15d-PGJ2 inhibits NF-κB and AP-1-mediated MMP-9 expression and invasion of breast cancer cell by means of a heme oxygenase-1-dependent mechanism

  • Activation of peroxisome proliferator-activated receptor γ (PPARγ) serves as a key factor in the proliferation and invasion of breast cancer cells and is a potential therapeutic target for breast cancer. However, the mechanisms underlying this effect remain largely unknown. Heme oxygenase-1 (HO-1) is induced and over-expressed in various cancers and is associated with features of tumor aggressiveness. Recent studies have shown that HO-1 is a major downstream target of PPARγ. In this study, we investigated the effects of induction of HO-1 by PPARγ on TPA-induced MMP-9 expression and cell invasion using MCF-7 breast cancer cells. TPA treatment increased NF-κB /AP-1 DNA binding as well as MMP-9 expression. These effects were significantly blocked by 15d-PGJ2, a natural PPARγ ligand. 15d-PGJ2 induced HO-1 expression in a dose-dependent manner. Interestingly, HO-1 siRNA significantly attenuated the inhibition of TPA-induced MMP-9 protein expression and cell invasion by 15d-PGJ2. These results suggest that 15d-PGJ2 inhibits TPA-induced MMP-9 expression and invasion of MCF-7 cells by means of a heme oxygenase-1-dependent mechanism. Therefore, PPARγ/HO-1 signaling-pathway inhibition may be beneficial for prevention and treatment of breast cancer.

An investigation Of IntraFraction Motion Correction For Lung Stereotactic Body Radiation Therapy By Using IntraFraction Cone Beam Computed Tomography (폐암 환자의 정위적 체부 방사선 치료 시 IntraFraction CBCT를 이용한 치료 중 자세 오차 교정에 대한 고찰)

  • Purpose: The purpose is to correct for position errors caused by long treatment times. By correcting the target motion that can occur during lung SBRT using IntraFraction CBCT. Methods and materials: We analyzed retrospectively the IFM data of 14 patients with two treatment arc in the treatment plan for lung cancer with stereotactic radiotherapy. An IntraFraction Motion was applied to the Arccheck phantom to acquire the Gamma index data. Results : IntraFraction Motion during the first treatment arc is in the left-right(LR), superiorinferior(SI), anterior-posterior(AP) directions were $0.16{\pm}0.05cm$, 0.72 cm(max error), $0.2{\pm}0.14cm$, 1.26 cm, $0.24{\pm}0.08cm$, 0.82 cm and rotational directions was $0.84{\pm}0.23^{\circ}$, $2.8^{\circ}$(pitch), $0.72{\pm}0.23^{\circ}$, $2.5^{\circ}$(yaw), $0.7{\pm}0.19^{\circ}$, $2^{\circ}$(roll). IntraFraction Motion during the second treatment arc is in the LR, SI, AP directions were $0.1{\pm}0.04cm$, 0.37 cm, $0.14{\pm}0.17cm$, 2 cm, $0.12{\pm}0.04cm$, 0.5 cm and rotational directions was $0.45{\pm}0.12^{\circ}$, $1.3^{\circ}$, $0.37{\pm}0.1^{\circ}$, $1^{\circ}$, $0.35{\pm}0.1^{\circ}$, $1.2^{\circ}$. Gamma index pass rates were $82.64{\pm}10.51%$, 48.4 %. Conclusions : In this study, we examined the validity of IntraFraction Motion correction in lung SBRT and the efficiency of IntraFraction CBCT. Due to the nature of SBRT treatment, IFM may increase due to the increased treatment time. It is believed that the increase in IFM with the increase in treatment time can be improved with the use of FFF Beam and additional position correction using CBCT during treatment.

The Efficacy of Needle-Acupuncture and Moxibustion at BL18 on Hepatic Injury in Dogs (간유(肝兪)(BL18)에 대한 자침 및 뜸 요법의 개 간 손상 회복에 미치는 효과)

  • The present study was performed to elucidate the recovery effect of needle-acupuncture (needle-AP) and moxibustion treatments at Gan Shu (BL18) on $CCl_4$ induced hepatic injury in dogs, Total 9 clinically healthy experimental dogs (1 to 2 years old, 2.3-5.3kg body weight) were divided into control (3 dogs), needle-AP (3 dogs) and moxibustion (3 dogs) groups, respectively. Hepatic iniw was induced by intraperitoneal injection with $CCl_4$ (olive oil : $CCl_4=1:1,\;1ml/kg$, once/day). As far the treatments in each group, control group was not treated at all after induction of the hepatic injury till the end of experiment. Needle-Af group was treated by AP at BL18 (once/day, 20 minutes) from the next day of induction of the hepatic injury for 11 days. Moxibustion group was also treated with commercial moxa at BL18 (once/day, 20minutes) for 11days. The changes of the serum alanine transaminase(ALT), aspartate transaminase(AST) and gamma glutamyl transpeptidase (GGT) activities were investigated on pre, 0, 1, 3, 5, 7 and 11 days after hepatic injury, respectively. Histopathological changes were also investigated in the liver tissues on day 11 in experimental and control groups. The results obtained in the present study were as follows. In needle-AP group, they showed significant lower values on the 1st (p<0.05) and $3^{rd} day (p <0.05)$ in serum ALT activities, and the $11^{th}$ day in serum GGT activities, compared with those of control group, respectively. However, the significances were not detected in serum AST activities of needle-AP group by comparison with those of control group. In moxibustion group, they showed significant low values on the $1^{st}(p<0.05)$, 3rd day (p<0.05) and $5^{th}$ day (p<0.05) in serum ALT activities, compared with those of control group, respectively. However, significances were not detected in serum GGT and AST activities of moxibustion group by comparison with those of control group. Among the experimental groups, the significant low values were found on $1^{st}\;and\;3^{rd}$ day in serum ALT activities (p<0.05), and $3^{rd}\;and\;5^{th}$ day in serum GGT activities (p<0.05) in needle-AP group, compared by those of moxibustion group, respectively. In addition, the marked recovery findings of histopathological changes in experimental groups were found, compared by those of control group: mild vacuolar degeneration and necrosis findings in needle-AP group, and moderate vacuolar degeneration and necrotic findings in moxibustion group were found, in contrast to severe changes such as accumulation of bile juice, vacuolar degeneration and necrotic findings observed in control group. In conclusion, needle-AP and moxibustion treatments at BL-18 were effective for the recovery for $CCl_4$ induced hepatic injury and needle-AP was more effective than that of moxibustion in dogs.

The Effect of Phaseolus Angularis Shell on Soyang-in Metabolic Syndrome with Obesity (적소두(赤小豆) 외피(外皮)(Phaseolus angularis shell)의 고지방식이로 유도된 비만 동물모델에서 항비만, 항고지혈증 효과)

  • Objectives This experimental study was designed to investigate the effect of Phaseolus angularis shell on metabolic syndrome. Methods Each 5 C57BL/6J mice were randomly assigned to normal diet group, high-fat diet(HFD) control group, high-fat diet plus 15.6 mg/kg/day of Orlistat(HFD-Orlistat) group, high-fat diet plus 100mg/kg/day of Phaseolus angularis shell extract(HFD-PAS_E) group. Weight, the blood chemical and hematologic parameter was med. The mRNA expression was assayed through Reverse transcriptase polymerase chain reaction(RT-PCR). Results In HFD-PAS_E group, the body weight gain, weight of liver, and the level of LDL-Cholesterol were significantly decreased and the level of HDL-Cholesterol were significantly increased. The size of adipocyte in HFD-PAS_E group was smaller than HFD group's. In HFD-PAS_E group, the expression of leptin, PPAR-${\gamma}$, AP2/FABP4 mRNA in liver adipocyte tissue was decreased, the expression of Adiponectin, UCP-2 mRNA in liver adipocyte tissue was increased and the expression of Leptin, C/EBP-a, AP2/FABP4 mRNA in epididymal adipocyte tissue was decreased. Conclusion These results suggest that Phaseolus angularis shell has inhibitory effects on metabolic syndrome by reducing the body weight and the levels of lipid contents in high-fat-diet induced obese mice.

The Effects of Prunus Armeniaca Linne Var Fractions on Th2 Cytokine Expression and Atopic Dermatitis of NC/Nga Mouse (행인(杏仁) 분획물이 Th2 cytokine 발현과 NC/Nga mouse의 아토피 피부염에 미치는 영향)

  • Objectives PRAL (Prunus armeniaca Linne Var) has been known to suppress allergic reaction. However, the cellular target and its mechanism of action were unclear. This study was designed to investigate the effect of PRAL on RBL-2H3 mast cell, which is PMA-Ionomycin-induced activated in vitro and the effect of PRAL on the MNC/Nga mice that are DNCB-induced activated in vivo. Methods In this study, IL-4, IL-13 production were examined by ELISA analysis; IL-4, IL-13, IL-31, IL-31Ra, $TNF-{\alpha}$ and GM-CSF mRNA expression were examined by Real-time PCR; manifestations of AP-1 and MAPKs transcription factors were examined by western blotting in vitro. Then skin rashes have been evaluated and verified the distribution of mast cells by H&E and toluidine blue. Also, WBC, eosinophil and neutrophil, IgE level in serum, $IFN-{\gamma}$, IL-4, IL-5 in the splenocyte culture supernatant, the absolute cell numbers of $CD4^+$, $CD8^+$, $Gr-1^+CD11b^+$, $B220^+CD23^+$, $CD3^+CD69^+$ in the Axillary Lymph Node (ALN), PBMCs and dorsal skin and IL-5, IL-13, IL-31, IL-31Ra in the dorsal skin by Real-time PCR were all evaluated from the NC/BNga mice. Results As a result of this study, the mRNA expression of IL-4, IL-13, IL-31, IL-31Ra and $TNF-{\alpha}$ and IL-4, IL-13 production, shown in ELISA analysis, were suppressed by PRAL. Results from the western blot analysis showed decrease on the expression of mast-cell-specific transcription factors, including AP-1 and p-JNK, p-ERK. Histological examination showed that infiltration levels of immune cells in the skin of the AD-induced NC/Nga mice were improved by PRAL orally adminstration. Orally- administered PRAL group also showred decreased level of IgE in the serum. This group has shown decreased the level of IL-4, IL-5, but shown elevated $IFN-{\gamma}$ level in the splenocyte culture supernatant. The same group also has shown decreased cell numbers of $CD4^+$, $CD8^+$, $CD3^+CD69^+$ in the ALN, and $CD4^+$, $Gr-1^+CD11b^+$ in the dorsal skin. PRAL oral adminstration increased cell numbers of $CD4^+$, but decreased cell numbers of $CD8^+$, $Gr-1^+CD11b^+$, $B220^+CD23^+$ in the PBMCs. Conclusions Obtained results suggest that PRAL can regulate molecular mediators and immune cells that are functionally associated with atopic dermatitis (AD) induced in the NC/Nga mice. This may play an important role in recovering AD symptoms and suppressing pruritus.

Study on the surface modification of zirconia with hydrophilic silanes (친수성기를 가진 실란을 이용한 지르코니아의 표면의 개질 연구)

  • Since microzirconia has excellent thermal and mechanical properties with high chemical and electrical resistance, it can be used in various fields. When the surface of zirconia becomes hydrophilic, its dispersibility in water will be improved as well as the resistance to most hydrophobic contaminants will be increased. In this study, we investigated the introduction of a hydrophilic groups on the microzircornia surface through hydrolysis and condensation reactions with two different silanes containing hydrophilic functional groups, such as ${\gamma}$-aminopropyltrimethoxysilane (APS) and ${\gamma}$-ureidopropyltrimethoxysilane (UPS) at different pH and concentration conditions. A covalent bond formation between the surface hydroxyl groups of zirconia and that of hydrolyzed silanes was confirmed by ninhydrin test and FT-IR spectroscopy. However, the presence of Si on the surfaces of both silane modified microzirconias was unable to detect by SEM/EDS technique. In addition, particle size analysis results provide that the size of microzirconia was changed to smaller or bigger than that of original zirconia due to crushing and aggregation during the modification process. The water dispersibility was improved for only APS modifed zirconia (AS-2 and AS-3) under neutral pH condition, but the water dispersibility and stability for all cases of 0.5~2% UPS modifed zirconia (US series) were much improved.

Anti-obesity Effect of Yeoldahanso-tang on Obesity Mice (고지방식이로 유도된 비만 생쥐모델에서 열다한소탕의 항비만효과)

  • Objectives This study was conducted to evaluate the effects of Yeoldahanso-tang on obesity in rats induced by high fat diet experimentally. Methods The experiment was conducted with rats divided into 5 groups. Rats were evaluated for change of weight, hematologic and serum biochemical parameters. Results Yeoldahanso-tang group showed significant reductions in FER, body weight, adipose tissue weight and size. The level of Creatinine, glucose, ALP, T-cholesterol, Triglyceride, LDL-cholesterol, Leptin and IGF-1 of Yeoldahanso-tang group was significantly lower than those in HFD-CTL group. The level of HDL-cholesterol, free fatty acid and Adiponectin in Yeoldahanso-tang group was significantly higher than those in HFD-CTL group. As compared with HFD-CTL group, AMPK-${\alpha}1$, UCP2 and adiponectin mRNA in liver of Yeoldahanso-tang group were significantly increased and AP2/FABP4, AMPK-${\alpha}2$ and PPAR-${\gamma}$ mRNA in liver of Yeoldahanso-tang group was significantly decreased. Conclusion These results suggest that Yeoldahanso-tang has inhibitory effects on obesity in high fat diet induced obese mice.

Investingation on the Relationship Scoliotic Curve and BMI, Cervical Lordosis Lumar Lordosis and Ferguson Angle in Spinal Scoliosis Patient (척추측만증 환자의 척추만곡도에 관한 고찰)

  • Objectives: To invesgate correlation between the scoliotic curve and BMI, cervical lordosis, lumbar lordosis and Ferguson angle in spinal scoliotic patient. Methods: The study was composed of 14 scoliosis patients beyond cobb'a angle $10^{\circ}$ (sample I group) and 15 scoliosis patients over cobb'a angle $10^{\circ}$ (sample II group). The patients were evaluated with X-ray findings of full spine AP and lateral views and statistically analyzed. Results: 1. Sample II group showed a significant decrease in BMI as compared with Sample I group(P<0.05). 2. Scoliotic curve had s negative relationship with $BMI({\gamma}=0.406)$ 3. Scolotic patients had a lower cervical angle than normal man. Conclusion: 1. The larger the scoliotic angle, the lower BMI 2. There are no concemed scoliotic curve and cervical lordosis, lumbar lordosis and Ferguson angle.

Construction of voxel head phantom and application to BNCT dose calculation (Voxel 머리팬텀 제작 및 붕소중성자포획요법 선량계산에의 응용)

  • Voxel head phantom for overcoming the limitation of mathematical phantom in depleting anatomical details was constructed and example dose calculation for BNCT was performed. The repeated structure algorithm of the general purpose Monte Carlo code, MCNP4B was applied for yokel Monte Carlo calculation. Simple binary yokel phantom and combinatorial geometry phantom composed of two materials were constructed for validating the voxel Monte Carlo calculation system. The tomographic images of VHP man provided by NLM(National Library of Medicine) were segmented and indexed to construct yokel head phantom. Comparison of doses for broad parallel gamma and neutron beams in AP and PA directions showed decrease of brain dose due to the attenuation of neutron in eye balls in case of yokel head phantom. The spherical tumor volume with diameter, 5cm was defined in the center of brain for BNCT dose calculation in which accurate 3 dimensional dose calculation is essential. As a result of BNCT dose calculation for downward neutron beam of 10keV and 40keV, the tumor dose is about doubled when boron concentration ratio between the tumor to the normal tissue is $30{\mu}g/g$ to $3{\mu}g/g$. This study established the voxel Monte Carlo calculation system and suggested the feasibility of precise dose calculation in therapeutic radiology.

GEMINI NEAR-IR PHOTOMETRY OF THE ARCHES CLUSTER NEAR THE GALACTIC CENTER

  • We present Near-IR photometry of the Arches cluster, a young and massive stellar cluster near the Galactic center. We have analyzed the high resolution (FWHM $\~$ 0.2") Hand K' band images in the Galactic Center Demonstration Science Data Set, which were obtained with the Gemini/Hokupa's adaptive optics (AO) system. We present the color-magnitude diagram, the luminosity function and the initial mass function (IMF) of the stars in the Arches cluster in comparison with the HST/NICMOS data. The IMF slope for the range of 1.0 < log (M/M$\bigodot$) < 2.1 is estimated to be ${\Gamma} = -0.79 {\pm} 0.16$, in good agreements with the earlier result based on the HST/NICMOS data [Figer et al. 1999, ApJ, 525, 750]. These results strengthen the evidence that the IMF of the bright. stars close to the Galactic center is much flatter than that for the solar neighborhood. This is also consistent with a recent finding that the IMFs of the bright stars in young clusters in M33 get flatter as the galactocentric distance decreases [Lee et al. 2001, astro-ph 0109258]. It is found that the power of the Gemini/ AO system is comparable, with some limits, to that of the HST/NICMOS.
