Clinical Application of $^{18}F-FDG$ PET in Hepatocellular Carcinoma (간세포암에서 $^{18}F-FDG$ PET의 임상 이용)

  • Hepatocellular carcinoma is the most common primary tumor in the liver. FDG PET has been applied for staging and treatment planning of hepatocellular carcinoma. It could reflect tumor prognosis because glucose metabolism assessed by FDG PET is known to have correlations with the differentiation and aggressiveness of the tumor. Although the ability of FDG PET to detect well-differentiated or low grade tumors and intra-hepatic lesions is not good, it is expected to playa major role in pre-surgical assessments for liver transplantation because it is useful in detecting extra-hepatic lesions and unexpected distant metastases with a better diagnostic performance than other conventional imaging modalities. Additionally, FDG PET has an advantage to screen other cancers through whole body scanning. As a new tracer for PET, Acetate demonstrates higher sensitivity and specificity to FDG in evaluating hepatocellular carcinoma. It thus seems that simultaneous use of Acetate PET with FDG PET could be helpful in diagnosis, especially detecting extra-hepatic metastases.

Effects of volatile fatty acids on microalgae growth and N, P consumption in the advanced treatment process of digested food waste leachate by mixotrophic microalgae (Mixotrophic microalgae에 의한 음폐수 소화액 고도처리에 있어 유기산이 microalgae의 성장 및 질소, 인 제거에 미치는 영향)

  • Acetate, propionate, butyrate are the major soluble volatile fatty acids metabolites of fermented food waste leachates. This work investigate the effects of volatile fatty acid on the growth rate and $NH_4-N$, $PO_4-P$ removal efficiency of mixotrophic microalgae Chlorella vulgaris to treat digested food waste leachates. The results showed that acetate, propionate and butyrate were efficiently utilized by Chlorella vulgaris and microalgae growth was higher than control condition. Similar trends were observed upon $NH_4-N$ and $PO_4-P$ consumption. Volatile fatty acids promoted Chlorella vulgaris growth, and nutrient removal efficiencies were highest when acetate was used, and butyrate and propionate showed second and third. From this work it could be said that using mixotrophic microalgae, in this work Chlorella vulgaris, fermented food waste leachates can be treated with high efficiencies.

Effect of Riboflavin on Lipid Metabolism (Riboflavin 이 지질대사에 미치는 영향)

  • The effect of riboflavin feeding on lipid emtabolism in rat were studied. 70 male Sprague-Dawley rats were divided into 7 diet group ; control, riboflavin, cholesterol, MLHP (methyl linoleate hydroperoxide) cholesterol plus MLHP, cholesterol plus riboflavin, cholesterol plus MLHP plus riboflavin. After 4 weeks formular feeding sera of animals were collected, then checked cholesterol, triglyceride, phospholipid levels and lipoprotein by use of cellulose acetate papter. The results were as follows ; 1) Marked increase of triglyceride and phospholipid levels were found in the group feeding with cholesterol, cholesterol plus MLHP. 2) Combined riboflavin feeding prevented the increase of triglyceride and phospholipid levels in the group of cholesterol, cholesterol plus MLPH. 3) Also combined riboflavin feeding prevented the increase of $\beta$-lipoprotein fraction in cholesterol feeding group. Our results demonstrate that riboflavin has an inhibitory effects on the derangements of lipid metabolism due to administration of cholesterol, cholesterol plus MLHP.

  • The fractions of Uncariae Ramulus et Uncus seemed to be closely related with the levels of amino acids and other components which concerns with formation and metabolism of neurotransmitters in brain. The pretreatments of methanolic extract and its fractions prohibited the pentylenetetrazole (PTZ) induced convulsion. In such cases, lowered levels of ${\gamma}-aminobutyric$ acid and glutathione in brain were significantly recovered. And also the increased levels or activities of lipid peroxide, ${\gamma}-aminobutyric$ acid aminotransferase, xanthine oxidase, aldehyde oxidase, superoxide dismutase, catalase and glutathione peroxidase by PTZ-convulsion were lowered to normal state.

  • The ethyl acetate fraction of Uncariae Ramulus et Uncus, which showed anticonvulsant effects against pentylenetetrazole (PTZ) treated mice, were subjected to column chromatography to isolate ursolic acid and hyperin from active eluate. Hyperin decreased the elevated activities of GABA-T and xanthine oxidase and lipid peroxide level dose-dependently in PTZ treated mice brain tissue in vitro, but no effect on superoxide dismutase activity. The effects on such enzyme and component seemed to be related with biosynthesis or metabolism of neurotransmitters.

Anticancer Effects of Gut Microbiota-Derived Short-Chain Fatty Acids in Cancers

  • Short-chain fatty acids (SCFAs), such as butyrate, propionate, and acetate produced by the gut microbiota have been implicated in physiological responses (defense mechanisms, immune responses, and cell metabolism) in the human body. In several types of cancers, SCFAs, especially butyrate, suppress tumor growth and cancer cell metastasis via the regulation of the cell cycle, autophagy, cancer-related signaling pathways, and cancer cell metabolism. In addition, combination treatment with SCFAs and anticancer drugs exhibits synergistic effects, increasing anticancer treatment efficiency and attenuating anticancer drug resistance. Therefore, in this review, we point out the importance of SCFAs and the mechanisms underlying their effects in cancer treatment and suggest using SCFA-producing microbes and SCFAs to increase therapeutic efficacy in several types of cancers.

Changes in Esterase Isozyme Activity after Selective Diets and Insecticides in Monochamus saltuarius (Gebler) Larva (북방수염하늘소(Monochamus saltuarius) 유충의 먹이와 살충제 처리에 따른 소화 효소의 활성 변화)

  • Esterase isozymes were extracted from final instar larvae of M. saltuarius treated with selective diets and inhibitors. Twenty esterase isozymes were separated on 12% native-PAGE gel and stained with three different substrates(${\alpha}$-naphthyl acetate, ${\beta}$-naphthyl acetate, or ${\alpha}$-naphthyl butyrate). Interestingly, the isozymes of Est7(${\alpha}$-naphthyl acetate and ${\alpha}$-naphthyl butyrate) and Est6(${\beta}$-naphthyl acetate) were specifically activated in final instar larvae fed with the bark of Pinus koraiensis. However, we could not find any band from substrate ${\beta}$-naphthyl stearate. The esterase activities of Est3, Est6, and Est7 were inhibited by organophosphate and carbamate insecticides. In addition, The esterase activities of Est4, Est6, and Est7 were also inhibited by eserine. However, inhibition of esterase activities in methoprene, bornyl acetate, linal, cineol, and citral was not observed. However, It is necessary to reconfirm these results in vivo.

Screening of Anti-atherogenic Substances from Insect Resources (곤충자원으로부터 항동맥경화 활성물질 탐색)

  • Lipoprotein-associated phospholipase $A_2\;(Lp-PLA_2)$ is a potential biomarker of coronary heart disease and plays an important proinflammatory role in the progression of atherosclerosis. Also acyl-CoA: cholesterol acyltransferase (ACAT) and oxidized low-density lipoprotein (LDL) playa key role in atherosclerosis, respectively. And so, the inhibitory activities of the methanol extracts of 42 insect resources were examined on $Lp-PLA_2$, ACAT, and LDL oxidation for screening of anti-atherogenic substances. Among them, the methanol extracts of Eurydema rugosa significantly inhibited all of upper three activities. Several kinds of tested insects having high inhibitory effect with the methanol extracts were extracted with n-hexane, ethyl acetate, and acetone, and their inhibitory activities were tested.

Liquid Chromatography-tandem Mass Spectrometry for Quantification of Dioscin in Rat Plasma

  • Dioscin is a biologically active steroidal saponin with anticancer and hepatoprotective effects. A rapid, selective, and sensitive liquid chromatographic method with electrospray ionization tandem mass spectrometry was developed for the quantification of dioscin in rat plasma. Dioscin was extracted from rat plasma using ethyl acetate at acidic pH. The analytes were separated on a Halo C18 column using gradient elution of acetonitrile and 0.1% formic acid and detected by tandem mass spectrometry in selected reaction monitoring mode. The standard curve was linear ($r^2$ = 0.998) over the concentration range of 1-100 ng/mL. The lower limit of quantification was 1.0 ng/mL using 50 ${\mu}L$ of plasma sample. The coefficient of variation and relative error for intra- and inter-assay at four QC levels were 1.3 to 8.0% and -5.4 to 10.0%, respectively. This method was applied successfully to the pharmacokinetic study of dioscin after oral administration of dioscin at a dose of 29.2 mg/kg in male Sprague-Dawley rats.

Studies on Protein Profiles and Isozymes in Germinating Seeds (종자발아에 있어서 Protein Profile과 Isozyme에 관한 연구)

  • The purpose of this experiment was to study one side of germination physiology based on that protein profiles and protease relating to protein metabolism, that peroxidase, catalase, $\alpha$-amylase, $\beta$-amylase, and malate dehydrogenase involved in the carbohydrate metabolism of seed germination. All these experiments were divided into the two groups with and without acetone treatment, and were carried out. The protein bands of each germinating stage between the groups treated with and without acetone showed certain basic pattern in polyacrylamide gel disc electrophoresis. However, there was a little difference in the number of protein band, optical density, and migration velocity between two groups. The isozyme bands of peroxidase, and catalase between two groups in polyacrylamide gel disc electrophoresis did not show the numeral difference, but the optical density of certain germinating stage treated with acetone was higher than the group untreated with it and it showed their enzyme activity. The $\alpha$-amylase and $\beta$-amylase activities which involved in starch metabolism of seed germination were higher in the treated group than the other. On one hand, the protease activity of hydrolase occurred in the seeds for germination was also higher, more or less in the treated group than in the other. The isozyme band pattern of malate dehydrogenase in TCA cycle of energy metabolism pathway was very different between two groups growing for 72 hours with and without acetone treatment in cellulose acetate electrophoresis. It indicated that two isozyme bands of malate dehydrogenase was high. Consequently these experimental results mentioned above indicated that acetone treatment before sowing had an effect on dissolving certain complexed lipid substance involved in the seed coats, the activity of carbohydrate hydrolase increased with water absorption which was most comfortable in its germination, dissolved glycerin and fatty acid became certain energy source, and they stimulated the acceleration of respiration metabolism.
