Synthesis, Design and Polymerization of 5-membered exo methylene Cyclic Acetals (이중결합을 가지는 5원환 고리화합물의 합성 및 중합)

  • 5원환 고리아세탈 화합물인 4-methylenes(4,5)을 합성하고 저온조건 하에서 중합반응을 조사한 결과, 선택적인 개환중합이 일어났다. 보통의 양이온 개시제를 사용한 결과 $-78^{\circ}C$에서 가교체가 얻어졌으며, $CH_3SO_3H$를 개시제로 사용하면 같은 조건에서도 주사슬과 곁사슬에 이중결합을 가지는 개환중합체가 얻어진다. 이 중합체는 반응성을 가지는 불포화기를 포함하고 있으므로 prepolymer로써 널리 사용될 수 있다.

Comparison of Photoaddition Reactions of Aromatic Carbonyl Compounds with Silyl Thioketene Acetal vs. Silyl Ketene Acetal

  • Photoaddition reactions of aromatic carbonyl compounds with silyl thioketene acetals have been explored. The results of this study show that the acetonphenone react with dimethyl substituted silyl thioketene acetal competitively via either single electron transfer (SET)-desilylation or [2+2]-cycloaddition pathways to produce b-hydroxyester and oxetanes. In contrast, photochemical reactions of the benzaldehyde with dimethyl substituted silyl thioketene acetal mainly lead to the formation of oxetanes arising by [2+2] cycloaddition. A comparison of the results with those of silyl ketene acetal revealed that replacement of sulfur atom in ${\alpha}$-silyl donor substrate bring about dramatic changes in chemoselectivities as well as excited state reaction mechanism.

  • Acid and base catalyzed intramolecular cyclizations of N-benzoylthioureidoacetal, contain-ing four functional groups adjacent to thiourea such as benzocarbamoyl, acetal, thioure and amide, were investigated. The condensation reaction of N-benzoyl thiocarbamoylgly-cine amide in the presence of 10% aqueous NaOH provided 1-(2,2-dimethoxy)ethyl-imi- dazolidine-2-thione exclusively. In the presence of pyridine, it was transformed to 2- thiohydantoin. N-Benzoyl thiocarbamoyl glycine amide was completely transformed to an iminothiazolidine exclusively in the presence of Lewis acid such as borontrifluoride ether-ate or trimethylsilyl iodide. 1-(2,2-Dimethoxy)ethyl-imidazolidine-2-thione was transformed to imidazole[2,1-b]thiazole and pyrazino[5,1-a]imidazole in the presence of $BF_3$.$ET_2$O and formic acid, respectively.

  • Aliphatic aldehyde polyoxyethylene glycol acetals were synthesised through the reaction of aliphatic aldehydes such as caproic aldehyde, capryl aldehyde, capric aldehyde and lauric aldehyde with excess diethylene glycol, triethylene glycol and tetraethylene glycol, respectively. The acetal formation, in which water was azeotropically distilled by adding benzene to the reaction system, was gained a good yield of acetal type compounds. This reaction is found pseudo first order reaction at various temperatures such 70, 80, 90 and $97^{\circ}C$. Also these activation energies of reaction of acetal type products such as caproic aldehyde diethylene glycol acetal, capryl aldehyde diethylene glycol acetal, capric aldehyde diethylene glycol acetal, lauric aldehyde diethylene glycol acetal, caproic aldehyde triethylene glycol acetal and caproic aldehyde tetraethylene acetal were 17.3, 19.6, 21.2, 21.6, 15.5 and 14.7 Kcal/mole.

Physiological Responses of Oxygen-Tolerant Anaerobic Bifidobacterium longum under Oxygen

  • In order to investigate what kind of response anaerobic bifidobacteria has on oxygen stress, five oxygen-tolerant bifidobacteria were isolated from human fecal samples. All were temporarily identified as Bifidobacterium longum through an analysis of carbohydrate utilization patterns and cellular fatty acid profiles. In the presence of oxygen, the lag phase became extended and the cell growth was suppressed. Bifidobacterial cell was able to remove dissolved oxygen in an early stage of growth and to overcome oxygen stress to a certain extent. The cell became long n size and showed a rough surface containing many nodes which were derived from abnormal or incomplete cell division. Cellular fatty acid profiled changed remarkably under a partially aerobic condition, so that the carbon chain of cellular fatty acid became short. All the dimethyl acetals originated from plasmalogen were reduced, any cyclopropane fatty acid, 9, 10-methyleneoctadecanoic acid ($C_{19:0}cyc9,10$), was increased remarkably. Oxygen stress induced a 5.5 kD protein in B. longum JI 1 of the oxygen-teolerant bifidobacteria, that was named Osp protein, and its N-terminal amino acid sequence was as follows: unknown amino acid-Thr-Gly-Val-Arg-Phe-Ser-Asp-Asp-Glu. Therefore, the oxygen-tolerant bifidobacteria seemed to defend against oxygen stress byincreasing the content of short fatty acid and cyclopropane fatty acid, and induction of an oxygen stress protein, but not the plasmalogen.

  • Acetals of ${\alpha},{\beta}$-unsaturated aldehydes reacted readily with aryl bromides in the presence of palladium catalyst and triethylamine to form aryl conjugated enals. Acrolein diethyl acetal and methacrolein diethyl acetal were reacted with phenyl bromides with substituents such as methyl and isopropyl groups at $100^{\circ}C$. The reaction products yields except the reaction of o-bromotoluene with methacrolein diethyl acetal. The products were identified by proton nuclear magnetic resonance and infrared spectroscopy. In the reverse combination of reactants to prepare aliphatic 2,4-dienals in good yield of above 50%, 3-bromopropenal dimethyl acetal and (E)-3-bromo-2-methylpropenal diethyl acetal were used as vinylic halide reactants and 1-alkenes and ethyl acrylate as olefin reactants.

Changes of Oxidative Enzymes and Fatty Acid Composition of Bifidobacterium adolescentis and B. longum under Anaerobic and Aerated Conditions. (산소의 Stress에 따른 Bifidobacterium adolescentis와 Bifidobacterium longum의 산화효소의 활성과 세포 지방산 조성의 변화)

  • To study the oxygen tolerance mechanism of bifidobacteria, we have studied the growth of cells, the activities of the enzymes which were related with oxygen, such as catalase, superoxide dismutase(SOD), NADH oxidase, and NADH peroxidase, and cellular fatty acid compositions of Bifidobacterium adolescentis and B. longum under anaerobic and aerated (microaerobic and aerobic) conditions. B. longum grew relatively well under the microaerobic conditions, whereas the growth of B. adolescentis was inhibited under the same aerated conditions. B. adolescentis had extremely low level of NADH oxidative enzymes while B. longum had the relatively high level of NADH oxidative enzymes, whose activities were dramatically increased from 3.7 to 11.4 times by microaerobic condition but not in B. adolescentis. The activity of SOD was unexpectedly high in B. adolescentis compared with in B. longum under anaerobic and aerated conditions. The activities of catalase were not detected in all samples tested in this study. We also found that normal $C_{l6:0}$ and $C_{18:1}$ were the major fatty acids in B. adolescentis and B. longum under anaerobic and aerated conditions. 2.2-14.1% $C_{l9:0}$ cyclo fatty acid was detected only in B. longum and the fatty acid was increased by the addition of the aeration. The $C_{l9:0}$ cyclic fatty acid was identified as a cis 9, 10-methylene octadecanoic acid, which was different from lactobacillic acid in the cyclized site. 6.6%-24.6% of dimethyl acetals (DMA) which came from plasmalogen were observed in the B. adolescentis and B. longum grown under anaerobic condition, and the components were notably decreased in the cells grown under the aerated conditions. It is believed that NADH oxidative enzymes play an important role to detoxify oxygen metabolites of Bifidobacteriurn spp. under anaerobic and microaerobic conditions. Independently from oxidative enzymes, it seems that oxygen stress may induce the change of the level of cellular fatty acids showing an increase of $C_{l9:0}$ cyclo in B. longum and a decrease of $C_{l8:1}$ of plasmalogen in B. longum and B. adolescentis to adapt in environment.
