Characterization of Acetylcholinesterase from Korean Electric Ray and Comparison with Torpedo Californica

  • This study has been undertaken to examine the acetylcholinesterase (AChE) of electric organ from korean electtric ray(Narke japonica). Korean electric ray was caughted at Chungmu sea and transported to the laboratory, where electric organs were removed and stored at $-70^{\circ}C$ until used. Acelycholinesterase(AChE) of electric organ was purified by affinity column that was prepared with dicaproyl-methylpyridinium linked to Sepharose 4B. Upon purification, the specific activities in Ellman unit were increased by 52 and 39 times for high salt soluble AChE (HSSE, 870.86 $\DeltaOD/min/geam$ of tissue) and detergent soluble AChE(DSE, 105.42 .$\DeltaOD/min/geam$ of tissue), respectively. Each subunit of AChE separated by SDS polyacrylamide gel electrophoresis(SDS-PAGE)was transferred to immonilon P by western boltting and detected by mAbs raised against each subunit of AChE from electric organ og Torpedo califomica. Collagenic tails of AChE from Torpedo califomica, likewise 103Kd protein of AChE from Narke japonica was detected by monoclonal antibody specific to 103Kd of AChE from Torpedo califomica. However, molar ratio of three subunits of AChE from Narke japonica is different from that of Torpedo calicormica. Furthermore, catalytic subunit of AChE from Narke japonica was not identified by monoclnal antibody specific to catalytic subunit of AChE from Torpedo californica. These results showed differences in molecular structure of AChE from Narke japonica and AChE from Torpedo califormica eventhough they showed same enzymatic activities.

뇌조직의 리포푸신, 아세틸콜린 및 그 관련효소 활성에 미치는 실크 피브로인의 영향 (Effects of Silk Fibroin Powder on Lipofuscin, Acetylcholine and Its Related Enzyme Activities in Brain of SD Rats)

  • This study was designed to investigate the effects of silk fibroin (Mw 500) powder (SFP) on lipofuscin, acetylcholine (ACh) and its related enzyme activities in brain of rats. Sprague-Dawley (SD) male rats (160$\pm$10 g) were fed basic diet (control group), and experimental diets (SFP-2.5 and SFp-5.0 groups) added 2.5 and 5.0 g/kg BW/day for 6 weeks. In case of liver membranes, lipofuscin (LF) levels resulted in a considerable decreases (11.5% and 13.8%, respectively) in SFP-2.5 and SFP-5.0 groups compared with control group. But in case of brain as the most sensitive organ, LF levels were remarkably inhibited about 18.3% and 21.7% in SFP-2.5 and SFP-5.0 groups compared with control group. Acetylcholine (ACh) levels were considerable decrease (3.0% and 9.2%, respectively) in brain membranes of SFP-2.5 and SFP-5.0 groups compared with control group. choine acetyltranferase (ChAT) activities as a synthesis enzyme of ACh, and acetylcholinesterase (AChE) activities as a hydrolysis enzyme resulted in a slight increases (2.4% and 3.0%, 4.6% and 6.3%, respectively), but significance difference between ChAT and AChE activities by SFP administration could be not obtained. Monoamine oxidase-B (MAO-B) activities were significantly inhibited (9.5% and 12.6%, respectively) in brain of SEP-2.5 and SFP-5.0 groups compared with control group. These results suggest that inhibiting effects of LF accumulation and MAO-B activity of silk fibroin(SFP) may play a pivotal role in protecting learning memory impairments by attenuating a various age-related changes for improvement of brain function.

In case of liver membranes, lipofuscin (LF) levels resulted in a slight decreases (4.6% and 11.5%, respectively) in SWP-200 and SWP-400 groups compared with control group. But in case of barin as the most sensitive organ, LF levels were remarkably inhibited about 16.7% and 20.0% in SWP-200 and SWP-400 groups compared with control group. There were no significant differences in acetylcholine (ACh) syntheses as a very important neurotransmitter, and choline acetyltransferase (ChAT) activities as a synthesis enzyme of ACh, and acetylcholinesterase (AChE) activities as a hydrolysis enzyme, which were concerned in transmission of neuron through synapses in brain of SWP-200 and SWP-400 groups compared with control group. Monoamine oxidase-B (MAO-B) activities were significantly inhibited (about 10.2%) in brain of SWP-400 groups compared with control group. These results suggest that inhibiting effects of LF accumulation and MAO-B activity of silkworm powder (SWP) may play a pivotal role in attenuating a various age-related changes for improvement of brain function.

도토리 급여가 치매모델 마우스 뇌조직의 아세틸콜린 및 관련효소 활성에 미치는 영향 (Effects of Acorn (Quercus acutissima CARR.) Supplementation on the Level of Acetylcholine and Its Related Enzyme Activities in the Brain of Dementia Mouse Model)

  • 고령화 현상에 의한 노인 인구의 증가에 따라 노인성 질환연구의 중요성이 대두되고 있으며, 특히 노인성 질환 중 가장 문제되는 치매(dementia)에서 노인성 치매(Alzhemier's disease)는 복합적 인 원인에 의 해서 나타나므로 다양한 관점에서의 연구가 필요하다. 따라서 본 연구에서는 마우스에 scopolamin(30 mg/kg BW)을 투여하여 기억${\cdot}$학습장애를 유발하고, 뇌조직의 아세틸콜린 및 그 관련 효소에 미치는 도토리 분말의 영향을 분석하였다. 뇌의 가장 중요한 신경전달 물질로서 아세틸콜린 생성에 미치는 도토리 분말 투여효과를 비교하였을 때, AP-5및 AP-10 투여군에서 $4.2\%$$11.3\%$의 ACh 합성이 증가하는 경향이 나타났으며, ACh의 분해효소인 acetylcholinesterase(AChE)의 활성은 AP-5 및 AP-10의 도토리 분말 투여군에서 $13.5\%$$17.6\%$로서 AChE효소활성이 유의적으로 감소하였다. 그리고 카테롤아민계 신경전달물질의 파괴에 관계하는MAO-B효소의 활성은 AP-5 및 AP-10의 도토리 분말 투여군에서 $10.0\%$$12.7\%$로서 도토리 분말 투여에 의해 MAO-B효소활성이 현저하게 억제되었다. 따라서 도토리는 AChE 효소의 활성을 저해하여 ACh의 농도를 높일 뿐만 아니라 MAO-B효소의 활성을 효과적으로 억제하여 카테콜아민계 신경전달물질을 잘 보존함으로써 치매 예방에 도움이 될 것으로 기대된다.
