당근 세포배양으로부터 체세포배 발생에 미치는 아스콜빈산의 효과 (Effects of Ascorbate on Somatic Embryogenesis in Carrot Cell Cultures)

  • 당근의 배양세포로부터 체세포배의 발생과정에 미치는 아스콜빈산 및 dehydroascorbic acid의 영향을 밝히기 위하여 본 실험을 시도하였다. 비배발생세포의 배양에 처리된 아스콜빈산은 세포증식을 촉진시켰을 뿐인데 dehydroascorbic acid는 세포증식을 억제시키면서 배발생세포로 전환시킨 효과가 있었다. 배발생세포의 배양에 처리된 아스콜빈산은 체세포배 발생을 억제시켰지만 dehydroascorbic acid는 체세포배발생을 촉진시켰다. 그러나 이와 같은 발생촉진은 구상배에서 중단되므로 성숙에는 오히려 저해적이었다. 이상의 결과로부터 당근의 캘러스배양에 dehydroascorbic acid를 처리하여 빠른 시일내에 배발생캘러스를 확보한 다음 dehydroascorbic acid 첨가 배발생 배지에서 초기배발생기간 배양 후 MS 기본배지로 옮겨 배양하면 고빈도의 체세포배생산 실험계가 확립될 것으로 판단된다.

Whole-cell Biotransformation of Chlorella Oil Hydrolysates into Medium Chain Fatty Acids

  • A synthetic pathway, which consisted of fatty acid double bond hydratase, alcohol dehydrogenase, and Baeyer-Villiger monooxygenase, was applied to Chlorella oil to produce ester fatty acids, which can be hydrolyzed into medium chain fatty acids. Since linoleic acid is a major fatty acid constituent of Chlorella oil, a fatty acid double bond hydratase from Lactobacillus acidophilus NBRC13951, which is able to convert linoleic acid into 13-hydroxyoctadec-9-enoic acid, was used. Recombinant Escherichia coli expressing the fatty acid double bond hydratase from L. acidophilus NBRC13951 successfully transformed linoleic acid in Chlorella oil hydrolysates into 13-hydroxyoctadec-9-enoic acid with approximately 60% conversion yield. 13-Hydroxyoctadec-9-enoic acid was further converted into ester fatty acids by the recombinant E. coli expressing a long chain secondary alcohol dehydrogenase and a Baeyer-Villiger monooxygenase. The resulting ester fatty acids were then hydrolyzed into medium chain fatty acids by a lipase. Overall, industrially relevant medium chain fatty acids were produced from Chlorella oil hydrolysates. Thereby, this study may contribute to biosynthesis of medium chain fatty acids from microalgae oils as well as long chain fatty acids.

고농도 에탄올 내성균 Acetobacter sp. FM-10을 이용한 초산 발효조건 검토 (Investigation of the Condition of Acetic Acid Fermentation with High Concentration Ethanol Resistant Acetobacter sp. FM-10)

  • The fermenting conditions for acetic acid production with Acetobacter sp. FM-10 which could grow in the medium containing 10% ehtanol were investigated. Initial concentration of acetic acid in broth medium affected greatly to the fermentation speed. For example , the acetic acid production increased proportionally by the increasing of initial concentration was higher that 1.0%. When the cultivation was started with broth medium containing 5% ethanol, the additional adding ethanol during the fermentation was not significantly increased the acidity of the medium. The acidity of the medium containing 10% ethanol was reached to 8.3% after shaking than static cultivation by about 10 days with 150 rpm shaking speed. Acetic acid production with shaking cultivation was faster the static cultivation by abot 10 days under the same condition except shaking. In acetic acid fermentation with the batch style fermentor , the optimum fermentation condition was 700 rpm of agitation speed and 5L/min air flow rate in 3L culture medium .

가축분뇨를 이용한 SCP 생산 균주의 분리 및 균체 단백질 생산

  • From the soil collected from provincial area of South Korea, a microorganisms which have been shown good growth in the minimal poultry feces extract medium was isolated. Supplement of glucose to the poultry feces extract medium helped the complete uptake of soluble protein by microorganism. Uric acid in the poultry feces extract medium could be completely degraded during the microbial growth. Maximum cell growth (3.8 $\times$ 10$^{9}$ CFU/ml) obtained at 36 hours of incubation after inoculation. Uric acid was degraded faster in minimal medium than in the glucose complement medium. VFA (volatile fatty acid), which are known as major compounds of poultry feces odor, were almost removed from the minimal poultry feces extract medium. Glucose supplement to the minimal medium enhanced the growth of microbial cells. Addition of 4% of glucose and 4% of neopeptone to the minimal poultry feces extract medium helped the maximal growth of cells.

부레옥잠을 이용한 Lactobacillus spp.의 젖산 생산 (Production of Lactic Acid from Water Hyacinth by Lactobacillus spp.)

  • Lactic acid fermentations were conducted using water hyacinth. It is known that the pretreatment and enzyme hydrolysis process optimize the potential of water hyacinth. Lactic acid produced by using lactic acid bacteria. All cells were grown at $37^{\circ}C$ and initial pH 5.5. Lactic acid production was measured by HPLC. All Lactobacillus strains could produce lactic acid from pretreated water hyacinth. The highest lactic acid was achieved when lactic acid fermentation was carried out by L. delbrueckii for D-form and L. helveticus for L-form lactic acid production. The lactic acid concentration was 10.70 g/L by L. delbrueckii and it converted glucose in the medium to lactic acid, almost perfectly. Lactic acid production became higher when fermentation was carried out at a controlled pH 5.5. Lactic acid yield and productivity were 0.52 g/g and 0.19 g/L/h for L. helveticus, while L. delbrueckii was 0.64 g/g and 0.27 g/L/h. This study showed that water hyacinth medium could be alternative medium which can replace the complex and expensive medium for growing Lactobacillus strains in production of lactic acid.

Stabile Fermentation of Citric Acid Using Immobilized Saccharomycopsis lipolytica

  • The effects of media composition on citric acid fermentation using surface immobilized Saccharomycopsis lipolytica were studied. The use of the standard medium for these organisms resulted in rapid decrease of citric acid production and a transformation of immobilized cell morphologies from a yeast-type to a mycelium-type. When the standard medium was enriched with vitamins, trace minerals, a growth factor and ammonium to form a Vigorous Stationary Phase (VSP) fermentation type medium, relatively stable citric acid production (10 mg/lㆍh) was obtained. Using the VSP type medium, the surface immobilized cells also retained their yeast-type form.

  • A mono-type glycerine fatty acid ester compound was chemically synthesized using medium-chain fatty acids with antibacterial activity, and the physicochemical quality and antibacterial activity of the synthesized glycerine medium-chain fatty acid ester compound were measured. At a reaction molar ratio of MCT(medium chain triglyceride) to glycerine of 1:2.5, 48.15% mono ester was produced. The physicochemical analysis of the synthesized glycerine fatty acid ester compound showed an acid content of 0.38~0.60%, which tended to increase slightly as the glycerine molar ratio increased. The saponification value decreased as the synthesized molar ratio of glycerine increased from 218 to 284. The number of bacteria was measured to confirm the degree of antibacterial activity of glycerine medium-chain fatty acid esters against food poisoning bacteria, Bacillus cereus and Salmonella typhi. The number of bacteria significantly decreased as the MCT:glycerine molar ratio increased. In particular, the antibacterial effect between the treatment groups was the highest when at an MCT:glycerine molar ratio of 1:2.5.

비타민류가 곰팡이의 유지생산에 미치는 영향 (Effect of Vitamins on Lipid Accumulation of Molds)

  • Influence of vitamins on the felt and lipid production of Aspergilus niger var. macrosporus, Aspergillus fumigatus and penicillium notatum were investigated after 10 days of incubation at $30^{\circ}C$ under static culture condition. The felt of molds were lower in the media containing vitamins than control medium. The lipids produced by each strains were highest yields in media containing vitamins. Among of these Aspergillus niger var. macrosporus and Aspergillus fumigatus produced. 4.6g and 5.28g in medium containing inositol 2.5mg/l and 0.5mg/l, respectively; while Penicillium notatum produced 1.51g in a medium containing thiamine 10mg/l. The major fatty acid of lipids were palmitic acid, stearic acid, oleic acid and linoleic acid. Aspergillus niger var. macrosporus was lower palmitic acid in media containing vitamins than control medium, While linoleic acid was higher in media containing thiamine and inositol. Aspergillus fumigatus was lower oleic acid in media containing vitamins than control medium, While linoleic acid was higher in media containing vitamins. Peniillium notatum was lower palmitic acid, oleic acid and stearic acid in media containing vitamins but linoleic acid was higher. The composition of fatty acid of lipids changed depending on the media containing vitamins but hardly found a certain tendency except linoleic acid which was higher in a media containing thiamines and inositols. The degree of unsaturation of fatty acids in the lipids were comparatively higher in media containing vitamins than control medium.

수은(水銀)이 Lactobacillus bulgaricus의 젖산 생성(生成)과 생육조해작용(生育阻害作用)에 대한 양파 추출물(抽出物)의 효과 (Effect of Onion Extracts on the Growth and lactic Acid Production of Lactobacillus bulgaricus in Medium Containing Mercury)

  • Onion extracts were tested for effects on the growth and lactic acid production of Lactobacillus bulgaricus in the medium containing mercury. The media containing mercury were added with onion extracts and inoculated with the bacterium and then incubated at $39^{\circ}$ for four days. All of the onion extracts examined increased the growth and lactic acid production of the bacterium in the medium containing mercury. At the addition of the edible portion extract of onion to the medium containing 5 ppm, 10 ppm and 20 ppm mercury, the higher the concentration of the onion extract was added, the greater the increasing effect on the growth and lactic acid production. The brown peel extract of onion increased the growth and lactic acid production of the bacterium in the medium containing mercury at all concentrations. The higher the concentration of the extract added, the more effective the increasing effect. The onion solutions of edible portion and brown peel extracted at $90^{\circ}\;to\;100^{\circ}$ showed more desirable effects than those extracted at the room temperature. Among four kinds of the onion extracts, the brown peel solution extracted at $90^{\circ}\;to\;100^{\circ}$ was the most effective in increase of the growth and lactic acid production in the medium containing mercury.

Urease 생산 젖산균의 탐색을 위한 한천 배지 (Agar Medium for Screening of Urease-Producing Lactic Acid Bacteria)

  • An agar medium(HY) was developed to detect the urease-producing lactic acid bacteria. HY medium was prepared with the addition of tryptone, glucose and tween 80 to the supernatant of autoclaved skim milk and yeast extract mixture. There was no difference in eumeration of lactic acid bacteria between the HY and commercial media, such as M17, MRS and BCP agar. The urease activity of Streptococcus salivarius subsp. thermophilus was detected on the HY agar medium contained urea by the color change of bromocresol purple as the pH indicator, but not on the commerical agar media. Furthermore, it was succeeded to screen the urease activity of bacteria in skim milk used as a raw material in dairy product manufacture. Therefore, HY medium was proved to be suitable for the screening of urease-producing lactic acid bacteria.
