Extractions of Surface-Active Substances from Defatted Rapeseed Meal (Brassica napus L.) by Supercritical Carbon Dioxide (초임계 CO2 유체 추출법을 이용한 탈지 유채박 중 표면활성물질 추출의 최적화)

  • In this study, an attempt is being made to extract surface-active substances from defatted rapeseed cakes by supercritical carbon dioxide fluid. Independent variables for the extraction process, being formulated by D-optimal design, are pressure (150~350 bar), temperature ($33{\sim}65^{\circ}C$ and co-solvent (ethanol, 50~250 g). The dependent variables of the extraction yield, the content of neutral lipids, phospholipids and glycolipids in the extracts were analyzed upon the results through the response surface methodology. As for the extraction yield, it was found to increase with increasing independent variables, among which the co-solvent proved to be a major influencing parameter. Similar trends were found for the content of surface-active substances (i.e, phospholipids and glycolipids) in the extracts, except for the content of neutral lipids. Regression equations were suggested to coincide well with the results from the experiments. Extraction conditions are being optimized to maximize the extraction yields, the content of phospholipids, and glycolipids were 350 bar (pressure), $65^{\circ}C$ (temperature) and 228.55 g (co-solvent), respectively.

Identification of Antioxidative Substances in Allium fistulosum L. by GC-MS (GC-MS에 의한 대파의 항산화 물질 동정)

  • The ethyl acetate-soluble acidic fraction of juice, hot water, and MeOH extract of Allium fistulosum L. showed DPPH radical-scavenging activity. Each fraction was purified through silica gel adsorption column chromatography, and the active substances in the juice and hot water extract were identified as succinic acid, fumaric acid, 4-hydroxybenzoic acid, 4-hydroxy-3-methoxybenzoic acid, 4-hydroxycinnamic acid, and 4-hydroxy-3-methoxycinnamic acid. For the MeOH extract, 4-hydroxybenzoic acid, 4-hydroxy-3-methoxybenzoic acid, 4-hydroxycinnamic acid, and 4-hydroxy-3-methoxycinnmic acid were identified as the active substances by GC-MS. The contents of these compounds were determined by GC analysis, and their anti-oxidative activities were measured using the DPPH radical-scavenging assay. The results obtained showed that 4-hydroxy-3-methoxycinnamic acid was the dominant antioxidant in Allium fistulosum L.

Searching of Antimicrobial Active compounds from Microalgae (해양 미세조류로부터 항균성 물질의 탐색)

  • As a part of the investigation on useful compounds from microalgae, and its recently that marine planktonic algae have been recognized as potial sources of antibacterial and antifungal substances, we searched for antimicrobial active compounds from the extracts of six microalgae-Lyngbya sp., Tetraselmis sp., Microcystis sp., Chlorella sp., Navicula sp. and Rhalassiosira sp.-treated with several solvents. There were two active species-Lyngbya ap., Teraselmis sp.- in the antimicrobial activity test to bacteria, yeast and molds, especially the activity existed in the extracts by ethyl acetate of supernatants to the microalgae incubatio. and there won’t any activity in two diatoms to the test microorganism.

Biological Activities of Phytosterols and Their Variations in Crops (작물 함유 Phytosterol의 변이와 생리활성)

  • Plant fats and oils contain phytosterols as naturally occurring constituents, which are present in pure or esterified form, or conjugated as glycosides. The predominant phytosterol is sitosterol and minor components are campesterol, stigmasterol, $\Delta^7$-avenasterol and brassicasterol. They play a key role in cell membrane function of plants as cholesterol in animals. Phytosterols have been known for many years for their properties for reducing blood cholesterol levels, as well as their other beneficial health effects for cancer preventive biological-active substances. The objective of this paper is to present a description of the recent advances in phytosterols for future research.

Comparison of Active and Passive Sampler for Determining Temperal and Spatial Concentration Assessment of the Main Volatile Organic Compounds Concentration in Shihwa Industrial Complex (시화산업단지에서 주요 휘발성유기물질의 시간적, 공간적 농도 파악을 위한 능동식과 수동식 시료채취기 비교)

  • In this study, we measured the concentrations of volatile organic compounds (VOCs) in Shiwha area by using active and passive sampler. We did a comparative analysis of the characteristics of the active sampler and passive sampler. In the case of the passive sampler, the average TVOC concentration of the industrial area was 1.86 times higher than that of the residential area. In the case of the active sampler, the average TVOC concentration of the industrial area was 1.07 times higher than that of the residential area. When using the passive sampler, the concentration of VOCs in the industrial area was noted to be higher than the concentration found in the residential area. However, when we used the thermal desorption tube, the concentration of residential area was higher rather than that of industrial area in some substances such as trichloroethylene, toluene, ethylbenzene, and xylene. Toluene was a larger percentage of the overall BTEX ratio. In case of the passive sampler, the relative ratio of toluene, ethylbenzene, and xylene was higher in the industrial area than in the residential area. In contrast in case of the thermal desorption tube, the ratio of these substances was higher in the residential area rather than in the industrial area. The passive sampling in this study showed an appropriate method to analyze the temporal and spatial concentrations of air contaminants. This assessment would prove to be useful for its observance of standards or epidemical study.

  • The traditional concept of ginseng quality was investigated in relation to historical experiences. traditional quality measure and mordern analytical method. The traditional concept appears to be based on the original Korean thought of oneness in life and universe. The outside appearance such as shape and size in traditional quality measure includes the inside quality. Since certain shape and size define specific tissues. cells and biologically active substances in cells the traditional measure will he a map for analytical method to find active principles. Traditional method suggests that the balance among biologically active compounds seems to he more important than the large amount of one active compound and that the mode of active compounds in ginseng for the homeostasis of human body is the multicompound to multitarget system. Traditional method strongly suggests the importance of nitrogen compounds. especially soluble protein and heat stable protein for the balance of active principles since nitrogen compounds are more abundant in the central part (xylem-pith) that grows faster than the outer part (cortex-epidermis). The balance of physiologically active principles appeares to be meaningful in relation to the difference in traditional use of Panax species.

  • Bacteriocins from lactic acid bacteria have attracted much attention in recent years because of their useful worth in increasing safety and extending shelf life of foods. These substances show an inhibitory effect against some food spoilage bacteria and food-borne pathogens. The inhibitory effect fo the bacteriocin produces by lactic acid bacteria against Listeria monocytogenes(L. monocytogenes) was examined in this study. The culture supernatants of 5 kinds of bacteria among the 10 kinds of testes lactic acid bacteria had the inhibitory activity against Listeria sp., various Gram positive and Gram negative bacteria. Bacteriocin produced by Lactobacillus plantarum(Lact. plantarum) LMG 7945 was the most active toward L. monocytogenes. Bacteriocin production of the Lact. plantarum LMG 7945 cultured on MRS broth was increased late logarithmic phase over early stationary phase. This bacteriocin was stable at heat treatment and acidic pH relatively; The activity was retained after heating at 121$^{\circ}C$ for 15min and was active in the pH range of 2~4 but was lost above pH 5.

  • The rice lamina inclination test indicated the presence of brassinosteroid-like active substances in immature Vicia faba seeds. Two of these were identified as castasterone and brassinolide by GC/MS and GC/SIM, respectively. Another active principle was identified as methyl 4-chloroindole-3-acetate by GC/MS and HPLC.

  • In relation to the global increase trend of obesity population, there is a demand for the development of foods having high functional activity by mass-extracting anti-obesity active substances using mulberry product such as leaf and fruit. Therefore, we evaluated the antiobesity efficacy according to varieties by using the mulberry leaves and fruit extracts. At the same time, high active varieties were selected. For this purpose, the effects of the extracts of the mulberry leaf and fruit on 3T3-L1 adipocyte differentiation were examined. As a result, in the case of mulberry leaves, the lipid accumulation inhibitory rate of 'Cheongolppong' was higher than that of the control at $500{\mu}g/ml$ treatment. And in the case of the extract of mulberry fruit, 'Daesim' showed the highest lipid accumulation inhibitory rate compared with the control at 50 times of diluted extract.
