Antioxidant Activity and Whitening Effects of Acteoside and Isoacteoside (Acteoside와 Isoacteoside의 항산화 작용 및 미백 효과)

  • This study was performed to investigate the effects of acteoside and isoacteoside isolated from Clerodendron trichotomum Thunberg on melanin production in B16 melanoma cells. In DPPH radical scavenging activity, acteoside and isoacteoside had a potent anti-oxidant activity in a dose-dependent manner. Both acteoside and isoacteoside dose-dependently inhibited silica-induced ROS (reactive oxygen species) generation in B16 melanoma cells. They significantly inhibited tyrosinase activity and melanin production in MSH-stimulated B16 melanoma cells. The inhibitory effect of acteoside was more potent than that of isoacteosidee. In Western blot of tyrosinase, acteoside inhibited MSH-induced tyrosinase expression in B16 melanoma cells, which is related to the inhibitory action of acteoside on tyrosinase activity and melanin production. These results show that acteoside and isoacteoside from Clerodendron trichotomum Thunberg has a potent antioxidant activity and whitening activity. The underlying mechanism of acteoside on whitening activity may be due to the inhibition of tyrosinase activity and tyrosinase expression.

Prediction and Identification of Biochemical Pathway of Acteoside from Whole Genome Sequences of Abeliophyllum Distichum Nakai, Cultivar Ok Hwang 1ho (미선나무 품종 옥황 1호의 유전체를 활용한 Acteoside 생화학 합성과정 예측 및 확인)

  • Whole genome sequence of Abeliophyllum distichum Nakai (Oleaceae) cultivar Ok Hwang 1 Ho, which is Korean endemic species, was recently sequenced to understand its characteristics. Acteoside is one of major useful compounds presenting various activities, and its several proposed biochemical pathways were reviewed and integrated to make precise biochemical pathway. Utilizing MetaPre-AITM which was developed for predicting secondary metabolites based on whole genome with the precise biochemical pathway of acteoside and the InfoBoss Pathway Database, we successfully rescued all enzymes involved in this pathway from the genome sequences, presenting that A. distichum cultivar Ok Hwang 1 Ho may produce acteoside. High-performance liquid chromatography result displayed that callus of A. distichum cultivar Ok Hwang 1 Ho contained acteoside as well as isoacteoside which may be derived from acteoside. Taken together, we successfully showed that MetaPre-AITM can predict secondary metabolite from plant whole genomes. In addition, this method will be efficient to predict secondary metabolites of many plant species because DNA can be analyzed more stability than chemical compounds.

  • Acteoside는 일명 Verbascoside로 불리우는 Phenylpropanoid glycoside에 속하는 화합물로 이미 보고되어진 생리활성으로는 Cytotoxic effect, Anti bacterial effect, Immunosuppressive effect, C-AMP Phospaestrase의 저해활성등 여러가지 생리활성이 보고되어 있다. 저자등은 마주송이풀의 성분을 규명하는 가운데 acteoside가 주성분임을 확인하게 되었고 이 식물이 한의서에서 관절염등에 사용되어 진다는 점에서 소염활성을, 식물을 건조할때 흑색으로 변하는데 착안하여 iridoid glycoside의 존재가 추정되어 iridoid glycoside의 일반 생리활성인 담즙분비 촉진작용과 간장보호 작용을 검색하던 중 phenylpropanoid glycoside인 acteoside에서도 담즙분비촉진 작용과 간장보호작용의 지견을 얻었기에 보고하고자 한다.

The Effect of Acteoside on Histamine Release and Arachidonic Acid Release in RBL-2H3 Mast Cells

  • The effect of acteoside, a phenylpropanoid glycoside isolated from Clerodendron trichotomum Thunberg, on histamine and arachidonic acid release was investigated in RBL 2H3 cells. Histamine was dose-dependently released from RBL 2H3 cells by melittin, arachidonic acid and thapsigargin. In extracellular $Ca^{2+}-free$ solution, basal secretion of histamins increased by two fold. The response of histamine release to melittin and thapsigargin in $Ca^{2+}-free$ solution was significantly decreased, whereas the response to arachidonic acid was significantly increased as compared with those in normal solution. Acteoside inhibited histamine release induced by melittin, arachidonic acid and thapsigargin in a dose-dependent manner in the presence or absence of extracellular $Ca^{2+}$. However, the inhibitory activity of acteoside was more potent in normal solution than that in $Ca^{2+}-free$ solution. These data suggest that inhibitory mechanism of acteoside on histamine release may be related to extracellular $Ca^{2+}$. On the other hand, acteoside significantly inhibited arachidonic acid release and prostaglandin $E_2$ production Induced by $0.5\;{\mu}M$ melittin. It is possible that acteoside may be developed as an anti-inflammatory agent.

Induction of Differentiation in HI-60 Human leukemia cells by Acteoside.

  • In previous reports, we exhibited that acteoside showed significant cytotoxicity against various cancer cells. In this study we investigated that acteoside is capable of inducing differentiation in HL -60 human leukemia cell line. After being treated with acteoside, the growth curve was decreased remakably in a dose- and time-dependent manner, and cell doubling time was delayed. Exposure of cells to 20 $\mu\textrm{g}$/m$\ell$ acteoside induced differentiation of HL-60 cells to monocyte/macrophage-like cells by cell surface antigen expression. The percentage of NBT reducing activity was increased in a time-dependent manner. In addition. the protein lever of p21 and p16 increased and ppRb decreased in western biot analysis. Theas results suggest that acleoside possess the activity of inducing differentiation in HL-60 cells.

  • The protective effects of acteoside, a phenylethanoid glycoside, on cabon tetrachloride-induced hepatotoxicity and the possible mechanisms involved in this protection were investigated in mice. Pretreatment with acteoside prior to the administration of carbon tetrachloride significantly prevented the increased serum enzymatic activities of alanine and aspartate aminotransferase in a dose-dependent manner.   • Determination of acteoside in Pedicularis resupinata var. oppositifolia has been studied using ion pair liquid chromatography. Sample was extracted with 40mL methanol for 4 hrs. The extract was cleaned up by using Sep-Pak $C_{18}$ catridge and 8mL aqueous methanol eluent(methanol 50%, water 50%, phosphate buffer pH=8.0). Its determination was performed by means of IP-HPLC with a Hamilton PRP-1 polystyrene-divinylbenzene reversed phase column($15cm{\times}4.6mm$ i. d., $5{\mu}m$) and an aqueous methanol eluent(methanol 60%, water 40% phosphate buffer pH=8.2) containing of $5.0{\times}10^{-3}M$ tetrabutylammonium bromide. The established method was applied to the sample that was collected in area of Pyung Chang gun. As a result, its content ranges showed to be 0.062~0.076%.

  • In this study, we evaluated the antioxidant activity and anti-inflammatory effects of Abeliophyllum distichum (A. distichum) leaves that were prepared via air-drying. Fresh and air-dried A. distichum leaves were examined via 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical, and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging assay and measurements of the reducing power. The suppression effects on inflammation of the leaves were analyzed by a western blot and RT-PCR on LPS-induced RAW 264.7 cells. As a result, the antioxidant activity of the fresh leaves was found to be more effective than that of the air-dried leaves. Also, the fresh leaves were more effective in suppressing the protein and mRNA levels of iNOS and COX-2 than the air-dried leaves, thereby indicating the better anti-inflammatory effects. In addition, the contents of phenolic compounds and acteoside were analyzed by high-performance liquid chromatography (HPLC). The results showed that the acteoside content decreased with the use of the air-drying method, while there was no change in the content of phenolic compounds. Therefore, this study indicated that fresh A. distichum leaves potential antioxidant and suppression activities of various factors that are involved in the production of NO, which were found to be better than those of air-dried A. distichum leaves. These biological activities were also found to be independent of the content of phonolic compounds and were assumed to be directly or indirectly related to the content of acteoside.
