BACKGROUND: The commercial biopesticides containing Chenopodium ambrosioides L. extracts which have been registered as one of the ingredients of the commercial biopesticide by the organic agriculture materials, and have been widely used in Republic of Korea. However, the quantitative analysis method of the active substances for the commercial biopesticides containing C. ambrosioides L. extract has not been conducted.METHODS AND RESULTS: To analyze the quantitative analysis of ascaridole, carvacrol, and p-cymene as active substances of C. ambrosioides L. extract, hydrophilic lipophilic balance cartridge was used for solid phase extraction. The active substances were analyzed by the gas chromatography with flame ionization detector. The limit of quantitation values of ascaridole, carvacrol, and p-cymene were 10, 5, and 2 mg/L, and the recovery rates were 96.3, 84.0, and 82.5% in liquid products and 98.3, 99.1, and 97.3% in solid products, respectively. The total content of ascaridole, carvacrol, and p-cymene in the commercial biopesticides was ranged from 0.08 to 12.75%.CONCLUSION: From these results, this method was suitable for the quantitative analysis of the active substances of commercial biopesticides containing C. ambrosioides L. extract.
To estimate the effect of long-term fertilization on metabolically active bacterial communities in a rice field, RNA was extracted from endosphere (rice root), rhizosphere, and bulk soil that had been subjected to different fertilization regimes for 59 years, and the 16S rRNAs were analyzed using the pyrosequencing method. The richness and diversity of metabolically active bacteria were higher in bulk soil than in the endosphere and rhizosphere, and showed no significant difference between non-fertilized and fertilized plots. Weighted UniFrac analysis showed that each compartment had characteristic bacterial communities and that the effect of long-term fertilization on the structure of bacterial community was more pronounced in bulk soil than in the endosphere and rhizosphere. The 16S rRNAs affiliated with Alphaproteobacteria and Firmicutes were more abundant in the endosphere than in bulk soil while those affiliated with Chloroflexi and Acidobacteria were more abundant in bulk soil than in the endosphere. Several dominant operational taxonomic units (clustered at a 97% similarity cut-off) showed different frequencies between non-fertilized and fertilized plots, suggesting that the fertilization affected their activities in the rice field.

We previously isolated and cloned a cDNA of abaecin from the Bombus ignitus. In an effort to produce a large amount of soluble abaecin at low cost, we successfully expressed the peptide in Escherichia coli that are highly sensitive to its mature form. For this, we fused the peptide encoding 39 amino acids of mature B. ignitus abaecin to the thioredoxin gene together with a C-terminal 6xHis tag. An enterokinase cleavage site was introduced between the 6xHis tag and mature abaecin to allow final release of the recombinant peptide. A high yield of 9.6 mg soluble fusion protein from 200 ml of bacterial culture was purified by $Ni^{2+}$-charged His-Bind resin affinity column, and 1.4 mg of pure active recombinant abaecin was readily obtained by enterokinase cleavage, followed by affinity chromatograph. The molecular mass of recombinant abaecin peptide was determined by Tricin-SDS-PAGE analysis. The recombinant abaecin exhibited antibacterial activity against Gram-negative bacteria.
The effect of ischemia/reperfusion-induced neuronal damage on the memory impairment were investigated using active avoidance and Morris water maze tasks in Wistar rats. Focal ischemia was induced by 1 h occlusion of the right middle cerebral artery (MCA) of Wistar male rats. Reperfusion was induced by releasing the occlusion and restoring the blood circulation for 24 h. The acquisition and preservation memory tested by active avoidance showed a significant difference between the sham and ischemia/reperfusion group. The water maze acquisition performance was also significant difference between sham and ischemia/repefusion groups in both latency and moving distance. The infarction volume was increased by the ischemia/reperfusion. Furthermore, the cresyl violet staining of the ischemia/reperfusion brain showed severe neuronal damage (pyramidal cell loss) in the cortex in addition to the striatum lesion of brain. This study shows that pyramidal cell damage in the cortex lesion may be partially related to memorial disturbance in the ischemia/reperfusion brain injury.
To investigate the significant indicators Improving the undisturbed memory in animal behavior, we employed several behavioral methods (learning, relearning in radial maze, and active avoidance) with ginseng components. Results showed that the repeated intranasal administration of $Rb_1$ and total saponins from Korean red ginseng induced direct effects on the brain mechanisms in rats, and improved the spatial memory during the learning, relearning and retention in the 12-arm radial maze test. The intranasal treatment of the total saponins also effectively improved the disturbed memory (amnesia) by pentylentetrazole, and simultaneously protected the brain by decreasing the severity of motor epileptic seizures. The intraperitonial administration of polysaccharide fraction of Korean red ginseng could improve avoidance behavior (amount of the total ecapes) in the active-avoidance test. Objectives : The purpose of this study was to examine the anti-allergic effect in vivo, and to observe single toxicity in mice of Armeniacae Semen herbal acupuncture solution (ASHA). Methods : We investigated anti DNP IgE-mediated passive cutaneous anaphylaxis in rodents and compound 48/80-induced active systemic anaphylatic shock in mice after treatment at both BL13 with ASHA of 25 ${\mu}{\ell}$(mice) or 50 ${\mu}{\ell}$(rats) 3 times for 5 days. To ascertain safety and toxicity of ASHA, we examined single toxicity test. In single test, three groups were treated with different dosages of ASHA (ASHA250, ASHA500 and ASHA1000) according to on Korean Food and Drug Administration, respectively. We observed attentively motality, abnormal clinical sign, body weight change, and organ weight of mice after ASHA treatment. Results : ASHA inhibited passive cutaneous anaphylaxis and active systemic anaphylatic shock by oral administration. During toxicity experiment period, there was no difference in body weight change, and organ weight among different dose groups. Death were not found in single test i.p. group. (ASHA250, ASHA500 and ASHA1000). Several individuals of single test i.p. group were observed yellow brown discharge around anus in early period after administration. Conclusions : These results indicate that ASHA have inhibition effects on passive cutaneous anaphylaxis and active systemic anaphylatic shock, and suggest that has some toxicity in high dosage.
This study was performed to examine the antigenic potential of purified bee venom (Apis mellifera L., PBV) collected using bee venom collector. Antigenic potential of PBV was examined by active systemic anaphylaxis (ASA) in guinea pigs. PBV was subcutaneously administered at 0.025 and 0.05 mg/kg and also as a suspension with adjuvant (Freund's complete adjuvant, FCA). Ovalbumin (OVA) as a suspension with adjuvant was used to introduce positive control response. In the weight measurement and clinical observation, experimental groups didn't show any significant changes compared with control group. In the autopsy of body, the abnormalities of lung were detected only in the positive control. In the ASA test, experimental groups didn't show any symptoms of anaphylaxis like piloerection, hyperpnea and staggering gait. These results suggested that PBV didn't have antigenic potential in guinea pig.
토양에는 생존하지만 현 기술로는 배양이 불가능한 미생물로부터 천연 항생제를 탐색하기 위해 한국 토양 DNA 단편들을 가진 cosmid library을 대장균에서 제작하였고 약6만개의 clone들을 대상으로 항세균 활성을 보여주는 YS92B를 최종 선발하였다. YS92B클론 배양액의 ethyl acetate추출액은 다양한 병원성 세균의 성장을 in vitro에서 강력히 저해하였다(Listeria monocytogenes, Bacillus subtilis, Pseudomonas syringae, Xanthomonas campestris pv. oryzae, Staphylococcus epidemis). 이 항균활성의 주 물질인 YS92B-VII는 ethyl acetate추출, Sephadex LH20 column chromatography와 HPLC(High Performance Liquid Chromatography)에 의해 순차적으로 분리하였으며 이 과정에서 주 활성물질은 각 피크를 항균검정으로 추적하여 최종 정제하였다. 이 물질은 NMR(Nuclear Magnetic Resornance)에 의한 구조 분석 결과에서 탄소 12개의 포화지방산인 lauric acid가 tyrosine에 결합된 N-lauroyl tyrosine임을 최종 확인하였다. 따라서 본 보고는 한국 토양에서 유래한 고유의 N-acyl amino acid synthase(NAS)유전자가 대장균에 발현되어 생산되는 N-acyl amino acid tyrosine의 특성을 밝히는 것이다.
The inhibitory activities of Amberlite active fraction, which was obtained from methanol soluble fraction of freeze dried slikworm urine, on the rat intestinal glycosidase-catalyzed enzymatic reaction were examined in in viro and in vivo experiments. Amberlite active fraction showed significant inhibitory effects on the hydrolysis of o-glycosidic bond, especially $\alpha$-1,4 bond. On the other hand, the inhibition on the hydrolysis of $\beta$-glycosidic bond was very weak. Oral administration of Amberlite active fraction resulted in a dose-dependent decrease in the blood glucose after an oral maltose load, and postprandial hyperglycemia in carbohydrate-loaded mice was suppressed by Amberlite active fraction at 60 mgHg in decreasing order of maltose, starch, sucrose and lactose. 60 mg/kg of Amberlite active fraction lowered the blood glucose level markedly after 18, 35, and 60 min after an oral maltose load and the antihyperglycemic activity was maintained upto 90 min. In alloxan-induced hyperglycemic mice, Amberlite active fraction at a dose of 100 mg/kg also significantly lowered blood glucose after an oral maltose load, and its efficacy was almost equivalent to that of acarbowe.
