Identification of GATA2 and AP-1 Activator Elements within the Enhancer VNTR Occurring in Intron 5 of the Human SIRT3 Gene

  • Human SIRT3 gene contains an intronic VNTR enhancer. A T > C transition occurring in the second repeat of each VNTR allele implies the presence/absence of a putative GATA binding motif. A partially overlapping AP-1 site, not affected by the transition, was also identified. Aims of the present study were: 1) to verify if GATA and AP-1 sites could bind GATA2 and c-Jun/c-Fos factors, respectively; 2) to investigate whether such sites modulate the enhancer activity of the SIRT3-VNTR alleles. DAPA assay proved that GATA2 and c-Jun/c-Fos factors are able to bind the corresponding sites. Moreover, co-transfection experiments showed that the over-expression of GATA2 and c-Jun/c-Fos factors boosts the VNTR enhancer activity in an allelic-specific way. Furthermore, we established that GATA2 and c-Jun/c-Fos act additively in modulating the SIRT3-VNTR enhancer function. Therefore, GATA2 and AP-1 are functional sites and the T > C transition of the second VNTR repeat affects their activity.

Tree Diversity Changes over a Decade (2003-2013) in Four Inland Tropical Dry Evergreen Forest Sites on the Coromandel Coast of India

  • Forest tree diversity inventory and its periodical monitoring are important to understand changes in tree population structure and to provide information useful for biodiversity conservation and reserve management. In a long-term forest dynamics program in Indian tropical dry evergreen forest, this communication deals with tree diversity changes at decadal interval. The initial inventory of tree diversity was carried out in 2003, in four tropical dry evergreen forest sites - (much disturbed sites Shanmuganathapuram - SP and Araiyapatti - AP and moderately disturbed sites - Karisakkadu - KR and Maramadakki - MM) on the Coromandel Coast of peninsular India, by establishing four 1ha permanent plots, one in each site. In 2013, the four plots were re-inventoried for tree diversity (${\geq}10cm\;gbh$) changes which yielded 56 species from 46 genera and 26 families. The studied forest sites are threatened by disturbance due to multiple reasons; cutting of trees inside of the forest, grazing by goats, construction of temple approach road, and some aspects cultural attachment of local people like constructing new, additional strctures of temple by denuding a portion of forest etc.. Tree species richness over a decade increased by four species in site SP, two species in site AP, and one species in site KR, but decreased by one species in site MM. Tree density decreased drastically by 480 (28.92%) and 102 (12.63%) stems $ha^{-1}$ respectively in sites SP and AP, but moderately increased by 82 (12.09%) stems $ha^{-1}$ in site KR and 26 (3.46%) stems $ha^{-1}$ in site MM. Tree basal area declined in site KR from $21.6m^2$ to $20.26m^2ha^{-1}$ and in site SP from 21.1 to $20.38m^2ha^{-1}$, but increased from $19.1m^2$ to $19.43m^2$ and from 15.5 to $18.63m^2ha^{-1}$ in sites AP and MM respectively. Three tree species (Allophylus serratus, Maytenus emarginata and Ehretia pubescens) were lost out of the 57 species recorded in 2003, and two species (Jatropha gossypiifolia and Streblus asper) were new additions in ten years. The long-term forest monitoring data will be valuable to understand forest dynamics and for conservation and management of this and similar tropical forests.

A Study on Clinical Classification and Characteristic of Children with Recurrent Abdominal Pain (만성(慢性) 반복성(反復性) 복통(腹痛)을 주증(主症)으로 하는 환아(患兒)의 임상적(臨床的) 특징(特徵)에 관한 연구(硏究) -기능성 복통을 중심으로-)

  • Purpose : This Study was conducted to evaluate clinical characteristic of children with recurrent abdominal pain (RAP) and to be classified by its six subtype in the Oriental Pediatric Text Book and to find out relationship of western classification. Methods : Patients who visited Dong-Eui Oriental Medical hospital from August, 2001 to October, 2002 due to RAP were included. According to questionnaire and history taking, RAP was classified by its six subtype based on Oriental medical theory. Results : 1. Patients with RAP were more internalized, have a close relation with their parents, and have strong desires of success, but social intercourse is low. 2. 76% of Patients have a less desire to eat and 67% of Patients have a diarrhea or constipation. 3. According to questionnaire, first abdominal pain was their $3{\sim}5$ ages most, cause of occurrence was more 'eating cold foods' most, time of AP (abdominal pain) was $1{\sim}2$ hours after eating and no characteristic most, site of AP was the umbilicus most, shape of AP was impotent pain most, cause of reduce pain was abdominal massage and defection most. 4. frequency of RAP's type, AP caused by diet(食積腹痛) is 45.5%, AP caused by cold(寒腹痛) is 29.1%, AP caused by cold in internal organs of deficiency(臟腑虛冷腹痛) is 12.7%, stagnation of qi and stasis of blood(氣滯血瘀腹痛) is 10.9%, AP caused by internal diet and external cold(內食外寒腹痛) is 1.8%. There is no AP caused by parasites(蟲腹痛). 5. During clinical classifications of RAP, cause of occurrence was most important cause of reduce pian, defection practice was helpful for diagnosis, but shape of AP, site of AP was not helpful. 6. With relationship of Oriental classification and western classification, AP caused by diet is similar to dysmotilitylike dyspepsia and irriTable bowel syndrome. AP caused by cold is similar to irriTable bowel syndrome. AP caused by cold in internal organs of deficiency is similar to unspecified dyspepsia. stagnation of qi and stasis of blood and AP caused by internal diet and external cold is not like to western classification. Conclusion : RAP in Childhood is most occurred by food and cold. there is few AP caused by stagnation of qi and stasis of blood and internal diet and external cold. So the study on subclassification and clinical Manifestations of RAP in Childhood is more performed.

Transcriptional Regulation of the Murine Dopamine Receptor Regulating Factor (DRRF) Gene (생쥐 도파민 수용쳬 조절인자 (DRRF) 유전자의 전사조절)

  • The murine dopamine receptor regulating factor (DRRF) gene is transcribed from a TATA-less promoter that has several putative Sp1 binding sites. The present investigation identifies functional transcription factors that modulate the expression of this gene, In the $D_2-expressing$ NB41A3 cells, Spl potently activates transcription from the DRRF promoter in pCAT-DRRF-1153/+17, but DRRF effectively inhibits it. Deletion of the 31 bp fragment between -1153 and -1122 decreased transcription down to about $60\%$. This fragment contains a functional API binding site. In addition, deletion of the 129 bp region between -901 and -772 further decreased transcription. The latter region has a functional AP2 binding site. Using a DRRF_AP1 (bases -1153 to -1121) probe, a specific retarded band was observed, and the unlabeled AP1 consensus competitor could effectively compete away this retarded band. In addition, using a DRRF_AP2 (bases -873 to -846), a specific retarded band was observed, and the unlabeled AP2 consensus competitor could effectively compete away this retarded band. The present observations suggest that Spl and DRRF regulate the DRRF promoter and that both API and AP2 also modulate this gene.

Expression Site of Protoporphyrinogen Oxidase Influences on Herbicide Resistance in Transgenic Rice (형질전환 벼에서 Protoporphyrinogen Oxidase의 발현 위치가 제초제 저항성에 미치는 영향)

  • The effect of Protox expression site on herbicidal resistance was investigated in wild-type and transgenic rice plants imposed by peroxidizing herbicide oxyfluorfen. The transgenic rice systems involved the plastidal expression of Arabidopsis protoporphyrinogen oxidase (Protox; AP line) and the dual expression of Myxococcus xanthus Protox in chloroplasts and mitochondria (TTS line). The oxyfluorfen-treated TTS4 line showed the lower levels of cellular leakage and malonyldialdehyde and the sustained capacity of 5-aminolevulinic acid synthesis, compared to the oxyfluorfen-treated AP and wild-type lines. During oxyfluorfen action, the TTS4 line had greater herbicide resistance than the AP1 line, indicating that the dual expression of M. xanthus Protox in chloroplasts and mitochondria prevented the accumulation of photodynamic protoporphyrin IX more effectively than the expression of Arabidopsis Protox only in chloroplasts. These results suggest that the ectopic expression of Protox in mitochondria greatly contributes to the herbicidal resistance in rice plants.

SIMULATED AP1000 RESPONSE TO DESIGN BASIS SMALL-BREAK LOCA EVENTS IN APEX-1000 TEST FACILITY

  • As part of the $AP1000^{TM}$ pressurized water reactor design certification program, a series of integral systems tests of the nuclear steam supply system was performed at the APEX-1000 test facility at Oregon State University. These tests provided data necessary to validate Westinghouse safety analysis computer codes for AP1000 applications. In addition, the tests provided the opportunity to investigate the thermal-hydraulic phenomena expected to be important in AP1000 small-break loss of coolant accidents (SBLOCAs). The APEX-1000 facility is a 1/4-scale pressure and 1/4-scale height simulation of the AP1000 nuclear steam supply system and passive safety features. A series of eleven tests was performed in the APEX-1000 facility as part of a U.S. Department of Energy contract. In all, four SBLOCA tests representing a spectrum of break sizes and locations were simulated along with tests to study specific phenomena of interest. The focus of this paper is the SBLOCA tests. The key thermal-hydraulic phenomena simulated in the APEX-1000 tests, and the performance and interactions of the passive safety-related systems that can be investigated through the APEX-1000 facility, are emphasized. The APEX-1000 tests demonstrate that the AP1000 passive safety-related systems successfully combine to provide a continuous removal of core decay heat and the reactor core remains covered with considerable margin for all small-break LOCA events.

Quantitative Assay for the Binding of Jun-Fos Dimer and Activator Protein-1 Site

  • The Jun and Fos families of eukaryotic transcription factors form heterodimers capable of binding to their cognate DNA enhancer elements. We are interested in searching for inhibitors or antagonists of the binding of the Jun-Fos heterodimer to the activator protein-1 (AP-1) site. The basic-region leucine zipper (bZIP) domain of c-Fos was expressed as a fusion protein with glutathione S-transferase, and allowed to form a heterodimer with the bZIP domain of c-Jun. The heterodimer was bound to glutathione-agarose, to which were added radiolabeled AP-1 nucleotides. After thorough washing, the gel-bound radioactivity was counted. The assay is faster than the coventional electrophoretic mobility shift assay because the gel electrophoresis step and the autoradiography step are eliminated. Moreover, the assay is very sensitive, allowing the detection of picomolar quantities of nucleotides, and is not affected by up to 50% dimethylsulfoxide, a solvent for hydrophobic inhibitors. Curcumin and dihydroguaiaretic acid, recently known inhibitors of Jun-Fos-DNA complex formation, were applied to this Jun-GST-fused Fos system and revealed to decrease the dimer-DNA binding.

Natural Ripening versus Artificial Enhancing of Silty Reclaimed Tidal Soils for Upland Cropping Tested by Profile Characterization

  • This study was performed to produce basic data for silty reclaimed tidal lands and to develop techniques of environmentally-friendly utilization in agricultural system. We chose the two sites in Saemangeum reclaimed tidal lands, one (Site I) has been treated with cultivating green manure and conducting the desalinization process through submergence since April, 2007 and the other (Site II) has been under natural condition without artificial treatment. In situ and ex situ physic-chemical properties were determined and comparisons were made for soil profiles examined at these two sites in April 2009. Surface soil of Site I had lower EC and higher field saturated hydraulic conductivity than those of Site II, uncultivated land. Especially, exchangeable sodium content was lowest in Site I Ap1 layer than in other layers. This is probably due to flooding desalination and green manure cultivation. Besides, Ap1 and A2 layers of soil profile in Site I showed brighter soil color and more root observation than those of Site II. This is probably due to green manure cultivation. By the large, for high cash upland crops and intensive agricultural use of silty reclaimed tidal land, site-specific soil ripening such as flooding desalination and green manure cultivation could be useful.

Inhibitory Effect of Amentoflavone of Selaginella Tamariscina on MMP-9 Expression through NF-${\kappa}$B and AP-1 in Macrophage Raw 264.7 cells

  • The French paradox has been attributed to the antioxidant properties of flavonoids present in the red wine. Amentoflavone(AF) is a bi-flavonoid compound with anti-fungal and anti-inflammatory activities. We isolated AF from Selaginella tamariscina, and studied its effects on nuclear factor-B(NF-B)-mediated MMP-9 gene expression in RAW264.7 cells. AF blocked the lipopolysaccharide(LPS)-induced expression of MMP-9. Zymographic and immunoblot analyses showed that AF suppressed LPS-induced MMP-9 expression in a dose-dependent manner. To clarify the mechanistic basis for its inhibition of MMP-9 induction, we examined the effect of AF on the transactivation of MMP-9 gene by luciferase reporter activity using -1.59 kb flanking region. AF potently suppressed the reporter gene activity. This inhibition was characterized by down-regulation of MMP-9, which was transcriptionally regulated at NF-B site and activation protein-1 (AP-1) site in the MMP-9 promoter, two important nuclear transcription factors that are involved in MMP-9 expression. These findings indicate the efficacy of AF in inhibiting MMP-9 expression through the transcription factors NF-B and AP-1 on LPS-induced RAW264.7 cells.

Inhibitory Effect of Paeoniflorin on Fos-Jun-DNA Complex Formation and Stimulation of Apoptosis in HL-60 Cells

  • The Fos-Jun heterodimers are part of the regulatory network of gene expression and nuclear proteins encoded by proto-oncogenes. The activation of Fos-Jun is important in the transmission of the tumor-promoting signal from the extracellular environment to the nuclear transcription mechanism. To search for the inhibitors of the Fos-Jun DNA complex formation, several natural products were screened and water-soluble paeoniflorin reduced the binding activity of the Fos-Jun heterodimer. This active compound was purified by silica gel column chromatography and HPLC. The electrophoresis mobility shift assay and reverse-phase HPLC test showed that paeoniflorin reduced the AP-l function. The cytotoxic effect of paeoniflorin was observed in HL-60. These results indicate that paeoniflorin blocks the Fos-Jun heterodimer-binding site of the AP-l DNA and it also has cytotoxic effects on human leukemia cell lines.
