질산화 작용이 있는 Aeromonas hydrophila의 동정 및 특성 (Identification and Characterization of Aeromonas hydrophila Producing Nitrification Capability)

  • 폐수처리 중 생물학적 처리에 활용할 수 있는 질소분해 능력을 가진 미생물을 분리하여 동정하고자 경기도내 하천 6지점에서 채취한 시료로부터 60개 균주를 선별하였다. 형태학적, 생화학적 및 배양학적 실험결과 Bergey\`s mannual of systematic bacteriology의 색인을 통하여 Aeromonas hydrophila로 동정하였다. Aeromonas hydrophila(AH-1), (AH-3), (AH-4), (AH-6) 균이 질산화 능력이 우수하였다. 4개 균주모두 amoxillin, ampicillin. cephalothin 과 ticarcillin에 내성을 나타내었다. 본 실험에서 분리한 Aeromonas hydrophila의 질산화의 최적조건은 균 농도 1.0$\times$$10^{6}$cells/ml, 배양온도 37$^{\circ}C$로 나타났다.

  • 정보통신기술의 발달에 따라 컴퓨터, 스마트폰과 같은 스마트 기기가 대중화됨에 따라 개인 블로그나 SNS 등을 기반으로 하여 개인이 다양한 콘텐츠를 생산하고 공유하는 커뮤니케이션 플랫폼인 1인 미디어가 활성화 되었다. 효율적으로 1인 방송시스템을 구축하기 위해서는 기존 방송 시스템의 체계는 유지하면서 개인의 협소한 공간을 효율적으로 활용할 수 있는 방안을 요구하게 된다. 따라서 본 논문에서는 각각의 방송장비들을 노트북, 어플과 같은 스마트 기기와 연동시켜 효율적이고 편리한 1인 미디어 방송 시스템을 구축하기 위한 방법을 제안하였다.

Replacing alfalfa hay with amaranth hay: effects on production performance, rumen fermentation, nutrient digestibility and antioxidant ability in dairy cow

  • Objective: The aim of this research was to explore the effects of dietary substitution of alfalfa hay by amaranth hay on production performance, rumen fermentation, nutrient digestibility, serum biochemical parameters and antioxidant ability in dairy cows. Methods: A total of 45 healthy Holstein cows with same parity and similar milk yield and body weight were randomly divided into 3 groups: control diet without amaranth hay (CON) or 50% and 100% alfalfa hay replaced by an equal amount of amaranth hay (dry matter basis, AH1 and AH2, respectively). All the cows were fed regularly 3 times a day at 06:30, 14:30, and 22:30 and had free access to water. The experiment lasted for 60 d. Results: The dry matter intake of CON and AH1 groups was higher (p<0.05) than that of AH2 group. Compared with AH1 group, the milk yield of AH2 group was reduced (p<0.05). Moreover, dietary substitution of alfalfa hay by amaranth hay increased (p<0.05) milk fat, ammonia nitrogen and acetate concentrations. However, the crude protein digestibility of AH2 group was lower (p<0.05) than that of CON group, while an opposite tendency of serum urea nitrogen was found between two groups. The neutral detergent fiber digestibility of AH1 group was increased (p<0.05) when compared to AH2 group. Amaranth hay treatment increased (p<0.05) the serum concentration of glutathione peroxidase in dairy cows. Compared with CON group, the malonaldehyde activity of AH1 group was decreased (p<0.05). Conclusion: Dietary replacing alfalfa hay with amaranth hay (50% ratio) in dairy cows did not affect production performance but improved their antioxidant ability.

Particulate Matter-Induced Aryl Hydrocarbon Receptor Regulates Autophagy in Keratinocytes

  • Particulate matter (PM), which refers to the mixture of particles present in the air, can have harmful effects. Damage to cells by PM, including disruption of organelles and proteins, can trigger autophagy, and the relationship between autophagy and PM has been well studied. However, the cellular regulators of PM-induced autophagy have not been well characterized, especially in keratinocytes. The Aryl Hydrocarbon Receptor (AhR) is expressed in the epidermis and is activated by PM. In this study, we investigated the role of the AhR in PM-induced autophagy in HaCaT cells. Our results showed that PM led to AhR activation in keratinocytes. Activation of the AhR-target gene CYP1A1 by PM was reduced by co-treatment with ${\alpha}$-naphthoflavone (${\alpha}-NF$), an AhR inhibitor. We also evaluated activation of the autophagy pathway in PM-treated keratinocytes. In HaCaT cells, treatment with PM treatment led to the induction of microtubules-associated proteins light chain 3 (LC3) and p62/SQSTM1, which are essential components of the autophagy pathway. To study the role of the AhR in mediating PM-induced autophagy, we treated cells with ${\alpha}-NF$ or used an siRNA against AhR. Expression of LC3-II induced by PM was decreased in a dose dependent manner by ${\alpha}-NF$. Furthermore, knockdown of AhR with siAhR diminished PM-induced expression of LC3-II and p62. Together, these results suggest that inhibition of the AhR decreases PM-induced autophagy. We confirmed these results using the autophagy-inhibitors BAF and 3-MA. Taken together, our results indicate that exposure to PM induces autophagy via the AhR in HaCaT keratinocytes.

Molecular Characterization of Some Antilisterial Bacteriocin Genes from Enterococcus faecium and Pediococcus pentosaceus

  • Food bio preservation is of major interest in the food industry. Many types of antimicrobial compounds can be produced by lactic acid bacteria (LAB), including bacteriocins. Bacteriocins increase the shelf-life of food by decreasing some food-borne diseases. In this study, a multi-coding sequence of bacteriocin genes was used for primer design to produce bacteriocin genes in Enterococcus faecium AH2 strain and Pediococcus pentosaceus AH1. Multi-coding sequences were aligned to detect conserved sequences in the bacteriocin gene. Eight genes encoding proteins involved in bacteriocin production were isolated and sequenced, including six from E. faecium AH2 (entA, entI, entF, entR, orfA2, orfA3) and two from P. pentoceseus AH1 (papA, pedB), and all gene sequences were deposited in the Gen Bank database under accession numbers LC064146-LC064151, LC101300, and LC101789, respectively. P. pentosaceus AH1 and E. faecium AH2 strains displayed bacteriocin activities of $2610AU\;mL^{-1}$ and $690AU\;mL^{-1}$ and inhibition zones of 26 mm and 19 mm, respectively. Overexpression of entA in E. faecium AH2 increased the bacteriocin and antimicrobial activities.

  • 유튜브에 대한 관심이 급증하면서 1인 방송시스템에 대한 수요가 증가하고 있다. 장비를 구축하는데 생기는 문제 중 가장 중요한 요소는 경제적인 부분이다. 따라서 본 논문에서는 스튜디오의 상용화와 경제성, 전문성의 상향평준화에 초점을 맞춘 스튜디오 구성을 제안해 누구나 손쉽게 1인 방송을 할 수 있도록 한다. 또한, 본 논문에서 제안한 새로운 1인 방송 시스템을 통해 '크로마키' 기능을 추가로 구현해봄으로서, 매번 같은 장소에서 촬영하는 1인 미디오 스튜디오의 특성을 보완해 1인 방송 제작자들이 더욱 다양한 컨텐츠를 만들어낼 수 있도록 한다.

Ligand and Dimerization Dependent Transactivation Capability of Aromatic Hydrocarbon Receptor

  • The aromatic hydrocarbon receptor (AhR) is a cytosolic protein that binds the environmental pollutant, dioxin. The liganded AhR translocates into the nucleus where it heterimerizes with a constitutive nuclear protein, AhR nuclear translocator (Arnt). The N-terminal regions of both AhR and Arnt contain basic helix-loop-helix (bHLH) and Per-AhR-Arnt-Sim (PAS) motifs that are required for DNA binding, dimerization, and ligand binding whereas the C-terminal regions of both AhR and Arnt contain transactivation domains. Here, results from the mammalian two-hybrid system indicate that Arnt can make a homodimer but AhR cannot. In the presence of dioxin, the interaction between AhR and Arnt is stronger than that of the Arnt homodimer, suggesting that Arnt prefers to make a heterodimer with the liganded AhR rather than a homodimer. Transfection analyses using the GAL4-driven reporter system suggest that AhR's N-terminal region represses its own transactivation domain, as well as exogenous transactivation domains such as Sp 1 and VP16. Interestingly, the repressed transactivation domains of AhR are activated by ligand-dependent heterodimerization with Arnt. These observations suggest that heterodimerzation with Arnt is necessary not only for DNA binding but also for activation of the repressed transactivation capability of AhR.

Bacillus subtilis AH18의 고추역병 방제능과 $Siderophore_{AH18}$의 구조분석 (Structural Identification of $Siderophore_{AH18}$ from Bacillus subtilis AH18, a Biocontrol agent of Phytophthora Blight Disease in Red-pepper)

  • Bacillus subtilis AH18균주는 auxin, siderophore 그리고 cellulase를 동시에 생산하는 PGPR 균주이자 생물방제균주로 항진균성 siderophore의 특성을 확인한 결과 catechol type의 siderophore로 확인하였다. $Siderophore_{AH18}$ 의 정제는 Amberlite XAD-2, sephadex LH-20 column chromatography 그리고 HPLC를 통해 정제 및 정제여부를 확인하였으며, GC-MS, $^1H$-NMR, 그리고 $^{13}C$-NMR을 통해 구조 및 분자량을 확인하였다. 그 결과 B. subtilis AH18이 생산하는 siderophore은 분자량 883의 bacillibactin임을 확인하였으며, 포자발아억제활성을 나타냄을 확인하였다. B. subtilis AH18은 P. capsici에 의한 고추역병을 효과적으로 방제하였으며(방제력 55%), 이는 bacillibactin에 의한 효과가 포함되리라 추측된다.

Sub 1GHz Wireless LAN

  • 본고에서는 1GHz 이하 주파수에서 동작하는 Wireless LAN(WLAN) 표준으로, IEEE 802.11ah PHY/MAC 표준 규격에 대해서 알아 본다. IEEE 802.11ah 표준은, Smart Grid, M2M, IoT 와 같이 실내, 실외에서 많은 수의 단말들을 최소한의 전력소모로 데이터 통신을 하기 위한 목적으로 개발되었다. WLAN의 서비스 범위가 1Km 이상으로 늘어나게 되면, 그 동안 WLAN 에서 심각하게 다루지 않았던 Hidden Node 문제가 단말의 전력 소모, throughput performance 을 결정하는 중요한 요소로 등장하게 된다. 특별히 IEEE 802.11ah 에서는, 많은 수의 단말들간에 Hidden Node 을 고려한 Restricted Access Window 라는 새로운 channel access 기법이 제안되었고, 이에 본 고에서는 IEEE 802.11ah 에 새롭게 지원되는 MAC 기능들에 대해 중점적으로 살펴 보도록 한다.

유기전해액 $LiMn_{2}O_{4}$/Lithium 전지의 전기화학적 특성 (Electrochemical Characteristics of $LiMn_{2}O_{4}$/Lithium Cells in Organic Electrolyte)

  • The electrochemical properties of LiM $n_2$ $O_4$as a cathode and an anode for the lithium secondary battery were evaluated. When LiM $n_2$ $O_4$ material was used as the cathode with the current collector of aluminum, the 1st specific capacity and the 1st Ah efficiency in LiM $n_2$ $O_4$/lithium cell were 123 mAh/g and 91.7%, respectively The anodic properties of LiM $n_2$ $O_4$ material was also evaluated in the LiM $n_2$ $O_4$/1ithium cell with the current collector of copper. It showed that the LiM $n_2$ $O_4$ was useful as the anode for the lithium secondary battery. During the 1st discharge, a potential plateau was observed at the potential of 0.3 $V_{Li}$ Li+/. The 1st specific charge capacity and the 1st specific discharge capacity were 790 mAh/s and 362 mAh/g, respectively. Therefore, the 1st Ah efficiency was 46%. The discharge capacity was gradually faded with the charge-discharge cycling to about 50th cycles. Thereafter, the discharge capacity was stabilized to about 110 mAh/g.
