석청포 정유의 진정 및 진통효과 (Sedative and analgesic effects of essential oil of Acorus gramineus Soland in mouse)

  • The rhizomes of the Acorus gramineus Soland have been used as sedatives, analgesics, stomachics and anthelmintics in chinese medicine. It is known that the rhizomes of the Acorus gramineus Soland contains the essential oil about 0.5~0.8% and this essential oil contains asarone about 86%. The asarone possess many pharmacological properties similar to those of reserpine and chlorpromazine. Sedative and analgesic effects of essential oil of Acorus gramineus Solaad in mouse was observed. The essential oil of Acorus gramineus Soland decreased the frequency of ambulation on mouse in proportion of concentration. ${\alpha}_2$ receptor antagonist(yohimbine hydrochloride) and opioids receptor antagonist(naloxone hydrochloride) were markedly decreased in frequency of ambulation. The essential oil of Acorus gramineus Soland decreased writhing syndrome in mouse and ${\alpha}_2$ receptor antagonist(yohimbine hydrochloride), opioids receptor antagonist(naloxone hydrochloride) were not increased above effects. In conclusion, these experimental results show that the essential oil of Acorus gramineus Soland have sedative and analgesic effects, but it did not antagonized ${\alpha}_2$ receptor antagonist(yohimbine hydrochloride) and opioids receptor antagonist(naloxone hydrochloride).

Evaluation of the Effects of Acorus gramineus and Acorus tatarinowii Extracts on a Rat Model of Arterial Thrombosis induced by Ferric Chloride

  • We investigated the antithrombotic effect of Acorus gramineus Soland (A. gramineus) from Korea and Acorus tatarinowii Schott (A. tatarinowii) from China in a rat model of arterial thrombosis induced by ferric chloride ($FeCl_3$). Thirty minutes prior to a 35% $FeCl_3$ application, Sprague Dawley rats were intraperitoneally injected with saline, A. gramineus and A. tatarinowii (100 mg/kg), respectively. Occlusion time of rats injected with A. gramineus was delayed significantly compared to that of the vehicle and A. tatarinowii. Thrombus weight was meaningfully decreased in rats injected with A. gramineus compared to the vehicle. Additionally, A. gramineus inhibited collagen fiber damage in vessel compared to the vehicle, but A. tatarinowii did not show a significant effect. Our results show that A. gramineus and A. tatarinowii from the same genus have different antithrombotic effects, and especially A. gramineus has a better antithrombotic effect than A. tatarinowii.

창포 정유가 토끼의 적출장관 운동에 미치는 영향 (Effect of Essential Oil of Acori Rhizoma on Motility of Isolated Rabbit Jejunum Segment)

  • 같은 천남성과에 속하면서도 정유의 함유성분 및 함량이 서로 다른 수창포 정유와 석창포 정유가 토끼 적출장관 운동에 미치는 영향과 차이점, 또 어떠한 작용양식에 의하여 이루어지는가를 알아보고저 carbachol pilocarpine, histamine 및 barium chloride등과의 상호작용을 비교, 검토하였던 바 다음과 같은 결론을 얻었다. 1. 수창포 정유와 석창포 정유는 모두 토끼 적출장관 운동을 이완시켰으며, 농도를 증가시킬수록 이완정도는 현저하였고, 장관운동의 이완되는 정도는 두 정유 모두 같은 농도에서는 거의 비슷하였다. 2. ED50에 해당하는 농도는 수창포 정유는 0.0045% 이었고, 석창포 정유는 0.0035%이었다. 3. 수창포 정유와 석창포 정유에 의해 이완된 토끼의 적출장관은 carbachol pilocarpine, barium chloride에 의하여 정유 투여전의 상태로 회복하였으나, histamine에 의해서는 부분적인 회복을 보였다. 4. 수창포 정유와 석창포 정유는 carbachol pilocarpine, histamine 및 barium chloride에 의하여 수축된 토끼의 적출장판을 이완시켰다. 5. 이상의 결과로 보아서 수창포 정유와 석창포 정유는 다같이 향신경성작용 및 향근육성작용에 의해 토끼 적출장관 운동을 이완시킨다고 사료된다.

  • The hexane fractions from methanolic extracts of Anetheum graveolens L. (Umbelliferae) and Acorus gramineus Soland. (Araceae) revealed potent inhibitory activities against the resistance of multi-drug resistant Staphylococcus aureus SA2 when combined with ampicillin (Am) or chloramphenicol (Cm). As active principles, carvone and the liquid mixture containing carvone from Anetheum graveolens L. and a liquid mixture mainly consisting of benzoic acid phenyl-methyl ester (benzyl benzoate) from Acorus gramineus Soland. were identified. They showed resistance inhibition at the level of 20-50${\mu}g$/ml when combined with 100 or ${\mu}g$/ml of Am or Cm, respectively.

석창포의 약리(藥理)에 대한 문헌적(文獻的) 고찰(考察) (The Bibiographical Investigation of effect of Acorus gramineus Soland)

  • 1. 목적(目的) 및 방법(方法) 음양오행적(陰陽五行的) 기미론(氣味論)에 입각(立脚)한 기존약물(旣存藥物)들을 사상의학내(四象醫學內)의 각 체질병증(體質病證)에 사용코자 했을때 아직까지도 그 약물(藥物)의 정확한 체질소속(體質所屬)과 사상의학적(四象醫學的) 약리(藥理)를 설명하지 못하고 혼돈(混沌)과 논쟁(論爭)에 빠져 있는 경우가 많았다. 이에 본 연구(硏究)는 석창포(石菖浦)이라는 약물(藥物)을 통해 약물(藥物)의 체질소속(體質所屬)과 체질병증하(體質病證下)에서의 용약정신(用藥精神) 그리고 체질약물(體質藥物)의 공통속성(共通屬性)을 객관화(客觀化)하며 각 다른 약물(藥物)의 약리규명(藥理糾明)의 객관화(客觀化) 연구(硏究)에 그 실마리를 찾고자 한다. 문헌적(文獻的) 고찰(考察)의 방법(方法)을 위주로 현재 사용되는 석창포(石菖浦)의 종류(種類)와 학명(學名)을 살펴보고 각 문헌(文厭) 상(上)에서 기술된 석창포(石菖浦)의 약효(藥效)를 검토하였고 필요에 따라 다른 약물(藥物)을 인용(引用)하였으며 동의보감(東醫寶鑑)과 사상처방(四象處方) 중에 석창포(石菖浦)을 단방(單方)으로 사용하거나 포함한 방제(方劑)가 있는 문(門)을 종합 정리하여 그를 통해 석창포(石菖浦)의 체질소속(體質所屬)과 효능(效能)을 비교 검토하였다. 2. 결과(結果) 및 결론(結論) 기존 본초론적(本草論的)으로 석창포(石菖浦)는 기미(氣味)가 신온(辛溫)하여 향(香)이 있고 약용부위(藥用部位)가 뿌리여서 상초(上焦)인 폐(肺)와 상하초(中下焦)인 간(肝)를 겸(薦)하여 상승(上昇)하며 조습거담(燥濕祛痰) 및 분리청탁(分別淸獨)하여 기석(氣液)의 저체(沮滯)와 착종(錯綜)을 균조(均調)한다. 이런 약리(藥理)는 단미(單味)를 떠나 사상의학내(四象醫學內)의 체질병증(體質病證)에 소속되는 과정(過程)중에서 태음인(太陰人)의 기석(氣液)의 분리청탁(分別淸獨)작용을 통한 폐이호(肺以呼), 간이급(府I以吸)의 승강조절(昇降調節)의 용약정신(用藥精新)을 찾을 수 있고 착종폐기지참오균조(錯綜肺氣之參伍均調)의 약성(藥性)을 가짐을 알 수 있다. 기능(機能)과 구조(構造)를 동시에 개선(改善)시키며 단순 치습(治濕)이 아닌 기파지기(氣波之氣)의 호산흡취(呼散吸聚)관계로만 설명 가능함을 또한 확인 할 수 있었다. 향후 사상체질(四象體質) 약물의 객관화(客觀化)에 문헌적(文獻的) 고찰(考察)의 방법은 그 유용성 및 가치가 크다 할수 있다.

  • Although hypoxic/ischemic injury is thought to contribute to the incidence of Alzheimer disease (AD), the molecular mechanism that determines the relationship between hypoxia-induced β-amyloid (Aβ) generation and development of AD is not yet known. In this study, we investigated the protective effects of Acorus gramineus Soland. (AGS) on oxygen-glucose deprivation/reoxygenation (OGD/R)-induced A β production in SH-SY5Y human neuroblastoma cells. Pretreatment of these cells with AGS significantly attenuated OGD/R-induced production of reactive oxygen species (ROS) and elevation of levels of malondialdehyde, nitrite (NO), prostaglandin E2 (PGE2), cytokines (TNF-α, IL-1β and IL-6) and glutathione, as well as superoxide dismutase activity. AGS also reduced OGD/R-induced expression of the apoptotic protein caspase-3, the apoptosis regulator Bcl-2, and the autophagy protein becn-1. Finally, AGS reduced OGD/R-induced Aβ production and cleavage of amyloid precursor protein, by inhibiting secretase activity and suppressing the autophagic pathway. Although supporting data from in vivo studies are required, our results indicate that AGS may prevent neuronal cell damage from OGD/R-induced toxicity.

  • Scutellariae baicalensis GEORGt Acorus gramineus SOLAND and Gastrodia elata BLUME are traditional medicines used in the treatment of incipient stoke. In this study we investigated their effects on various aspects of neuronal differentiation in single or composite forms. Water-extracts of these medicines showed neuroprotective effects on cultured rat cortical neurons in normoxia and hypoxia. To understand the mechanism for neuroprotection we carried out various cell biological assays. They stimulated initial differentiation of neuronal development (transition from stage 1 to 2), and increased the number of spines and the length and number of dendritic processes. These effects were best manifested in the experimental group, which were given a mixture of the three kinds of extracts (p<0.01). To assess improvement of brain functions we carried out Morris water-maze tests for the mice that were fed on these extracts instead of water for 4 weeks. The experimental groups, especially those which were given the mixture of the three kinds of extract, showed significant (p<0.01) enhancement in memory as early as one day after the learning trial. These results indicate that these three kinds of extracts have synergistic effects on neuronal protection and improvement of brain functions.

ICR 마우스를 이용한 초석잠, 석창포 단독추출물 및 복합추출물의 단회경구투여 독성시험 (Single Dose Oral Toxicity Test of Water Extracts of Stachys sieboldii and Acorus gramineus, and their Mixture in ICR Mice)

  • Stachys sieboldii Miq. (SSM) and Acorus gramineus Soland. (AGS) have been used as traditional medicines for thousands of years in parts of Asia, including Korea, China, and Japan. Recent researches on SSM and AGS have documented a wide spectrum of therapeutic properties, including anti-inflammatory, anti-oxidative, neurodegenerative disease effects. However, the toxicity and safety of SSM and AGS, and their mixture (medicinal herber mixture, MHMIX) were not confirmed. Therefore, this study was performed to evaluate the acute toxicity and safety of SSM, AGS and MHMIX. SSM, AGS and MHMIX were orally administered at a dose of 5,000 mg/kg in ICR mice. Animals were monitored for the mortality and changes in the body weight, clinical signs and gross observation during the 14 days after dosing, upon necropsy. We also measured parameters of organ weight, clinical chemistry, and hematology. No dead and no clinical signs were found during the experiment period after administration of a single oral dose of SSM, AGS and MHMIX. There were no adverse effects on clinical signs, body weight, or organ weight and no gross pathological findings in any treatment group. Therefore, LD50 value of SSM, AGS and MHMIX may be over 5,000 mg/kg and it may have no side toxic effect to ICR mice. The results on the single-dose toxicity of SSM, AGS and MHMIX indicate that it is not possible to reach oral dose levels related to death or dose levels with any harmful side effects.

종자 및 기내배양을 이용한 석창포 증식 (Propagation of Acorus gramineus from Seeds and In vitro Culture)

  • 뿌리에 진통, 진정, 건위, 구충 효능이 있다고 알려진 석창포(Acorus gramineus Soland)의 확대재배를 위하여 현재 분근증식만으로 한정되어 있는 번식법을 개선하고자 종자번식과 기내배양에 의한 번식 방법을 검토하였다. 종자번식을 위한 수확적기는 종피가 연녹색을 띠었을 때가 적합하였으며, 종자의 보관은 $5^{\circ}C$에서 30일까지 저장하는 것이 발아율을 높게 유지할 수 있었다. 석창포의 기내발아에는 $2.0mg{\cdot}L^{-1}$ NAA와 $0.1mg{\cdot}L^{-1}$ BA를 MS 기본배지에 첨가하여 다수의 신초를 유도한 후, $0.1mg{\cdot}L^{-1}$ BA 단독 처리 및 $0.5mg{\cdot}L^{-1}$ NAA와 $1.0mg{\cdot}L^{-1}$ BA 혼합처리에서 발근을 유도하는 것이 효과적이었다. 석창포 육묘상토로는 vermiculite보다 원예상토가 생육이 우수하였다.

석창포에 의한 발기부전 개선 효과 (Effect of Acorus Gramineus on the Relaxation of Corpus Cavernosum Smooth Muscle)

  • The aim of the present study is to investigate whether an ethanol extract of Acorus gramineus Soland (EAG) augments penile erection in vitro and in vivo experiment. Preconstructed with phenylephrine (PE) in isolated endothelium-intact rabbit corpus cavernosum, EAG relaxed penile smooth muscle in a dose-dependent manner, which was inhibited by pretreatment with NG-nitro-L-argininemethylester (L-NAME), a nitricoxide synthase inhibitor, and 1H-[1,2,4]-oxadiazole-[4,3-${\alpha}$]-quinoxalin-1-one (ODQ), a soluble guanylylcyclase (sGC) inhibitor, respectively. EAG-induced relaxation was significantly attenuated by pretreatment with tetraethylammonium (TEA), a nonselective $K^+$ channel blocker. EAG increased cGMP levels of the rabbit corpus cavernosum in a concentration-dependent manner without changes in cAMP levels. In addition, EAG caused increase of peak intracavernous pressure (ICP), ICP/MAP ratio and area under the carve (AUC) in SD rats. Taken together, these results suggest that EAG augments penile erection via NO-cGMP system and $K^+$ channels in corpus cavernosum.
