  • In the course of evaluation of hepatoprotective components against alcohol-induced toxicity from Aloe spp., the methanol extract was found to cause a significant inhibition of rat liver cytosolic alcohol dehydrogenase activity. Systematic fractionation of active tractions monitored by bioassay led to isolation of four compounds; aloe-emodin, aloenin, ethylidene-aloenin and ${\beta}-sitosterol$, which were estimated as active principles for inhibition of c-ADH and c-ALDH activities in vitro.

Lactobacillilus brevis를 이용한 다시마 발효물의 알코올 분해 활성 (Alcohol Metabolizing Activity of Fermented Sea Tangle Juice)

  • Alcohol metabolizing activity of fermented sea tangle juice (FSYJ) using Lactobacillilus brevis BJ20 were evaluated by measuring relative alcohol dehydrogenease (ADH) and aldehyde dehydrogenase (ALDH) activities. According to the results of MTI assay. the fermented sea tangle juice by Lactobacillilus brevis BJ20 appeared safe in the cytotocxity. The relative ADH activity of FSTJ showed 124% at 10 mg/mL, which increased with increasing concentration. The relative ALDH activity showed, however, insignificant difference (P>0.05) between concentrations of FSTJ up to 50 mg/mL. These results suggested that fermented sea tangle juice by L. brevis BJ20 could be used as a potential material for metabolizing alcohol.

귀뚜라미의 물 및 메탄올 추출물이 알코올 대사에 미치는 효과 (Effects of Water and Methanol Extracts of Cricket (Gryllus bimaculatus) on Alcohol Metabolism)

  • The cricket has been used in East Asia as crude drugs for treating fever and hypertension, and is presently reared as a pharmaceutical insect in China and a food for animals. For the purpose of evaluating protective extracts against alcohol-induced toxicity, the extracts of the cricket (Gryllus bimaculatus) were examined in animal models acutely administered alcohol by the cricket in ICR-mice. Water and methanol extracts from the cricket, were found to cause a significant decrease (37%) in the blood ethanol concentration as well as enhancement of liver mitochondrial alcohol dehydogenase (ADH) and acetaldehyde dehydogenase (ALDH) activitieson on a single intraperitoneal administration in mice. Futhermore methanol extract was demonstrated to exhibit more potent enhancing activity on ethanol metabolism than water extract. These results suggest that water/alcohol extract of G. bimaculatus may be used as a food for reducing the toxicity of alcohol.

Two novel mutations in ALDH18A1 and SPG11 genes found by whole-exome sequencing in spastic paraplegia disease patients in Iran

  • Hereditary spastic paraplegia is a not common inherited neurological disorder with heterogeneous clinical expressions. ALDH18A1 (located on 10q24.1) gene-related spastic paraplegias (SPG9A and SPG9B) are rare metabolic disorders caused by dominant and recessive mutations that have been found recently. Autosomal recessive hereditary spastic paraplegia is a common and clinical type of familial spastic paraplegia linked to the SPG11 locus (locates on 15q21.1). There are different symptoms of spastic paraplegia, such as muscle atrophy, moderate mental retardation, short stature, balance problem, and lower limb weakness. Our first proband involves a 45 years old man and our second proband involves a 20 years old woman both are affected by spastic paraplegia disease. Genomic DNA was extracted from the peripheral blood of the patients, their parents, and their siblings using a filter-based methodology and quantified and used for molecular analysis and sequencing. Sequencing libraries were generated using Agilent SureSelect Human All ExonV7 kit, and the qualified libraries are fed into NovaSeq 6000 Illumina sequencers. Sanger sequencing was performed by an ABI prism 3730 sequencer. Here, for the first time, we report two cases, the first one which contains likely pathogenic NM_002860: c.475C>T: p.R159X mutation of the ALDH18A1 and the second one has likely pathogenic NM_001160227.2: c.5454dupA: p.Glu1819Argfs Ter11 mutation of the SPG11 gene and also was identified by the whole-exome sequencing and confirmed by Sanger sequencing. Our aim with this study was to confirm that these two novel variants are direct causes of spastic paraplegia.

알코올 의존 환자에서의 Aldehyde Dehydrogenase II와 CYP2E1 유전자 다형성과 임상적 특성간의 연관성 (Association of Genetic Polymorphisms of Aldehyde Dehydrogenase II and CYP2E1 and Clinical Characteristics of Patients with Alcohol Dependence)

  • Objective:This study was to explore the relation of genetic polymorphisms of ALDH2 and CYP2E1 to clinical characteristics of alcoholic patients and alcohol induced liver damage. Methods:The genotype and allele frequencies of 128 male hospitalized patients who met DSM-IV criteria for alcohol dependence were compared with 128 healthy male control subjects. The genetic informations of ALDH2 and CYP2E1 were identified with the technique of polymerase chain reaction and restriction fragment length polymorphism. The clinical characteristics of the alcoholic patients were assessed and analyzed in relation to the family history of alcoholism. For the relation of CYP2E1 genetic polymorphism to the liver damage, the blood levels of various liver function indicators such as ALT, AST, and protein were checked out. Results:1) The alcoholic patients with the family history of alcoholism had the earlier onset of age (p=0.001), the longer duration of illness(p=0.045), and higher NCA scores(p=0.018) than those without the family history of alcoholism. 2) Most alcoholic patients were homozygous for $ALDH2^*1$, compared to control subjects.(p=0.000) 3) There was no difference of CYP2E1 distribution between alcoholic patients and control subjects. However, alcoholic patients having mutant c2 allele showed higher alcoholism severity scores(p=0.004) and more hospitalizations(p=0.014) than those having c1 allele. 4) There was no relationship between CYP2E1 genotype and the functional abnormalities of the liver. Conclusion:This study suggests that $ALDH2^*1$ is highly related with alcohol dependence. Also mutant c2 allele of CYP2E1 is correlated with the severity of alcoholism and the number of hospitalization. But genetic polymorphim of CYP2E1 seems to have no relation to liver damages.

천연소재복합물의 알코올 및 아세트알데히드 분해활성 및 호기중 알코올 농도 감소 (Natural herbal extract complex induces the degradation of alcohol and acetaldehyde and reduces the breath alcohol concentration)

  • 본 연구는 천연소재 12종의 페놀성 화합물 함량, 라디칼 소거능, 알코올탈수소효소(ADH), 아세트알데히드탈수소효소(ALDH) 활성도를 측정하고 상승효과를 가지도록 배합한 복합물의 숙취해소에 미치는 효과를 규명하는데 목적이 있다. 우수한 개별 소재를 선정하고 배합비를 달리하여 효능 평가를 통해, 배합비가 효능에 미치는 상관관계를 분석하였다. 8종의 추출물 중 보이차, 소나무, 갈근 순으로 ADH 활성이 높았고, 갈근은 배합량과 효능 간의 양의 상관관계, 보이차, 소나무 등은 음의 상관관계를 가지는 것으로 나타났다. 8종의 소재로 배합한 F1~F7 을 대상으로 ADH, ALDH 활성평가를 통해 F.7에서 상승효과가 가장 높은 것을 확인하여 최적 배합비로 도출하였다. F.7를 섭취한 시험군에서 알코올 섭취 30분 후부터 호기 알코올 농도가 섭취 직후 호기중 알코올 농도 대비 58%로 유의적으로 감소되기 시작하였고, 120분 후 27%로 호기중 알코올 농도가 감소되었다. 음주 후 발생하는 숙취증상 설문 결과 대조군에 비해 시험군에서 음주 후 발생하는 피곤함과 갈증에서 유의적인 개선효과 (p<0.05)가 있었다.

홍합과 여러 식물의 열수 추출물의 알코올분해효소에 미치는 영향과 항산화 및 ACE 저해 활성 (Effect of Hot Water Extracts of Blue Mussel and Several Plants on Alcohol Metabolizing Enzymatic, Antioxidant, and Angiotensin Converting Enzyme Inhibitory Activities)

  • 홍합을 함유한 숙취 해소용 식품 개발의 근거를 확보하기 위하여 12종류의 열수 추출물을 제조하였다 - A(겨우살이); B(냉이); C(칡); D(콩나물); E(헛개나무); F(홍합); G(홍합과 겨우살이); H(홍합과 냉이); I(홍합과 칡); J(홍합과 콩나물); K(홍합과 헛개나무). C 추출물이 alcohol dehydrogenase(ADH)와 acetaldehyde dehydrogenase(ALDH)의 활성을 가장 크게 증가시켰으나, 홍합과 혼합한 경우에는 상승 효과를 보이지 않았다. 칡을 함유한 경우를 제외하고는 H 추출물이 높은 ADH($237.4{\pm}1.7%$)와 ALDH($136.5{\pm}2.1%$) 활성을 보였다. 또한, H 추출물은 가장 높은 DPPH 라디칼 소거능($93.9{\pm}0.1%$)과 안지오텐신 전환효소(ACE) 저해능($42.7{\pm}1.6%$)을 나타내었다. 홍합과 냉이를 혼합한 경우에는 alcohol dehydrogenase 활성과 ACE 저해능이 상승되는 시너지 효과를 확인하였다.

발효한약추출물 HP-1이 알코올을 투여한 쥐의 알코올 대사에 미치는 영향 (Effect of Fermented Herbal Extracts, HP-1 on Enzyme Activities and Gene Expressions Related to Alcohol Metabolism in Ethanol-loaded Rats)

  • Recently, much attention has been paid to developing various kinds of fermented herbal extracts, a new type of traditional herbal medicine, in the field of Korean traditional medicine. The fermentation of medicinal herbs is intended to exert a favorable influence on digestability, bioavailavility and pharmacological activity of herbal extract in the gastrointestinal tract. It also produces a number of fermentation products that intensify the nutritional and pharmacological aspects of the medicinal herbs. In order to develop a functional beverage of alleviating the aftereffects of the previous drinks, the extracts (HP-1) of fermented herbal mixture, including Artemisia capillaris Thunb., Lonicera japonica Thunberg, and Hovenia dulcis Thunb., were prepared and the medicinal effect as a hangover cure was evaluated in ethanol-loaded rats. The enzyme activities of alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase-2 (ALDH2) were analyzed by measuring the concentration of blood ethanol and acetaldehyde. The mRNA expression of ADH and ALDH2 was also investigated through RT-PCR analysis. In the HP-1-treated group, the concentration of blood ethanol was significantly reduced at one hour after loading of ethanol, as compared to that in the saline-treated group. The reduced ethanol was converted to acetaldehyde, which resulted in rapid increase in acetaldehyde concentration in an hour. Acetaldehyde was started to decrease at 5 hours after ethanol loading. It implies that HP-1 is highly effective to stimulate the activities of ADH and ALDH2. The HP-1 treatment also activated the mRNA expression of ADH and ALDH. This study suggests that fermented herbal extract, HP-1 can be used as a functional beverage of alleviating the alcohol-induced hangover symptoms by stimulating the activities and gene expression of hepatic alcohol metabolizing enzymes.
