노화촉진마우스에 있어서 참깨박 및 들깨박의 메탄올 추출물이 인지기능 및 항산화능에 미치는 영향 (Effect of Defatted Sesame and Perilla Methanol Extracts on Cognitive Function and Antioxidant Activity in SAMP8 Mice)

  • 본 연구는 탈지 참깨 및 탈지 들깨의 메탄올추출물이 기억과 학습능력 감퇴의 특성을 가지고 있는 노화촉진마우스 SAMP8의 인지기능 및 항산화체계에 미치는 영향을 검토하기 위하여 각 추출물 0.3% 첨가 식이로 3개월간 섭취시킨 후 passive avoidance test로 인지기능변화를 측정하였고, 뇌중 acetylcholinesterase 활성, 지질과산화물 및 항산화효소활성을 분석하였다. 그 결과, 탈지 참깨 및 탈지 들깨의 메탄올 추출물이 노화촉진으로 감소한 step through latency가 증가하였으나, 혈액과 뇌조직의 anetylcholinesterase 활성에는 영향을 미치지 못하였다. 체내 지질 과산화물은 SAMP8 대조군에서 유의하게 증가하였으나, 탈지 참깨와 탈지 들깨의 메탄올 추출물을 첨가함으로써 유의하게 감소하였으며, SOD와 GPx의 활성은 증가하였다. 따라서 탈지 참깨와 탈지 들깨의 메탄올 추출물은 노화촉진마우스의 인지 기능을 개선시키며, 이는 cholinergic 신경전달체계의 변화보다는 체내 지질 과산화물 억제 및 항산화효소 활성을 통한 메카니즘으로 생각된다.

소 반추위 메타게놈에서 새로운 carboxylesterase 유전자 클로닝 및 유전산물의 특성 (Cloning and Characterization of a Novel Carboxylesterase Gene from Cow Rumen Metagenomic Library)

  • 한우의 반추위에서 게놈 DNA를 분리하여 메타게놈 은행을 구축한 다음 carboxylesterase를 암호화하는 유전자를 클로닝 및 유전자를 선별하였다. 선별된 유전자의 DNA 염기서열 및 아미노산 서열을 분석하고 유전산물의 생화학적인 특성을 조사하였다. est1R 유전자는 2,465 bp로 366개의 아미노산 잔기를 가진 단백질을 암호화하였으며 이 단백질의 이론적인 분자량은 61,166 Da이었다. Est1R단백질은 PNB carboxylesterase (P37967), acetylcholinesterase (1EEAA) 및 chain A (1H23A)와 각각 5.9%, 6.1%, 6.1% 상동성을 가지고 있었다. 이러한 검색 결과 기존의 알려진 lipase 및esterase와의 상동성이 낮은 것으로 보아 새로운 그룹의 효소로 추정된다. Est1R효소의 최적 pH는 7.0 근방이었으며 최적 온도는 $40^{\circ}C$ 부근이었다. 한편 10% 유기용매를 함유한 기질의 효소활성측정에서 대조구에 비해 methanol은 95%의 상대적인 활성을 가진 반면에 hexane 용액에서는 그 활성이 반으로 감소하였다. 따라서 유기용매 농도의 작용성에 따라 이 효소의 산업적 이용성도 가능하리라 추정된다.

10가지 식용식물 추출물의 가바, 글루탐산 함량, 아세틸콜린에스테라제 억제 및 라디컬 소거능 비교 (A Comparative Study of GABA, Glutamate Contents, Acetylcholinesterase Inhibition and Antiradical Activity of the Methanolic Extracts from 10 Edible Plants)

  • 본 실험에서는 식용 가능한 10종 식물의 인지기능 개선 관련 효능을 검토하기 위하여 추출물의 AChE 억제활성과 DPPH, ABTS 라디컬 소거활성을 비교하였으며, GABA 및 글루탐산의 함량을 측정하였다. 그 결과, GABA 함량은 오미자 추출물에서 14.8mg/g으로 가장 높게 나타났으며, 천궁과 하수오 추출물에서도 각각 3.1 mg/g과 5.4 mg/g으로 비교적 높게 나타났다. AChE 저해활성은 오미자, 헛개나무, 측백엽, 가시오가피 추출물이 1 mg/mL에서 각각 33, 26.6, 20.7%, 그리고 17.8%의 저해율을 나타내어 10종 추출물 중 높게 나타났다. 또한, DPPH 및 ABTS에 대한 라디컬 소거능은 헛개나무, 측백엽, 가시오가피, 오미자 추출물에서 매우 강하게 나타났다. 글루타민산은 석창포 외의 9가지 추출물에서 고르게 존재하는 것으로 나타났다. 본 연구에서 검토한 10종 추출물 중 특히 오미자는 뇌의 대사를 향상시켜 뇌기능을 촉진시키는 GABA의 함량이 높은 것과 함께 AChE 억제 활성도 가장 높았으며, 라디컬 소거효과도 비교적 높게 나타나 뇌기능 개선을 위한 뛰어난 소재로 나타났다. 또한, AChE 억제 활성과 라디컬 소거능이 높게 나타난 헛개나무, 가시오가피 및 측백엽의 인지기능 개선을 위한 기능성식품 소재로서의 활용 가능성을 확인하였다. 본 연구 결과를 통하여 인지기능 개선 효능과 관련하여 사용되는 지표 중 AChE 억제 활성과 식물 추출물의 라디컬 소거활성과의 상관관계를 나타내는 경향을 확인하였으며, GABA 및 체내에서 GABA로 전환되는 글루탐산 함량이 직접적인 AChE 억제 효과를 나타내지는 않았다.

유산균 발효에 의한 손바닥선인장 줄기추출물의 지표물질 함량 변화 분석 (Analysis of Marker Components of Fermented Opuntia ficus-indica var. saboten Stem Extracts)

  • 손바닥선인장(Opuntia ficus-indica var. saboten)은 제주도 자생식물로 섭취가 가능하고 항염 및 항산화, 인지개선 등 다양한 효능이 보고되었다. 본 연구에서는 손바닥선인장 줄기추출물의 지표물질 함량과 인지개선 효능을 높이기 위해 유산균 발효를 수행하였으며, 비발효추출물과 발효추출물 간 지표물질 함량 차이를 비교하고 acetylcholinesterase (AChE)의 저해 활성도를 평가하여 유산균 발효가 인지장애 개선에 미치는 효과를 연구하였다. 지표물질로는 손바닥선인장 줄기 중 함량이 높은 dihydrokaempferol(DHK)과 항산화 효능이 우수하다고 알려진 quercetin-3-methyl ether (3-MeQ)를 선정하였으며, 함량은 HPLC-PDA (280 nm, 360 nm)로 측정하였다. 실험 결과, 발효추출물의 DHK 함량은 비발효추출물에 비해 16.7 % 증가하였고, 3-MeQ 함량은 L. plantarum과 L. fermentum 발효추출물에서 각각 28.6 %, 21.4 % 증가하였다. 또한, 발효추출물의 AChE 저해 활성이 비발효추출물 ($IC_{50}=28.35mg/mL$)보다 높았으며, 발효추출물 중에서도 L. plantarum 발효추출물 ($IC_{50}=12.56mg/mL$)이 L. fermentum 발효추출물 ($IC_{50}=17.71mg/mL$)보다 높은 AChE 저해 활성을 보였다. 따라서 손바닥선인장 줄기추출물은 유산균 발효에 의해 지표성분 함량이 증가되었으며, AChE 저해 활성이 높아지고 이로 인해 인지장애 개선 효과가 나타난 것으로 사료된다. 이러한 결과는 효능이 향상된 고부가가치의 손바닥선인장 제품 개발에 유용한 자료로 활용될 수 있을 것이다.

면화자 정유의 기억력 손상 완화 효과 (Gossypii Semen oil alleviates memory dysfunction in scopolamine-treated mice)

  • Objectives : Gossypium arboreum (cotton) is traditionally used to treat various health disorders. However, anti-amnesic effect of G. arboreum has not been reported. The objective of this study was to investigate in-vivo the anti-amnesic effects along with in vitro antioxidant and acetylcholinesterase (AChE) inhibition potential in G. arboreum seed essential oil. Methods : The essential oil of G. arboreum obtained by solid phase microextraction (SPME) techniques were identified by gas chromatography-mass spectroscopy (GC-MS). 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) assay were performed to determine the antioxidant activity at various concentrations (312.5, 625, 1250, 2500, 5000, 10000 ㎍/㎖. Y-maze, passive avoidance and Morris water maze tests were carried out to evaluate improved effect on scopolamine (1 mg/kg)-induced memory dysfunction at the dose level of 50, 100 and 200 mg/kg. Donepezil (5 mg/kg) was used as a positive drug control. We performed acetylcholinesterase (AChE) activity assay in ex vivo. Results : Five volatile compounds were identified in G. arboreum. The assays of DPPH and ABTS revealed that G. arboreum increased antioxidant activity in a dose-dependent manner. G. arboreum ameliorated the percent of spontaneous alternation in the Y-maze test, shortened step-through latency in the passive avoidance test, and increased swimming time in the target zone in the Morris water maze test. In addition, G. arboreum inhibited the AChE activity. Conclusions : Based on these findings, G. arboreum may aid in the prevention and treatment of learning and memory-deficit disorders through antioxidant and AChE inhibitory activities.

LDH/ AChE and LDH/BChE Ratios (Paralichthys olivaceus) as Biomarkers of Coastal Pollution on Coast of Korea.

  • This study was designed to develop biomarkers of coastal pollution using biochemical indices of flounder (Paralichthys olivaceus) by changes in lactate dehydrogenase (LDH) activity in the serum and cholinesterase activities in brain membranes. For this purpose acetylcholiesterase (AChE) activity, butyrylcholinesterase (BChE) activity, LDH/AChE ratio and LDH/BChE ratio of cultured flounders at 5 different sites on the southern coast of Korea were compared to those of wild flounders caught in the Pohang, eastern coast of Korea as a control group. Relatively high LDH activities were measured in the serum of flounders cultured on the southern coast of Korea (0.101-0.145 unit) than those in the Pohang control group (0.093 unit). AChE activities were significantly low $(about\;10-20\%)$ in brain membranes of cultured flounders compared to those in the Pohang control group. The ratios of LDH/AChE and LDH/BChE were consistently higher $(136-178\%,\; 155-214\%)$ in cultured flounders than those of Pohang control group. Thus, we propose that the ratios of LDH/AChE and LDH/BChE in flounders could be applicable for the diagnosis of marine pollution.

Comparative Study of Korean White, Red, and Black Ginseng Extract on Cholinesterase Inhibitory Activity and Cholinergic Function

  • This study evaluated cholineresterase inhibitory activity of Korean white ginseng extract (WGE), red ginseng extract (RGE), and black ginseng extract (BGE) and the cholinergic effect on scopolamine (SCOP)-induced amnesic mice. WGE, RGE, and BGE inhibited acetylcholineserase (AChE), as well as butyrylcholineserase (BuChE) in a concentration-dependent manner. BGE presented strong inhibition of AChE with an $IC_{50}$ value of 1.72 mg/mL, followed by WGE (5.89 mg/mL), RGE (6.30 mg/mL), respectively. The inhibitory activity of the three ginseng extracts on BuChE showed similar values among the groups. To better understand the mechanisms of the possible effect of ginseng extract on the cholinergic function, this study assessed the expression of the cholinergic markers of choline acetyltransferase (ChAT) and AChE using western blot and RT-PCR analysis in the brains of amnesic mice. Treatment with ginseng extracts led to inhibition of AChE expression and, the activation of ChAT expression in the hippocampus and the cerebral cortex of amnesic mice as induced by SCOP. The results suggest that ginseng extracts including BGE, appear to modulate the metabolism of acetylchoine (ACh), which would greatly increase synaptic ACh levels and most potently revert SCOP-induced amnesia.

Effects of Erythropoietin on Memory Deficits and Brain Oxidative Stress in the Mouse Models of Dementia

  • The present study was undertaken to explore the potential of erythropoietin in memory deficits of mice. Memory impairment was produced by scopolamine (0.5 mg/kg, $i.p.$) and intracerebroventricular streptozotocin (i.c.v STZ, 3 mg/kg, $10{\mu}l$, $1^{st}$ and $3^{rd}$ day) in separate groups of animals. Morris water-maze test was employed to assess learning and memory. The levels of brain thio-barbituric acid reactive species (TBARS) and reduced glutathione (GSH) were estimated to assess degree of oxidative stress. Brain acetylcholinesterase enzyme (AChE) activity was also measured. Scopolamine/streptozotocin administration induced significant impairment of learning and memory in mice as indicated by marked decrease in Morris water-maze performance. Scopolamine/streptozotocin administration also produced a significant enhancement of brain AChE activity and brain oxidative stress (an increase in TBARS and a decrease in GSH) levels. Treatment of erythropoietin (500 and 1,000 IU/Kg i.p.) significantly reversed scopolamine- as well as streptozotocin-induced learning and memory deficits along with attenuation of those-induced rise in brain AChE activity and brain oxidative stress levels. It may be concluded that erythropoietin exerts a beneficial effect in memory deficits of mice possibly through its multiple actions including potential anti-oxidative effect.

Development of Cholinesterase Inhibitors Using (a)-Lipoic Acid-benzyl Piperazine Hybrid Molecules

  • A series of hybrid molecules between (${\alpha}$)-lipoic acid (ALA) and benzyl piperazines were synthesized and their in vitro cholinesterase [acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE)] inhibitory activities were evaluated. Even though the parent compounds did not show any inhibitory activity against cholinesterase (ChE), all hybrid molecules showed BuChE inhibitory activity. Some hybrid compounds also displayed AChE inhibitory activity. Specifically, ALA-1-(3-methylbenzyl)piperazine (15) was shown to be an effective inhibitor of both BuChE ($IC_{50}=2.3{\pm}0.7{\mu}M$) and AChE ($IC_{50}=30.31{\pm}0.64{\mu}M$). An inhibition kinetic study using compound 15 indicated a mixed inhibition type. Its binding affinity ($K_i$) value to BuChE is $2.91{\pm}0.15{\mu}M$.

Protective Effect of Physostigmine and Neostigmine against Acute Toxicity of Parathion in Rats

  • The effects of physostigmine and neostigmine on the parathin induced toxicity were examined in adult female rats. Physostigmine $(100\;{\mu}g/kg,\;ip)$ or neostigmine $(200\;{\mu}g/kg,\;ip)$ inhibited acetylcholinesterase (AChE) and cholinesterase (ChE) activities in blood, brain and lung when the enzyme activity was measured 30 min after the treatment. At the doses of two carbamates equipotent on brain AChE, neostigmine showed greater inhibition on peripheral AChE/ChE. The enzyme activity returned to normal in 120 min following the carbamates except in the lung of rats treated with neostigmine. Carbamates administered 30 min prior to parathion (2 mg/kg) antagonized the inhibition of AChE/ChE by parathion when the enzyme activity was measured 2 hr following parathion. Neostigmine showed greater protective effect on peripheral AChE/ChE. The effect of either carbamate on AChE/ChE was not significant 2 hr beyond the parathion treatment. Carbamates decreased the mortality of rats challenged with a lethal dose of parathion (4 mg/kg, ip) either when treated alone or in combination with atropine (10 mg/kg, ip). Lethal action of paraoxon (1.5 mg/ks ip), the active metabolite of parathion was also decreased by the carbamate treatment indicating that the protection was not mediated by competitive inhibition of metabolic conversion of parathion to paraoxon. The results suggest that carbamylation of the active sites may not be the sole underlying mechanism of protection provided by the carbamates.
