양식 참전복(Haliotis discus hannai) 열성패 추출물의 제조 및 품질특성 (Processing and Quality Characteristics of a Cultured Recessive Small-sized Abalone Haliotis discus hannai Extract)

  • To determine whether there are differences in the food component characteristics of the cultured recessive small-sized abalone Haliotis discus hannai (SA; 30-40 each/kg) and middle-sized abalone (MA; 10-15 each/kg), the proximate compositions, fatty acid and total amino acid compositions of these two species were evaluated. Additionally, extraction methods were performed on the SA to asses the quality characteristics of the resulting. In terms of proximate composition, MA had lower moisture and carbohydrate levels and higher crude protein contents than SA. The total amino acid contents of MA and SA were 15,734.4 and 11,379.1 mg/100 g, respectively, the major amino acids were glutamic acid, aspartic acid, serine, glycine, alanine, leucine, arginine and lysine, and the major fatty acids were 16:0, 18:0, 18:1n-9, 18:1n-7, 20:4n-6, 20:5n-3, and 22:5n-3. The pH levels and total nitrogen and amino nitrogen contents of the hot-water extract (WE) and scrap enzyme hydrolysate (SE) samples from the SA were 6.32 and 6.05, 1.36% and 1.52%, and 342.1 and 403.1 mg/100 g, respectively. The extraction yields and free amino acid contents from SA were 1,317 and 440 mL/kg, and 8,721.1 and 9,070.7 mg/100 g, respectively, and the concentrations of major components were as follows: arginine, glycine, glutamic acid, alanine and lysine. Additionally, the complex extract (WE+SE) was superior to the traditional extract (WE) in terms of extraction yield, amino-nitrogen content, and organoleptic qualities but not odor.

  • This study was conducted to investigate effects of dietary pigment sources on shell color of juvenile abalone, Haliotis discus hannai. Three replicate groups of the abalone, average weight 173 mg, were fed the diets containing various pigment sources such as Porphyra powder, Spirulina, yeast astaxanthin and paprika extract for 16 weeks. Survival and weight gain were not affected by dietary pigment sources (P>0.05). Shell color of abalone fed diets containing Porphyra powder and Spirulina was approached to yellow-red and orange which are similar to shell color of wild abalone. However, shell color of abalone fed the diets containing yeast astaxanthin and paprika extract were similar to that of control diet showing bright green. Porphyra powder and Spirulina contain not only fat-soluble pigment such as chlorophyll and carotenoids but also water-soluble pigment such as phycoerythrin and phycocyanin. These results would be useful information to change shell color of abalone in aquaculture.

한국산 전복을 이용한 교잡종 개발 (Molecular and Physiological Aspects of Breeding Program for Development of Hybrids between Abalones Distributed in the Coast of Korea)

  • 전복은 전복과 전복속에 속하는 복족류 연체동물로, 전 세계적으로 어업에 의한 식용전복의 공급은 감소하고, 양식에 의한 전복생산은 증가하고 있다. 한국, 일본, 중국을 포함해 전 세계적으로 전복은 중요한 해양 식량자원이며, 양식 산업에 있어서도 그 비중이 증가되고 있긴 하나, 그 느린 성장 속도가 가장 큰 문제점 중의 하나로 여겨진다. 선별, 종내교잡, 종간교잡, 배수체 등과 같은 여러 진보된 기술의 도입을 통하여 전복의 주요 양식 품종을 개발하기 위한 연구가 시도되고 있다. 양식용 전복품종개발을 위하여, 한국 해안에 서식하고 있는 전복은 6종(Haliotis discus hannai, 북방전복; Haliotis discus discus, 둥근전복; Haliotis makada, 왕전복; Haliotis gigantea, 말전복; Haliotis diversicolor diversicolor, 마대오분자기; Haliotis diversicolor supertexta, 오분자기) 간의 교배를 통한 교배종 개발이 유용한 기술 중의 하나가 될 수 있으며, 한국, 중국, 그리고 일본에 서식하는 종 간의 교배종 연구가 활발히 진행 되어 오고 있다. 유용한 잠재적 형질을 가진 한국 산 전복들의 계통학적 관계와 이들의 종 판별에 대해서 살펴볼 것이며, 이들 종들로부터 개발된 분자 마커의 현황과, 성공적인 양식용 교잡종 개발에 있어서 고려해야 할 유전적 사항들에 대하여 알아보고자 한다.

Behavioral analysis of Pacific abalone, Haliotis discus hannai, reveals its feeding preference and attraction potential for brown alga, Sargassum horneri

  • The Pacific abalone, Haliotis discus hannai, is a highly valued and industrially important aquaculture species with growing demands of the expanding abalone aquaculture industry. To explore the feasibility of using the brown alga, Sargassum horneri, as a potential substitute for abalone feed, it is important to identify the feed preference and attractant effect of S. horneri on Pacific abalone. Our experiments indicated that the feeding-associated movement of abalone could be detected using a video tracking system under indirect illumination with dim red light. To further analyze the attraction potentials of various test materials, preference analysis was performed using Avicel-coated glass plates with ground powders of various seaweeds (e.g., S. horneri, Saccharina japonica, and Undaria pinnatifida) and commercial abalone feed, together with coffee waste. Heat map analysis indicated greater attraction by the kelp S. japonica than by S. horneri and commercial feed, which showed similar preference levels. Feeding preference based on the area of Avicel eaten by abalone showed a significant preference for U. pinnatifida over S. horneri (feeding area: 68.6 ± 20.1% vs. 37.5 ± 22.4%, p < 0.05). Additionally, the feeding area was significantly greater for plates with S. japonica than for plates with S. horneri (44.0 ± 16.6% vs. 22.6 ± 15.4%, p < 0.05). There was no significant difference in feeding area between commercial feed and S. horneri (31.7 ± 11.6% vs. 31.6 ± 20.2%, p > 0.05). The methanol extracts attracted abalone in the following order: U. pinnatifida > S. horneri > S. japonica > commercial feed > coffee waste. To determine the attractive effects of the components of methanol extracts, mixtures of methanol extracts of commercial feed with increasing amounts of S. horneri were examined. The results showed a significant increase in feeding preference upon addition of S. horneri up to 50% and 75%, suggesting its potential for use as an appetite-enhancing feed additive. This study identified conditions that can be successfully used to monitor the movement of Pacific abalone; the results of preference analysis confirmed that abalone exhibited similar attraction and feeding preference for S. horneri, compared with commercial feed.

Expression Profile of Heat Shock Protein Gene Transcripts (HSP70 and HSP90) in the Nerve Ganglia of Pacific abalone, Haliotis discus hannai Exposed to Thermal Stress

  • Heat shock proteins (HSPs) are highly conserved cellular proteins that contribute to adaptive responses of organisms to a variety of stressors. In response to stressors, cellular levels of HSPs are increased and play critical roles in protein stability, folding and molecular trafficking. The mRNA expression pattern of two well-known heat shock protein transcripts, HSP70 and HSP90 were studied in two tissues of nerve ganglia, cerebral ganglion and pleuropedal ganglion of Pacific abalone (Haliotis discus hannai). It was observed that both HSP70 and HSP90 transcripts were upregulated under heat stress in both ganglion tissues. Expression level of HSP70 was found higher than HSP90 in both ganglia whereas cerebral ganglion showed higher expression than pleuropedal ganglion. The HSP70 and HSP90 showed higher expression at Day-1 after exposed to heat stress, later decreased at Day-3 and Day-7 onwards. The present result suggested that HSP70 and HSP90 synthesize in nerve ganglion tissues and may provide efficient protection from stress.

북방전복 Haliotis discus hannai의 부착생물 제거에 따른 생물학적 영향 (Biological Effect with Remove the Attached Organisms of Haliotis discus hannai)

  • 본 연구에서는 한국 남해안의 해상가두리에서 북방전복 Haliotis discus hannai 부착생물의 종류와 밀도에 관한 자료의 축적과 부착생물 제거에 따른 영향을 평가하고자 하였다. 해상가두리에서 북방전복의 부착생물은 굴, 태형동물, 따개비가 우점하였으며, 이들의 평균 부착생물 면적비는 약 57.5%였다. 성장률과 간췌장의 건강도는 대조구에 비해 부착생물 제거구에서 좋은 결과를 보였다. 따라서 전복의 부착생물 제거는 개체의 성장과 출하 시 상품성 향상을 위해 필요한 것으로 판단된다.

근친교배에 의한 북방전복 (Haliotis discus hannai)의 근교약세 현상 (Inbreeding depression of Pacific abalone, Haliotis discus hannai by inbreeding mating experiments)

  • 북방전복의 근친교배로 인한 근교약세현상의 유무를 명확히 하기 위하여 근친교배 2가계 (AA 및 BB)와 비근친교배 (ww1 및 ww2) 2가계를 같은날 동시에 생산하여 유생단계부터 동일한 수조에 혼합 사육관리한 후 친자확인 기술을 통하여 4개월째 및 10개월째 각각의 가계별 생존율 및 성장을 비교분석하였다. 성장의 경우, 4개월째 근친교배구 AA가계만이 비근친교배구 ww1 및 ww2가계보다 유의적으로 낮은 값으로 나타났지만, 10개월째는 근친교배구 AA가계 및 BB가계 모두 비근친교배구 ww1 및 ww2가계보다 유의적으로 낮은 값을 나타냈다. 또한 생존율에 있어서도 근친교배구가 비근친교배구에 비하여 낮은 값을 나타내었다. 이것은 한세대의 근친교배(근교계수 : 0.25) 에 있어서도 근교약세현상이 나타난다는 것을 보고하고 있다.

급격한 수온 스트레스에 따른 전복, Haliotis discus hannai 치패의 생리학적 연구 (Physiological Studies on Acute Water-temperature Stress of Juvenile Abalone, Haliotis discus hannai)

  • 본 연구는 전복, Haliotis discus hannai 치패를 이용하여 급격한 수온변화 스트레스에 따른 간부위에서의 항산화효소 및 HSP70 mRNA의 변화를 조사하였다. 실험구는 10, 15, 20(대조구), 25 및 $30^{\circ}C$로 설정하였으며, 측정 시간은 0, 6, 12, 24 및 48h후에 측정하였다. 그 결과 HSP70 mRNA의 발현은 다른 실험구들과 비교하여 $30^{\circ}C$ 실험구에서만 유의적으로 높게 발현되었으며, SOD의 경우 모든 실험구에서는 증가하는 경향을 나타내었으나, 고수온에서 12 h 이후 급격한 상승을 나타내었고, 저수온에서는 수온 스트레스 자극 직후에 증가한 후 회복되는 경향을 나타내었다. CAT에서는 실험 개시 후부터 고수온 실험구에서 지속적인 상승을 나타내었다. 실험기간 중의 생존율은 $30^{\circ}C$ 실험구를 제외한 모든 실험구에서 $100\%$의 생존율을 나타내었으며, $30^{\circ}C$ 실험구에서는 $92\%$의 생존율을 나타내었다. 이상의 결과로, 20"C에서 순치된 전복은 저수온 스트레스에 대한 생리학적 방어기작이 약 $5^{\circ}C$ 내외의 고수온 스트레스에 대한 생리학적 방어기작과 비교하여 보다 빠르게 작용하여 안정화하는 것으로 나타났으며, 일정 수온 이상의 고수온에 노출되었을 경우에는 생체 내 과도한 생리학적 방어 기작이 작용하여 항산화효소 및 HSP70 발현이 증가하는 것으로 나타났다.

Molecular cloning of metal-responsive transcription factor-1 (MTF-1) and transcriptional responses to metal and heat stresses in Pacific abalone, Haliotis discus hannai

  • Background: Metal-responsive transcription factor-1 (MTF-1) is a key transcriptional regulator playing crucial roles in metal homeostasis and cellular adaptation to diverse oxidative stresses. In order to understand cellular pathways associated with metal regulation and stress responses in Pacific abalone (Haliotis discus hannai), this study was aimed to isolate the genetic determinant of abalone MTF-1 and to examine its expression characteristics under basal and experimentally stimulated conditions. Results: The abalone MTF-1 shared conserved features in zinc-finger DNA binding domain with its orthologs; however, it represented a non-conservative shape in presumed transactivation domain region with the lack of typical motifs for nuclear export signal (NES) and Cys-cluster. Abalone MTF-1 promoter exhibited various transcription factor binding motifs that would be potentially related with metal regulation, stress responses, and development. The highest messenger RNA (mRNA) expression level of MTF-1 was observed in the testes, and MTF-1 transcripts were detected during the entire period of embryonic and early ontogenic developments. Abalone MTF-1 was found to be Cd inducible and highly modulated by heat shock treatment. Conclusion: Abalone MTF-1 possesses a non-consensus structure of activation domains and represents distinct features for its activation mechanism in response to metal overload and heat stress. The activation mechanism of abalone MTF-1 might include both indirect zinc sensing and direct de novo synthesis of transcripts. Taken together, results from this study could be a useful basis for future researches on stress physiology of this abalone species, particularly with regard to heavy metal detoxification and thermal adaptation.

Inclusion of Distillers Dried Grain as Partial Replacement of Wheat Flour and Soybean Meal in the Diet of Juvenile Abalone Haliotis discus hannai

  • An 8-week feeding experiment was conducted to determine the influence of dietary distillers dried grain (DDG) on the growth and body composition of juvenile abalone Haliotis discus hannai. Five diets were formulated to contain 0% (DDG0), 15% (DDG15), 30% (DDG30), 45% (DDG45), and 60% (DDG60) DDG, and three replicate groups of abalone (average body weight: $3.6{\pm}0.21$ g) were fed one of the experimental diets at a feeding rate of 5% body weight per day once daily (17:00 h) for 8 weeks. Survival, shell length, and shell width of juvenile abalone were not affected by dietary DDG levels (P > 0.05). Weight gains of juvenile abalone fed DDG15 and DDG30 diets were not different compared to DDG0, but abalone fed DDG45 and DDG60 diets gained less weight than those fed DDG0 (P < 0.05). Soft body weight/body weight ratio of juvenile abalone fed the DDG60 diet was lower than that of those fed the DDG0 diet (P < 0.05), but proximate composition of the soft body was not affected by dietary DDG levels (P > 0.05). The results of this experiment suggest that DDG is a good replacement for wheat flour and soybean meal, and can be used up to 30% in the diet to maintain the growth performance of the juvenile abalone.
