  • Some pharmacological activities of acanthoic acid, isolated from Acanthopanax koreanum root was investigated in animals. It is revealed that the compound had analgesic and anti-inflammatory activities without actions on central nervous system and showed inhibition of lipid peroxidation. The anti-inflammatory activity might be related to inhibition of prostaglandin E$_2$ synthesis in exudates of inflammation.

남부오가피잎의 성분 및 정량 (Constituents and Quantitative analysis from the Leaves of Acanthopanax divaricatus f. nambunensis)

  • From the water fraction of the MeOH extract, three compounds, 1,3,4,5-terrahydroxycyclo-hexanecarboxylic acid 3-(3,4-dihydroxycinnamate) (chlorogenic acid), $quercetin 3-O-{\beta}-D-galactopyranoside(hyperoside)$, and 1 (R)-hydroxy-3,4-seco-lup-4(23), 20(30)-dien-3,$11{\alpha}-olactone-{\alpha}-L-rhamnopyrnaosy(1{\rightarrow}4)-{\beta}-D-glucopyanosy(1{\rightarrow}6)-{\beta}-L-glucopyrnaosyl$ ester (chiisanoside) were isolated and their strutures determinated by $^1H-NMR,{\;}^{13}C-NMR$, IR, and FAB-Mass. Chlorogenic acid and Chiisanoside had been quantitated by HPLC from eight Acanthopanax species per 10 g A. koreanum 19.82, 4.17 mg, A. nambunensis 65.00, 1.86mg, A. chiisanense 27.19, 8.17mg, A. sessiliflorum 7.49, 5.88 mg.

  • We investigated the mechanism of immunomodulatory action of polysaccharide isolated from cell culture of Acanthopanax senticosus (ASP). ASP was found to increase directly proliferation and differentiation of B cells, and cytokine production of macrophages, but not proliferation and cytokine production of T cells.(omitted)

  • Many polysaccharides isolated from plants are considered to be biological response modifiers and have been shown to enhance various immune responses in vivo and in vitro. Here we demonstrate that a polysaccharide isolated from cell culture of Acanthopanax senticosus (AS) has a unique mode of immunostimulation with regard to its cell-type specificity. AS was found to markedly increase polyclonal IgM antibody production and the proliferation of B cells. and to activate iNOS transcription and NO production in macrophages. (omitted)

Analysis of Flavonoid Contents in the Fruits of Acanthopanax Species using HPLC

  • Analysis of flavonoid contents in the fruits of Acanthopanax species (A. chiisanensis, A. divaricatus, A. koreanum, A. senticosus, and A. sessiliflorus) was conducted by high performance liquid chromatography. A Discovery$^{(R)}$ C18 ($4.6{\times}250$ mm, 5 ${\mu}m$) column was used with a gradient mobile phase of water and acetonitrile (90 : 10 to 60 : 40 for 60 min) and UV detection was conducted at 350 nm. The contents of rutin, hyperin, quercetin, afzelin, and kaempferol were 0.063~0.540, 0.494~7.480, 0.584~0.704, 0.388~0.567, 0.190~0.471 mg/g, respectively, in the fruits of Acanthopanax species. Total content of flavonoids in the fruits of Acanthopanax species was highest in those of A. chiisanensis. Furthermore, hyperin was the most abundant compound in the fruits of Acanthopanax species. Consequently, our results demonstrate that the fruits of Acanthopanax species containing flavonoids have promising potential as a new income source of agriculture and industry in medicinal natural products, health supplements, and beverages.

Inhibition of Lung Inflammation by Acanthopanax divaricatus var. Albeofructus and Its Constituents

  • In order to find potential therapeutic agents on lung inflammatory conditions, the extracts of Acanthopanax divaricatus var. albeofructus were prepared and its constituents were isolated. They include lignans such as (+)-syringaresinol (1), acanthoside B (2), salvadoraside (3) and acanthoside D (4), lariciresinol-9-O-${\beta}$-D-glucopyranoside (5) and phenylpropanoids such as 4-[(1E)-3-methoxy-1-propenyl]phenol (6), coniferin (7), and methyl caffeate (8). The extracts and several constituents such as compound 1, 6 and 8 inhibited the production of inflammatory markers, IL-6 and nitric oxide, from IL-$1{\beta}$-treated lung epithelial cells and lipopolysaccharide (LPS)-treated alveolar macrophages. Furthermore, the extracts and compound 4 significantly inhibited lung inflammation in lipolysaccharide-treated acute lung injury in mice by oral administration. Thus it is suggested that A. divaricatus var. albeofructus and its several constituents may be effective against lung inflammation.

오갈피나무의 계통별, 부위별, 추출용매별 생리활성물질 분석 및 기능적 특성 (Functional Properties and Biological Activity of Breeding Lines, Parts, and Various solvents from Acanthopanax)

  • This study was evaluated to biological activity of breeding lines from Acantopanax( A. sessiliflorus: ASF, A. koreanum: AKN, A. chiicanensis: ACS, A. senticosus: AST) and parts(root, stem, fruit, and leaf) and various extracted solvents( 100% water, 100% EtOH, 50% EtOH). Total polyphenol content of AKN root in 100% water extracts was high detected 464.46 mg/100 g. Total flavonoid content in the leaf was significantly higher than in other parts. The content of total sugars was high in the 50% EtOH extracts and fruit. The major free amino acids were arginine in all extracts. The content of arginine was detected in the root of AKN(1.807 mg/100 mg). Contents of eleutheroside B, E were high detected in 100% water extracts. Antioxidative capacity in the leaves of AKN was the higher than other extracts($EC_{50}=84.8{\mu}g/mL$). The results would be useful for understanding of the physiological properties of AKN extracts.

오가피(五加皮) Ethanol Extract의 실험적(實驗的) 신성고혈압(腎性高血壓)에 미치는 영향(影響) (Effect of Acanthopanax Ethanol Extract on Experimental Renal Hypertension in Rats)

  • This study was carried out for the purpose of observing the effect of Korean Acanthopanax Radicis Cortex on renal hypertension and to clarify the mechanism of this effect, making use of its ethanol extract. Adult male or female rats, weighing 180-250g, were divided into 3 groups; the first for normotensive control, the second for hypertensive control and the third for hypertensive Acantopanax-treatment. Rats in the normotensive and hypertensive control group were administered 0.9% saline subcutaneously only, whereas those in the Acanthopanax-treated hypertensive group were administered 50mg/kg Acanthopanax ethanol extract subcutanously once a day. Changes of original blood pressure, and responses of blood pressure to various 4gents(norepinephrine, angiotensin, acetylcholine, serotonin and histamine) were recorded for each group on the initial, 18th, 32nd and 46th days of the experiment. The results obtained in this experiment are as follows: 1) The initial blood pressure was $102.6{\pm}7.6mmHg$ on the average. The blood pressures of the normotensive control group were not observed to alter significantly at any period in the course of the experiment. 2) The mean blood pressures in the hypertensive control group were recorded at $120.3{\pm}10.4mmHg$ on the 18th day, at $134.5{\pm}9.2$ on the 32nd day and at $138.8{\pm}8.3$ on the 46th day, thus revealing significant elevation in comparison with the corresponding normotensive control group blood pressures. On the other hand, the mean blood pressures in Acanthopanax-treated hypertensive group on the 18th, 32nd and 46th days were $118.3{\pm}9.7,\;129.9{\pm}8.3\;and\;120.2{\pm}8.3mmHg$ respectively. The blood pressures of the hypertensive-Acanthopanax group recorded. on the 46th day revealed a significant difference as compared with those of the corresponding hypertensive control group. 3) On the 46th day of this experiment, the responses of blood pressure to acetylcholine in the hypertensive-Acanthopanax group were suppressed significantly as compared with those of the hypertensive control group, and in the latter group, angiotensin was decreased markedly as compared with the corresponding normotensive control group. In contrast, pressor action of norepinephrine and depressor action of serotonin and histamine did not differ significantly among the three groups. These results suggest that Acanthopanax ethanol extract suppresses the induction of renal hypertension by means of a cholinergic action such as that caused by acetylcholine.
