Effect of Fermented Benincasa hispida cong. Extract on Promotion of Osteoblast Differentiation and Inhibition of Osteoclast Generation (동과 발효물의 조골세포 분화 촉진 및 파골세포 생성 억제 효과)

  • The bones of the human body support the structures of the body and provide protection for a person's internal organs. Bone metabolic diseases are on the rise due to a significant increase in life expectancy over a short period of time. Therefore, we investigated the osteoblast differentiation promoting and osteoclastogenesis inhibitory activities of fermented Benincasa hispida cong. (HR1901-BS, HR1901-BSaf). We evaluated the alkaline phosphatase (ALP) activity of MC3T3-E1 mouse calvarial-derived osteoblasts. We also evaluated expression of ALP, osteocalcin (OCN), and runt-related transcription factor 2 (Runx2), which regulate osteoblast differentiation. To assess effects on osteoclast formation, tartrate-resistant acid phosphatase (TRAP) activity in RAW264.7 cells was analyzed. ALP activity increased by 121-136% and 140-156%, respectively in the presence of HR1901-BS and HR1901-BSaf. Expression of osteoblast differentiation factor also increased significantly. We also confirmed that HR1901-BS and HR1901-BSaf decreased TRAP activity in osteoclasts by 35-47% and 23-39%, respectively. Our results showed that fermented Benincasa hispida cong. (HR1901-BS, HR1901-BSaf) increase bone mineralization and osteoblast differentiation activity in MC3T3-E1 cells, and inhibit bone resorption activity in RAW264.7 cells. In conclusion, fermented Benincasa hispida cong. (HR1901-BS, HR1901-BSaf) can be used as an effective natural resource for preventing and treating bone-related diseases.

The Effect of a Convergence Approach of Home Environment Modification and Task-oriented Intervention on Fall Efficacy and Activity Level of Person with Stroke in the Community (가정환경 수정과 과제기반 중재의 융복합 접근이 지역사회 뇌졸중 환자의 낙상 효능감 및 활동 수준에 미치는 영향)

  • The purpose of this study is to determine the effect of a convergence approach, which applies both home environment modification applied with assistive technology and task-oriented intervention using a fall safety checklist, on the fall efficacy and activity level of stroke patients. 41 persons with stroke(21 experimental groups and 20 control groups) were divided into experimental and control groups. The experimental group was applied environmental modification and the task-oriented intervention using the fall safety checklist for 4 weeks. The control group was provided only the environmental modification. Before and after the intervention, the Korean Activity Card Sort (KACS) and the Korean Falls Efficacy Scale-Korean version (FES-K) were used for evaluation. There was no significant difference in fall efficacy and activity level change between the experimental group and the control group. Both the experimental group and the control group significantly improved the fall efficacy and activity level after the intervention. By applying the results of this study to the clinical field, it will be possible to support stroke patients to live safely and actively participate in the community.

Comparison of Muscle Activities in Upper Extremity Muscles During the Knee-flexed and Standard Push-ups With Different Hand Shape (무릎 팔굽혀펴기와 표준 팔굽혀펴기 시 손 모양에 따른 어깨 근육의 근활성도 비교)

  • Background: Push-up are effective exercises for shoulder stability. Previous studies have documented the effects of support plane and hand position and width on muscle activities during a push-up. Objects: This study aimed to investigate the changes in muscle activities in the upper extremity when performing the standard and the knee-flexed push-up with different hand shapes. Methods: A total of twenty-six healthy males participated in this study. Three different hand shapes (finger abduction, finger adduction, and fists) and two types of push-up posture (standard and knee-flexed push-up) were set as the independent variables. Electrograms were used to measure the muscle activity of the upper trapezius (UT), triceps brachii (TB), pectoralis major (PM), and serratus anterior (SA). Each participant performed the randomly assigned push-up to the sound of the metronome. The mixed-effect linear regression model was used to detect the changes in muscle activities after changing the hand shape and push-up posture. Statistical significance was set at α = 0.05. Results: The UT muscle activity was statistically significantly higher when performing push-up with fists than finger abduction (p = 0.035) or finger adduction (p = 0.044). During the standard push-up, the muscle activity in all muscles was that the push-up with fists showed the highest muscle activity compared to the finger abduction (p < 0.01) and finger adduction (p < 0.01). Regardless of the shape of the hand, UT had the lowest muscle activity compared to other muscles (p < 0.001). In contrast, the SA muscle had the highest muscle activity among four muscles during the standard push-up. Conclusion: Based on the results of this study, we suggest hand shape is related to the difficulty level of push-up either in the standard or the knee-flexed push-up, especially in the push-up with fists. In addition, knee push-up can be recommended as shoulder muscle-strengthening exercises for individuals with low shoulder muscle strength.

Exploring Physical Activity Promotion Strategies for Older Residents in a Small Town Based on Socio-ecological Model: A Mixed Method Inquiry (사회생태학적 분석을 통한 중소규모 A 도시 거주 노인 신체활동 촉진전략 모색 - 혼합연구기법으로)

  • The purpose of this study was to understand barriers and problems associated with physical activity of older adults in a small town and to provide improvement plans based on socio-ecological model. This study employed three independent phases including survey, accelerometer study, and photovoice. Three phases were analyzed based on nesting design among diverse mixed method designs. Among 134 older participants, 73 older individuals reported that their physical activity per week was less than 150min in the first phase. Among the 73 older individuals, 40 older individuals participated in the second phase. In the second phase, 32 older individuals does not meet at least 150min of moderate-to-vigorous physical activity(MVPA) suggested by DHHS. Finally, 6 older adults participated in the third phase. Five major themes emerged including 1) lack of proper information and service regarding physical activity, 2)lack of support from people close to them, 3)some limits due to chronic disease, 4)lack of appropriate facility, and 5)lack of opportunity due to existing inactive lifestyle. Based on these five themes, this study attempted to provide the improvement plans with socio-ecological model.

LIGHT-DEPENDENT CELLULAR LEAKAGE FROM CUCUMBER COTYLEDON DISCS TREATED WITH $\delta$-AMINOLEVULINIC ACID, OXYFLUORFEN, AND ROSE BENGAL

  • When cucumber (Cucumis sativus L.) cotyledon discs were floated on $\delta$-aminolevulinic acid, oxyfluorfen, or rose bengal solution under light condition following 20 h dark incubation, rapid electrolyte leakage from the tissues occurred. The electrolyte leakage from the tissues was dependent on the compounds treated, their concentrations, and the duration of light exposure to the tissues. Dark incubation before exposure to continuous white light enhanced electrolyte leakage from the tissues treated with the compounds and reduced lag period for the activity of the compounds. Electrolyte leakage from the treated tissues was greatly influenced by the light intensity to which they were exposed. Higher light intensities stimulated electrolyte leakage and reduced lag period. Porphyrin biosynthesis inhibitors, gabaculine and 4,6-dioxoheptanoic acid, completely inhibited electrolyte leakage from the oxyfluorfen-treated tissues. Protection against the activity of $\delta$-aminolevulinic acid from electrolyte leakage was complete with 4,6-dioxoheptanoic acid, but not with gabaculine. However, gabaculine and 4,6-dioxoheptanoic acid gave no such protection against rose bengal activity. In summary, our results indicate that $\delta$--aminolevulinic acid, oxyfluorfen, and rose bengal exert their effects by causing electrolyte leakage from the treated tissues in a similar manner, except that oxyfluorfen has an apparent lag period for its action on electrolyte leakage increase. All above compounds require preincubation of treated tissues in darkness and subsequent light exposure with a high intensity for their maximal activities. Our results also support that in the presence of light, $\delta$-aminolevulinic acid and oxyfluorfen cause cellular damage through the indirect generation of singlet oxygen from accumulated tetrapyrroles of porphyrin pathway, whereas rose bengal causes cellular damage through the direct generation of singlet oxygen.

Effects of Yeoldahanso-tang, a Sasang Constitutional Herbal Prescription for Taeeumin, on the Adipogenesis in 3T3-L1 Cells (태음인(太陰人) 열다한소탕(熱多寒少湯)이 3T3-L1 지방전구세포(脂肪前驅細胞) 분화(分化)에 미치는 영향(影響))

  • Objectives : Although Yeoldahanso-tang (YDHST) has been widely used for treatment of obesity and its related diseases such as hyperinsulinemia and hypertension for Taeeumin, no scientific evidence has reported yet to support its ability to work against these metabolic disorders. Our study was aimed to investigate the anti-obesity effect of YDHST extract on the cellular differentiation of 3T3-L1 preadipocytes into adipocytes. Methods : 3T3-L1 preadipocytes were differentiated into adipocytes by adding insulin, dexamethasone and 3-isobutyl-1-methylxanthine (IBMX) for 8 days in the absence or presence of YDHST extract. Anti-obesity effects of YDHST extract were evaluated by Oil Red O staining, glycerol-3-phosphate dehydrogenase (GPDH) activity, triglyceride contents, and leptin production. Results : YDHST extract remarkably prevented lipid accumulation with no cytotoxicity in the differentiated 3T3-L1 cells. In addition, YDHST extract decreased contents of triglyceride 3T3-L1 adipocytes. Consistently, YDHST extract caused a significant inhibition of GPDH activity and leptin production in a dose-dependent manner. Conclusions : Our findings suggest that Sasang constitutional herbal formula YDHST for Tae-eumin has anti-obesity activity by regulation of the adipogenesis process in vitro. Additional study will be required to further confirm the inhibitory effect on adipocyte differentiation by using in vivo animal model.

Global Healthcare Supporting System and Activity in some General Hospitals according to Hospital Social Responsibility in Korea (사회적 책임에 따른 일부 종합병원의 해외보건의료지원체계와 활동)

  • The purpose of this study was to investigate current status of 19 general hospitals which performed the hospital social responsibility (HSR) to global health issues. The survey focused on the global healthcare supporting system & activity (including the administration, information, materials, finance, and human resources, et al.), and the government's policy in Korea. Based on the analysis of survey data, the main global health issues of general hospitals were activities of overseas volunteers (56%) and aids of the developing countries (19%). Also, general hospitals have mainly supported into eastern Asia (49%). And then, the most important goal of general hospitals related these activities kept the founding principle of a hospital for global health issues (53%). The second ranking was the social responsibility (32%) and the interchange between Korean and foreign hospitals (32%). There were many differences between general hospitals about the global healthcare supporting system & activities for HSR to global health issues. In conclusion, we suggest that most of general hospitals in Korea would be gotten prior setting the global issues for unmet-need of the developing countries around the world in the near future. This study also served as the effective way by partnership on global health issues of general hospitals for recognizing the HSR as hospital governance.

The Effect of GagamSohabhwang-won(Jiajiansuhexiang-yuan) Essential Oil on Cell Activity and Anti Oxidation (가감소합향원(加減蘇合香元) 향기액이 세포활성에 미치는 영향과 항산화효과에 관한 연구)

  • Objectives : The glial cell, located in between the blood vessel and nerve cell, takes charge of the cell support, nutrition supply, elimination of body waste, and cell action. GagamSohabhwangwon(GGSH), a chinese traditional medicinal prescription has been used orally for the treatment of seizures, infantile, convulsion, stroke and so forth. This paper examines the effect of the GagamSohabhwangwon(GGSH) essential oil on cell activity and anti oxidation. Methods : MTT assay methods were employed to measure the cell activity based on the amount of the GagamSohabhwangwon(GGSH) essential oil by using primarily cultivated glial cell. In addition, this paper measured a viability of the glial cell after a protein active retarder control to confirm the multiplication of the cell and examined the cell extinction by the active oxygen, an extinction shielding effect with different amount of the GagamSohabhwangwon(GGSH) essential oil to observe anti oxidation. Furthermore, this paper measured a viability of the cell and phosphorylation(phosphorylation) of the protein which affects the multiplication of the glial cell. Results : When controlling the amount of the GagamSohabhwangwon(GGSH), there was a multiplication effect of the primary glial cell, the multiplication of the cell was dependent on the density of the GagamSohabhwangwon. The multiplication power of the primary glial cell was suppressed by PKA inhibiter (H89). In compliance with the active oxygen the extinction of the primary glial cell was dependent on the density of the GagamSohabhwangwon, there was a shielding effect of the cell extinction when GagamSohabhwangwon(GGSH) was preprocessed. When inducing the multiplication of the primary glial cell, phosphorylation of the Akt, BDNF, CREB, ERK and ERM were increased. Conclusions: Based on the results, GagamSohabhwangwon essential oil will have the effect which activates the nervous system cell and protects the cell through anti oxidation.

Ultrastructure of Vascular Meristems in the Rhizome of Botrychium ternatum (고사리삼 지하경의 유관속 분열조직 미세구조)

  • To elucidate the origin of secondary growth in the rhizome of B. ternaturn, the developmental changes of vascular cambium was observed in ultrastructural features. The vascular cambium was gradually differentiated from procambium as in seed plants, but the cambial activity did not persist very long so that the cambial cells became a dormant state like fossil cryptogams. Dense cytoplasm of procambial cells became progressively sparse during the growth, and the tiny vesicles were fused to form numerous small vacuoles and then a few large vacuoles. These gradual changes and the occurrence of storage materials which was associated with the developmental stages might support the progressive differentiation of the cambial cells. In addition, the cessation of cambial activity could be indicated by the facts that late vascular cambial cells accumulate large lipid bodies and show very small peripheral cytoplasm and unlikely thickened cell wall, compared to other meristematic cells. Therefore. the vascular cambium showed the characteristics of both seed plants and fossil cryptogams from the view point of cambial ontogeny and activity.tivity.

  • This study is focused on the gifted education program, "EEE" of Columbia Public Schools in Missouri. This program is recommended to follow NAGC Pre-K-Grade 12 Gifted programming standards(NAGC, 2010) and Missouri school laws(MDESE, 2012b), but is allowed to run autonomically without any support in a federal or state level. The characteristics of EEE are as follows: ${\cdot}$ emphasizing not only on the cognitive development but also on the social and affective/emotional development of the gifted students ${\cdot}$ encouraging each student's own interest by allowing him/her to select his/her major and minor ${\cdot}$ the variety of classes ${\cdot}$ the call-up class - discriminating from regular classes ${\cdot}$ the interdisciplinary approach - connecting many subjects around the main idea ${\cdot}$ the activity-based learning such as hands-on activities, projects, and simple experiments ${\cdot}$ using the individual activity as well as pair or group activity In special, this paper also contains an example of program about mathematics and suggests some implications for gifted education programs in Korea.
