개불, Urechis unicinctus, 건조중의 핵산관련물질의 변화 (DEGRADATION OF NUCLEOTIDES AND THEIR RELATED COMPOUNDS IN GAE-BUL, URECHIS UNICINCTUS, DURING SUN DRYING)

  • 개불, Urechis unicinctus, 건조중의 핵산관계물질의 변화를 음이온교환칼럼크로마토그래피로 실험하여 다음과 같은 결과를 얻었다. 살아있는 개불에는 AMP가 건물량기준으로 $6.8\mu\;mole/g$ 로서 가장 함량이 높았고, IMP는 존재하지 않았다. 건조중 ATP, ADP, AMP는 감소하고 inosine과 hypoxanthine은 증가하였으나, AMP이외의 다른 pattern은 함량이 적고 개불에서와 마찬가지로 건조한 개불에 있어서도 AMP가 건물량기준으로 $5\mu\;mole/g$로서 함량이 가장 많았다.

개장년도에 따른 골프코스 토양의 미생물 활성 (Microbial Activity of Golf Course Soils with Different Open Year)

  • The relationship between microbial activity and physico-chemical characteristics of golf course soils were investigated in Kyonggi Province with different open years. The increasing tendency of soil PH with depth seemed to the result of mineral leaching. Exchangeable $Ca^2$+ , $Mg^2$+ and K+ showed relatively higher concentration in lower horizon indicating the leaching of minerals. The minerals were heavily leached in older golf course. There was a significant positive correlation between pH and $Ca^2$+.The contents of organic matters, total nitrogen and phosphorus were high in higher horizon.Dehydrogenase activity(DHA) and the content of ATP were determined as an index of soil microbial activity. DHA value was significantly different with golf club as well as golf courses ac-cording todifferent open year. This indicates that DHA was affected by several fertilizertreatments rather than herbicide pesticide treatments. DHA was positively correlated with water holding capacity and total nitrogen. The content of ATP was high in fairway and in golf course with older open year, showing the similar tendency with the results of DHA.

Effect of Biphenyl Dimethyl Dicarboxylate on Chemical-Induced Hepatotoxicity

  • To know the mechanism of biphenyl dimethyl dicarboxylate (DDB) in the protection of chemically induced hepatotoxicity, the activity of glutamic pyruvic tran.saminase (GPT) and the level of lipid peroxidation metabolite (malondialdehyde, MDA) and ATP content in hepatocytes were determined in serum and primarily cultured hepatocytes. For in vibo study, rats were pretreated with DDB (300 mg/ kg, p.o.)for 7 days. DDB pretreatment efficiently reduced the elevation of serum GPT activity induced by carbon tetrachloride (1.6 ml/kg, s.c.) and acetaminophen administration (1500 mg/kg, i.p.). In ex vivo study, hepatocytes were isolated from the rats pretreated with DDB (300 mg/kg, p.o.)for 7 days and cultured for 12 hrs before inducing cytotoxicity with chemicals. The MDA formation and the GPT release induced by adriamycin $(1\times10^{-4} mg/ml)$ and cisplatin $(2\times10^{-4} mg/ml)$ were markedly decreased in the hepatocytes from the rats pretreated with DDB as compared to vehicle only. However, DDB pretreatment did not prevent the decrease of ATP contents of hepatocytes induced by cisplatin and adriamycin. In in vitro experiment, DDB was pretreated in primary cultured hepatocytes for 3 days. DDB enhanced the decreases of ATP contents induced by cisplatin and adriamycln. These results suggest that DDB may protect the hepatocytes from injury induced by hepatotoxlcants through inhibiting the lipid peroxidation.

Compositions of Extractive Nitrogenous Constituents and Their Monthly Variation for Fresh Capsosiphon fulvescens

  • To elucidate the composition of extractive nitrogenous components in the fresh Capsosiphons fulvescens cultured off the southern coast of Korea, and to determine the monthly variation of these nitrogenous components, extract samples collected monthly from December to March at Jangheung-gun, Jeonnam Province were analyzed for total nitrogen, free and combined amino acids, ATP and related compounds, betaines, trimethylamine oxide (TMAO) and trimethylamine (TMA). The content of extractive nitrogen was 1,090~1,233 mg/100 g on dry basis. The number of 21~25 ninhydrin-positive substances was detected in the analysis of free amino acids, and their total amount was 3,710~4,788 mg/100 g on dry basis. Among them, free proline, asparagine, glutamic acid, alanine, taurine and glutamine were found to be abundant. The combined amino acids amounted to 1,573~2,121 mg/100 g in total and the total amount of ATP and related compound was 33.8~84.0 mg/100 g ($1.06{\sim}2.46\;{\mu}mol/g$) on dry basis. Betaine, glycinebetaine, $\beta$-alaninebetaine, $\gamma$-butyrobetaine, homarine and trigonelline were detected in most of samples. Levels of free and combined amino acids, ATP and related compounds fluctuated from sample to sample, with their contents higher in December and January and lower in March.

Nitric Oxide-cGMP-Protein Kinase G Pathway Contributes to Cardioprotective Effects of ATP-Sensitive $K^+$ Channels in Rat Hearts

  • Ischemic preconditioning (IPC) has been accepted as a heart protection phenomenon against ischemia and reperfusion (I/R) injury. The activation of ATP-sensitive potassium $(K_{ATP})$ channels and the release of myocardial nitric oxide (NO) induced by IPC were demonstrated as the triggers or mediators of IPC. A common action mechanism of NO is a direct or indirect increase in tissue cGMP content. Furthermore, cGMP has also been shown to contribute cardiac protective effect to reduce heart I/R-induced infarction. The present investigation tested the hypothesis that $K_{ATP}$ channels attenuate DNA strand breaks and oxidative damage in an in vitro model of I/R utilizing rat ventricular myocytes. We estimated DNA strand breaks and oxidative damage by mean of single cell gel electrophoresis with endonuclease III cutting sites (comet assay). In the I/R model, the level of DNA damage increased massively. Preconditioning with a single 5-min anoxia, diazoxide $(100\;{\mu}M)$, SNAP $(300\;{\mu}M)$ and 8-(4-Chlorophenylthio)-guanosine-3',5'-cyclic monophosphate (8-pCPT-cGMP) $(100\;{\mu}M)$ followed by 15 min reoxygenation reduced DNA damage level against subsequent 30 min anoxia and 60 min reoxygenation. These protective effects were blocked by the concomitant presence of glibenclamide $(50\;{\mu}M)$, 5-hydroxydecanoate (5-HD) $(100\;{\mu}M)$ and 8-(4-Chlorophenylthio)-guanosine-3',5'-cyclic monophosphate, Rp-isomer (Rp-8-pCPT-cGMP) $(100\;{\mu}M)$. These results suggest that NO-cGMP-protein kinase G (PKG) pathway contributes to cardioprotective effect of $K_{ATP}$ channels in rat ventricular myocytes.

Extracellular ATP Stimulates $Na^+\;and\;Cl^-$ Transport through the Activation of Multiple Purinergic Receptors on the Apical and Basolateral Membranes in M-1 Mouse Cortical Collecting Duct Cells

  • The mammalian cortical collecting duct (CCD) plays a major role in regulating renal NaCl reabsorption, which is important in $Na^+$ and $Cl^-$ homeostasis. The M-1 cell line, derived from the mouse cortical collecting duct, has been used as a mammalian model of the study on the electrolytes transport in CCD. M-1 cells were grown on collagen-coated permeable support and short circuit current $(I_{sc})$ was measured. M-1 cells developed amiloride-sensitive current $5{\sim}7$ days after seeding. Apical and basolateral addition of ATP induced increase in $I_{sc}$ in M-1 cells, which was partly retained in $Na^+-free$ or $Cl^--free$ solution, indicating that ATP increased $Na^+$ absorption and $Cl^-$ secretion in M-1 cells. $Cl^-$ secretion was mediated by the activation of apical cystic fibrosis transmembrane regulator (CFTR) chloride channels and $Ca^{2+}-activated$ chloride channels, but $Na^+$ absorption was not mediated by activation of epithelal sodium channel (ENaC). ATP increased cAMP content in M-1 cells. The RT-PCR analysis demonstrated that M-1 cells express $P2Y_2,\;P2X_3\;and\;P2Y_4$ receptors. These results showed that ATP regulates $Na^+$ and $Cl^-$ transports via multiple P2 purinoceptors on the apical and basolateral membranes in M-1 cells.

지질의 물리학적 성상이 $Ca^{2+}$-ATPase 활성도에 미치는 영향 (The Effects of Physical States of Phospholipids on $Ca^{2+}$-ATPase Activity of Biological Membranes)

  • 세포막을 구성하고 있는 지질의 물리학적 성상이 단백질의 세포막 속으로의 삽입과정 및 단백질의 기능에 미치는 영향을 관찰하기 위하여 골격근의 근세망(SR)으로부터 $Ca^{2+}-ATPase$ 단백질을 분리한 후 이를 세포막의 주 구성성분인 포스파티딜콜린(PC)과 포스파티딜에타노라민(PE)의 혼합지질과 재조합(reconstitution)시켰다. 이와같이 인공적으로 재조합된 구조물에서 $Ca^{2+}-ATPase$의 기능을 측정하기 위하여 칼슘지시색소인 아르세나죠III(AIII)를 이용한 분광방법과 방사선동위원소를 이용한 여과법으로 칼슘흡수율을 측정하였고 또한 ATP 가수분해 능력을 측정하였다. 실험결과 칼슘의 흡수율은 포스파티딜코린의 함량이 많은 혼합지질과 재조합시킬 때에 증가하였고, ATP 가수분해 능력은 포스파티딜함량이 25%까지는 포스타피딜코린의 양에 비례하여 증가하였으나 50%이상에서는 약간 감소하는 경향을 보였다. 한편 지질세포막속으로 단백질이 삽입되는 양은 포스파티딜 함량이 25%일 때 최고의 값을 보였으며 함량이 그 이하 또는 이상일 때는 감소하였다. 이상의 실험결과로보아 단백질의 기능은 세포막이 "bilayer" 구조를 갖출때에 증가하고 세포막속으로 단백질이 삽입되는 양은 세포막이 "non-bilayer" 구조를 형성할 때에 증가함을 알 수 있다.

생선회의 육질향상에 관한 연구 1. 생선회 육질향상에 미치는 저온 브라인 온도의 영향 (Improvement of Sliced Raw Fish Texture 1 Effect of Cold Brine Temperature on Sliced Raw Fish Texture)

  • To improve the sensory quality of sliced raw fish, the effect of cold brine temperature on the texture properties was investigated. Fishes were immersed in cold brine by various condition and killed instantly by spiking at the head. The onset of rigor-mortis was accelerated by decrease of immersion temperature. Also, the time reaching full rigor was shortened remarkably too. However, the rigor index of samples immersed in the cold brine decreased more than that of samples killed instantly. Rigor index was the highest in samples killed instantly, followed by $0^{\circ}C,\;-5^{\circ}C,\;-10^{\circ}C,\;-12.5^{\circ}C,\;-15^{\circ}C\;and\;-20^{\circ}C$ in that order. The breaking strength of samples immersed in the cold brine increased slowly but did not increase through all the conditions in the samples. Breaking strength of all samples immersed in the cold brine decreased significantly after reaching the maximum values (p<0.05). The lactate content of muscle was increased by the decreased brine temperature, however ATP content decreased. Minimum lactate and maximum ATP content in fish muscle showed at $-12.5^{\circ}C$. Those results demonstrate that optimal temperature for improvement of sliced raw fish quality by cold brine was $-12.5^{\circ}C.$

시스플라틴에 의한 $LLC-PK_1$의 알파-메틸글루코스 흡수 감소 기전 (Mechanism of Inhibition of ${\alpha}$-Methylglucose Uptake by Cisplatin in $LLC-PK_1$)

  • We have previously shown that determination of glucose uptake using ${\alpha}$-methylglucose(${\alpha}$-MG) is very sensitive and rapid parameter for the assessment of loss of cellular fu nction in renal cell line($LLC-PK_1$). The present study was designed to elucidate the mechanism of inhibition of ${\alpha}$-MG uptake and the intracellular site of toxic action of cisplatin(CIS). $LLC-PK_1$ cells were exposed to various concentrations(5 ${\mu}$M-l00 ${\mu}$M) of CIS for 5 hrs or 24 hrs and ${\alpha}$-MG uptake was determined. Mitochondrial function was evaluated by measuring intracellular ATP content and MTT reduction. The activities of marker enzymes for the basolateral membrane(Na$^+$-K$^+$ ATPase) and brush border membrane (alkaline phosphatase: ALP) were also measured. CIS treatment significantly inhibited the ${\alpha}$-MG uptake in a time- and dose-dependent manner above 25 ${\mu}$M for 5 hrs. Intracellular ATP content and MTT reduction were affected by 24 hr-treatment of 50 ${\mu}$M CIS. The activities of Na$^+$-K$^+$ ATPase and ALP were significantly decreased at 10 ${\mu}$M and 5 ${\mu}$M of CIS for 24 hrs, respectively. The incubation with CIS for 5 hrs had no effects on the intracellular ATP content, MTT reduction and the activities of marker enzymes up to 100 ${\mu}$M. These results partly indicate that inhibition of ${\alpha}$-MG uptake by CIS may not be attributed to the disturbance of mitochondrial function or inhibition of the activity of Na$^+$-K$^+$ ATPase and can be resulted from direct effect of CIS on the Na$^+$/glucose cotransporter in brush border membrane. This study shows that additional mechanistic information, indicating the intracellular site of nephrotoxic action, can be gained by coupling the ${\alpha}$-MG uptake and ATP content or the activity of Na$^+$-K$^+$ ATPase.

Bacillus subtilis ATP 조해(阻害) Ribonuclease에 관한 연구 (ATP-Inhibited Ribonuclease of Bacillus subtilis)

  • ATP-저해 ribonuclease는 Bacillus subtilis와 같은 Bacillus속에서만 발견 되는 것으로서 이들 효소의 생리적 역활을 규명할 목적으로 Bacillus subtilis 균주를 사용하여 ATP저해성 negative ribonuclease변이주의 취득을 시도하고 선정된 균주가 생산하는 ribonuclease의 정제 및 몇 가지 성질에 대하여 실험한 결과는 아래와 같다. 1. Bacillus subtilis로 부터 ATP-저해 ribonuclease negative mutant의 취득을 시도한 결과 시험된 1817주의 균중에서 101균주가 변이주주로 선정 되었다. 2. 수개의 개별적인 column chromatography에 의하여 당균주가 생산하는 ATP-저해성 ribonuclease의 정제를 시도한 결과 DEAE-Sephadex A-50 column chromatography을 이용한 경우는 30배의 정제 효과와 99%의 회수율을 나타냈고 Sephadex G-75 column의 경우는 20배의 정제 효과와 98%의 회수율을 각각 나타냈다. 3. pH 5침전, 유안염석, Sephadex G-75 및 CM Sephadex를 연속적으로 통과시켜 당효소에 대한대량정제 시험을 시도한 결과 최초의 step(crude extract) 과정에 비하여 60배의 정제효과를 나타냈다. 4. 본정제 효소액에 대하여 전기영동을 실시한 결과 비정제 효소액에 비하여 매우 감소된 수의 담백 pattern을 나타냈다. 5. 당균주가 생산하는 ATP저헤 ribonuclease는 single stranded RNA를 분해하여 2',3'-cyclic AMP, 2',3'-cyclic CMP, 2',3'-cyclic GMP, 2',3'-cyclic UMP와 몇종의 미지물을 생성하였고 또한 본효소는 double stranded RNA를 분해하지 않었다.
