In this study, we show that Mycobacterium avium subsp. paratuberculosis MAP1305 induces the maturation of bone marrow-derived dendritic cells (BMDCs), a representative antigen presenting cell (APC). MAP1305 protein induces DC maturation and the production of pro-inflammatory cytokines (Interleukin (IL)-6), tumor necrosis factor (TNF)-${\alpha}$, and IL-$1{\beta}$) through Toll like receptor-4 (TLR-4) signaling by directly binding with TLR4. MAP1305 activates the phosphorylation of MAPKs, such as ERK, p38MAPK, and JNK, which is essential for DC maturation. Furthermore, MAP1305-treated DCs transform naive T cells to polarized $CD4^+$ and $CD8^+$ T cells, thus indicating a key role for this protein in the Th1 polarization of the resulting immune response. Taken together, M. avium subsp. paratuberculosis MAP1305 is important for the regulation of innate immune response through DC-mediated proliferation of $CD4^+$ and $CD8^+$ T cells.
This study was carried out to analyse the microbio\logical contamination and biogenic amines(BA) content in Korea traditional soybean paste and commercial soybean paste. The results of microbio\logical analysis through Korean traditional soybean pastes($L1{\sim}L4$) were $7.8{\pm}0.1\;\log\;CFU/g{\sim}7.9{\pm}0.1\;\log\;CFU/g$, commercial soybean pastes($H1{\sim}H6$) were $6.2{\pm}0.1\;\log\;CFU/g{\sim}7.4{\pm}0.1\;\log\;CFU/g$ for APC (Aerobic Plate Count), and $L1{\sim}L4$, H5, H6 soybean pastes were $2.3{\pm}0.4\;\log\;CFU/g{\sim}2.6{\pm}0.1\;\log\;CFU/g$ for Bacillus cereus. But other microorganism was not dectected. Among biogenic amines, PUT(putrescine), TYR(tyramine), HIS(histamine), PHE(2-Phenylethylamine) were dectected high level and CAD(cadaverine), TRY(trypramine), AGM(agmatine) were dectected medium level and SPD(spermidine), SPM(spermine), NOR(noradrenaline), SER(serotonin) were dectected low level. Dectected contents of biogenic amines were higher in commercial soybean paste compared to the traditional soybean paste.
The changes in quality characteristics and shelf-life of vacuum-shrink packaged Bierschinken after stuffing in fibrous casing and stored at 5 and $10^{\circ}C$ were investigated The total aerobic plate count (APC) of Bierschinken was initially <2.00 $log_{10}CFU/cm^2$, however increased gradually over storage period The APCs of Bierschinken maintained at the level of 4.86 and 5.13 $log_{10}CFU/cm^2$ after 35 days at 5 and $10^{\circ}C$, respectively. pH tended to decrease with the extension of storage period The values of TBA and VBN tended to increase with storage time, of which trend was pronounced at $10^{\circ}C$ rather than at $5^{\circ}C$. According to the sensory evaluation, Bierschinken had the marketing value till 28 days irrespective of storage temperature. The shelf-life of Bierschinken tested might be preferentially restricted by the occurrence of off-odor and off-flavor at the end of storage period.
Health functional foods refer to food which manufactured in the form of a tablet, capsule, granule or pill, using materials and ingredients with useful function for a human body in Korea. It needs to be confirmed as safe. Microbiological analyses on 37 samples of botanical raw materials used in the health functional food were performed. Microbiological analyses and probability of irradiation using PSL on 4 samples of aloe powder products were studied. In health functional food ingredients APC was $10^{3}-10^{6}\;CFU/g$ and coliform counts were $10^{2}-10^{5}\;CFU/g$. Among 37 samples B. cereus were found in 12 samples. Four samples of aloe powder products were tested for possible irradiation using preliminary PSL, Two samples of aloe powder products showed positive on preliminary PSL test for irradiation.
Effects of sanitizers and disinfectants on microorganisms in food-processing plants were investigated. Chlorine and hydrogen peroxide were most effective, showing approximately 2-3 log reductions, among products tested, whereas alcohol showed less than 1 log reduction. Before using sanitizer and disinfectant in food processing plants, aerobic plate, coliforms, and E. coli counts were about $10^1-10^5,\;10^1-10^2$, and below $10\;CFU/cm^2$, respectively. After use of sanitizer, APC decreased to $10^1-10^2$. The result to confirm sanitizing effectiveness for 2 months using showed that Alcohol compound, QAC, hydrogen peroxide compound were effective against Staphylococcus aureus ATCC 6538 and E. coli ATCC 10536 at recommended usage concentrations, whereas effectiveness of chlorine compound decreased from 8 to 4 log 2 weeks after opening of product. Although sanitizers and disinfectants approved by law showed 5 log reduction in vitro for S. aureus ATCC 6538 and E, coli ATCC 10536, sanitizing effectiveness in food processing plants was very low, and effectiveness decreased once used product was stored.
Background: Synergistic antitumor effects of the combined chemoimmunotherapy based on dendritic cells have been reported recently. The aim of this study is to search new applicability of gefitinib into the combination treatment through the confirmation of gefitinib effects on the monocyte derived dendritic cells (moDCs); most potent antigen presenting cell (APC). Methods: Immature and mature monocyte-derived dendritic cell (im, mMoDC)s were generated from peripheral blood monocyte (PBMC) in Opti-MEM culture medium supplemented with IL-4, GM-CSF and cocktail, consisting of TNF-${\alpha}$ (10 ng/mL), IL-$1{\beta}$ (10 ng/mL), IL-6 (1,000 U/mL) and $PGE_2$ ($1{\mu}/mL$). Various concentrations of gefitinib also added on day 6 to see the influence on immature and mature MoDCs. Immunophenotyping of DCs under the gefitinib was performed by using monoclonal antibodies (CD14, CD80, CD83, CD86, HLA-ABC, HLA-DR). Supernatant IL-12 production and apoptosis of DCs was evaluated. And MLR assay with $[^3H]$-thymidine uptake assay was done. Results: Expression of CD83, MHC I were decreased in mMoDCs and MHC I was decreased in imMoDCs under gefitinib. IL-12 production from mMoDCs was decreased under $10{\mu}M$ of gefitinib sinificantly. Differences of T cell proliferation capacity were not observed in each concentration of geftinib. Conclusion: In spite of decreased expressions of some dendritic cell surface molecules and IL-12 production under $10{\mu}M$ of gefitinib, significant negative influences of gefitinib in antigen presenting capacity and T cell stimulation were not observed.

The results of a recent case study for material characterizations and structural evaluation to design asphalt overlay thickness of William P Hobby airport in Houston, Texas are presented herein. The existing runway 12R-30L of Hobby airport consisted of thick asphalt overlay over Portland Cement Concrete (PCC) and the localized surface shoving as evident in the closure of surface groove has been observed recently. Using the field cored asphalt concrete mixtures, measurements of percent air voids, asphalt content and aggregate gradation were conducted to find out the causations of surface shoving and groove closure. The FAA layered elastic program, LEDFAA was utilized to evaluate pavement structural conditions for new asphalt overlay. Two different composition assumptions for existing pavement were made to evaluate the pavement as followings: 1) APC, Asphalt Concrete Overlay over PCC pavement and 2) AC, Asphalt Concrete pavement. Based on laboratory testing results, a ratio of percent passing #200 to asphalt content ranged 1.1 to 2.2, which is considered a high ratio and a tendency of tender mix design was observed. Thus, the localized surface shoving and groove closure of the runway 12R-30L could be attributed to the use of excessive fine contents and tender mix design. Based on the structural evaluation results, it was ascertained that the analysis assuming the pavement structure as AC pavement gives more realistic structural life when the asphalt overlay is thicker enough compared to PCC layer because the existing PCC pavement under asphalt overlay acts more like a high quality base material.

The study was conducted to evaluate the effects of spray-dried plasma (SDP) supplementation during late gestation and lactation on productive performance and immune responses of sows and their litters. Twelve sows (227.78 ± 2.16 kg average body weight; 2.0 average parity) were randomly allotted to two dietary treatments: a basal diet (CON) and the basal diet supplemented with 1% SDP. Sows were fed experimental diets from d 30 before farrowing to weaning of their piglets. Blood samples were collected from sows on d 1, 3, and 7 of lactation and from two randomly selected nursing pigs per litter on d 3 and 7 after birth, and d 1, 3, and 7 after weaning. Productive performance and immune responses of sows and their piglets were measured. There was a trend of less body weight loss in sows supplemented with SDP (p < 0.10) during the lactation period and a trend of greater (p < 0.10) average daily gain in SDP piglets compared to those in the CON group. Sows in the SDP group tended to have lower (p < 0.10) serum concentrations of tumor necrosis factor-α (TNF-α), transforming growth factor-β1 (TGF-β1), and cortisol on d 3 and lower serum concentration of TNF-α on d 7 compared with sows in CON group. In comparison with CON piglets, piglets from SDP sows tended to have lower (p < 0.10) serum concentrations of TNF-α, TGF-β1, and cortisol on d 7 after birth, lower (p < 0.10) serum TNF-α and C-reactive protein on d 3 and 7 after weaning, and greater (p < 0.10) average daily gain after weaning. Moreover, weaned pigs from sows fed SDP had significantly lower (p < 0.05) serum concentrations of cortisol and TGF-β1 on d 3 and 7 postweaning, respectively, than CON piglets. In conclusion, SDP supplementation in sow diets from late gestation to weaning improved the productive performance of sows and their offspring; the beneficial effects of SDP may be mediated in part through modulation of immune responses of both sows and piglets.
This paper present an 8kbps ACLMS-MPC(Amplitude Compensation and Least Mean Square - Multi Pulse Coding) coding method integrated with ACFBD-MPC(Amplitude Compensation Frequency Band Division - Multi Pulse Coding) and LMS-MPC(Least Mean Square - Multi Pulse Coding) used V/UV/S(Voiced / Unvoiced / Silence) switching, compensation in a multi-pulses each pitch interval and Unvoiced approximate-synthesis by using specific frequency in order to reduce distortion of synthesis waveform. In integrating several methods, it is important to adjust the bit rate of voiced and unvoiced sound source to 8kbps while reducing the distortion of the speech waveform. In adjusting the bit rate of voiced and unvoiced sound source to 8 kbps, the speech waveform can be synthesized efficiently by restoring the individual pitch intervals using multi pulse in the representative interval. I was implemented that the ACLMS-MPC method and evaluate the SNR of APC-LMS in coding condition in 8kbps. As a result, SNR of ACLMS-MPC was 15.0dB for female voice and 14.3dB for male voice respectively. Therefore, I found that ACLMS-MPC was improved by 0.3dB~1.8dB for male voice and 0.3dB~1.6dB for female voice compared to existing MPC, ACFBD-MPC and LMS-MPC. These methods are expected to be applied to a method of speech coding using sound source in a low bit rate such as a cellular phone or internet phone. In the future, I will study the evaluation of the sound quality of 6.9kbps speech coding method that simultaneously compensation the amplitude and position of multi-pulse source.
