Effect of Triol Fraction of Ginseng Saponin on Filamentous Formation through Reduction in C-AMP Concentration in Bacillus subtizis. (Botcillus subtilis에서 인삼 saponin의 triol fraction에 의한 C-AMP농도 감소가 filament생성에 미치는 효과)

  • Bacillus subtilis grown on medium containing 0.1% trios fraction, 0.005% sodium deoxycholate, and 0.05% taurocholate respectively was shown to grow as filamentous form and contain 2.3 x 10-5M, 1 x 10-5M, and 2.3${\times}$10-5M of intracellular C-AMP, respectively. The concentration was 3 or 4 times lower than that of the control. But concentrations of extracellula: C-AMP were similar to that of the control. Such decrease in C-AMP concentration was shown to parallel with decrease in autolysin activity indicating 40% and 20% lower activity of Bacillus subtilis grown on medium containing 0.05% and 0. l% triol fraction respectively than that of the control, The activity was also shown to be in inverse proportion with the formation of filament .

  • In the present study, hormonal changes in uterine tissue and circulation were evaluated during the implantation period in rats in order to understand the mechanism by which implantation takes place. The results obtained were as follows. 1. Concentrations of serum estradiol and progesterone were significantly increased on days 4 and 5. 2. Concentration of estrogen receptor reached maximum on day 5 when implantation normally occurred in rats. On the other hand, progesterone receptor was gradually decreased, reaching the lowest on day 5. 3. Uterine PGs and cAMP concentrations were significantly increased on day 5. 4. Uterine PGs and cAMP concentrations in implant sites were significantly greater than those in non-implant sites. It is, therefore, concluded that prostaglandins and cAMP in uterine tissue as well as circulating ovarian steroid hormones were increased during the period of implantation, suggesting that these hormones might be actively involved in the process of implantation in rats.

Effect of PEO viscoelasticity on carbon dioxide absorption in aqueous PEO solution of AMP

  • Carbon dioxide was absorbed into aqueous polyethylene oxide (PEO) solution containing AMP in a flat-stirred vessel to investigate the effect of non-Newtonian rheological behavior of PEO on the rate of chemical absorption of $CO_2$, where the reaction between $CO_2$ and AMP was assumed to be a first-order reaction with respect to the molar concentration of $CO_2$ and AMP respectively. The liquid-side mass transfer coefficient, which was obtained from the dimensionless empirical equation containing the properties of viscoelasticity of the non-Newtonian liquid, was used to estimate the enhancement factor due to chemical reaction. PEO with elastic property of non-Newtonian liquid made the rate of chemical absorption of $CO_2$ accelerate compared with Newtonian liquid based on the same viscosity of the solution.

Saikosaponin Contents and cAMP Phosphodiesterase Inhibitory Activities in Bupleuri Radix Extracts According to Extraction Conditions (추출조건에 따른 시호건근 추출액 중의 Saikosaponin함량 및 cAMP Phosphodiesterase 저해활성)

  • Two major oleananesaponins in Bupleuri radix, saikosaponin $\alpha$ and d, were extracted at various solvent compositions and leaching temperatures. Solvent composition was varied at 0 to 100%(v/v) of ethanol-water and methanol-water, whereas leaching temperature was adjusted to te range of 25~45$^{\circ}C$. The most effective extractant and leaching temperature were 70% ethanol and 45$^{\circ}C$, respectively. However, no major differences in saikosaponin content and cAMP phosphodiesterase inhibition were found at various leaching times. The cAMP phosphodiesterase inhibitions were alo the highest when 70% ethanol was used as the extractant.

Epac: new emerging cAMP-binding protein

  • The well-known second messenger cyclic adenosine monophosphate (cAMP) regulates the morphology and physiology of neurons and thus higher cognitive brain functions. The discovery of exchange protein activated by cAMP (Epac) as a guanine nucleotide exchange factor for Rap GTPases has shed light on protein kinase A (PKA)-independent functions of cAMP signaling in neural tissues. Studies of cAMP-Epac-mediated signaling in neurons under normal and disease conditions also revealed its diverse contributions to neurodevelopment, synaptic remodeling, and neurotransmitter release, as well as learning, memory, and emotion. In this mini-review, the various roles of Epac isoforms, including Epac1 and Epac2, highly expressed in neural tissues are summarized, and controversies or issues are highlighted that need to be resolved to uncover the critical functions of Epac in neural tissues and the potential for a new therapeutic target of mental disorders.

  • Carbon dioxide was absorbed into water-in-oil (w/o) emulsion composed of aqueous 2-amino-2-methyl-1-propanol (AMP) droplets as a dispersed phase and benzene solutions of polybutene and polyisobutylene as a continuous phase in a flat-stirred vessel to investigate the effect of non-Newtonian rheological behavior on the rate of chemical absorption of $CO_2$, where the reaction between $CO_2$ and AMP in the aqueous phase was assumed to be a pseudo-first-order reaction. It was expressed that PIB with elastic property made the rate of chemical absorption of $CO_2$ accelerated by comparison of mass transfer coefficient of $CO_2$ in the non-Newtonian liquid with that in the Newtonian liquid.

The Removal of Carbon Dioxide using AMP+HMDA in Absorption/Regeneration Continuous Process (흡수/재생 연속공정에서 AMP+HMDA를 이용한 이산화탄소의 제거)

  • Increasing amounts of anthropogenic $CO_2$ emitted to the atmosphere are believed to be a significant factor in global climate change. Hence, the method of chemical absorption has been suggested to separate and recover acid gases such as $CO_2$. In this study, the characteristics of absorption and regeneration of $CO_2$ for the absorbent which adding HMDA (hexamethylenediamine) into AMP (2-amino-2-methyl-1-propanol), hindered amine, was investigated in lab-scale absorption/regeneration reactor. As a result of this study, the removal efficiency of $CO_2$ increased when adding $5.9\%,\;11.7\%\;and\;23.4\%$ HMDA into $30\%$ AMP respectively. Also, the removal efficiency of $CO_2$ increased $6.5\%,\;8.4\%,\;10.3\%$ respectively as compared to AMP alone when the gas flow rate was 7.5 SL/min. In addition, all absorbents used in the study revealed the high stripping efficiency, which was almost $99\%$, at the temperature of $110^{\circ}C$. Thus, the regeneration tower should be operated at $110^{\circ}C$. At this time, the concentration of exhausted $CO_2$ was higher than $99\%$.

Effects of Sipyukmiyukieum on DNA Synthesis, cAMP Synthesis and MHC-class II Expression of FRTL-5 Thyroid Cells (십육미류기음(十六味流氣飮)이 FRTL-5 갑상선 세포의 DNA와 cAMP의 합성 및 MHC-class II의 발현에 미치는 영향)

  • Objective: Graves' disease encompasses hyperthyroidism and diffused goiter associated with auto-antibodies to the thyroid stimulating hormone(TSH) receptors. In clinical environment, treatments of Graves' disease have many side effects such as recurrence and hypothyroidism. We've studied the effects of Sipyukmiyukieum on DNA synthesis, cAMP synthesis, and MHC-class II expression of FRTL-5 thyroid cells were studied. Methods: DNA synthesis was investigated by using BrdU staining and cAMP synthesis by ELISA kit, and expression of $interferon-{\gamma}$ activated MHC class II by Flow cytometer. Results: After introduction of Sipyukmiyukieum, significant inhibition of DNA synthesis. cAMP synthesis, and expression of $interferon-{\gamma}$ activated MHC class II of FRTL-5 thyroid cells was observed. Conclusions: Judging from these results, Sipyukmiyukieum has potential as a potent herbal treatment for inhibiting the enlargement of goiter, synthesis of abnormal thyroidal hormones, and autoimmuine responses of Graves' disease.
