Actinomycin D Induces Phosphorylation of STAT3 through Down-Regulation of SOCS3 in Renal Cancer Cells (신장암 세포주에서 actinomycin D에 의한 SOCS3 발현 감소를 통한 STAT3 활성화)

  • Actinomycin D is a natural antibiotic that is used in anti-cancer chemotherapy and is known as a transcription inhibitor. Interestingly, actinomycin D induces phosphorylation of signal transducers and activators of transcription 3 (STAT3) in renal cancer Caki cells. In this study, we examined the molecular mechanism of actinomycin D-induced STAT3 phosphorylation. Treatment with actinomycin D induced phosphorylation of STAT3 (Tyr705) in a dose- and time-dependent manner. However, actinomycin D did not induce phosphorylation of STAT3 (Ser727), STAT1 (Tyr701) and STAT1 (Ser727). Moreover, actinomycin D-induced STAT3 phosphorylation was caused by decreased protein and mRNA levels of SOCS3, but not by JAK2 and SHP-1. In addition, other transcription inhibitor (5,6-dichloro-1-b-D-ribofuranosyl benzimidazole; DRB) also induced phosphorylation of STAT3 (Tyr705). Taken together, the present study demonstrates that transcriptional inhibitors (actinomycin D and DRB) induce phosphorylation of STAT3 (Tyr705) in Caki cells by down-regulation of SOCS3.

Synthetic Cannabinoid-Induced Immunosuppression Augments Cerebellar Dysfunction in Tetanus-Toxin Treated Mice

  • Synthetic cannabinoids are one of most abused new psychoactive substances. The recreational use of abused drug has aroused serious concerns about the consequences of these drugs on infection. However, the effects of synthetic cannabinoid on resistance to tetanus toxin are not fully understood yet. In the present study, we aimed to determine if the administration of synthetic cannabinoids increase the susceptibility to tetanus toxin-induced motor behavioral deficit and functional changes in cerebellar neurons in mice. Furthermore, we measured T lymphocytes marker levels, such as CD8 and CD4 which against tetanus toxin. JWH-210 administration decreased expression levels of T cell activators including cluster of differentiation (CD) $3{\varepsilon}$, $CD3{\gamma}$, CD74p31, and CD74p41. In addition, we demonstrated that JWH-210 induced motor impairment and decrement of vesicle-associated membrane proteins 2 levels in the cerebellum of mice treated with tetanus toxin. Furthermore, cerebellar glutamatergic neuronal homeostasis was hampered by JWH-210 administration, as evidenced by increased glutamate concentration levels in the cerebellum. These results suggest that JWH-210 may increase the vulnerability to tetanus toxin via the regulation of immune function.

Evaluation of Optimum Mix Proportion and Strength of Volcanic Ash based Geopolymer (화산재 기반 지오폴리머의 최적배합 도출 및 강도 특성)

  • This study determined the optimum mix proportions for volcanic-ash-based geopolymer by analyzing the flow, setting time, and compressive strength. $Na2SiO_3$ and NaOH were used as alkali activators, and NaOH concentrations of 2, 4, 6, and 8M were used for different experimental cases. The A/B ratios examined were 0.25, 0.3, 0.35, 0.4, and 0.45, and the ratios of volcanic ash to blast furnace slag binder were 7:3, 6:4, and 5:5. In the experiment, the flow and setting time tended to decrease and the compressive strength increased as the molarity of NaOH in the geopolymer increased. The optimum molarity of NaOH was determined to be 4M. As the A/B ratio increased, the setting time decreased and the compressive strength increased. The most advantageous A/B ratio for the setting time and strength was 0.35. Increasing the ratio of volcanic ash resulted in a longer setting time and lower compressive strength. The optimum binder ratio was chosen as 6:4 based on the setting time and compressive strength. Thus, 4M of NaOH, an A/B ratio of 0.35, and binder ratio of 6:4 are considered as the proper parameters for the volcanic-ash-based geopolymer.

Effect of Fillers on High Temperature Shrinkage Reduction of Geopolymers (충전재에 의한 지오폴리머의 고온수축 감소효과)

  • Geopolymers produced from aluminosilicate materials such as metakaolin and coal ash react with alkali activators and show higher fire resistance than portland cement, due to amorphous inorganic polymer. The percentage of thermal shrinkage of geopolymers ranges from less than 0.5 % to about 3 % until $600^{\circ}C$, and reaches about 5 ~ 7 % before melting. In this study, geopolymers paste having Si/Al = 1.5 and being mixed with carbon nanofibers, silicon carbide, pyrex glass, and vermiculite, and ISO sand were studied in order to understand the compressive strength and the effects of thermal shrinkage of geopolymers. The compressive strength of geopolymers mixed by carbon nanofibers, silicon carbide, pyrex glass, or vermiculite was similar in the range from 35 to 40 MPa. The average compressive strength of a geopolymers mixed with 30 wt.% of ISO sand was lowest of 28 MPa. Thermal shrinkage of geopolymers mixed with ISO sand decreased to about 25 % of paste. This is because the aggregate particles expanded on firing and to compensate the shrinkage of paste. The densification of the geopolymer matrix and the increase of porosity by sintering at $900^{\circ}C$ were observed regardless of fillers.

  • Material of the gasket for lithium ion battery requires the chemical resistance, the electrical insulting property, the compression set, the anti-contamination level and the low temperature resistance. We compounded ethylene propylene diene monomer (EPDM), which showed widely different solubility parameter index, with adjusting the amount of metal oxide as an activator. We did long-term test and compression set against an electrolyte with consideration for operating conditions in lithium-ion battery. In these tests, we checked the physical, chemical characteristics and the effect to lithium ion battery with different kinds of activators. In case of rubber with ZnO as an activator, through 1000 h depositing test in propylene carbonate which is one of representative solvents, we could get the satisfying characteristics and result. However, $Zn^{2+}$ had eluted in the ion elution test. So, ZnO should be limited in EPDM compound for the gasket material in lithium-ion battery.

  • Cement is pointed out as the main cause of carbon dioxide emission in the construction industry. Many researchs are underway to use blast furnace slag, an industrial by-product, as a substitute for cement to reduce carbon dioxide emitted during the manufacturing the cement. When blast furnace slag is used as a substitute for cement, it has advantages such as long-term strength and chemical resistance improvement. However, blast furnace slag has a problem that makes initial strength low. This is due to the impermeable film on the surface created during the production of blast furnace slag. The created film is known to be destroyed in an alkaline environment, and based on this, previous studies have suggested a solution using various alkali activators. But, alkali activator is dangerous product since it is a strong alkaline material. And it has the disadvantage in price competitiveness. In this study, an experiment was conducted to improve the initial hydration reactivity of the blast furnace slag to secure the initial strength of the mortar substituted with the blast furnace slag and to check whether the carbonation resistance was increased. As a result of the experiment, it was confirmed that the mortar using alkaline water showed higher strength than the mortar using tap water, and there were more hydration products generated inside. In addition, it was confirmed that the mortar using alkaline water as a compounding water had high carbonation resistance.

Anti-melanogenic Effects of Cnidium japonicum in B16F10 Murine Melanoma Cells (B16F10 피부 흑색종세포에서 갯사상자 추출물의 멜라닌 합성 저해 효과)

  • Melanin is a pigment produced by melanocytes to protect the skin from external stimuli, mainly ultraviolet (UV) rays. However, abnormal and excessive production of melanin causes hyperpigmentation disorders, such as freckles, age spots, and discoloration. Natural cosmeceuticals are a new trend for treating or preventing hyperpigmentation due to fewer side effects and biocompatibility. In this context, the current study focused on Cnidium japonicum, a halophyte with several uses in folk medicine, to evaluate its potential as a skin-whitening agent. The effect of C. japonicum extract (CJE) on melanin production was analyzed in melanogenesis-stimulated B16F10 melanoma cells. The results showed that CJE successfully inhibited the oxidation of tyrosine and L-DOPA by tyrosinase and subsequently decreased the production of the key enzymes responsible for melanin production: tyrosinase, tyrosinase-related protein-1, and protein-2. This effect was confirmed by decreased intracellular and extracellular melanin levels in B16F10 melanoma cells after CJE treatment. Further experiments to elucidate the action mechanism revealed that CJE treatment suppressed melanin production by inhibiting the activation of glycogen synthase kinase 3 β (GSKβ)/β-catenin and protein kinase A (PKA)/cAMP-response element binding protein (CREB) pathways, which are the upstream activators of melanogenesis. In conclusion, the present study suggests that C. japonicum is a potential natural source of bioactive substances for the development of novel cosmeceuticals that can act against hyperpigmentation.

The Regulation of Stress Responses by Non-tandem CCCH Zinc Finger Genes in Plants (식물에서 non-tandem CCCH zinc finger 그룹 유전자에 의한 스트레스 반응 조절)

  • In plants, there are many CCCH zinc finger proteins consisting of three cysteine residues and one histidine residue, which bind to zinc ions with finger configuration. CCCH-type zinc finger proteins are divided into tandem CCCH-type zinc finger (TZF) and non-TZF proteins: TZF proteins contain exactly two tandem CCCH-type zinc finger motifs whereas non-TZF proteins have fewer or greater than two CCCH-type zinc finger motifs. The functions of TZF genes, especially plant-specific RR-TZF genes, have been well studied in several plants, whereas the functional roles of non-TZF genes have not been adequately researched compared to TZF genes. Many non-TZF genes have been identified as being involved in the responses to biotic and abiotic stresses, such as pathogen, high salt, drought, cold, heat, and oxidative stresses. Some non-TZF proteins bind to RNA and are involved in the post-transcriptional regulation of stress-responsive genes in the cytoplasm. In addition, other non-TZF proteins act as transcriptional activators or repressors that regulate the expression of stress-responsive genes in the nucleus. Despite these studies, stress signal transduction and upstream and downstream genes of non-TZF genes have not been sufficiently researched, suggesting that additional studies of the functions of non-TZF genes' functions in plants' stress responses are needed. In this review, we describe non-TZF genes involved in biotic abiotic stress responses in plants and their molecular functions.

Molecular Characterization of Hanwoo Glucose Transporter 4 Gene (한우 Glucose Transporter 4 유전자의 분자생물학적 해석)

  • The uptake of glucose for metabolism and growth is essential to most animal cells and is mediated by glucose transport protein. In the glucose transport protein family, GLUT4 plays a key role in cellular glucose uptake stimulated by insulin in skeletal muscles and adipose tissue in rodents and human. In this studies, we reported the identification, characterization, and expression of Hanwoo GLUT4 gene. The Hanwoo GLUT4 cDNA includes a 1527 bp open reading frame encoding a protein of 509 amino acids. The GLUT4 amino acid sequences of the Hanwoo show strong conservation with the corresponding sequences reported in other species. The highest mRNA expression of GLUT4 was detected in heart and lower expression was detected in rib meat, sirloin, and colon. We confirmed the expression of GLUT4 in the subcutaneous and small intestinal adipose tissue using RT-PCR. To investigate the expression of GLUT4 in the bovine intramuscular adipose differentiation, fibroblast-like cells were isolated from the sirloin of Hanwoo bull aged 12 months by collagenase digestion of minced tissue and cultured with activators of PPAR gamma. We identified that GLUT4 mRNA expression decreased during differentiation of preadipocytes into adipocyte in Korean cattle. These results indicated that function of GLUT4 in bovine adipose tissue was different from that of mouse and human.

A study on treatment effects of different activator types in Angle's Class II div.1 malocclusion patients (2급 1류 부정교합 환자에서 activator유형에 따른 치료 효과에 관한 연구)

  • This research was carried out to compares the treatment effects of Horizontal and Vertical type activators in Angle's Class II div. 1 maloccusion patients with mandibular retrusion dand to find out whether different treatment effects or growth pattern were observed between sexes in each study groups. The results were as follows: 1. In Horizontal activator group, forward positioning of mandible and vertical increase in anteror face as examplified by increase of LAFH and AEM were observed when pre and post-treatment datas were evaluated. 2. Males samples in Horizontal activator group showed increase in mandiular length accmpanied by posterior positioning of maxilla, wheras female samples in Horizontal activator group showed increase in mandibular body length, labial inclination of mandibular incisors and increase in lower anterior facial height .3. In vertical activator group, increase in AFH, LAFH, PFH and LPFH were observed when pre and post treatment datas were evaluated. 4. Male samples in Vertical activator group showed increase in mandibular body length and anterior and posterior facial heights, whereas females samples of Vertical activator group showed mainly increase in anterior facial height. 5. When pre and post treatment datas of Horizontal and Vertical activator groups were compared, skeletal difference were mainly observed in pretreatment datas but dental difference were observed in post treatment datas ,indicating that two actiators differ only in their effects to dental variables. 6. Difference between sexes were noted after treatment although no difference were observed between sexs in each groups before treatmentt. This indicates that inherent growth effects in each sex exerts more influence 1km appliances used for treatment.
