Effect of Different Seasons on Cross-Bred Cow Milk Composition and Paneer Yield in Sub-Himalayan Region

  • The study was designed to evaluate the seasonal influences on cross-bred cow milk composition and paneer yield in Dhauladhar mountain range of sub-himalayan region. Fifty samples from each season were collected from a herd of $Jersey{\times}Red\;Sindhi{\times}Local$ cross-bred cows during summer (April-June), rainy (July-September) and winter (November-February) and analyzed for fat, total solids (TS) and solids not fat (SNF). Paneer was prepared by curdling milk at $85{\pm}2^{\circ}C$ with 2.5 per cent citric acid solution. Overall mean for fat, TS and SNF content of milk and paneer yield were 4.528, 13.310, 8.754 and 15.218 per cent respectively. SNF and TS content varied among seasons being highest in winter (8.983% and 13.639%) followed by summer (8.835% and 13.403%) and lowest in rainy season (8.444% and 12.888%). Paneer yield was lowest (14.792%) in rainy season and highest (15.501%) in winter season.

Isolation and Identification of Weissella kimchii from Green Onion by Cell Protein Pattern Analysis

  • This study was conducted to investigate the potential origin of Weissella species, which were found in ingredients of kimchi, such as salted Chinese cabbage, radish, green onion, red pepper powder, pickled shrimps, garlic, and ginger. Ten strains of Weissella species (Weissella thailandensis, W. kimchii, W. koreensis, W. minor, W. halotolerans, W. hellenica, W. kandleri, W. confusa, W. viridescens, and W. paramesenteroides) and lactic acid bacteria isolated from ingredients of kimchi were analyzed by SDS-PAGE of whole-cell proteins. Several strains with patterns identical to those of Weissella kimchii were isolated from green onion. On the basis of biochemical characteristics and 16S rDNA sequence comparisons, these strains were identified as Weissella kimchii, suggesting green onion as a major origin of Weissella kimchii found in kimchi.

Biochemical Characteristics and Antibiotic Resistant Patterns of Klebsiella pneumoniae (Klebsiella pneumoniae 분리균의 생화학적 특성과 항생물질 내성유형 연구)

  • One hundred and thirty-six strains of Klebsiella pneumoniae were isolated from clinical specimen taken from pediatric patiants at 6 different hospitals and identified, characterized and investigated on the patterns of antibiotic resistance. The 136 strains showed the positive reactions in the 17 tests; Voges-Proskauer, ONPG, cirate utilization, KCN degradation, productions of lysine decarboxylase, acid and gas from glucose, utilizations of malonate, manitol, rhamnose, salicin, sucrose, raffinose, arabinose, lactose, sucrose, inositol and raffinose, but the strains showed the negative reactions in the 8 tests; production of $H_2S$, indole, arginine dehydrolase, ornithine decaraoxylase, phenylalanine deaminase, motility, methly red and gelatin liquefaction. 41 did not utilize dulcitol, and 32 did not utilize adonitol. The 36 out of them produced klebecin. The 136 strains were resistant to ampicillin, 2 to gentamicin, 14 to cephalothin, 16 to chloramphenicol, 8 to kanamycin, 13 to streptomycin, and 17 to tetracycline. Twenty strains were resistant to 2 kinds of antibiotics 5 strains to 3, 4 to 4 and 1 to 6 and 7.

  • From crude drug preparation(Soshiho-Tang) ginseng sapogenins were identified by TLC and $ginsenoside-Rb_1$ was determined quantitatively by HPLC. Panaxadiol, pandaxatriol, acid-hydrolysates of ginseng saponin, were identified by TLC with benzene/acetone(4 : 1, v/v). Rf values of which were measured as 0.26 and 0.14, respectively. The content of $ginsenoside-Rb_1$ was determined by HPLC on $Lichrosorb-NH_2$ column with $CH_3CN/H_2O/n-BuOH$(80 : 20 : 10, v/v). Its recovery rate in the extract granules, was as relatively low as $19.8{\pm}1.4%$ compared to the content in raw red ginseng.

Effect of Thyroid hormone on Lipogenesis in Rat White and Brown Adipocytes Culture System

  • Thyroid hormone(T3) stimulates hepatic lipogenesis by increasing expression of genes, indluding acetyl-CoA carboxylase and fatty acid synthase. S14 protein, which is thougth to be involved in lipid metabolism , appears to respond in parallel . Effect of T3 on lipogenesis in white and brown adipose tissue are less clear, and may be complicated by indirect effects of the hormone. We developed an adipocytes system where the indirect effects of thyroid hormone are abolished and direct effects of T3 on lipogenesis could be tested. Fat accumulation was mesured by Oil-Red O staining. Insulin clearly enhanced fat accumulation by 2-fold . Isobutylemethylxanthie(IBMX) apeared to inhibit insulin -stimulated fat accumulation. Dexamethasone increased insulin-stimulatedfat accumulation about 1.3-fold. confluent adipocytes were cultured in serum-free medium or medium containing 10% fetal calf serum or 10% fetal calf serum stripped of thyroid hormone and lipogenesis, assessed by the incorporation of 3H2O , was measured. Medium without serum or supplemented with T3-depleted serum did not amplify the stimulatory effect of T3 on lipogenesis compared to medium containing 10% fetal calf seru. Dexamethasone alone led to a decrease inlopogenesis of about 50 % in white adipocytes and 25% in brown adipocytes. However, dexamethasone amplified the lipogenic respnse to T3 by about 30% in whit eadipocytes and 60% in brown adipocytes. T3(1$\mu$M) stimulated lipogenesis and acetyl-CoA carboxylase and fatty acid syntase mRNA levels up to 2 -fold in both types of adipocytes. It seems that these adipocytes systems are as useful model to study the effects of hormones on lipogenic gene expression as well as lipogenesis.

In Vitro Skin Irritation Test of Anti-Inflammatory Drugs (소염진통제 약물에 대한 In vitro 피부자극 시험연구)

  • In vitro skin iritation of anti-inflammatory drugs was investigated in terms of the cytotoxicity method to human skin fibroblast cells. Five anti-inflammatory drugs (Diclofenac, Naproxen, Meclofenamic acid, Ibuprofen and Fnoprofen) which are commercially available as oral preparations or injections were tested. The cytotoxicity of 5 chemicals was evaluated by using MTT[tetrazolium salt 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide] assay. NRU (neutral red uptake) assay and Alamar Blue assay after fibroblast cells had been exposed to the chemicals for 24 hours or 489 hours. The $IC_{50}$ values of the chemicals showed the comparative strength of cytotoxicity as following order of Meclofenamic acid>Diclofenac>Fenoprofen>Ibuprofen>Naproxen. The values of $IC_{50}$ determined by Alamar Blue assay were lower than those of MTT and NRU assay. These data suggest Alamar Blue assay can be useful method for assessing in vitro skin irritation potential of anti-inflammatory drugs.

  • There are several main antibacterial susceptibility tests, such as agar dilution method, broth dilution method and disk diffusion technique. Especially, for minimal inhibitory concentration (MIC) test, agar dilution method has been widely used. But that method is so complicated and bothering that it's difficult to treat a large amount of strains. On the other hand, modified broth dilution method(add 1% glucose and 0.018% phenol red as a pH indicator to broth) is fast and easy to perform. Most of all, it can visualize the result by color. The MICs of 22 antibiotics Including penicillins, aminoglycosides, cephalothin, chloramphenicol, lincomycin, ceftiofur, vancomycin and quinolones, erythromycin, colistin. sul-fadimethoxine, trimethoprim for arcanobacterium pyogenes 14 strains, actinobacillus pleuropneu-moniae 41 strains and pasteurella multocida 37 strains, which were collected from porcine during 1996 ∼ 1999, were determined by modified broth dilution method. Actinobacillus pleuropneumoniae was highly susceptible to all kinds of quinolones such as ciprofloxacin, enrofloxacin and norfloxacin and to all aminoglycosides, like gentamicin, apramycin, kanamycin and ampicillin, cephalothin and ceftiofur. But It was quite resistant to solfadimethoxin, colistin and vancomycin. Pasteurella multocida was found to have high susceptibility to ampicillin, cephalothin, chlorampenicol and gentamicin but had mid-degree susceptibility to other aminoglycosides. In addition, it was susceptible to norfloxacin and nalidixic acid, but not to newer fluoroquinolone like ciprofloxacin and enrofloxacin and it was resistant to colistin and kanamycin. Arcanobacterium pyogenes was highly susceptible to most of quinolones such as cipoofloxacin, enrofloxacin and norfloxacin and gentamicin and penicillin G. But it also obtained high resistance against the early quinolone, nalidixic acid and aminoglycosides such as amikacin, apramycin and kanamycin and erythromycin, chlorampenicol, tetracyclin and vancomycin.

Genome re-sequencing to identify single nucleotide polymorphism markers for muscle color traits in broiler chickens

  • Objective: Meat quality including muscle color in chickens is an important trait and continuous selective pressures for fast growth and high yield have negatively impacted this trait. This study was conducted to investigate genetic variations responsible for regulating muscle color. Methods: Whole genome re-sequencing analysis using Illumina HiSeq paired end read method was performed with pooled DNA samples isolated from two broiler chicken lines divergently selected for muscle color (high muscle color [HMC] and low muscle color [LMC]) along with their random bred control line (RAN). Sequencing read data was aligned to the chicken reference genome sequence for Red Jungle Fowl (Galgal4) using reference based genome alignment with NGen program of the Lasergene software package. The potential causal single nucleotide polymorphisms (SNPs) showing non-synonymous changes in coding DNA sequence regions were chosen in each line. Bioinformatic analyses to interpret functions of genes retaining SNPs were performed using the ingenuity pathways analysis (IPA). Results: Millions of SNPs were identified and totally 2,884 SNPs (1,307 for HMC and 1,577 for LMC) showing >75% SNP rates could induce non-synonymous mutations in amino acid sequences. Of those, SNPs showing over 10 read depths yielded 15 more reliable SNPs including 1 for HMC and 14 for LMC. The IPA analyses suggested that meat color in chickens appeared to be associated with chromosomal DNA stability, the functions of ubiquitylation (UBC) and quality and quantity of various subtypes of collagens. Conclusion: In this study, various potential genetic markers showing amino acid changes were identified in differential meat color lines, that can be used for further animal selection strategy.

Physiological Responses of the Ark Shell Scapharca broughtonii (Bivalvia: Arcidae) to Decreases in Salinity

  • The ark shell (or 'blood clam') Scapharca broughtonii is a filter-feeding bivalve that has red blood and inhabits waters approximately 10m in depth off the southern coast of South Korea. This study was part of a larger research project investigating the causes of death and restoration of shellfish resources, which are important aquaculture products in South Korea. We examined physiological responses related to survival, respiration, excretion, and amino acid changes as a result of changes in salinity. The 9-day median lethal salinity ($LS_{50}$) was 16.5 psu with confidence limits of 14.9-18.1 psu. At $25^{\circ}C$, the oxygen consumption and ammonia-nitrogen excretion rates were increased with decreases in salinity. Although the osmolality of individuals was acclimated within 2 h at 26.4 psu and 12 h at 19.8 psu, it took more than 5 days at a salinity of 13.2 psu, whereas no individuals acclimated and all died at a salinity of 6.6 psu. Of the amino acids present in the blood, taurine and alanine increased in response to decreased salinity. Tissues of the gill and the mid-gut gland were affected by decreasing salinity. These data will provide important fundamental information for examining the causes of mass mortality of shellfish in the summer.

Effects of Extraction and Processing Methods on Antioxidant Compound Contents and Radical Scavenging Activities of Laver (Porphyra tenera)

  • Laver is one of the most consumed edible red algae seaweeds in the genus Porphyra. Laver is primarily prepared in the form of dried, roasted, and seasoned products. We investigated the total polyphenol and flavonoid contents of laver products, and evaluated the in vitro antioxidant properties of solvent extracts from commercially processed laver products. Significant differences in the concentration of phenolic compounds were found among differently processed laver. The total phenolic content for laver extracts ranged from 10.81 mg gallic acid equivalent (GAE)/g extract to 32.14 mg GAE/g extract, depending on extraction solvent and temperature. Laver extracts contained very few flavonoids (0.55 mg catechin equivalent/g extracts to 1.75 mg catechin equivalent/g extracts). 2,2-Diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS), hydroxyl radical, and superoxide anion scavenging assays were used to determine the radical scavenging capacities of laver extracts. These assays revealed that the processing method and extraction condition affected the antioxidant potentials of laver. Antioxidant activity of dried laver, roasted laver, and seasoned laver increased in a concentration-dependent manner ($100{\sim}1,000{\mu}g/mL$). The radical scavenging activities of $37^{\circ}C$ and $100^{\circ}C$ water extracts were lower than that of a $37^{\circ}C$ 70% ethanol extract. The highest radical scavenging capacity was observed in the $37^{\circ}C$ 70% ethanol extracts of dried laver, roasted laver, and seasoned laver. Overall, these results support that notion that laver contains bioactive compounds, such as polyphenols and flavonoids, which may have a positive effect on health.
