MEAT SPECIATION USING A HIERARCHICAL APPROACH AND LOGISTIC REGRESSION

  • Food adulteration is a serious consumer fraud and a matter of concern to food processors and regulatory agencies. A range of analytical methods have been investigated to facilitate the detection of adulterated or mis-labelled foods & food ingredients but most of these require sophisticated equipment, highly-qualified staff and are time-consuming. Regulatory authorities and the food industry require a screening technique which will facilitate fast and relatively inexpensive monitoring of food products with a high level of accuracy. Near infrared spectroscopy has been investigated for its potential in a number of authenticity issues including meat speciation (McElhinney, Downey & Fearn (1999) JNIRS, 7(3), 145 154; Downey, McElhinney & Fearn (2000). Appl. Spectrosc. 54(6), 894-899). This report describes further analysis of these spectral sets using a hierarchical approach and binary decisions solved using logistic regression. The sample set comprised 230 homogenized meat samples i. e. chicken (55), turkey (54), pork (55), beef (32) and lamb (34) purchased locally as whole cuts of meat over a 10-12 week period. NIR reflectance spectra were recorded over the wavelength range 400-2498nm at 2nm intervals on a NIR Systems 6500 scanning monochromator. The problem was defined as a series of binary decisions i. e. is the meat red or white\ulcorner is the red meat beef or lamb\ulcorner, is the white meat pork or poultry\ulcorner etc. Each of these decisions was made using an individual binary logistic model based on scores derived from principal component or partial least squares (PLS1 and PLS2) analysis. The results obtained were equal to or better than previous reports using factorial discriminant analysis, K-nearest neighbours and PLS2 regression. This new approach using a combination of exploratory and logistic analyses also appears to have advantages of transparency and the use of inherent structure in the spectral data. Additionally, it allows for the use of different data transforms and multivariate regression techniques at each decision step.

  • 금속 규소와 구리 촉매가 함유된 접촉물과 메탄올의 반응에 의한 메톡시실란의 합성에서의 TMOS 반응 생성속도를 산출하였다. 활성 자리 수의 변화에 따른 영향을 제거하기 위해서 유속 전이 기술을 사용하여 주입되는 메탄올의 유속을 반응도중 급격히 변화시켰다. 실험 결과 TMOS 생성속도에 영향을 미치는 인자는 반응에 참여하는 메탄올 농도가 아닌 접촉물질의 사용량임을 확인하였으며, 이를 바탕으로 TMOS 생성 메카니즘에서 접촉 물질의 표면에서 중간생성물이 형성되는 반응 단계가 반응 율속단계라고 추정되었다. 최적 공정조건에서 규소 1 g당 최대 TMOS 생성속도는 $210^{\circ}C$에서 0.030 (g/min)이었으며, 이때의 활성화 에너지는 값은 8.5 kcal/mol, 반응 생성속도 상수의 온도 의존성은 식 $k=4.09{\times}10^4\;exp$ ($-4.73{\times}10^3/T$)로 나타났다.

Disruption of Sphingolipid Metabolism as a Potential Mechanism of Fumonisin Inhibition of Cell Growth in $LLC-PK_1$ Cells

  • Fumonisins are a family of mycotoxins produced by the fungus Fusarium moniliforme which is a common contaminant in corn. Fumonisins are potent inhibitors of sphingosine and sphinganine N-acyltransferase (ceramide synthase), key enzymes in sphingolipid metabolism. The purpose of this study was to provide the evidence that the elevated levels of free sphingoid bases (primarily sphinganine) and depletion of complex sphingolipids were closely related to the inhibition of cell growth in LLC-$PK_1$ cells exposed to fumonisin $B_1$$(\leq 35 {\mu}M)$. Concentrations of fumonisin $B_1$ between 10 and $35 {\mu}M$ were known to inhibit cell growth without cytotoxicity in $LLC-PK_1$ cells (Yoo et al. Toxicol. Appl. Pharmacol. 114, 9-15, 1992). Cells exposed to 35$\mu M$ fumonisin B$_1$ for 48 and 72 hr developed a fibroblast-like (elongated and spindle-shaped) appearance and were less confluent than normal cells. At between 24 and 48 hr after exposure to fumonisin $B_1$ cells were beginning to show the inhibition of cell growth and at 72 hr the number of viable cells in fumonisin-treated cultures was about 50% of concurrent control cultures. During the 24 hr lag period preceding inhibition of cell growth, the free sphinganine levels in cells exposed to $35 {\mu}M$ fumonisin $B_1$ were highly elevated (approximately 230 fold higher than normal cells). The elevated levels of free sphinganine were $435\pm14$$pmoles/{10^6}$ cells at 48 hr and approximately TEX>$333\pm11$$pmoles/{10^6}$ cells in cells exposed to $35{\mu}M$ fumonisin$B_1$ at 72 hr, while the levels of free sphinganine in normal cells were less than 2$pmoles/{10^6}$ cells. Under the same condition, depletion of intracellular complex sphingolipids as a consequence of fumonisin inhibition of de novo sphingolipid biosynthesis and turnover pathway was appeared. Content of free sphingold bases in dividing cells was more elevated than in confluent cells at 24-48 hr after cells were exposed to $20{\mu}M$ fumonisin $B_1$. The dividing cells were showing the inhibition of cell growth at 48-72 hr and $20{\mu}M$ fumonisin $B_1$. The results of this study support the hypothesis that the inhibition of cell growth is very well related to the disruption of sphingolipid metabolism in $LLC-PK_1$ cells.

기화성방청제 Dialkylamine (di-)nitrobenzoates 합성 및 방청성능 (Synthesis and Performance of Dialkylamine (di-)nitrobenzoates for Vapor Corrosion Inhibitor)

  • 방청제로 dialkylaminc (di-)nitrobenzoates 14개 화합물을 합성하고 첨가제 효과와 함께 방청성능을 조사하였다. 합성화합물은 원소분석, FT-IR, $^1H$-NMR 분석으로 확인하였으며, 합성한 화합물과 첨가제 [$(NH_4)_2CO_3$, $NaHCO_3$]의 방청성능은 방청제를 1 M $Na_2SO_4$ 용액에 1% (w/v) 용해시켜 potentiostatic method로 분극실험하여 조사하였다. 철에 대해 dialkylamine 3, 5-dinitrobenzoates보다 dialkylamine 4-nitrobenzoates 부동태화 전류밀도 ($i_p$)가 작았으며, diethylamine 4-nitrobenzoate 용액에서 낮은 값 ($i_p$; $4.78mA/cm^2$)을 나타내었다. 비철에 대한 $i_p$값은 dialkylamine 3, 5-dinitrobenzoates가 작았고 dipropylamine 3, 5-dinitrobenzoate와 hexamethyleneimine 3, 5-dinitrobenzoate가 낮은 값 ($i_p$; 36, $37mA/cm^2$)을 나타냈다. 방청제에 $(NH_4)_2CO_3$$NaHCO_3$ 첨가효과는 철에 대해 높은 부식억제 효과를 나타냈으나, 비철에서는 좋은 결과를 얻지 못하였다. 철용 방청제 diethylamine 4-nitrobenzoate와 $(NH_4)_2CO_3$, $NaHCO_3$의 최적 혼합비율은 4 : 6과 5 : 5이며, 부동태화 전류밀도 ($i_p$)는 0.26, $0.85mA/cm^2$의 최소값을 각각 나타내었다.

  • 2-Bromooctanoic acid (2-BrOA) is known to block the formation of polyhydroxyalkanoic acid (PHA) in Pseudomonasfluorescens BM07 without any influence on the cell growth when grown on fructose, but it inhibits the cell growth when grown on octanoate (OA) (Lee et al., Appl. Environ. Microbiol. 67: 4963- 4974, 2001). We investigated the role of 2-BrOA in the PHA synthesis of the bacterium grown with mixtures of fructose and fatty acids. OA, 11­phenoxyundecanoic acid (1 1-POU), and 5-phenylvaleric acid (5-PV) were selected as model substrates. When supplemented with 50 mM fructose, all these carboxylic acids suppressed the formation of PHA from fructose, however, the ~-oxidation coenzyme A monomers derived from the carboxylic acids were efficiently polymerized, but the conversion yield [(mol of carboxylate substrate converted into PHA)/(mol of carboxylate substrate in the feed)] was low (e.g., maximally $\~53\%$ for 5 mM 11-POU). Addition of 2-BrOA (up to 5 mM) to the mixed carbon sources raised the conversion yield sensitively and effectively only at low levels of the acid substrates (e.g., 2 mM 1 1-POU or 5 mM OA): For instance, $100\%$ of 2 mM ll-POU were converted into PHA in the presence of 5 mM 2-BrOA, whereas only $\~10\%$ of the 1 1-POU were converted in the absence of 2-BrOA. However, at highly saturated suppressing levels (e.g., 5 mM ll-POU), 2-BrOA inhibitor showed no significant additional effect on the conversion ($60- 70\%$ conversion irrespective of 2-BrOA level). The existence of competitive and compensative relationship between 2­BrOA and all the carboxylic acid substrates used may indicate 'Present address: Section on Brain Physiology and Metabolism, Bldg. 10, Rm. 6N202, National Institute on Agmg, National Institute of Health, Bethesda, MD 20892, U.S.A. that all the acid substrate-derived inhibiting species bind to the same site as the 2-BrOA inhibiting species does. We, therefore, suggest that 2-BrOA can be used for efficiently increasing the yield of conversion of expensive substituted fatty acids into PHA and then substituted 3-hydroxyacids by hydrolyzing it.

  • 알콕시실란 중의 하나인 tetramethyl orthosilicate (TMOS)를 금속 규소와 메탄올을 출발물질로 구리계 촉매상에서 기상 반응시키는 직접합성법으로 제조할 때, 규소와 구리촉매 및 금속 염화물 조촉매로 이루어진 접촉물질의 제조 방법 및 온도가 생성물의 수율 및 선택도에 미치는 영향을 검토하였다. 이때 구리 촉매와 함께 Zn, Sn, Cd계 화합물을 조촉매로 사용하여 그 효과를 비교하였다. 구리 공급원으로 제일염화구리를 사용하고 염화아연을 조촉매로 사용한 2원 촉매계가 가장 적합하였으며, 구리/규소 = 7 wt %, 아연/구리 = 7 wt % 조성에서 함침법을 사용하여 규소와 혼합한 후 $380^{\circ}C$에서 활성화시킨 접촉물질을 제조하여 TMOS 합성에 사용하였을 때, 반응온도 $220^{\circ}C$에서 평균선택도 87.2% 규소소모율 69.2%를 나타냈다.

국내산 침엽수 추출물의 Trichoderma spp.에 대한 항균활성 (Antifungal activity of extracts from Chamaecyparis obtusa and Pseudotsuga menziesii against Trichoderma spp.)

  • 본 연구는 국내산 침엽수로부터 천연 항균화합물 개발을 목표로 미송 및 편백으로부터 추출물을 확보하여 푸른곰팡이병의 원인균인 Trichoderma spp. 5종을 대상으로 항균활성을 테스트하고 GC/MS를 이용하여 항균화합물을 탐색하였다. 미송 추출물은 Trichoderma spp.에 대해 1,000 ppm에서 저해 활성을 나타냈으며, 4,000 pp에서는 T. viride>T. harzianum>T. aggressivum 순으로 70.1, 67.3, 64.7%의 높은 항균활성을 나타냈다. 편백 추출물도 1,000 ppm에서 항균활성을 나타냈고 4,000 ppm에서 T. viride>T. harzianum>T. aggressivum 순으로 63.2, 59.3, 59.1%의 높은 항균활성을 나타냈다. 그러나 1,000 ppm의 미송추출물과 1,000 ppm의 편백추출물을 각각 9:1, 8:2, 6:4, 2:8 (w/w) 비율의 혼합제제로 만들어 Trichoderma spp.에 처리하였을 때 미송추출물 1,000 ppm 및 편백추출물 1,000 ppm 각각의 단일추출물보다 낮은 항균활성을 나타냈다. Trichoderma spp. 5종에 대해 높은 항균활성을 나타내는 미송추출물에서는 2-Isopropoxy-ethylamine 46.5%, epifluorohydrin 8.6%, trans-2,3-Di-methyloxirane 7.6%, (IR)-(-)-Myrtenal 6.0%, 2-Methoxy-4-Vinylphenol 3.9%, benzaldehyde 2.8%가 함유되어 있었고, 편백추출물에서는 ${\alpha}$-Terpinenyl acetate 14.9%, Sabinene 10.9%, dl-Limonene 9.6%, ${\alpha}$-Terpinolene 7.5%, ${\alpha}$-Pinene 7.1%가 함유된 것이 확인되었다.

비교 분자 유사성 지수분석(CoMSIA) 방법에 따른 1-(5-methyl-3-phenylisoxazolin-5-yl)methoxy-2-chlore-4-fluorobenzene 유도체들의 Protox 저해 활성에 관한 이해 (Understanding the Protox Inhibition Activity of Novel 1-(5-methyl-3-phenylisoxazolin-5-yl)methoxy-2-chloro-4-fluorobenzene Derivatives Using Comparative Molecular Similarity Indices Analysis (CoMSIA) Methodology)

  • 새로운 5-methyl-3-phenylisoxazolin-5-yl)methoxy-2-chloro-4-fluorobenzenes 유도체들의 구조 변화에 의한 벼(Orysa sativa L.)와 논피 (Echinochloa crus-galli) 뿌리와 줄기 부위의 protox 저해활성에 대한 3차원적 구조-활성관계(3D-QSAR)에 근거하여(성낙도, 등 (2004) 한국응용생명화학회지 47(3), 351-356) 비교분자 유사성 지수분석(CoMSIA) 방법으로 연구하였다. 두 초종의 부위별 protox 저해활성에 관한 CoMSIA 모델들은 수소결합 주게장이 제외된 입체장, 정전기장, 소수성장, 수소결합 받게장 등으로 조합된 CoMSIA장과 부가적 설명 인자로서 LUMO 분자 궤도장, 몰라 굴절을(MR) 및 쌍극자 능율(DM) 등이 추가된 조건에서 유도되었다. 방제 대상인 논피에 대한 모델이 벼에 대한 모델보다 양호하였으며 논피에 대한 모델은 cross-validated $r^2\;_{cv.}$$(q^2=0.871{\sim}0.913)$과 non cross-validated $r^2\;_{ncv.}$$(0.936{\sim}0.920)$ 그리고 PRESS 값$(0.255{\sim}0.273)$에 근거하여 매우 좋은 예측성을 나타내었다. 그리고 protox 저해 활성은 분자의 입체장$(5.4{\sim}15.7%)$ 및 소수성장$(68.0{\sim}84.3%)$과 높은 상관성을 보였다. 이같은 CoMSIA 분석결과, 논피에 대한 선택적인 protox 저해활성은 C-phenyl 고리상 ortho-위치가 steric bulky 할수록 클 것으로 예상되었다.

As a result, accelerated senescence and enzymatic browning OCCUI', Recent l'esearch on minimally processed fl'uits and vegetables has mainly focused on methods to inhibit browning, due to ban on use of sulfur dioxide, In order to retard or prevent these physiological changes, val'ious al ternatives, reducing agents. acidulants, chelating agents and inol'ganic sal ts have been evaluated for use on fresh cut fl'ui ts. Al though some agents were effective replacement for sulfur dioxide. consum$\textregistered$I'S demandless use of chemical on such products. Shel~ life of minimally processed products has been extended by inhibition of metabolic reactions associated with loss of quality and by inhibition of aerobic spoilage caused by wide variety of microorganisms. Appl ication of ~I.-\ packaging, including vacuum packaging, retards the rate of respiration, prevents growth of aerobic spoilage organisms, inhibits oxidation and color deterioration. Tissue softening is another major problem in minimally processed products because enzymes re 1 a ted to ce 11 wa 11 degrada t i on are not inactivated. Various treatments have been investigated for retardation of the softening of sliced products. Some studies have concentrated on the application of an active packaging system with ~I, l. packaging and calcium infi 1 tration as possible measures to retain firmness of processed products. In my opinion, one important step for production of minimally processed frui ts wi th favorabl e color of cut surface and firm texture is the selection of better cultivar. As the view, changing tendency of fresh color by apple cultivars and relationship between the tendency and PPO activity will be discussed in the seminar. In addition to the topic, research result on quality enhancement of fresh apple slices by heat shock treatment will be introduced.
