백김치의 Amaranth투여 흰쥐에 대한 독성억제효과 (Inhibitive Effects of Baik-Kimchi Against Amaranth Toxicity in Rats)

  • 백김치의 생체내 Am독성억제효과를 확인하기 위하여 Am 식이에 10% 백김치(동결 건조분말) 첨가사료로 사육한 흰쥐 의 성장회복능을 실험한 결과를 요약하면 다음과 같다. Am 식이군의 섭취량이 양적으로 다른 군에 비해 적었으며, 기본 식이군 생배추식이군, 백김치식이군은 거의 같은 섭취량을 보였다. 기본식이군의 체중이 사육종료일에 289.4 g에 달하는데 비해 Am식이군은 229.8 g으로 기본식이군의 79.4%에 불과해 체중증가 현상이 크게 저조하였다. 그러나 백김치를 첨가한 식이군의 경우는 설사증상이 없어지고 체중도 기본 식이군의 경우와 비슷한 수준까지 회복상태를 보여 백김치의 Am에 대한 독성억제효과를 나타내었다. 변의 총배설량은 백김치식이군(1737.99 g/3 weeks)이 기본식이군의 약 15배, 생배추식이군의 약 1.6배로 백김치가 변부피를 증가시키는 데 가장 효과적인 것으로 확인되었다. 대장과 맹장의 무게는 생배추 및 백김치식이군에서 유의적 (P<0.05)인 증가를 나타내었다. 또한 신장의 무게는 Am식이군에서 유의적인 감소를 나타내어 Am의 독성이 신장과 깊은 상관성이 있음을 시사하고 있었다. Am식이군의 단백질(50.26%) 및 지질소화율 (58.37%)이 백김치식이군의 단백질(77.30%)및 지질소화율 (92.12%)에 비하여 매우 낮았는데, 이것은 이들 영양소의 낮은 소화율이 횐쥐의 체중저하에 직접적인 영향을 미친 것으로 생각되었다. 백김치의 수용성 및 불용성식이섬유는 소화관을 통과한 형태 변화가 뚜렷하게 관찰되었다. 변의 배설 형태는 백김치식이군의 변이 가장 부피가 크고 일정한 크기를 보였다.

Speciation and Solubility of Major Actinides Under the Deep Groundwater Conditions of Korea

  • The speciation and solubility of Am, Np, Pu and U have been analyzed by means of the geochemical code MUGREM, under the chemical conditions of domestic deep groundwater, in order to support the preliminary safety assessment for a Korean HLW disposal concept. Under the conditions of groundwaters studied, the stable solid phase is AmOHC $O_3$(s) or Am(OH)$_3$(s), soddyite((U $O_2$)$_2$ $SiO_2$.2$H_2O$) or N $a_2$ $U_2$ $O_{7}$ (c), Np(OH)$_4$(am), and Pu(OH)$_4$(am) for Am, U, Np, and Pu, respectively. The dominating aqueous species are as follows: the complexes of Am(III), Am(OH)$_2$$^{+}$ and Am(C $O_3$)$_2$$^{[-10]}$ , the complexes of U(VI), U $O_2$(OH)$_3$$^{[-10]}$ and U $O_2$(C $O_3$)$_3$$^{4-}$, the complexes of Np(IV), Np(OH)$_4$(aq) and Np(OH)$_3$C $O_3$, and the complexes of Pu(IV), Pu(OH)$_4$(aq) and Pu(OH)$_3$C $O_3$$^{[-10]}$ . The calculated solubilities exist between 1.9E-10 and 1.3E-9 mol/L for Am, between 5.6E-6 and 1.2E-4 mol/L for U, between 3.1E-9 and 1.3E-8 mol/L for Np, and between 6.6E-10 and 2.4E-10 mol/L for Pu, depending on groundwater conditions. The present solubilities of each actinide agree well with the results of other studies obtained under similar conditions.s.

폴리에틸렌 옥사이드가 암포테리신-B의 응집 특성 및 독성에 미치는 영향 (The Effect of Polyethylene Oxide on the Aggregation State and Toxicity of Amphotericin B)

  • Amphotericin B (AmB) is a drug of choice for the treatment of systemic fungal diseases, but its use is considerably limited due to a high incidence of toxicity, particularly nephrotoxicity. It has been demonstrated that the toxicity of AmB is caused by self-aggregated species of the drug and that unaggregated (monomeric) drug is nontoxic but still expresses antifungal activity. Poly (ethylene oxide) (PEO) is a water-soluble polymer, which may impact the aggregation state of AmB. We have studied the aggregation state of AmB as a function of PEO molecular weight and concentration. At 3,000 and 8,000 g/mole, there was minimal or no change of critical aggregation concentration (CAC) of AmB regardless of the concentration of polymer. By contrast at 20,000 g/mole, the CAC of AmB strikingly increased to 24.3 and $37.5\;{\mu}M$ at 5.0% and 10 % w/v of polymer, respectively. The critical overlap concentration (COC) of PEO 20,000 g/mole was 5.5%. It appears that an interaction between monomeric AmB and polymer coil increases above the COC, competing with self-aggregation of the drug. Accordingly, the degree of aggregation of AmB stayed low and the toxicity became less. There was no such effect at 3,000 and 8,000 g/mole of PEO, owing perhaps to small dimensions in comparison to AmB. Based upon these findings, less toxic AmB formulation may be developed by a pharmaceutical technique such as solid dispersion system containing both AmB and PEO 20,000 g/mole.

아스팔라톤의 토끼 위장관 점막 투과 및 효소적 분해 (Permeation and Enzymatic Degradation of Aspalatone in Gastrointestinal Tract of Rabbit)

  • To evaluate the site-specific permeation of aspalatone (acetylsalicylic acid maltol ester, AM) through gastrointestinal tract, the enzymatic degradation and permeation studies were carried out using gastric, duodenal and jejunal mucosae of rabbits. It was found that $15.2{\pm}11.4%$, $11.6{\pm}5.2$ and $0.8{\pm}0.6%$ of the donor dose of AM, salicylmaltol (SM) and aspirin (ASA) permeated through the upper gastric mucosa after 8 hr of permeation, respectively. After 8 hr of AM permeation, SM and ASA were measured to be $15.0{\pm}1.7$ and $2.6{\pm}0.8%$ of the dose in the donor solutions, respectively, and salicylic acid (SA) was not detected even after 6 hr, suggesting a very low gastric damage. For the gastric mucosa, the increase of donor dose from 100 to $1,000\;{\mu}g/ml$ increased the permeation flux dose-dependently (r=0.9905). For the duodenal and jejunal mucosae, however, AM was fully degraded into SM and SA due to the esterase activities within 30 min. AM and ASA were not detected in the receptor solution. This result indicates that AM is not a prodrug of ASA. Addition of potassium fluoride (0.5%) into the donor solution delayed the degradation of AM, but did not allow the permeation through duodenal mucosa even by the inhibition of esterase activity. The addition of $dimethyl-{\beta}-cyclodextrin$ and $2-hydroxypropyl-{\beta}-cyclodextrin$ (5%) into the donor solutions also did not show favorable effects on the permeation of AM through various mucosae. In comparison of permeation rates of AM and ASA through the upper gastric mucosa, the flux of ASA was 4.2 times faster than AM based on the molar concentration. ASA also was fully degraded in the donor solutions faced with duodenal and jejunal mucosae within 2 hr, and was not detected in the receptor solution, suggesting a slower metabolism compared with AM.

혼파초지에서 가축분뇨의 종류와 시용수준이 목초의 생산성 및 지력증진에 미치는 영향 (Effects of Animal Excreta Classification and Nitrogen Fertilizing Level on Productivity of Pasture Plants and Improvement of Soil Fertility in Mixed Grassland)

  • To establish the recycling system of animal manure(AM) for environmental preservation and improve the utilization of AM, this study was to investigate the effects of the types and nitrogen application rate of AM on herbage productivity, efficiency of nitrogen utilization, nutritive value and an increase of soil fertility and in mixed grassland. This sudy was arranged in split plot design. Main plots were the types of AM(Cattle feedlot manure, CFM; Pig manure fermented with sawdust, PMFS; cattle sluny, CS) and subplots were the application rate of animal manure, such as 100, 200 and 300kgNiha. I. DM yields of herbage were the highest with CS and decreased by application over ZOOkgNiha AM. 2. Crude protein(CP) ontent was the highest with CFM and followed by CS, and the lowest with PMFS, and increased as application rate of AM increased. 3. Nitrogen(N) yields of CS treatment was higher than that of CFM and CS. and increased significantly as application rate of AM increased(P<0.05). 4. The contents of NDF, ADF and TDN was hardly influenced by the types and application rate of AM. 5. Organic matter(0M) content in the soil was the highest with PMFS and followed by CFM and the lowest with CS. OM content increased significantly as application rate of AM increased(P<0.05). 6. Total nitrogen content of the soil was not affected by the type of AM, but increased significantly as application rate of AM increased(P<0.05). 형질전환 담배에서 Amaranthus 저장단백질인 AmA1 유전자의 발현 (Expression of AmA1 Gene Encoding Storage Protein of Amaranthus in Transgenic Tobacco)

  • Amaranthus hypochondriacus의 저장 단백질을 encoding 하는 AmA1 유전자를 RT-PCR 방법을 이용하여 분리하고 특성화하였다. AmAl 유전자를 담배에 형질전환 시키기 위해 CaMV 35S promoter와 3'NOS를 가지고 있는 식물 발현 vector에 subcloning 하고 이 재조합 vector를 이용하여 Agrobacterium-mediated형질전환 방법을 이용하여 담배에 도입시켰다 신초는 0.1 mg/L NAA, 1.0 mg/L BA, 100 mg/L kanamycin 그리고 250 mg/L cefotaxime이 첨가된 MS 선발 배지에서 선발했고, 선발된 신초는 식물 생장 조절제를 첨가 하지 않고 200 mg/L kanamycin과 250 mg/L cefotaxime이 첨가된 MS배지에서 뿌리를 유도하였다. 선발된 담배의 게놈내의 AmA1 유전자의 존재는 PCR 방법과 hybridization을 이용하여 확인되었고, AmA1 유전자의 발현은 RT-PCR방법과 Southern blot hybridization을 사용하여 확인되었다.

Complexation of Amphotericin B With Egg Phosphatidylcholine Liposomes

  • The complexation and physical characteristics of egg phosphatidylcholine (PC) liposome containing amphotericin B(AmB) were investigated through circular dichrosim(CD) spectra, the size distribution, the turbidity change, and the calcein release. CD spectra of AmB-containing egg PC mxture exhibited a positive peak around 330 nm indicative of complexation of AmB and four negative peaks. The positive peak increased up to $2.2{\;}millidegree/{\mu}g$ AmB as AmB contents increased up to 12% (w/w), suggesting that AmB-phospholipid complexation was promoted by the antibiotics. The effective diameter of liposomesa by dynamic light scattering decreased from 450 nm to 220 nm as the amount of AmB in liposomes increased from o to 30% (w/w). The complexation may be responsible for the reduction in size. On the other hand, at around 1 mN deoxycholate (DOC), the reltive turbidities of 5 and 10% (w/w) AmB-containing liposome suspension were less than 1 probably due to the soblubilization of the complex, while those of pure PC liposome suspension were larger than 1 at the same concentration. Deoxycholate-induced release of liposomes, indicating the intercalation of the drug into the bilayers. Therefore, it is concluded that in AmB/eggPC/water system, AmB-phospholipid complexcoexists with AmB-containing liposomes.

  • 적층제조(AM, 일명 3D프린팅)기술은 디자인 자유도가 높고 디지털화된 기술의 특성으로 품질데이터의 예측·관리가 용이한 형태로 존재한다. 이러한 이점으로 AM기술은 다양한 산업에 적용되고 있다. 특히 건축물과 기반시설을 AM기술로 제조하는 방법이 건설 산업에 제안되고 있다. 다만, 다소 부족한 기술의 역사와 품질 및 시공 관리방법의 미비, 제조상품의 인증과 같은 문제로 인해 기술사용이 제한되고 있다. 따라서 간접적인 형태로 AM기술을 활용하여 건축 상품제조를 구현하고 있다. 특히, 거푸집을 적층 제조하고 건축 소재를 투입하여 상품으로 구현하는 방법이 확인되고 있다. 본 연구에서는 대형크기의 거푸집을 적층제조하고 건축 상품을 구현하기 위해 하이브리드 압출적층제조를 활용한다. 또한, 적층제조과정에서 생산성과 경제성을 예측할 수 있는 인자를 확인한다. 결과적으로, 건축물의 구현결과와 생산 비용과 시간을 줄이기 위한 형상설계 최적화 방법을 제안한다.

상이한 에너지원을 이용하여 성장한 methylobacterium extorquens AM1내의 폴리아민 (Polyamines in methylobacterium extorquens AM1 grown on different energy sources)

  • Putrescine, spermidine, and spermine were found to persent in Methylobacterium extorquens AM1 growing on methanol, succinate, glucose, or nutrient broth as an energy source. Spermidine was found to be a major polyamine in cells growing on methanol or succinate, while putrescine to be the one in nutrient broth-grown cells. The overall content of polyamines in cells growing on glucose was less than that in cells growing on other substrates. Spermine was the most abundant polyamine in glucose-grown cells. Accumulation of polyamines in M. extorquens AM1 was maximal at the mid-exponential or early stationary phase during growth on each substrate. The effect of polyamines added into the medium on the polyamine composition in M. extorquens AM1 was variable. Each polyamine added into the nutrient broth medium was found to increase the amount of the respective polyamine in the cell. Exogeneously added polyamines had no effect on the growth of M. extorquens AM1.

Effects of Interspecific Interactions of Arbuscular Mycorrhizal Fungi on Growth of Soybean and Corn

  • Growth responses of Zea mays and Glycine max to colonization by mixture of combination of three species of arbuscular mycorrhizal (AM) fungi, two species of Glomus and a species of Scutellospora were compared. In Zea mays, plants inoculated with single species of AM fungi showed significantly higher in dry weight than non-mycorrhizal plant for all three AM fungal species. Also, growth of plants inoculated with spores of two species of AM fungi was significantly higher than nonmycorrhizal control except for plants inoculated with two Glomus species. When three species of AM fungi were inoculated, the plants showed the highest growth. In Glycine max, plants with single AM fungal species inoculation were not significantly different in plant growth from nonmycorrhizal plants. When the plants were inoculated with combination of two or more AM fungal species, their growth significantly increased compared to nonmycorrhizal plants. In both plant species, mycorrhizal root colonization by Scutellospora species was significantly lower than by Glomus species.
