Somatic Embryogenesis in Withania somnifera (L.) Dunal

  • Somatic embryos were formed from calli obtained from axillary shoots (raised from nodal segments of glasshouse-grown plants under aseptic conditions), internodal segments (from in vitro-raised plants), and root and coty-ledonary leaf segments (from in vitro-raised seedlings) after 8 weeks of initial culture. Embryo formation was the highest (97.33%) from cotyledonary leaf callus on Mura-shige and Skoog's (MS) medium containing kinetin (KN) (3 mg/L). Somatic embryo induction was lesser with different combinations of auxins while it increased to 100% in internodal segment and cotyledonary leaf calli with 6-benzyladenine (BA) (2mg/L) along with 2,3,5-triiodobenzoic acid (TIBA) (2mg/L). The shoots were induced from somatic embryos raised from root, coty-ledonary leaf and internodal segment calli grown on MS medium containing BA in combination with indole-3-acetic acid (IAA). Maximum of 66.67% cultures formed shoots on MS medium containing BA (1mg/L) in combination with IAA (2mg/L). The shoots raised from somatic embryos were rooted on MS medium supplemented with indole-3-butyric acid (IBA) (2mg/L). The plantlets transferred to the field showed 70% survival rate after one year.

In vitro propagation of Bambusa nutans Wall. ex Munro through axillary shoot proliferation

  • This communication describes for the first time an efficient and reproducible protocol for large-scale multiplication of Bambusa nutans. Nodal segments collected from field-grown clumps and cultured on Murashige and Skoog (MS) medium supplemented with $4.4{\mu}M$ benzylaminopurine (BA) and $2.32{\mu}M$ kinetin (Kin) gelled with 0.2% gelrite yielded 80% aseptic cultures with 100% bud-break. The in vitro-formed shoots obtained after bud-break were successfully multiplied in MS liquid medium supplemented with $13.2{\mu}M$ BA, $2.32{\mu}M$ Kin, and $0.98{\mu}M$ indole-3-butyric acid (IBA). Sub-culturing of shoots every 3 weeks on fresh multiplication medium yielded a consistent proliferation rate of 3.5-fold. Shoot clusters containing three to five shoots were successfully rooted with 100% success on half-strength MS liquid medium supplemented with $9.8{\mu}M$ IBA, $2.85{\mu}M$ indole-3-acetic acid (IAA), $2.68{\mu}M$ naphthaleneacetic acid (NAA), and 3% sucrose. Plantlets grown in vitro were acclimatized and subsequently transferred to the field. Inter-simple sequence repeat analysis has confirmed the genetic uniformity of the tissue-cultured plants up to 27 passages.

Growth Performance, Plasma Fatty Acids, Villous Height and Crypt Depth of Preweaning Piglets Fed with Medium Chain Triacylglycerol

  • A study was conducted to investigate the effects of feeding medium chain triacylglycerol (MCT) on growth performance, plasma fatty acids, villus height and crypt depth in preweaning piglets. A total of 150 new born piglets were randomly assigned into one of three treatments: i) Control (no MCT); ii) MCT with milk (MCT+milk); iii) MCT without milk (MCT+fasting). Body weight, plasma fatty acid profiles, villus height and crypt depth were measured. Final BW for the Control and MCT+fasting was lower (p<0.05) than MCT+milk. The piglets fed with MCT regardless of milk provision or fasting had greater medium chain fatty acids (MCFA) than the Control. In contrast, the Control had greater long chain fatty acid (LCFA) and unsaturated fatty acid (USFA) than the MCT piglets. The piglets fed with MCT regardless of milk provision or fasting had higher villus height for the duodenum and jejunum after 6 h of feeding. Similar observations were found in piglets fed with MCT after 6 and 8 days of treatment. This study showed that feeding MCT to the piglets before weaning improved growth performance, with a greater concentration of MCT in blood plasma as energy source and a greater height of villus in duodenum, jejunum and ileum.

Acetobacter xylinum에 의한 미생물 셀룰로오스의 생산을 위한 배지 최적조성 (The Optimal Medium Composition for the Production of Microbial Cellulose by Acetobacter xylinum)

  • Acetobacter xylinum A TCC 23769에 의한 미생물 생룰로오스의 생산을 위한 최적배지가 개발되었다. 미생물생룰로오스의 생산을 위한 각 영양성분의 최 척농도는 리터당 10 g peptone, 20 g yeast extract, 5g glucose, 1.56 g $Na_2HPO_4$, 1.8 g$KH_2PO_4$, 0.05 g $MaSO_4$, 0.002g $FeCl_3$, 5 g citric acid 그리고 10 mL ethanol로 결정되었다. 개발된 배지에서 수득한 생룰로오스의 생산성은 구연산과 에탄융의 상승효과 로 정치배양시 보고된 결과보다 매우 높은 포도당 단위그람당 0.446 그람이었다. 개발된 배지에서 정치배양시 세포성장 빛 셀룰로오스의 생산 또한 조사 되었다.

  • 농산 폐자원 중에서도 감귤 가공공정에 따라 부생되는 감귤과피를 발효 기질로서의 이용을 목적으로 분리균 Asp. niger를 사용하여 구연산 발효에 미치는 배양조건 및 영향인자를 조사하였다. 그 결과 고 체 배양에서의 최적조건 즉 감귤과피와 물을 동량 혼합하고 여기에 NH$_4$NO$_3$0.2%, MgSO$_4$7$H_2O$ 0.1%, Methanol 2.5%를 첨가하여 PH4.5로 조절한 다음포자 현탁액(1x$10^8$ spore/$m\ell$) 2%를 접종하여 균체증식 초기에 35$^{\circ}C$, 20시간 배양하고 4시간 저온 처리한 다음 구연산 생성기에서 3$0^{\circ}C$로 48시간 배양했을 때 당에 대해 80.4%의 높은 수율의 구연산을 얻었다.

남조류(Nostic pruniforme)의 질소고정능에 관한 연구 (Nitrogen Fixation of Blue Green Algae (Nostoc pruniforme))

  • Three kinds of organic matter such as glucose, oxalic acid, and ethanol were added to the media(N-free or $NO_{3}$-riched) and their effects on the nitrogen fixation of Nostoc pruniforme were measured by manometric technique through the experiments in vivo. 1) The organic matters used in this experiments showed effective results as a role of substrate for the fixation of atmospheric nitrogen. 2) In the nitrogen-free medium treated with the both of flucose nad ethanol, the highest nitrogen uptakes were detected in the treated of low concentrations (glucose ; 0.1%, 0.5%, ethanol : 0.1%, 0.5%). On the contrary, the highest nitrogen uptakes in $NO_{3}$-riched medium were measured at the treated of high concentrations (glucose ; 2%, 1%, ethanol ; 1.5%, 1.0%). 3) The highest nitrogen uptakes in N-free medium treated with oxalic acid were measured at the concentration of 2% and 1%, respectively. In the medium of $NO_{3}$-riched, the nitrogen uptakes were in the opposite directions.

Callus induction and plant regeneration of Lychnis wilfordii (Regel) Maxim a critically endangered plant in Korea

  • Lychnis wilfordii (Regel) Maxim is a rare and valued ornamental plant. Germination rate reached 46.6% when seeds were treated with $100mg{\cdot}l^{-1}$ GA (Gibberellic acid). The highest callus induction was observed in the leaf explants of the seedlings on MS medium containing specific concentrations of $0.5mg{\cdot}l^{-1}$ BA ($N^6$-benzyladenine) and $3.0mg{\cdot}l^{-1}$ NAA (a-naphthalene acetic acid). The adventitious shoot was formed in 97.3% of calli on 1/2 WPM (Woody Plant Medium) medium. Shoot elongation of in vitro propagated plantlets was no difference among various medium. The plantlets grew well after transferring to the pot. This in vitro propagation protocol should be useful for conservation of this endangered plant.

n-3 지방산이 유방암세포의 증시과 지질과산화 및 Oncogene 발현에 미치는 영향 (Effects of n-3 Fatty Acids on Proliferation of Human Breast Cancer Cells in Relatino to Lipid Peroxidation and Oncogene Expression)

  • To investigate the effects of n-3 fatty acids on breast cancer, MDA-MB231 human breast cancer cells were cultured in the presence of $\alpha$-linolenic (LNA), eicosapentaenoic(EPA), and docosahexaenoic acid (DHA) at a concentration of 0.5$\mu\textrm{g}$/ml in serum -free IMM medium. Cell growth was monitored and thiobarbituric acid reactive substances (TBARS), $\alpha$-tocopherol contents, and oncogene expression were measured. To compare the effects of n-3 fatty acids with other types of fatty acid, steraic (STA), olieic(OA). linoleic acid(LA) were used. After one day , cell growth was retarded most highly when DHA was in the medium. Cellular TBARS level measured after three days of culture was the highest with DHA in the medium and was also increased by LNA and EPA, compared with STA, OA and LA. Alpha-tocoopherol contents of cells were decreased by DHA but only modestly. There was non significant difference in $\alpha$-tocopherol contents in cells cultured in the presence of the other fatty acids. northern blot hybridization carried out with cells cultured during 24 hours showed that levels of erbB-2 mRNA were not altered by six different fatty acids in the medium but those of c-myc were transiently decreased in the early period by both n-6 and n-3 polyunsaturated fatty acids. The level of tumor suppressor gen p53 mRNA , however, was increased by DHA with time. It is concluded that the cytotoxicity of lipid peroxide and increased expression of tumor suppressor gene p53 are at least partly responsible for the inhibitory effect of DHA on growth of breast cancer cells.

고온, 한냉 및 산도가 토끼 적혈구막 투과성에 미치는 영향 (Influence of Heating, Cooling and Acidity on the Permeability of the Rabbit Erythrocyte Membrane)

  • Outward movement of hemoglobin and $K^+$ ion across rabbit erythrocyte membrane after heating, cooling and in acid medium was studied. One milliliter of rabbit blood was centrifuged and packed red cells were obtained. Packed red cells were resuspended by addition of 4 ml of 0.9% NaCl solution and were subjected to heating $(57^{\circ}C\;for\;5\;minutes)$ or cooling $(-4^{\circ}C{\sim}-8^{\circ}C\;of\;-10^{\circ}C{\sim}-11^{\circ}C\;for\;10\;minutes) $. For acid medium experiment packed ref cells were resuspended by addition of 4 ml of acid medium of PH 4.5 consisting of 0.01% glacial acetic acid in 0.85% NaCl solution and kept standing for 10 minutes. All red cell suspensions were centrifuged again and packed red cells were separated. This packed red cells were again suspended in 4 ml of NaCl solution of 0.8%, 0.7%, 0.6%, and 0.5% concentration respectively and kept standing for 20 minutes. The concentration of hemoglobin and $K^+$ in the supernatant of the above red cell suspensions were measured and the following results were obtained. 1. Outward movement of hemoglobin and $K^+$ was greatest in red cells subjected to heating. The movement paralled to the osmolal concentration gradient between extra- and intra-cellular phase of red cells. 2. In acid medium the outflux of hemoglobin and $K^+$ increased as compared to the control. 3. In red cells subjected to the cold of $-10^{\circ}C{\sim}-11^{\circ}C$ the outflux of hemoglobin and $K^+$ increased. Whereas in the environment of $-4^{\circ}C{\sim}-8^{\circ}C$ there was no change in the outflux of $K^+$. The he-moglobin outflux showed rather a decreased as compared to tile control.
