Load Fidelity Improvement of Piecewise Integrated Composite Beam by Construction Training Data of k-NN Classification Model (k-NN 분류 모델의 학습 데이터 구성에 따른 PIC 보의 하중 충실도 향상에 관한 연구)

  • Piecewise Integrated Composite (PIC) beam is composed of different stacking against loading type depending upon location. The aim of current study is to assign robust stacking sequences against external loading to every corresponding part of the PIC beam based on the value of stress triaxiality at generated reference points using the k-NN (k-Nearest Neighbor) classification, which is one of representative machine learning techniques, in order to excellent superior bending characteristics. The stress triaxiality at reference points is obtained by three-point bending analysis of the Al beam with training data categorizing the type of external loading, i.e., tension, compression or shear. Loading types of each plane of the beam were classified by independent plane scheme as well as total beam scheme. Also, loading fidelities were calibrated for each case with the variation of hyper-parameters. Most effective stacking sequences were mapped into the PIC beam based on the k-NN classification model with the highest loading fidelity. FE analysis result shows the PIC beam has superior external loading resistance and energy absorption compared to conventional beam.

Antioxidative Characteristics of Dihydroxyphenylalanine, Melanin and Enzymatic Browning Reaction Products of Tyrosine in a Model System (Dihydroxyphenylalanine, Melanin 및 Tyrosine의 효소적 산화반응생성물질의 항산화 특성)

  • Antioxidative characteristics of dihydroxyphenylalalnine(DOPA), melanin and enzymatic oxidation products of tyrosine(EOPTs) were studied in a model system. EOPTs were prepared by the tyrosine-tyrosinase reaction at pH 6.5 and $25^{\circ}C$ at various time intervals(0~120min). All EOPTs were brown in varied intensities with increased absorption at 200~210, 280, 310~320nm, and 450~490nm. EOPTs obtaiend at the early stage of the reaction(1~3min especially) showed a higher antioxidative activity than those from the later stage on the inhibition of peroxide, conjugated dienoic acid and malonaldehyde formations in linoleic acid autoxidation. Additionally among the substances of tyrosine, DOPA and melanin, DOPA showed the highest antioxidative activity while that of tyrosine was the lowest during the linoleic acid autooxidation. It was observed that DOPA and melanin had the ability of free radical scavenging, which may party contribute to their antioxidative activity.

Crystallographic and Magnetic Properties of Li0.5Fe2.5-χRhχO4 by Using Applied Field Mossbauer Spectrometer (외부자기장 뫼스바우어 분광기를 이용한 Li0.5Fe2.5-χRhχO4의 자기적 성질과 결정학적 구조에 관한 연구)

  • L $i_{0.5}$F $e_{2.5-{\chi}}$R $h_{\chi}$ $O_4$ ($\chi$ = 0.25, 0.50, 0.75, 1.00) has been prepared by solid state reaction. Crystallographic and magnetic properties were investigated by Mossbauer spectroscopy, SQUID magnetometry, and x-ray diffraction. The crystal structure is found to be a cubic spinel structure with space group Fd3m for all the samples. The lattice constant $a_{0}$ increases from 8.3365 $\AA$ to 8.3932 $\AA$ with increasing Rh concentration $\chi$. The migration of Li ion has been confirmed by x-ray patterns and the results of applied field Mossbauer analysis. The temperature dependence of the absorption area of each site was analyzed with the Debye model for the recoil-free fraction. The Debye temperature for the octahedral sites is almost as large as for the tetrahedral sites, thereby suggesting similar inter-atomic binding forces for the octahedral and the tetrahedral sites. The saturated magnetic moment and the Mossbauer spectra taken at 4.2 K under the applied field (6 T) show that the spin structure of L $i_{0.5}$F $e_{2.5-{\chi}}$R $h_{\chi}$ $O_4$ is compatible with the collinear Neel Model.

Simulating Carbon Storage Dynamics of Trees on the Artificial Ground (시뮬레이션을 통한 인공지반 교목의 탄소저장량 변화)

  • To successfully create a low-carbon landscape in order to become a low-carbon city, it is necessary to understand the dynamics of artificial greening's resources on a multi-scale. Additionally, the effects of carbon storage should be quantitatively evaluated. The purpose of this study is to simulate and evaluate the changes in carbon storages of artificial ground trees using system dynamics throughout a long-term period. The process consisted of analyzing the dynamics of the multi-scale carbon cycle by using a casual loop diagram as well as simulating carbon storage changes in the green roof of the Gangnam-gu office building in 2008, 2018, 2028, and 2038. Results of the study are as follows. First, the causal loop diagram representing the relationship between the carbon storage of the artificial ground trees and the urban carbon cycle demonstrates that the carbon storage of the trees possess mutual cross-scale dynamics. Second, the main variables for the simulation model collected 'Biomass,' 'Carbon storage,' 'Dead organic matter,' and 'Carbon absorption,'and validated a high coefficient of determination, the value being ($R^2$=0.725, p<0.05). Third, as a result of the simulation model, we found that the variation in ranking of tree species was changing over time. This study also suggested the specific species of tree-such as Acer palmatum var. amoenum, Pinus densiflora, and Betula platyphylla-are used to improve the carbon storage in the green roof of the Gangnam-gu office building. This study can help contribute to developing quantitative and scientific criteria when designing, managing, and developing programs on low-carbon landscapes.

Adsorption Characteristic of Brownish Dark Colored Compounds from the Hot Water Extract of Auricularia auricula Fruit Body (흑목이 버섯 자실체의 열수추출물로부터 흑갈색 색소 성분의 흡착 특성)

  • The crude polysaccharide fraction from fruit body of Auricularia auricula were obtained by using hot water extraction and ethanol precipitation. As the crude polysaccharide fraction contained the brownish dark colored compounds, the adsorption study of pigments from the crude polysaccharide using activated carbon was carried out. The pigment compounds showed an absorption characteristic with $\lambda_{max}$ of 230 nm and the absorbance at 230 nm was taken as color intensity. Adsorption capacity of pigment depended on increase of the activated carbon to sample loading ratio. The adsorption capacity increased with increase of pH and temperature in the pH range of 3.0-7.0 and temperature range of 25-40$^{\circ}C$, but decreased in the temperature range of 40-70$^{\circ}C$. The optimum capacity was obtained at addition of 16.7 mg activated carbon per mL sample solution (concentration = 3 mg/mL) at pH of 7.0 and temperature of 40$^{\circ}C$. Treatment for 10 min was sufficient to achieve the 80% decolorization and 1.25 fold purification of polysaccharide. Langmuir isotherm and pseudo second-order kinetic model provided the best fitting for adsorption of the brownish dark colored compounds onto powdered active carbon. The activation energies of adsorption from the Langmuir isotherm parameter in the ranges of 25-40$^{\circ}C$ and 40-70$^{\circ}C$ was -2.54 and 4.38 kcal/g, respectively. The results of low activation energy also indicated that the adsorption process was a physical adsorption which was controlled by diffnsion.

GEMS BrO Retrieval Sensitivity Test Using a Radiative Transfer Model (복사전달모델을 이용한 GEMS 일산화브로민 산출 민감도 시험)

  • To estimate errors in GEMS retrievals for bromine monoxide (BrO) total vertical column densities(VCDs), we perform a sensitivity test using synthetic spectra generated by a radiative transfer model. Hourly synthetic data are produced for 00-07 UTC on the first day of every month in Jul 2013- Jun 2014. Solution errors estimated by the optimal estimation method tend to decrease with increasing air mass factors (AMFs) but increase when AMFs are larger than 5. Interference errors induced by formaldehyde (HCHO) absorption appear to be larger with smaller BrO AMFs. Total BrO retrieval errors estimated by combining solution and interference errors show an average of 26.74±30.18% for all data samples and 60.39±133.78% for those with solar zenith angles higher than 80°. Due to interfering spectral features and measurement errors not considered in thisstudy, errorsin BrO retrievals from actual GEMS measurements may have different magnitudes from our estimates. However, the variability of errors assessed in this study is still expected to appear in the actual BrO retrievals.

Anti-obesity Effects and Safety of the Mixture of Herbal Extracts in 3T3-L1 Cells and HR-1 Mice Fed a High Fat Diet (3T3-L1 지방전구세포와 고지방 식이로 유도된 비만 HR-1 마우스 피부에 도포한 한약 추출 복합물의 항비만 효과 및 안전성 평가)

  • The aim of this study was to investigate whether a novel formulation of an herbal extracts has an inhibitory effect on obesity. To determine its anti-obesity effects, we performed anti-obesity-related experiments in vitro and in vivo. Thus, our present study was carried out to evaluate the anti-obesity effect of herbal extracts using a high fat diet (HFD)-induced obese mouse model and 3T3-L1 adipose cells. The effects of each herbal extracts on lipid accumulation in 3T3-L1 cells were examined using Oil Red O staining. Results showed that treatment with each herbal extracts at $10{\sim}100{\mu}g/ml$ had no effect on cell morphology and viability. Without evidence of toxicity, herbal extracts treatment decreased lipid accumulation compared with the untreated adipocytes controls as shown by the lower absorbance of Oil Red O stain. Futhermore, compared with control-differentiated mature adipocytes, each herbal extracts significantly inhibited lipid accumulation in mature 3T3-L1 adipocytes. In the HFD-fed obese mice, body weight, liver weight and white adipose tissue weights were significantly reduced by mixture of herbal extracts administration in mouse skin. Futhermore, we found that mixture of herbal extracts administration suppressed serum triglyceride (TG), and total cholesterol (TCHO) in HFD-induced obese mouse model. The mixture of herbal extracts of permeability was estimated by measuring the transepithelial electrical resistance (TEER) value in pig skin. The optimized formulations of herbal extracts (Test 3 formulation) showed skin permeation. However, test 1 formulation containing essential oil as enhancer showed maximum skin permeation. After confirming the enhanced skin permeability, in vivo studies were performed to assess whether skin irritation potential on the basis of a primary irritation index (PII) in rabbit skin. Reactions were scored for erythema/edema reactions at 24 h, 48 h and 72 h post-application. It was concluded that the test 1 formulation was not irritation (PII = 0). The present study suggests that the test 1 formulation might be of therapeutic interest with respect to the treatment of obesity.

Effects of Anticancer Drug Delivery based on Microbubble and Microbubble-Nanoparticle Complex using Low-Intensity Focused Ultrasound in Breast Cancer Animal Model (유방암 동물모델에서의 저강도 집속초음파를 이용한 마이크로버블 및 마이크로버블-나노물질 복합체 기반 항암제 전달 효율 검증)

  • Ultrasound sonication along with microbubble (MB) could enhance drug delivery to promote the absorption of anticancer drugs into cancers in a noninvasive and targeted manners. In this study, we verify the acute drug delivery enhancement (within an hour) of two representative focused ultrasound driven drug delivery enhancement methods (MB and Doxorubicin-coated Nanoparticle complex (MB-NP) based). Experiments were conducted using in vivo mouse model with MDA-MB-231 breast cancer cell line. Ultrasound generated by single-element 1 MHz focused ultrasound transducer was delivered in pulsed sonication consisted of 0.125 msec bursts at a pulse repetition frequency of 2 Hz for 20 seconds without a significant increase in local temperature (less than $0.1^{\circ}C$) or hemorrhage. Doxorubicin concentrations in tumors were improved by 1.97 times in the case of MB-NP, and 1.98 times by using Doxorubicin and MB separately. These results indicate anticancer drug delivery based on MB and MB-NP can significantly improve the effect of anticancer drugs delivered to tumors in a short time period by using low-intensity focused ultrasound.

Anti-aging effects of Korean Red Ginseng (KRG) in differentiated embryo chondrocyte (DEC) knockout mice

  • Background: The circadian rhythm is the internal clock that controls sleep-wake cycles, metabolism, cognition, and several processes in the body, and its disruption has been associated with aging. The differentiated embryo chondrocyte (Dec) gene is related to circadian rhythm. To our knowledge, there are no reports of the relationship between dec gene expression and KRG effect. Therefore, we treated Dec gene knockout (KO) aging mice with KRG to study anti-aging related effects and possible mechanisms. Methods: We evaluated KRG and expression of Dec genes in an ototoxicity model. Dec genes expression in livers of aging mice was further analyzed. Then, we assessed the effects of DEC KO on hearing function in mice by ABR. Finally, we performed DNA microarray to identify KRG-related gene expression changes in mouse liver and assessed the results using KEGG analysis. Results: KRG decreased the expression of Dec genes in ototoxicity model, which may contribute to its anti-aging efficacy. Moreover, KRG suppressed Dec genes expression in liver of wild type indicating inhibition of senescence. ABR test indicated that KRG improved auditory function in aging mouse, demonstrating KRG efficacy on aging related diseases. Conclusion: Finally, in KEGG analysis of 238 genes that were activated and 158 that were inhibited by KRG in DEC KO mice, activated genes were involved in proliferation signaling, mineral absorption, and PPAR signaling whereas the inhibited genes were involved in arachidonic acid metabolism and peroxisomes. Our data indicate that inhibition of senescence-related Dec genes may explain the anti-aging efficacy of KRG.

Comparative analysis of the in vivo kinetic properties of various bone substitutes filled into a peri-implant canine defect model

  • Purpose: Deproteinized bovine bone or synthetic hydroxyapatite are 2 prevalent bone grafting materials used in the clinical treatment of peri-implant bone defects. However, the differences in bone formation among these materials remain unclear. This study evaluated osteogenesis kinetics in peri-implant defects using 2 types of deproteinized bovine bone (Bio-Oss® and Bio-Oss/Collagen®) and 2 types of synthetic hydroxyapatite (Apaceram-AX® and Refit®). We considered factors including newly generated bone volume; bone, osteoid, and material occupancy; and bone-to-implant contact. Methods: A beagle model with a mandibular defect was created by extracting the bilateral mandibular third and fourth premolars. Simultaneously, an implant was inserted into the defect, and the space between the implant and the surrounding bone walls was filled with Bio-Oss, Bio-Oss/Collagen, Apaceram-AX, Refit, or autologous bone. Micro-computed tomography and histological analyses were conducted at 3 and 6 months postoperatively (Refit and autologous bone were not included at the 6-month time point due to their rapid absorption). Results: All materials demonstrated excellent biocompatibility and osteoconductivity. At 3 months, Bio-Oss and Apaceram-AX exhibited significantly greater volumes of formation than the other materials, with Bio-Oss having a marginally higher amount. However, this outcome was reversed at 6 months, with no significant difference between the 2 materials at either time point. Apaceram-AX displayed notably slower bioresorption and the largest quantity of residual material at both time points. In contrast, Refit had significantly greater bioresorption, with complete resorption and rapid maturation involving cortical bone formation at the crest at 3 months, Refit demonstrated the highest mineralized tissue and osteoid occupancy after 3 months, albeit without statistical significance. Conclusions: Overall, the materials demonstrated varying post-implantation behaviors in vivo. Thus, in a clinical setting, both the properties of these materials and the specific conditions of the defects needing reinforcement should be considered to identify the most suitable material.
