Lipoxygenase gives soybeans their grassy flavor, which can disrupt food processing efficiency. This study aimed to identify soybean genotypes with lipoxygenase deficiency among 1,001 soybean accessions and to develop kompetitive allele specific PCR (KASP) markers that can detect lipoxygenase mutations. Three lipoxygenase isozymes (Lox1, Lox2, and Lox3) were analyzed using a colorimetric assay based on a substrate-enzyme reaction. Among the 1,001 accessions examined, two (IT160160 and IT276392) exhibited a deficiency solely in Lox1, and one (IT269984) lacked both Lox1 and Lox2. IT160160 had a 74-bp deletion in exon 8 of Lox1 (Glyma13g347600), whereas IT276392 displayed a missense mutation involving the change of C to A at position 2,880 of Lox1. Moreover, we successfully developed four KASP markers that specifically target Lox1, Lox2, and Lox3 mutations. To validate the Lox1 KASP markers, we used two F2:3 populations generated through a cross between Daepung 2 (lipoxygenase wild type, maternal parent), IT160160, and IT276392 (null Lox1, paternal parent). The results revealed that the Daepung 2 × IT160160 group followed the expected 3:1 ratio according to Mendel's law, whereas the Daepung 2 × IT276392 group did not. Furthermore, a comparison between the colorimetric and KASP marker analyses results revealed a high agreement rate of 96%. KASP markers offer a distinct advantage by allowing the distinction of heterozygous types independent of other variables. As a result, we present an opportunity to expedite the lipoxygenase-deficient cultivar development.
The evaluation of soybean germplasm has mainly been carried out by morphological characters at Genetic Resources Division, Rural Development Administration (RDA). However, this information has been limited serving a diverse information for user and effectively managing the soybean germplasm. To resolve this problem, soybean collection conserved at RDA gene bank was profiled using nine soybean SSR (Simple Sequence Repeat) markers. Soybean SSR allele was confirmed using genescan and genotyper softwares of automatic sequencer for accurate genotyping of each accession and continuous accumulation of data. SSR profiling of soybean germplasm has been carried out from 2,855 (Satt458) to 4,368 (Satt197) accessions by locus. The number of allele revealed 267 with an average of 29.6 in total accession, and varied from a low of 21 (Satt532 and Satt141) to a high of 58 (Sat_074). Although the number of accessions of wild soybean is less than that of soybean landraces, Korean wild soybean is more variable than other soybean landraces populations in total number of alleles. However, Korean soybean landraces were more variable than Korean wild soybeans in 5 loci. In the allele frequency, wild soybean accessions showed an even distribution in all alleles and higher distribution in low ladder than in high ladder. Also, Korean soybean landraces revealed a high condensed frequency in Satt286 (202 bp, 232 bp), Chinese soybean landraces in Satt197 (171 bp) and Satt458 (173 bp), and Japanese soybean landraces in Sat_074 (244 bp) and Satt458 (170 bp). These SSR profile information will be provided as indications of redundancies or omissions of accessions and can aid in managing soybean collection held at RDA gene bank. The information on diversity analysis could help to enlarge the genetic diversity of materials in breeding program, and could be used to develop a core collection of soybean germplasm.

Total of fifty accessions of Brassica rapa with various morphological characteristics were used for production of double haploid plants though microspore culture in Brassica rapa. Among them, only 30 accessions induced embryos from microspores. The highest efficiency of embryo induction of 1.194 per bud was obtained from IT135449 of turnip type, while 3 accessions of sarson (winter oil) type did not generate embryo. The effect of heat shock periods for embryogenesis was also investigated with 4 accessions (IT135449; Turnip type, IT199710; Chinese cabbage type, IT212886; Pak choi type, IT218043; Summer oil type). The high productions of embryos were observed in IT135449, IT199710 and IT212886 when microspores were pre-cultured to $32^{\circ}C$ for 2 days. In IT218043, high embryogenesis was observed at the 3 days of heat shock treatment. The optimal condition of shoot regeneration for IT199710 was observed in MS medium supplemented with NAA $0.5mg{\cdot}L^{-1}$ and BAP $1mg{\cdot}L^{-1}$. In contrast, the IT135449 and IT212886 were observed high regeneration frequency in MS medium without plant growth regulators. All the plantlets regenerated from microspore-derived embryos have been successfully transplanted to soil, and bud self-pollinated seeds were produced from doubled haploid plants. This indicated that double-haploid genotype was likely generated naturally during embryogenesis process.
The objective of this research was to develop Near-Infrared Reflectance Spectroscopy (NIRS) model for amylose and protein contents analysis of large accessions of rice germplasm. A total of 511 accessions of rice germplasm were obtained from National Agrobiodiversity Center to make calibration equation. The accessions were measured by NIRS for both brown and milled brown rice which was additionally assayed by iodine and Kjeldahl method for amylose and crude protein contents. The range of amylose and protein content in milled brown rice were 6.15-32.25% and 4.72-14.81%, respectively. The correlation coefficient ($R^2$), standard error of calibration (SEC) and slope of brown rice were 0.906, 1.741, 0.995 in amylose and 0.941, 0.276, 1.011 in protein, respectively, whereas $R^2$, SEC and slope of milled brown rice values were 0.956, 1.159, 1.001 in amylose and 0.982, 0.164, 1.003 in protein, respectively. Validation results of this NIRS equation showed a high coefficient determination in prediction for amylose (0.962) and protein (0.986), and also low standard error in prediction (SEP) for amylose (2.349) and protein (0.415). These results suggest that NIRS equation model should be practically applied for determination of amylose and crude protein contents in large accessions of rice germplasm.

The interest in mini watermelon (Citrullus lanatus) with small fruits weighing 2-3 kg has increased by the increasing trend in one-person households and consequent tendency to consume small meals. Watermelon grafting onto cucurbit rootstocks is a very effective way to control soil-borne diseases, such as Fusarium wilt; however, this practice negatively impacts the fruit quality. This study was conducted to investigate the growth, fruit set, and fruit quality of mini watermelon grafted onto wild watermelon accessions (Citrullus spp.) resistant to Fusarium wilt. Five watermelon accessions (Galactica, IT 208441, PI 482322, PI 500303, and PI 593358) were evaluated as rootstocks for the mini watermelon "Ministar". Non-grafted "Ministar" and "Ministar" grafted onto "Shintozwa" (Cucurbita maxima D. C. moschata D.) or "Bullojangsaeng" (Lagenaria leucantha) were used as controls. The roots of the transplants grafted onto "PI 593358" and "Shintozwa" weighed more than those on other rootstocks. Additionally, the transplants on "PI 593358" showed better growth and fruit set in the field than the other transplants. However, the total soluble solid contents and fruit quality indices of the transplants on "PI 593358" and "Shintozwa" were lower, whereas the total fruit quality index of those on "PI 482322" was higher. Thus, the wild watermelon accessions tested can potentially be used as basic germplasm for developing watermelon rootstocks instead of cucurbit rootstocks. The most promising accession for this purpose was found to be "PI 482322".
Phylogenetic relationships among the Korean taxa of the genus Allium subgenus Rhizirideum and some related taxa were assessed on the basis of in sequences of nuclear ribosomal DNA. Twenty-eight accessions of the genus Allium L. consisting of subgenera Rhizirideum (19 taxa), Allium (5 taxa) and Amerallium (one taxon) were analyzed. The variation in the ITS region was informative at the levels of section except for sect. Reticulato- bulbosa which is known to be of multiple origin. The ITS 2 region was longer than the ITS 1 region, and all of the investigated Allium taxa were the same in length in the 5.8S region except for A. monanthum. Allium cyaneum var. cyaneum was the shortest (635 bp) and A. victorialis the longest (646 bp) among the investigated Korean taxa. The three morphologically similar taxa, A. thunbergii, A. sacculiferum that has been included in A. thunbergii, and A. deltoid- fistulosum, had the same ITS lengths of 641 bp, but were clearly distinguished in the phylogenetic analysis of their ITS sequences.
Gnetum (Gnetaceae, Gnetales) is a gymnosperm genus with ca. 35 species distributed in tropical forests around the world. Due to its dioecious habit and lack of diagnostic characters from vegetative tissue, the identification of Gnetum species is not easy without seeds or reproductive structures. To identify and verify their phylogenetic positions, we applied DNA barcoding to Cambodian Gnetum collections gathered between 2010 and 2015, with previously designed cp matK gene primers. We newly sequenced partial matK sequences from 72 Gnetum collections, 43 out of 72 from Cambodia, and analyzed 115 Gnetum accessions using the neighbor-joining method. The resulting neighbor-joining tree categorized Cambodian Gnetum samples into three clades of species: G. macrostachyum, G. montanum, and G. aff. gracilipes. The recognition of G. aff. gracilipes in Cambodia is reported here for the first time. Taxonomic information for the three recognized Cambodian Gnetum species is provided and the benefits of the taxonomic reevaluation assisted by DNA barcoding are emphasized in this work.
Volatile oil components were analysed in Perilla frutescens accessions collected from different regions in South Korea and identified chemotypes based on the major volatile oil components. Major components out of 30 compounds identified were limonene, perillaldehyde, perillaketone, isoegomaketone, beta-caryophyllene, beta-farnesene, myristicin, and dillapiole. P. frotescens collections were classified into four chemotypes : PA type (57.7% limonene and 19.8% perillaldehyde), PK type (89.8% perillaketone), ST type (82.4% sesquiterpene, as 54.5% beta-caryophyllene and 27.9% beta-farnesene) and PP type (40.3% phenylpropenes as 13.6% myristicin and 26.7% dillapiole) and 37.8% sesquiterpenes. The majorities of P. frutescens collections in this study belong to PA type (41.9%) and PK type(38.8%).
