차량 가속특성에 따른 이산화탄소(CO2) 배출량 비교 (CO2 Emission Considering Condition of Vehicle Acceleration)

  • PURPOSES : The present study aims to evaluate the added $CO_2$ emissions incurred from accelerating operation when to increase the speed up to the allowed level. METHODS : The methodology used are basically the relationship between emission rates and vehicle speeds or acceleration rate. These rates together are used to calculate the added $CO_2$ emissions incurred from accelerating operation. RESULTS : It was resulted that the all the emission rates are increasing proportionally to vehicle speeds or acceleration rates. Additionally, it was also resulted that allowable speeds increasing, the added emission rates are increasing rapidly. CONCLUSIONS : It may be concluded that if the allowable speed ranges are managed, $CO_2$ emissions during vehicle operation are much reduced. From this reason, it was found that the allowable speed during highway design and operation would be much necessary

  • This paper presents an accelerated life test for burnout of tungsten filament of incandescent lamp. From failure analyses of field samples, it is shown that their root causes are local heating or hot sports in the filament caused by tungsten evaporation and wire sag. Finite element analysis is performed to evaluate the effect of vibration and impact for burnout, but any points of stress concentration or structural weakness are not found in the sample. To estimate the burnout life of lamp, an accelerated life test is planned by using quality function deployment and fractional factorial design, where voltage, vibration, and temperature are selected as accelerating variables. We assumed that Weibull lifetime distribution and a generalized linear model of life-stress relationship hold through goodness of fit test and test for common shape parameter of the distribution. Using accelerated life testing software, we estimated the common shape parameter of Weibull distribution, life-stress relationship, and accelerating factor.

전기철도용 견인전동기의 복합가속열화 상태진단에 관한 연구 (A Study on the Complex Accelerating Degradation and Condition Diagnosis of Traction Motor for Electric Railway)

  • In this study, the stator form-winding sample coils based on silicone resin and polyimide were made for fault prediction and reliability estimation on the C-Class(200$\^{C}$ ) insulation system of traction motors. The complex accelerative degradation was periodically performed during 10 cycles, which was composed of thermal stress, fast rising surge voltage, vibration, water immersion and overvoltage applying. After aging of 10 cycles, the condition diagnosis test such as insulation resistance '||'&'||' polarization index, capacitance '||'&'||' dielectric loss and partial discharge properties were investigated in the temperature range of 20 ∼ 160$\^{C}$. Relationship among condition diagnosis tests was analyzed to find a dominative degradation factor and an insulation state at end-life point.

  • zymolyase(${\beta}-1$, 3-glucanase)의 효모(酵母) 세포벽(細胞壁)에 대한 용해작용(溶解作用)을 촉진(促進)시키는 인자(因子)를 zymolyase의 조효소(粗酵素)로부터 분리(分離)하여 Sephadex G-75 gel 여과(濾過) 및 조제용(調製用) PAG-전기영동법(電氣泳動法)에 의하여 전기영동적(電氣泳動的)으로 균일(均一)하게 정제(精製)하였다. SDS-PAG 전기영동법(電氣泳動法)에 의하여 측정한 정제(精製) 인자(因子)의 분자량(分子量)은 40,500이었고, 등전점(等電點)은 pH 9.6이었다. 정제(精製)된 촉진인자(促進因子)는 395개(個)의 아미노산 잔기(殘基)로 구성(構成)되는 단일(單一) polypeptide의 염기성(鹽基性) protease임이 밝혀졌으며, 이 protease의 흡광계수(吸光係數)($E_{208,cm}^{1%}$)는 11.9, 분자흡광계수(分子吸光係數)(${\varepsilon}_{280,}mole/l$)는 $4.83{\times}10^4$이었다.

  • 동물의 장기를 인간에게 이식하게 되면 초급성거부반응(Hyperacute rejection, HAR)이 일어난다. 초급성거부반응은 면역계의 구성요소 중 보체(complement)에 의해 일어나는 거부반응으로 돼지의 혈관세포 표면에 있는 $Gal{\alpha}$(1,3)Gal 당분자에 인간의 항체가 즉각 반응하기 때문에 일어나며, ${\alpha}1,3$-galactosyltransferase(${\alpha}1,3$-GT) 유전자는 돼지 혈관세포 표면의 $Gal{\alpha}$(1,3)Gal 당분자 생성에 관여한다. 따라서 인간에게 돼지의 장기를 이식하기 위해서는 ${\alpha}1,3$-galactosyltransferase 유전자를 제거하는 것이 필요한 것으로 알려져 있다. 본 연구실의 이전 연구에서, 시카고 미니돼지 귀체세포에서 상동 재조합(Homologous recombination)을 통해 ${\alpha}1,3$-galactosyltransferase 유전자가 제거된 체세포를 개발한 바 있으며, 이 체세포를 통하여 ${\alpha}1,3$-GT 유전자가 제거된 돼지도 생산된 바 있다. 본 연구에서는, human serum 처리 시 돼지 세포를 보호해 준다고 보고되고 있는 human complement regulator인 human Decay-accelerating factor(hDAF)와 human ${\alpha}1,2$-fucosyltransferase(hHT)유전자를 ${\alpha}1,3$-GT 유전자 위치에 gene targeting하여 동시에 hDAF와 hHT가 발현하는 체세포를 개발하였다. Knock-in vector는 hDAF와 hHT 두 유전자가 발현할 수 있도록 IRES로 연결하였으며, ${\alpha}1,3$-GT 유전자의 start codon을 이용하여 발현할 수 있도록 구축하였다. 구축한 vector는 electroporation을 통해 미니 돼지 체세포에 도입하였으며, PCR 결과, ${\alpha}1,3$-GT 유전자 위치에서 상동 재조합이 일어났음을 확인하였다. Positive-negative 선별 방법을 통해 얻은 gene targeting 된 체세포는 RT-PCR에 의해 hDAF와 hHT 유전자의 발현이 확인되었으며, 대조군(NIH minipig)에 비해 ${\alpha}1,3$-GT 유전자의 발현이 감소하였다. 또한 이들 세포에 100% human complement serum을 처리하였을 때 knock-in 세포가 대조군에 비해 30% 정도 더 높은 생존율을 보였다. 따라서 개발된 체세포는 이종간 장기이식을 위한 돼지 생산과 함께 이를 이용한 이종간의 장기 이식 시 초급성 거부반응을 억제하는 데 사용될 수 있을 것으로 생각된다.

Decay-Accelerating Factor Differentially Associates With Complement-Mediated Damage in Synovium After Meniscus Tear as Compared to Anterior Cruciate Ligament Injury

  • We have reported that anterior cruciate ligament (ACL) injury leads to the differential dysregulation of the complement system in the synovium as compared to meniscus tear (MT) and proposed this as a mechanism for a greater post-injury prevalence of post traumatic osteoarthritis (PTOA). To explore additional roles of complement proteins and regulators, we determined the presence of decay-accelerating factor (DAF), C5b, and membrane attack complexes (MACs, C5b-9) in discarded surgical synovial tissue (DSST) collected during arthroscopic ACL reconstructive surgery, MT-related meniscectomy, osteoarthritis (OA)-related knee replacement surgery and normal controls. Multiplexed immunohistochemistry was used to detect and quantify complement proteins. To explore the involvement of body mass index (BMI), after these 2 injuries, we examined correlations among DAF, C5b, MAC and BMI. Using these approaches, we found that synovial cells after ACL injury expressed a significantly lower level of DAF as compared to MT (p<0.049). In contrast, C5b staining synovial cells were significantly higher after ACL injury (p<0.0009) and in OA DSST (p<0.039) compared to MT. Interestingly, there were significantly positive correlations between DAF & C5b (r=0.75, p<0.018) and DAF & C5b (r=0.64 p<0.022) after ACL injury and MT, respectively. The data support that DAF, which should normally dampen C5b deposition due to its regulatory activities on C3/C5 convertases, does not appear to exhibit that function in inflamed synovia following either ACL injury or MT. Ineffective DAF regulation may be an additional mechanism by which relatively uncontrolled complement activation damages tissue in these injury states.

Monitoring of Deforestation Rate and Trend in Sabah between 1990 and 2008 Using Multitemporal Landsat Data

  • Deforestation is a major and very critical problem faced by many tropical countries including Malaysia. Sabah is the second largest state in Malaysia and its deforestation rate has been accelerating. This study was conducted to monitor the deforestation in Sabah in the last two decades with Landsat images of 1990, 2000 and 2008. Supervised classification with maximum likelihood algorithm was conducted using the Landsat data for monitoring deforestation. In total, between 1990 and 2008, Sabah lost half of its intact forest, or more than 1.85 million ha in less than two decades. Overall, the deforestation rate for all forest types combined for the last two decades was 1.6% per year. Deforestation seemed to be accelerating because the deforestation rate between 1990 and 2000 was 0.9% per year and it had increased to 2.7% per year between 2000 and 2008. The deforestation trend seemed to follow a negative exponential from 1990 to 2008. In contrast, the agricultural areas increased rapidly with a total of increment more than 1 million ha. This confirmed that agriculture especially establishment of commercial plantation was the major factor of deforestation in Sabah for the last two decades.

Methylthioisocyanate 합성반응 공정의 열적위험 특성 (Characteristics of Thermal Hazard in Methylthioisocyanate Synthesis Reaction Process)

  • Compared to a batch reactor, where all reactants are initially charged to the reactor, the semi-batch reactor presents serious advantages. The feed of at least one of the reactants provides an additional way of controlling the reaction course, which represents a safety factor and increases the constancy of the product quality. The aim of this study was to investigate the characteristics of thermal hazard such as a feed time, catalysis concentration and solvent concentration in methylthioisocyanate(MTI) synthesis reaction process. The experiments were carried out by the Multimax reactor system and Accelerating rate calorimeter(ARC). The MTI synthesis reaction process has many reaction factors and complicated reaction mechanism of multiphase reaction. Through this study, we can use as a tool for assessment of thermal hazard of other reaction processes by applying experiment method provided.
