30 days after blooming were collected. The results obtained were as follows: 1) Amino acids detected in the first stage were lysine, histidine, arginine, cystine, aspartic acid, threonine (including serine), glutamic acid, valine, methionine sulfoxide and methionine sulfone. 2) In the second stage (15 days after blooming) more amino acids such as glycine, isoleucine, tyrosine, phenylalaninc, and methionine were detected in addition to those in the first stage. More methionine was appeared, while the level of methionine sulfoxide and methionine sulfone was decreased. 3) In the 3rd stage leucine was first detected. The level of leucine was increased slowly as the seed was being ripened. After 4th stage methionine sulfoxide and methionine sulfone were not detected, while the level of methioniene was steadily increased. 4) After 20 days the levels of lecuine, valine. isoleucine, and methionine were increased, while the others were either decreased or remained at the same level.

  • These experiment were conducted to study the effects of various vitamins and amino acids upon the growth of Saccharomyces rouxii T9 and the results obtained were as follows. (1) As a growth factor, inositol, ribofavin, niacin and para amino benzoic acid were not required. (2) Though biotin, thiamin and Ca-pantothenate were adaptably required, vitamins required in absolute were not existed. (3) The growth rate of Saccharomyces rouxii T9 in the deficient group of biotin, thiamin, pyridoxine, riboflavin and Ca-pantothenate were more decreased on the media containing 26% of NaCl than containing none of NaCl in the earlystage of cultivation, while the growth rate showed the opposition tendency in the latter term of cultivation. (4) As an amino acid, methionine, tryptophan, serine, threonine, cystine, glycine, leucine and valine showed systematically the effects of addition in the compound media containing $(NH_4)_2SO_4$ as a nitrogen sources. (5) The growth rate of Saccharomyces rouxii T9 in the addition group of histidine, lycine, arginine, aspartic acid, proline and tyrosine were somewhat decreased on the media containing higher concentration of NaCl comparison with none NaCl media in the early stage of cultivation, showing the similar or increasing tendency in the latter term of cultivation.

THE TASTE COMPOUNDS OF BROILED DRIED SEA MUSSELS (배건담치의 정미성분에 관한 연구)

  • This study was attempted to evaluate the taste of broiled-dried sea mussel which is a traditional dried Sea food in Korea. The contents of such compounds as free amino acids, nucleotides and their related compounds, TMAO, betaine, and nonvolatile organic acids were analysed. From the results of analysis of nucleotides, the contents of AMP and ADP in broiled-dried Mytilus coruscus and Mytilus edulis appeared higher than other nucleotides and tended to increase slightly after cooking, drying, and storage which might be due to both their stability and the addition of degradation of nucleic acids during cooking. In the free amino acid composition of fresh samples abundant amino were taurine, glycine, serine, glutamic acid, alanine, arginine, threonine, tyrosine, lysine and valine in order. Such amino acids as histidine, leucine, methionine and isoleucine were poor and both proline and phenylalanine were merely trace. The free amino acid composition in the extract of Mytilus corucus and Mytilus edulis after broil-drying was not changed. These amino acids, taurine, glycine, feline, glutamic acid, alanine and arginine were abundant in dried samples as well as in the fresh. The total free amino acid was greatly reduced after cooking and drying ranging from $76.3\%$ to $79.7\%$ loss to that of tile fresh. The content of betaine shelved the same tendency as in total free amino acid while TMA slightly increased relating to TMAO decrease during broil-frying. Such nonvolatile organic acids as succinic, lactic, malic and fumaric acid were abundant in both fresh and broiled-dried samples whereas oxalic and pyruvic acid were poor. It is found that the taste compounds of broiled-dried Mytilus coruscus and Mytilus edulis were composed of amino acids as glycine, serine, alanine, glutamic acid, arginine, and betaine, TMAO, ADP, AMP, and organic acids such as succinic acid, lactic acid, malic acid and fumaric acid. No significant differences in the taste compounds between male and female as well as between Mytilus coruscus and Mytilus edulis was not observed.

  • 혐기성대상과정중 메탄생성균(methanogenic bacteria)에 의한 메탄생성시 주요 기질인 아세트산 (acetic acid)을 분해할 경우에 여러 가지 복합기질 중 아미노산 첨가에 의한 분해속도증가에 미치는 영향과 투입한 아미노산이 미생물에 의하여 생체량으로 합성되는 정도를 고찰하였다. 실험결과 메탄생성균은 glycine, serine, threonine, aspartic acid, trytophan 등의 혐기성미생물의 생체량합성에 필요한 물질을 투입할 경우에 아세트산의 분해속도가 증가하였으며, 여러 가지 아미노산을 혼합하여 주입한 결과 분해속도가 17% 향상되었다. 한편, 메탄생성균의 lysing에 의하여 생성된 유기물은 메탄이나 이산화탄소의 최종산물로 전환되기보다는 새로운 메탄생성균의 생체량을 형성하는데 직접 이용되었으며, 아세트산의 분해속도를 52% 증가시켰다. 단순기질(sole substrate)과 복합기질(complex substrate)의 분해는 미생물의 생체량합성에 필요한 여러 가지 중간대사산물간의 상호자극효과에 의하여 복합기질이 용이한 것으로 나타났으며, 유입기질내 활성이 강한 슬러지의 농도는 혐기성처리에 매우 중요한 부분을 차지하였다.

Toxicity and Taste Components of the Pufferfish, Sphoeroides annulayus (bull's eye puffer), from Mexico (멕시코산 황소눈복어 (Sphoeroides annulatus)의 독성 및 정미성분)

  • The present study was conducted for the data of toxicity and taste components of the pufferfish, Sphoeroides annulayus (bull's eye fuller), transported from Mexico. All other parts including muscle and skin were nontoxic ranging below $10\;{\mu/g$ except gonad, The amounts of IMP and ADP were $5.6\;{\mu}mol/g\;and\;2.7\;{\mu}mol/g$, and the ratio to the total ATP and its related compounds was $41.1{\%}$. The great portion of free amino acids in the muscle of the puffer was occupied by L-glycine, L-alanine, L-anserine, L-threonine and L-valine. Their amounts were $233.5 mg/100 g, 169.0 mg/100 g, 149.1 mg/100 g, 135.7 mg/100 g and 132.3 mg/100 g$. Their concentration ratio to total free amino acids were $14.28{\%},\;10.33{\%},\;9.12{\%},\;8.30{\%}\;and\;8.09{\%}$, respectively. The content was $50.12{\%}$ of the total free amino acids. In addition, the amounts of taurine and L-histidine were 119.3mg/100 g and 14,7 mg/100 g.

  • The purpose of this study was to investigate the physiochemical properties of Doenjang was fermented by added with fungi and protease. The moisture content and pH of Doenjang added with protease (WP) were lower than those of control w/o protease while the contents of titratable acidity, reducing sugar, and amino-type nitrogen in WP were higher than control. The ${\alpha}$-amylase activities of Doenjang added with single and mixed Protease B were the highest at 4 weeks of fermentation period and protease activity of WP was about 4 times higher than that of control. The 4-9 kinds of free amino acids (proline, isoleucine, leucine, and phenylalanine etc.) in WP was increased in comparison with control. The DPPH radical scavenging activity and total polyphenol content were higher in WP than control. Total aerobic bacterial and fungal numbers were decreased depending on fermentation time regardless of addition of protease. In conclusion, the protease can be used as additives improving the quality and taste of fermented Doenjang.
