염분변화에 따른 북방전복의 생리적 반응을 조사하기 위하여 고염분 및 저염분의 농도에 따라 생존율, 삼투질농도, 대사율 및 조직학적 반응을 분석하였다. 염분변화에 따른 북방전복의 생존율은 37 psu에서는 100%였다. 반면 19.8 psu에서 노출 6일째, 12.8 psu 이하에서는 노출 24시간 이내 모두 사망하여 북방전복의 7-days $LS_{50}$은 24.9 psu (20.1-28.2 psu)를 나타내었다. 염분변화에 따른 호흡률은 대조구에 비해 감소하였으며, 반면 암모니아질소 배설률은 증가하였다. 삼투질농도는 염분 26.8 psu이상에서는 1시간 이내 순치하였다. 북방전복 발근육의 조직학적 반응은 염분감소에 따라 혈림프동의 확장, 상피세포의 괴사 및 식세포 증가 등의 현상이 관찰되었다.
카제인과 덱스트린 및 대두유와 어유의 혼합유를 주요 단백질과 탄수화물 및 지질원으로 구성한 전복용 배합사료에 다양한 microalgae (Haeatococccus, Isochrysis galbana, Shizochytrium)와 갑각류성 기원의 첨가제(krill meal, shrimp head meal, red crab meal) 및 녹차부산물을 각각 2% 첨가한 실험사료를 준비하여 16주간 전복에게 공급하여 주었다. 그리고 다시마를 공급한 실험구를 대조구로 두었다. 실험 종료시 전복의 생존율은 유의적인 차이가 없었으나, 사료 첨가제를 첨가한 모든 실험사료에서 전복의 체중증가량은 다시마를 공급한 실험구에 비하여 유의적으로 우수하였다. 전복의 체중 증가량은 Spirulina, Shizochytrium 및 Isochrysis의 순으로 우수하였다. 전복의 체장 및 체중에 대한 가식부의 중량 비는 유의적인 차이가 없었다. 그러나 체폭은 다시마를 공급한 실험구에 비하여 첨가제를 첨가한 모든 실험구에서 유의적으로 우수한 것으로 나타났다. 전복 패각의 색채 개선효과는 Spirulina 첨가구에서 가장 뚜렷하였다. 실험사료내 다양한 microalge와 갑각류성 기원의 사료첨가제는 전복의 성장을 효율적으로 개선시켰으며, 사료내 Spirulina의 첨가는 전복 패각 색채 개선에 가장 효과적이었다.
참전복의 3배체 유도를 위하여 수정 12분후 및 32분 후에 저온$(0^{\circ}C\;,\;3^{\circ}C)$해수 혹은 고온$(35^{\circ}C\;,\;40^{\circ}C)$ 해수 속에서 여러 처리 지속 시간으로 3배체 유도 실험을 한 결과, 수정율은 실험군 사이에 유의한 차가 없었다. (P>0.05). 그러나 부화율과 정상발생율은 3배체구간 2배체 대조구보다 유의하게 낮았다. (P<0.05). $40^{\circ}C$의 고온처리구에서 수정율은 매우 낮았고$(0\~2.7\%)$ 부화되는 유생은 없었다. 3배체 유도율은 염색체수를 직접 계수해서 구했는데 가장 높은 3배체 유도율은 수정 12분 후에 $3^{\circ}C$ 해수에 15분 동안 처리했을 때 $(84.0\%)$였다. 참전복 2배체의 염색체 수는 2n=36이었고 3배체의 염색체 수는 3n=54이었다.
The effect of water temperature and feed on the growth, survival, and body composition of juvenile abalone (Haliotis discus hannai) fed an artificial formulated diet and a macroalgae (Laminaria japonica) for 12 weeks was investigated. Rearing temperature was maintained at $13{\circ}C,\;16{\circ}C,\;19{\circ}C$ by heated and natural sea water temperatures $(7.5-10.2{\circ}C).$ Shell growth, weight gain, and survival rate of the abalone were affected by water temperature and diet (P<0.05). Survival and growth rate of the abalone fed with the formulated diet and Laminaria at $19{\circ}C$ was significantly higher than those of the abalone fed the formulated diet or Laminaria at $13{\circ}C,\;16{\circ}C,$ and ambient temperatures (P<0.05). Moisture, crude protein, crude lipid, and ash contents of the soft whole body were affected mainly by diet than rearing water temperature (P<0.05). These indicate that a mixture of formulated diet and macroalgae could improve the growth of the abalone independent of water temperature and heated water could improve the growth of abalone during the winter season.
Background: The evaluation of suitable reference genes as normalization controls is a prerequisite requirement for launching quantitative reverse transcription-PCR (RT-qPCR)-based expression study. In order to select the stable reference genes in abalone Haliotis discus hannai tissues (gill and hepatopancreas) under heavy metal exposure conditions (Cu, Zn, and Cd), 12 potential candidate housekeeping genes were subjected to expression stability based on the comprehensive ranking while integrating four different statistical algorithms (geNorm, NormFinder, BestKeeper, and ${\Delta}CT$ method). Results: Expression stability in the gill subset was determined as RPL7 > RPL8 > ACTB > RPL3 > PPIB > RPL7A > EF1A > RPL4 > GAPDH > RPL5 > UBE2 > B-TU. On the other hand, the ranking in the subset for hepatopancreas was RPL7 > RPL3 > RPL8 > ACTB > RPL4 > EF1A > RPL5 > RPL7A > B-TU > UBE2 > PPIB > GAPDH. The pairwise variation assessed by the geNorm program indicates that two reference genes could be sufficient for accurate normalization in both gill and hepatopancreas subsets. Overall, both gill and hepatopancreas subsets recommended ribosomal protein genes (particularly RPL7) as stable references, whereas traditional housekeepers such as ${\beta}-tubulin$ (B-TU) and glyceraldehyde-3-phosphate dehydrogenase (GAPDH) genes were ranked as unstable genes. The validation of reference gene selection was confirmed with the quantitative assay of MT transcripts. Conclusions: The present analysis showed the importance of validating reference genes with multiple algorithmic approaches to select genes that are truly stable. Our results indicate that expression stability of a given reference gene could not always have consensus across tissue types. The data from this study could be a good guide for the future design of RT-qPCR studies with respect to metal regulation/detoxification and other related physiologies in this abalone species.
This study attempts to establish a system extracting and monitoring cultural grounds of seaweeds (lavers, brown seaweeds and seaweed fulvescens) and abalone on the basis of both KOMPSAT-2 and Terrasar-X data. The study areas are located in the northwest and southwest coast of South Korea, famous for coastal cultural grounds. The northwest site is in a high tidal range area (on the average, 6.1 m in Asan Bay) and has laver cultural grounds for the most. An semi-automatic detection system of laver facilities is described and assessed for spaceborne optic images. On the other hand, the southwest cost is most famous for seaweeds. Aquaculture facilities, which cover extensive portions of this area, can be subdivided into three major groups: brown seaweeds, capsosiphon fulvescens and abalone farms. The study is based on interpretation of optic and SAR satellite data and a detailed image analysis procedure is described here. On May 25 and June 2, 2008 the TerraSAR-X radar satellite took some images of the area. SAR data are unique for mapping those farms. In case of abalone farms, the backscatters from surrounding dykes allows for recognition and separation of abalone ponds from all other water-covered surfaces. But identification of seaweeds such as laver, brown seaweeds and seaweed fulvescens depends on the dampening effect due to the presence of the facilities and is a complex task because objects that resemble seaweeds frequently occur, particularly in low wind or tidal conditions. Lastly, fusion of SAR and optic spatial images is tested to enhance the detection of aquaculture facilities by using the panchromatic image with spatial resolution 1 meter and the corresponding multi-spectral, with spatial resolution 4 meters and 4 spectrum bands, from KOMPSAT-2. The mapping accuracy achieved for farms will be estimated and discussed after field verification of preliminary results.
전복 방류효과조사를 위한 효과적인 표지 개발을 위하여 치패에 알맞은 표지 탐색과 표지에 따른 성장과 생존율에 관하여 2000년 5월 25일부터 2001년 1월 27일까지 8개월간 조사하였다. 실험군 (결각군, 동선부착군, 볼트너트구조물부착군 그리고 너트접착군)별 표지부착률은 각장 2, 3cm 볼트너트부착군에서 89.9, $89.5\%$로 가장 높았고, 결각군이 $18.5\%$ 이하로 가장 낮게 식별되었다. 패각 내부에서 표지를 피복하여 표지의 안정성을 높이는 표지피복률은 각장 2, 3cm 볼트너트부착군은 각각 $100.0\%$, $96.6\%$로 나타난 반면, 동선부착군에서는 $17.1\%$ 이하로 나타났다. 각장 2, 3, 4와 5cm의 치패 크기에 따른 표지 실험군별 성장은 대조군와 유의한 차이를 보이지 않아 표지가 전복치패의 성장에 영향을 미치지 않았다. 생존율 역시 각장 2, 3, 4cm의 모든 실험군에서 $93.8\%$ 이상을 보였고, 대조군에 비해 유의한 차이를 보이지 않아 표지가 전복의 폐사에 미치는 영향은 없었다. 각장 5cm 실험군의 생존율은 대조군에 비해 각 실험구는 유의한 차이를 보였다. 이상의 결과에서 표지부착 및 피복률, 성장과 생존 등에서 우수한 결과를 보인 미세볼트너트표지를 부착한 각장 3cm실험군이 본 연구에서 가장 우수한 효과를 보였다.
