Effects of Diet of Korean Safflower(Carthamus tinctorious L.) Seed Powder on Bone Tissue in Rats during the Recovery of Rib Fracture (토종홍화씨의 급여가 실험동물의 늑골골절 회복중 골조직에 미치는 영향)

  • This study was carried out to investigate the effects of diet of Korean safflower(Carthamus tinctorious L.) seed powder on bone tissue during the recovery of rib-fracture in rats. Male Sprague-Dawley rats of 10 weeks old, weighing 370$\pm$5g, were divided into two groups including the control group(C group, AIN-76 semipurified diet) and safflower seed group(S group, AIN-76 semipurified diet+10% safflower seed powder) and were fed experimental diets for 12 days after adaptation period. After this period, the 9th right rib was fractured surgically and sham-operation was also performed. Rats were fed with experimental diets for up to 30 more days after rib-fracture. The degree of bone repair was evaluated during the recovery period at the 8th, 11th, 16th, 21st, 30th days after the surgical operation by microscopic observation of the fractured rib tissue. In callus formation, the portion of hyaline cartilage was noticably higher in S group than C group. The intracatilagenous ossification was observed at the 8th day in S group, but at 11th day in C group. The intramembranous ossification in callus was widely found over the 8th day to the 11th day in S group, but it was shown over the 11th day to the 16th day in C group. Bone resorption was also occured more rapidly in S group as indicated by large numbers of osteoclasts observed. At the 30th day, most of trabecular bones were disappeared in S group, whereas still shwon in C group over wide ranges of fractured ribs. These results imply that the supplementation of Korean safflower seed powder influences in the recovery of bone fracture by accelerating the process of bone repair.

Effects of Angelica gigas Nakai diet on lipid metabolism, alcohol metabolism and liver function of rats administered with chronic ethanol (당귀의 첨가 식이가 흰쥐의 지방대사와 알콜대사 및 간기능에 미치는 영향)

  • To investigate the effects of Angelica gigas Nakai diet on lipid metabolism, alcohol metabolism and liver function of rats administered with chronic ethanol, Sprague-Dawley male rats were fed either AIN-76 diet (control), control diet with ethanol, control plus Angelica gigas Nakai diet, or control plus Angelica gigas Nakai diet with ethanol for 30 days. On the 21st day, all of the rats were given an oral dose of ethanol and blood-ethanol concentration was monitored for the next 5 hours. The results obtained were: 1) Upon ethanol administration, the blood ethanol concentration was decreased from 2 hr significantly in the group of control plus Angelica gigas Nakai diet compared with control diet group; 2) The blood ethanol oxidation rate was increased in the group of control plus Angelica gigas Nakai diet with ethanol compared with control diet group or control plus Angelica gigas Nakai diet group. After 30 days, rats were sacrificed and then lipid and enzyme determinations in blood and liver were carried out. The results obtained were: 1) LDL-cholesterol in the blood of control plus Angelica gigas Nakai diet group was decreased significantly compared with control diet group; 2) Angelica gigas Nakai diet decreased liver triglyceride and total lipid and blood ${\gamma}-GTP$ level increased due to the chronic ethanol administration. These data suggest that Angelica gigas Nakai can have a recovery function on the symptoms of alcohol related diseases.

Diet of Red Ginseng-Cheonggukjang Improves Streptozotocin-induced Diabetes Symptoms and Oxidative Stress

  • Protective effects of cheonggukjang fermented with 20% red ginseng (RC) were observed in streptozotocin (STZ)-induced diabetic rats by measuring levels of blood glucose, serum lipid profiles, and hepatic reactive oxygen species generating and scavenging enzymc activities. RC diet was prepared by mixing with AIN-76 diet at the final concentration 2%, and it was fed to STZ-induced diabetic rats for 3 weeks. The RC dict was significantly improved body weight, feed efficiency ratio, levels of serum glucose, and serum and hepatic lipids in diabetes. The significantly elevated O type activity of xanthine oxidase in diabetes was also greatly decreased by the RC diet. The treatment of RC showed the improved hepatic glutathione s-transferase activities in the diabetic animals. The present study indicates that cheonggukjang fermented with red ginseng could ameliorate STZ-induccd diabetic symptoms such as aggravated blood glucose levels, serum lipid profiles, and even the conditions of oxidative stress.

  • To investigate the effects of Ixeris sonchifolia Hance diets on serum and hepatic lipid levles and enzyme activities in rats administered with ethanol chronically, Sprague-Dawley male rats were AIN-76 diet(control), control diet plus ethanol, control plus Ixeris sonchifolia Hance diet, or control plus Ixeris sonchifolia Hance diet plus ethanol for 30 days. Ixeris sonchifolia Hance diets significantly decreased the serum total cholesterol, triglyceride, LDL-cholesterol, and GOT levels that were increased due to the chronic ethanol administration. In addition, Ixeris sonchifolia Hance diets significantly decreased the liver triglyceride and total lipid levels that were increased due to the ethanol administration. The present findings, combined with previous data showing differences in the effects of cabbage diets having a high or a low level of GABA on the lipid levels and the serum Y-GPT activity of rats(Cha and OH[2000] J. Korean Soc. Food Sci. Nutr. 29, 500-505) raise the possibility that GABA in plants could have a nytraceutical role in the recovery of chronic alcohol-related diseases. The Effect of Dietary Calcium Level on Bone Mineral Density and Bone Mineral Content in Ovariectomized Female Rats (난소절제한 흰쥐에서 식이칼슘량이 골밀도에 미치는 영향)

  • This study was done to evaluate the effect of dietary calcium level (a diet which met 100% or twice the calcium level in AIN-76 diet) on preventing bone loss in ovariectomized rats. Forty Sprauge-Dawley female rats(body weight 200$\pm$5g)were divided into two groups. One group were ovariecotomized (Ovx) while the others received sham operation(Sham). Thereafter, each rat group was further divided into normal calcium diet(0.52%) and high calcium diet(1.04%) subgroups. All rats were fed on experimental diet and deionized water ad libitum for 8 weeks. The total body, spine and femur bone mineral densities and bone mineral contents were measured by Dual Energy X-ray Absorptiometry, Eight weeks following operation, ovariectomized rats fed a high calcium diet had a significantly higher total bone mineral content, total bone calcium content, spine bone mineral density, spine bone mineral content and femur bone mineral content than ovariectomized rats fed control calcium diet. The correlation between dietary calcium intake level and spine bone mineral density were positive, but there was no correlation between dietary calcium intake and femur bone mineral density. The findings from the present study demonstrated that bone loss due to ovarian hormonal deficiency can be partially prevented by a high calcium diet. Futhermore, these findings support the strategy of the use of a high calcium diet in the prevention of estrogen depleted bone loss(postmenopausal osteoporosis)

Sea Tangle Supplementation Alters Intestinal Morphology in Streptozotocin-induced Diabetic Rats and Lowers Glucose Absorption

  • This study examined whether dietary supplementation with sea tangle alters the intestinal morphology of streptozotocin-induced diabetic rats and affects the glucose absorption rate. Forty male Sprague-Dawley rats were divided into 2 groups and fed either a control (AIN76-based) diet or a sea tangle-supplemented diet. After 3 weeks, 10 rats in each group received an intramuscular injection of streptozotocin (45 mg/kg BW), and feeding was continued for 3 additional weeks. Dietary supplementation with sea tangle resulted in a lower fasting plasma glucose level compared with the control diet in diabetic rats. Scanning electron micrographs revealed serious damage to the jejunal villi of diabetic rats fed the control diet, whereas supplementation with sea tangle alleviated the damage. In a separate experiment, 20 male Sprague-Dawley rats were divided into 2 groups and fed either a control diet or a sea tangle-supplemented diet for 5 weeks, and fasted rats were subjected to in situ single-pass perfusion. The glucose absorption rate determined in the absence of digesta was decreased by 34% in the jejunum of rats fed a sea tangle diet compared with those fed a control diet. In conclusion, sea tangle supplementation lowered glucose absorption rate, altered intestinal morphology, and appeared to protect villi from damage caused by diabetes mellitus.

The Effect of Kimchi Intake on Production of Free Radicals and Anti-oxidative Enzyme Activities in the Liver of SAM (김치가 노화촉진 쥐 간의 유리기 생성 및 항산화효소 활성에 미치는 영향)

  • This study was performed to investigate the effect of kimchi intake on antiaging characteristics in liver of senescence-accelerated mouse (SAM) in terms of free radical production and anti-oxidative enzyme activities. Two hundred twenty SAM were divided into four groups and fed kimchi diet for 12 months. Experimental groups were kimchi free AIN-76 diet (control) group, Korean cabbage kimchi diet (KCK) group, mustard leaf added (30%) Korean cabbage kimchi diet (MKCK) group, and mustard leaf kimchi diet (MLK) group. Amount of freez-dried kimchi added to the diet was 5% that is equivalent to 50 g of fresh kimchi. Concentrations of total free radical, OH radical, $H_2O$$_2$in the liver significantly increased as aged (p<0.05). But those free radical concentrations from kimchi diet groups were lower than those of control (p<0.05). Among kimchi groups, MKCK and MLK groues showed greater inhibiting effect than KCK.   • Objectives : The purpose of this study is to investigate the effects of Polygonum multiflorum THUNB(何首烏) on decresing concentration of serum lipid which are triglyceride, total cholesterol, HDL-cholesterol, LDL-cholesterol. Thiobarbituric Acid Reactive(TBARS) in high cholesterol diet induced hyperlipidemia rats. Methods : Sprague-Dawley rats were divided into 3 groups, Normal group (supplied enough water and feeds only, Normal Group), high fat diet administered group(AIN-76 high fat diet #100496 used beef tallow for 4 weeks, Control Group) and Polygonum multiflorum THUNB(何首烏). administered group(AIN-76 high fat diet #100496 used beef tallow and Polygonum multiflorum THUNB(何首烏) extracts 85.0mg/200g 1time/day for 4 weeks, HSO Group). Rats were sacrificed and concentration of triglyceride, total cholesterol, HDL-cholesterol, LDL-cholesterol, TBARS were measured in rat's serum lipid. Results : There was no significant difference in the level of weigh between Control and HSO Group.(by Duncan test) Concentration of Total lipid has no significant difference in each group. (F=0.702, p=0.511, ANOVA test) Concentration of triglyceride has no significant difference in each group. (F=0.727, p=0.500, ANOVA test) Concentration of LDL-cholesterol was significantly different in each group.(F=.9.894. p=0.002, ANOVA test) It was significantly increased in the Control group compared with the Normal group and it significantly decreased in the HSO group compared with the Control group. Concentration of HDL-cholesterol has no significant difference in each group. (F=1.079, p=0.365, ANOVA test) TBARS values(Thiobarbituric Acid Reactive Substances) were significantly different in each group.(F=15.580, p=0.0001, ANOVA test) It was significantly increased in the Control group compared with the Normal group and significantly decreased in HSO group compared with the Control group. Conclusions : According to the above results, it is considered that Polygonum multiflorum THUNB(何首烏) is effective in decreasing concentration of serum lipid in high cholesterol diet induced hyperlipidemia rats.

  • Objectives: CRATAEGII FRUCTUS(山査) is known as the substance which decreases concentration of sarum lipid. The purpose of this study is to investigate the effects of CRATAEGII FRUCTUS(山査) on decreasing concentration of sarum lipid which are triglyceride, total cholesterol, HDL-cholesterol, LDL-cholesterol in rat high cholesterol diet inducede hyperlipide rats. Methods: Sprague-Dawley rats were divided into 3 groups, Normal group(supplied enough water and feeds only. Normal Group), high fat diet administered group(AIN-76 high fat diet #100496 used beef tallow for 4 weeks, Control Group) and CRATAEGII FRUCTUS(山査) administered group(AIN-76 high fat diet #100496 used beef tallow and CRATAEGII FRUCTUS(山査) extracts 85.0mg/200g 1time/day for 4 weeks. SS Group). Rats were sacrificed and concentration of triglyceride, total cholesterol, HDL-cholesterol, LDL-cholesterol were measured in rat's sarum lipid Results: There was no significant difference in the level of waight between each experimental group(F=0.243, p=0.787, ANOVA test), Concentration of triglyceride has no significant difference in each group(F=0.210, p=0.813, ANOVA test). There was significant increase of total cholesterol in the Control group compared to those of Normal but there was no significant difference in the SS group compared to the Control group(F=5.237, p=0.0l9, ANOVA test). There was a significant increase of HDL-cholesterol in the SS group compared to the Control group and decrease in Control group compared Normal group(F= 11.437, p=0.001, ANDYA test). Concentration of LDL -cholesterol was significantly different in each group(F= 16.377, p=0.0001, ANDYA test). It was significantly increased in the Control group compared with the Normal group and it has no difference in the SS group compared to the control group. TBARS values(Thiobarbituric Acid Reactive Substances) were significantly different in each group(F=5.242, p=0.0019, ANDYA test). It was significantly increased in the Control group compared with the Normal group and significantly decreased in SS group compared with the Control group. Conclusions: According to the above results, it is considered that CRATAEGII FRUCTUS(山査) is effective in decreaseing concentration of sarum lipid in high cholesterol diet induced hyperlipidemic rats.

Effect of Old Antler Extracts on the Galactosamine-induced Hepatotoxicity in Rats (녹각이 Galactosamine에 의해 유도된 간장해에 미치는 영향)

  • This study was conducted to investigate the biochemical efforts of old antler extracts on the enzyme activities in serum of galactosamine (GaIN)-induced rats. Male rats of Sprague-Dawley strain weighing the average of 110$\pm$10g were divided into four groups and each group was fed either AIN-76 diet or modified AIN-76 diet with old antler extracts (water extract, neutral extract, ether extract) for four weeks. CaIN (400mg/ kg of body weight) was injected to CaIN treatment groups intraperitoneally on the same day every week. CaIN treatment decreased growth performance of rats. But dietary supplementation of old antler extracts prevented this decrement. Serum aminotransferase activities were increased by CaIN treatment, but this increment was reduced by feeding the old antler extracts, and the effect of water extract was the most prominent of the old antler extracts. Activities of lactate dehydrogenase (LDH) and alkaline phosphatase (ALP) in serum of CaIN treatment groups were higher than those of control groups and were decreased by old antler extracts supplementation.
