Effect of Garlic and Medicinal Plants Composites on the Liver Function and Lipid Metabolism of Rats Administered with Ethanol During the Short-term (단기 알코올 투여 시 마늘과 한약재 복합물이 체내 지질 조성 및 간기능 회복에 미치는 영향)

  • This study was performed to observe the effect of hot-water extracts from garlic and 13 kinds of medicinal plants composites (GMP) on hyperlipidemia and hepatoprotective activity in rats administered with alcohol. Male Sprague-Dawly rats were fed an AIN-93 diet (Normal), a normal diet plus ethanol (control, 10 ml of 40% ethanoljkgjday), a control diet plus 0.5% garlic and 1.0% medicinal plants composites extracts (GMP-I), and a control diet plus 1.0% garlic and medicinal plants composites extracts (GMP-II) for 7 days. Blood glucose was higher than the control, but it was markedly decreased in the GMP-II group. Elevation total lipids, cholesterol, triglyceride and phospholipids in serum were markedly decreased in rats fed with GMP-I. GMP-II also inhibited the increase of lipid content in serum. Activities of GOT, GPT, $\gamma$-GTP and ALP in serum elevated by alcohol were significantly inhibited in the GMP group. TBARS content of serum was significantly decreased in GMP groups administered with garlic and medicinal plant extracts. Extracts of garlic and medicinal plants play an important role in recovering liver function in rats from alcohol induced damage.

Screening and Development of Novel Herbal Medicine for Bone Healing Treatment (골유합 촉진 치료제 개발을 위한 수종의 한약재 활성 검색)

  • Objectives : The differentiation of osteoblasts is controlled by various growth factors and matrix protein expressed in bone. The aim of this study was to investigate the effects of many herbs medicine(KHBJs) for bone healing that induces osteogenic activity in human osteoblast-like SaOS-2 cells. Methods : The osteogenic effects of KHBJs were evaluated by using cell proliferation(WST-8) assay, alkaline phosphatase(ALP) activity assay, colorimetric analysis of vascular endothelial growth factor(VEGF) expression in human osteoblast like SaOS-2 cell. Also, osteogenic activity of KHBJ fractions(KHBJB and KHBJR) by activity guided fractionation were evaluated. Results : About 7 KHBJs had effect on the proliferation of osteoblast like SaOS-2 cells, and dose-dependently increased alkaline phosphatase(ALP) activity. KHBJs markedly increased expression for VEGF. Fractionated KHBJs(KHBJB or KHBJR) not enhanced more than KHBJs on osteogenic activity in SaOS-2 cells. Conclusions: This study found that 7 KHBJs had effect on proliferation, ALP activity, and VEGF expression in osteoblast like SaOS-2 cells. These results propose that KHBJs can play an important role in osteoblastic bone formation, and may possibly lead to the development of bone-forming drugs.

Purification and biological activity of recombinant human bone morphogenetic protein-2 produced by E. coli expression system (E. coli 발현 시스템에 의해 생산된 recombinant human bone morphogenetic protein-2의 정제와 생물학적 활성)

  • Purpose: Bone morphogenetic protein-2(BMP-2) has been shown to possess significant osteoinducitve potential. There have been attempts to overcome a limitation of mass production, and economical efficiency of BMP. The aim of this study was to produce recombinant human BMP-2(rhBMP-2) from E. coli in a large scale and evaluate its biological activity. Materials and Methods: The E.coli strain BL21(DE3) was used as a host for rhBMP-2 production. Dimerized rhBMP-2 was purified by affinity chromatography using Heparin column. To determine the physicochemical properties of the rhBMP-2 expressed in E. coli, we examined the HPLC profile and performed Western blot analysis. The effect of the purified rhBMP-2 dimer on osteoblast differentiation was examined by alkaline phosphatase (ALP) activity and representing morphological change using C2C12 cell. Results: E. coli was genetically engineered to produce rhBMP-2 in a non-active aggregated form. We have established a method which involves refolding and purifying a folded rhBMP-2 dimer from non-active aggregates. The purified rhBMP-2 homodimer was characterized by SDS-PAGE as molecular weight of about 28kDa and eluted at 34% acetonitrile, 13.27 min(retention time) in the HPLC profile and detected at Western blot. The purified rhBMP-2 dimer stimulated ALP activity and induced the transformation from myogenic differentiation to osteogenic differentiation. Conclusion: rhBMP-2 was produced in E. coli using genetic engineering. The purified rhBMP-2 dimer stimulated ALP activity and induced the osteogenic differentiation of C2C12 cells.

Hematological Changes of Olive Flounder, Paralichthys Olivaceus Exposed to Aroclor 1254 (Aroclor 1254에 노출된 Olive flounder, Paralichthys of olivaceus의 혈액학적 변동)

  • Effects of Aroclor 1254, on hematological disturbance were investigated in the olive flounder, Paralichthys olivaceus. Flounders were exposed to waterborne PCB (3.0${\mu}g \; L^{-1}$) for 00 days. Blood samples were taken to determine their hematological disturbances. Plasma calcium concentration was significantly decreased in PCBs exposed flounder compared to the control group, while magnesium and inorganic phosphorus concentrations in PCBs exposed flounders were not significantly influenced during the test Belied. Although plasma total protein and albumin level were significantly reduced compared to the control group after 40 days, plasma glucose level was found to be significantly increased over the control group. Exposure to waterborne PCBs resulted in significant increase in the various enzyme activities, such as GOT, GPT and ALP in the flounder. The results of the present study led us to conclude that concentration of PCBs 3.0${\mu}g \; L^{-1}$ of the estuarine could markedly affect the hems tological distribution of olive flounder, Paralichthys olivaceus.

The Effect of Magnetism(Neodymium Magnet) on Activity of Osteoblast (뼈모세포의 활성도에 대한 자성의 효과에 관한 연구)

  • The object of this study is to observe the effects of magnetism on the osteoblasts using a neodymium magnet. The osteoblasts was cultured under magnetic fields of varying intensities to evaluate the effect of magnetism on the activity and alkaline phosphatase acitivty of the osteoblasts. Osteoblasts were cultured in the cell density of $10^4$ for the evaluation of cell proliferation and 105/ml for the evaluation of ALP activity under 0. 10, 100, 500, 1000, 2000, 4000 gauss for 24 hour. For evaluation of osteoblast morphologic changes under magnetic, osteoblasts were observed by inverted microscope and TEM. To elucidate if IGF-receptors are increased under the magnetic field, we investigated osteoblasts by immunofluoroscence staining. The results were as follows: In the varying intensities of magnetic fields, the degree of cell proliferation was the highest in the magnetic field of 10 gauss and this gradually decreased up to 1000 gauss. In the magnetic fields stronger than 1000 gauss, the degree of the cell proliferation decreased to an even lower level than that of the control group. The ALP activity and protein synthesis showed a similar increase pattern as the degree of cell proliferation compared to the control group but showed little difference. Under the microscope, morphological change of the cells ( decrease in length and increase in roundness) were observed but no peculiarity of cell distribution could be found according to the magnetic field line. In the proper intensity of magnetic fields (10 gauss), the cultured cells showed increase in number of IGF Receptors compared to that of the control group.

Protective Mechanism of Flavonoids Isolated from Rhus Vernicifiua on the Paraquat Toxicity Reducing Agent and its Inhibition Mechanism (옻나무 목부로부터 분리한 Favonoids가 Paraquat 독성의 경감기전 검색)

  • In this study, we investigated to elevate the modulatory effect of flavonoid(fustin, sulfuretin, 10 mg/kg) which was isolated from Rhus verniciflua Stokes(RVS) in male Sprague-Dawley rats for 2 weeks on the toxicity of paraquat. In the flavonoids pretreated groups, glutamic oxaloacetic transaminase, glutamic pyruvic transaminase, blood urea nitrogen, creatinine, malondialdehyde and alkaline phosphatase activity in serum and malondialdehyde, alkaline phosphatase activity and collagen in lung tissue which was induced paraquat toxicity were slightly decrease compared to the normal group. In the lung tissue of flavonoids pretreated groups, malodialdehyde value, G-6-phosphatase activity and collagen synthesis were recovered to tile normal values and alkaline phosphatase activity was increased. From these results, we concluded that flavonoids which were isolated from RVS is an effective agent to inhibit the pulmonary and internal toxicities and hence we concluded that acitive components of fustin and sulfuretin which were isolated from RVS might be removed free radicals induced by paraquat.

Effects of Dimethylformamide on Lipid Peroxide Level and Activity of Superoxide Dismutase in Human Serum (Dimethylformamide가 사람 혈청의 과산화지질 농도와 Superoxide dismutase 활성도 변화에 미치는 영향)

  • The variation in the enzyme activities of human liver usually represents the particular physiological conditions of each individuals. Thus, we investigated the variation in the activities of SOD, HR and LPO of (1) non-exposed workers (56 subjects), and (2) exposed workers to DMF (43 subjects) in synthetic leather process. Serum levels of enzyme activities of exposed workers (AST:$30.26{\pm}20.041$U/L, ALT:$32.72{\pm}23.393$U/L, GGT:$28.47{\pm}18.635$ U/L, ALP:$81.77{\pm}34.879$ U/L)were slightly higher than those in nonexposed workers (AST:$24.00{\pm}9.441$ U/L, ALT:$23.89{\pm}18.305$U/L, GGT:$21.95{\pm}17.970$U/L, ALP:$70.84{\pm}24.678$U/L), but only the level of ALT was statistically significant (p< 0.05). Serum levels of LDH, TRF and CHOL in non-exposed workers were slightly higher than those of exposed workers. However, they were not statistically significant (p > 0.05). Serum levels of HR and LPO of the exposed workers appeared to be reduced, but not those of the non-exposed workers. The SOD activities of exposed workers were also slightly higher than those of non-exposed workers, but the results were not statistically significant (p > 0.05). The level of HR was increased with age, but the SOD level was not. These results suggest that the intermittent exposure to DMF at time-weighted average (TWA) level (10 ppm/$m^3$) has affected on the activities of enzymes such as AST, ALT, TRF, but not on the generation of HR, activity of SOD. However, if high dose of DMF was used, there would be severe effects for the generation of HR and LPO.

Effects of Kanghwalsokdan-tang Gamibang Water Extract on Osteoclast Differentiation and Osteoblast Proliferation (강활속단탕가미방(羌活續斷湯加味方)이 파골세포 분화 및 조골세포 활성에 미치는 영향)

  • Objectives : This study was conducted to evaluate the effect of Kanghwalsokdan-tang Gamibang water extract (KSG) on osteoporosis. Methods : RANKL-stimulated RAW 264.7 was used to evaluate inhibitory effect of KSG osteoclast differentiation and gene expression. We counted TRAP (+) multinucleated cells and measured TRAP activity and mRNA expressions of osteoclastogenesis-related genes (NFATc1, MITF, JNK1, cathepsin K, MMP-9) to figure out the effect of KSG on osteoclast. Osteoblastogenesis was also determined in rat calvarial cell. Alkaline phosphatase (ALP) activity, bone matrix protein and collagen synthesis were measured by using murine calvarial cell. Results : KSG inhibited the differentiation of osteoclast precursor cell and expression of genes related osteoclastogenesis like NAFTc1, MITF, c-fos, JNK1, Cathepsin K, MMP-9 and TRAP. KSG increased cell division and function of osteoblast separated from the skull of a rat and ALP synthesis, biosynthesis of bone matrix protein and collagen. Conclusions : Reviewing these results, KSG has efficacy on osteoclast inhibition and osteoblast activation. After further study, KSG will be able to apply for osteoporosis treatment and prevention.

Effects of Ginseng Saponin on the Metabolites and Enzymes of Normal and Alloxan-intoxicated Rats (인삼사포닌이 정상및 Alloxan투여 흰쥐에서 수종의 대사물질및 효소활성에 미치는 영향)

  • In order to evaluate the mode of action of ginseng saponin. metabolic changes in the plasma of normal and aloxan-intoxicated rats were compared Normal groups were administered only ginseng saponin 0, 5, 50mg/kg for 16 days, but alloxan-intoxicated groups were administered alloxan 25mg/kg for 3 days in addition to ginseng saponin. (1) No significant change in the concentration of glucose, cholesterol, and triglyceride (TG) was observed in normal rats but great inhibitory effect, except elevation of TG, was observed in alloxan-intoxicated rats. However, blood urea nitrogen was elevated in both normal and alloxan- intoxicated rats by administration of ginseng saponin 50mg/kg, and it was considered to be due to the impaired kidney function caused by overdose toxicity. (2) In normal rats, COT and ALP actIn,its were not changed by administration of ginseng saponin but GPT activity was decreased significantly. In alloxan-intoxicated rats, ginseng saponin exerted inhibitory action on the elevation of COT, CPT and ALP activity. But administration of ginseng saponin Smghg was much more effective than administration of 50mg/kg. (3) There fore, we concluded that ginseng saponin has the adaptogenic activity showing little effect on normal metabolism but great preventive action on alloxan-intoxicated rats.

Studies of the Cordyceps militaris Extract Administration on the Metabolic Enzyme Activities in Hypercholesterolemia (동충하초 엑스의 고콜레스테롤혈증 대사효소 활성 변동에 관한 효과)

  • The objective of this study was to investigate the effects of the feeding Cordyceps militaris extract on the improvement of the free fatty acid, lipid peroxide, creatinine and enzyme (creatine phosphokinase; CPK, lactate dehydrogenase; LDH, alkaline phosphatase; ALP, lecithin cholesterol acyltransferase; LCAT) activities in the sera of dietary hypercholesterolemic rats (SD strain, male) fed the experimental diets for 5 weeks. Concentrations of free fatty acid, lipid peroxide and CPK, LDH, ALP activities in sera were fairly reduced in the Cordyceps militaris extract administration group (CHE) than in the hypercholesterolemic diet group (CHD). However, no significance was found in the effect of an creatinine concentration among the groups. The LCAT activity in serum was increased in the Cordyceps militaris extract administration (CHE) than in the hypercholesterolemic diet group (CHD). From these results, Cordyceps militaris extracts were effective on the improvement of the lipid components and metabolic enzyme activities in sera of dietary hypercholesterolemic rats.
