Effects of Turmeric (Curcuma longa L.) Supplementation on Blood Urea Nitrogen and Enzyme Activities in Dyslipidemic Rats (울금(Curcuma longa L.)이 이상지질혈증 흰쥐의 혈액요소질소(BUN) 및 효소활성에 미치는 영향)

  • The purpose this study was to investigate the influences of 5% turmeric (Curcuma longa L.) supplementation on enzyme activities such as aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), lactate dehydrogenase (LDH), amylase, lipase and catalase in serum of dyslipidemic rats. Sprague-Dawley (SD) rats (24 male) were divided into four groups, namely the ND group (normal-nondyslipidemic diet), NT group (normal-nondyslipidemic diet+5% turmeric), DD group (control-dyslipidemic diet), and DT groups (dyslipidemic diet+5% turmeric). Serum concentrations of blood urea nitrogen (BUN), creatinine and uric acid were significantly decreased (p<0.05) by turmeric supplementation diet. The activities of AST, ALT, ALP, LDH, amylase and lipase in sera of turmeric diet group were significantly decreased (p<0.05). The catalase activity in serum of turmeric supplementation group was significantly increased than dyslipidemic diet (p<0.05). In vivo experiment with dyslipidemic rats showed that ingestion of turmeric were effective in kidney and hepatic functional enzyme activities. Which suggests that turmeric material could be used for further studies as a potential source for nutraceutical foods.

Analysis of attachment, proliferation and differentiation response of human mesenchymal stem cell to various implant surfaces coated with rhBMP-2 (다양한 rhBMP-2 코팅 임플란트 표면에서의 인간 간엽 줄기세포의 부착, 증식, 분화반응 분석)

  • Purpose: In this paper we tried to evaluate the most appropriate surface for rhBMP-2 coating among 4 rough titanium surfaces. Materials and methods: We used machined surface as a control group and anodized, RBM and SLA surfaces as test groups. We coated rhBMP-2 on the 4 surfaces and with uncoated surfaces for each case, we cultured human mesenchymal stem cells on all 8 surfaces. 24 hours after we measured the stem cell' attachment with SEM, and on 3rd, 7th, and 14th days, we checked the cell proliferation and differentiation by using MTT and ALP activity assay. And on the 7th day after the culture, we performed RT-PCR assay to determine whether the expression levels of Type I collagen, osteocalcin, osteopontin were changed. Results: We observed with SEM that 4 rhBMP-2 coated surfaces exhibited wider and tighter cell attachment and more cell process spreading than uncoated surfaces. The anodized rhBMP-2 surface caused robustest effects. In MTT assay we could not find any meaningful difference. In ALP assay there was a significant increase (P<.05) in the ALP activity of anodized rhBMP-2 coated surface compared with that of the control (3rd and 14th days) and with that of the RBM rhBMP-2 coated surface (14th day). In RT-PCR assay there was increased expressions in the anodized rhBMP-2 coated surface for osteocalcin, and osteopontin. Conclusion: We found that the anodized rhBMP-2 coated surface were most prominent stem cell attachment and differentiation in compared to control and Machined rhBMP-2 coated, RBM rhBMP-2 coated surface.

Establishment of a Murine Model for Radiation-induced Bone Loss in Growing C3H/HeN Mice (성장기 마우스에서 방사선 유도 골소실 동물모델 확립)

  • Bone changes are common sequela of irradiation in growing animal. The purpose of this study was to establish an experimental model of radiation-induced bone loss in growing mice using micro-computed tomography (${\mu}CT$). The extent of changes following 2 Gy gamma irradiation ($2Gy{\cdot}min^{-1}$) was studied at 4, 8 or 12 weeks after exposure. Mice that received 0.5, 1.0, 2.0 or 4.0 Gy of gamma-rays were examined 8 weeks after irradiation. Tibiae were analyzed using ${\mu}CT$. Serum alkaline phosphatase (ALP) and biomechanical properties were measured and the osteoclast surface was examined. A significant loss of trabecular bone in tibiae was evident 8 weeks after exposure. Measurements performed after irradiation showed a dose-related decrease in trabecular bone volume fraction (BV/TV) and bone mineral density (BMD), respectively. The best-fitting dose-response curves were linear-quadratic. Taking the controls into accounts, the lines of best fit were as follows: BV/TV (%) = $0.9584D^2-6.0168D+20.377$ ($r^2$ = 0.946, D = dose in Gy) and BMD ($mg{\cdot}cm^{-3}$) = $8.8115D^2-56.197D+194.41$ ($r^2$ = 0.999, D = dose in Gy). Body weight did not differ among the groups. No dose-dependent differences were apparent among the groups with regard to mechanical and anatomical properties of tibia, serum ALP and osteoclast activity. The findings provide the basis required for better understanding of the results that will be obtained in any further studies of radiation-induced bone responses.

Protective Effects of Extracts of Hovenia dulcis Thunb on Hepatotoxicity in Carbon Tetrachloride Intoxicated Rats (지구자나무 추출물이 사염화탄소로 유발된 흰쥐의 간손상에 미치는 영향)

  • This study was performed to investigate the protective effects of Hovenia dulcis Thunb on hepatotoxicity in carbon tetrachloride-intoxicated rats. Male Sprague-Dawley rats (220~240 g) were used as experimental groups, which were divided into 7 groups; Control group, $CCl_4$-treated group, hexane fraction pretreated and $CCl_4$-treated group, chloroform fraction pretreated and $CCl_4$-treated group, ethylacetate fraction pretreated and $CCl_4$-treated group, butanol fraction pretreated and $CCl_4$-treated group, $H_2O$ fraction pretreated and $CCl_4$-treated group. After 6 days, the activities of aminotransferase, contents of cholesterol, TG and hepatic lipid peroxide content in chloroform fraction pretreated and $CCl_4$-treated group were significantly decreased (p<0.05) compared to the only $CCl_4$-treated group. The content of glutathione and activities of GST in chloroform fraction pretreated and $CCl_4$-treated group were also significantly increased (p<0.05) compared to the only $CCl_4$-treated group. In addition, activities of SOD, catalase and GSH-Px in chloroform fraction pretreated and $CCl_4$-treated group were significantly decreased (p<0.05) compared to the only $CCl_4$-treated group. These results indicated that the chloroform fraction of Hovenia dulcis Thunb methanol extract showed hepatoprotective effect in carbon tetrachloride-intoxicated rats.

Changes in s-GOT and s-ALP Activities in the Korean Native Cows Within Estrus Cycle (한우(韓牛)의 성주기(性週期)에 따른 혈청(血淸) GOT 및 혈청(血淸) ALP 활성도(活性度)의 변동상에 관하여)

  • Changes in serum glutamic oxaloacetic transaminase (s-GOT) activities, serum alkaline phosphatase activities and serum total protein amounts were investigated on seven Korean native cows having normal estrus cycle of 18~24 days after normal parturition, dividing estrus cycle into estrus (0~1 days), metestrus (2~6 days), diestrus (7~16 days), proestrus (17~20 days). The results were as follows. 1. Serum GOT activities at estrus ranged from 73.5 to 121.5 Ku with a mean of $89.14{\pm}17.16$ Ku, at metestrus 57 to 89 Ku with a mean of $67.01{\pm}10.81$ Ku, at diestrus 54 to 89.5 Ku with a mean of $67.05{\pm}10.05$ Ku and at proestrus 53 to 112 Ku with a mean of $73.00{\pm}20.05$ Ku. The activities were significantly increased at the estrus comparing with other stages (P<0.01). 2. Serum ALP activities at estrus ranged from 8.0 to 10.4 K-Au with a mean of $8.74{\pm}0.83$ K-Au, at metestrus 6.5 to 9.2 K-Au with a mean of$7.74{\pm}0.76$ K-Au, at diestrus 5.6 to 9.0 K-Au with a mean of $7.67{\pm}1.13$ K-Au, at proestrus 6.2 to 9.3 K-Au with a mean of $7.12{\pm}1.18$ K-Au. The significance was not recognized among the stages of estrus cycle. 3. Serum total protein amounts at estrus ranged from 6.45 to 8.0g/10dl with a mean of $7.25{\pm}0.57/100dl$, at metestrus, 6.37 to 7.9g/100dl with a mean of $7.65{\pm}0.50g/100dl$, fat diestrus, 6.56 to 8.67g/100dl, with a mean of $7.53{\pm}0.55g/100dl$ and at proestrus 5.94 to 7.71g/100dl with a mean of $6.54{\pm}0.65g/dl$. There was not significance among the stages of estrus cycle.

Improvements Caused by Silk Sericin Extract Derived from Silkworm in Blood Glucose and Lipid Concentration in Diabetic Rats (누에 유래 실크 세리신이 당뇨성 흰쥐의 혈당 및 지질 농도 개선에 미치는 영향)

  • The objective of this study was to assess improvements caused by silk sericin extract in blood glucose and lipid concentrations in the sera of streptozotocin(STZ, 55 mg/kg B.W., I.P. injection)-induced diabetic rats fed on experimental diets for 5 weeks. Serum concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free cholesterol, cholesteryl ester ratio, triglyceride(TG) and phospholipids(PL) were remarkably higher in the diabetic group(group BSW) and STZ(I.P.)-plus-silk sericin extract group(group ESS) than in the control group(group CG, basal diet-plus-water). However, the serum concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free cholesterol, cholesteryl ester ratio, TG and PL were lower in the ESS group than in the BSW group, whereas the ratio of HDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration in the ESS group were higher than in the BSW group. The activities of alkaline phosphatase(ALP) and aminotransferase(AST, ALT) in serum were lower in the ESS group than in the diabetic BSW group. The results shown above suggested that silk sericin extract effectively improves blood glucose and lipid concentrations in the sera of STZ-induced diabetic rats.

Effect of Siho (Bupleurum falcatum L.) Extracts on Liver Function in $CCl_4$ Intoxicated rats (시호추출물이 간장기능 회복에 미치는 영향)

  • Effect of Siho (Bupleurum falcatum L.) extracts on liver function were investigated in CCl4 intoxicated rats. Plasma $\alpha$-fetoprotein and total protein concentration showed a tendency to decrease in Siho extracts groups. However plasma albumin concentration showed no significantly different in all treatment groups. Activities of plasma asparate aminotransferase and alanine aminotransferase in Siho extracts groups showed a lower values than control group. Alkan phosphatase and lactate dehydrogenase activities showed a tendency to decrease in Siho extracts groups. However ${\gamma}$-glutamyl transferase activity showed no significantly different in all treatment groups. Concentration of plasma triglyceride and total cholesterol showed no significantly different in all treatment groups.

Effects of the Prunus persica Batsch var. davidiana Max. Extract on the Lipid Compositions and Enzyme Activities in Hyperlipidemic Rats (돌복숭아(Prunus persica Batsch var. davidiana Max.) 추출액이 고지혈증 흰쥐의 생체 내 지질성분 및 효소활성에 미치는 영향)

  • The purpose of this study was designed to observe the effects of the feeding Prunus persica Batsch var. davidiana Max. extract on the concentrations of the lipids and blood glucose in the S.D. rats fed the experimental diets for 5 weeks. Concentrations of total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, triglyceride(TG), phospholipid(PL) and blood glucose in serum were significantly higher in the cholesterol administration groups (groups BCG (cholesterol+water), BCPG (cholesterol+ Prunus persica 5.0 g% extract) than those in the control group (group BG, basal diet+water). But the concentrations of total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free-cholesterol, cholesteryl ester, TG, PL and blood glucose in serum were remarkably lower in the group BCPG than those in the group BCG. In the ratio of HDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration, Prunus persica 5.0 g% extract administration group was higher percentage than in the group BCG. The activities of aspartate aminotransferase (AST), alanine aminotransferase(ALT), lactate dehydrogenase(LDH) and alkaline phosphatase (ALP) in serum were rather lower in the Prunus persica 5.0 g% extract administration group(group BCPG) than in the cholesterol diet group(group BCG). From the above research, Prunus persica Batsch var. davidiana Max. were effective on the improvement of the blood glucose, lipid compositions in serum of dietary hyperlipidemic rats. And particularly, Prunus persica Batsch var. davidiana Max. was more effective therapeutic regimen for the control of metabolic derangements in adult disease.

Protective Effect of Semisulcospira libertina Extract on Induced Hepatitis in Rats (다슬기 열수 추출물이 간독성이 유도된 흰쥐에 미치는 보호 효과)

  • This study aimed to investigate the protective effect of Semisulcospira libertina extract on liver injury induced by D-galactosamine in rats. After the administration of S. libertina extract, the local fat degeneration and infiltration of inflammatory cells in liver tissues were significantly decreased and peripheral hemorrhages around portal triads and central necrosis around central veins were found to be protective. The elevated levels of plasma ALT, AST, and LDH, the ALP activation lipid peroxidation, and the lipid contents of a damaged liver were recovered in experimental rats administrated with S. libertina extract, suggesting its role in blood enzyme activation and lipid content restoration within damaged rat liver tissues. Moreover, the expression rate of TNF-α, which accelerates inflammation and induces tissue damage and necrosis, was significantly decreased. In addition, the activities of antioxidant enzymes were more effectively upregulated compared to those of the control group induced hepatotoxicity. All data showed that S. libertina extract has a preventive role against liver damages, such as inflammation and tissue necrosis, as instigated with D-galactosamine by improving the activities of blood enzymes and antioxidant enzymes and modulating the expression of inflammation factor, suggest S. libertina extract is an effective medicinal resource for the restoration of hepatotoxicity.
