Gene Expression of Enzymes Related to Glutathione Metabolism in Anticancer Drug-resistant L1210 Sublines (항암제 내성 L1210세포의 Glutathione 대사 관련효소 유전자의 발현 양상)

  • Glutathione(GSH) has a very important role in detoxification of cells and is closely related to antitumor drug-resistance of cancer cells. In order to evaluate the importance of glutathione metabolism in the drug-resistant cancer cells, the concentration of celluar GSH and activities of ${\gamma}$-glutamylcysteine synthetase(GCS), ${\gamma}$-glutamyl transpeptidase (GGT) and glutathione S-transferases(GST) in the adriamycin, vincristine, or cisplatin resistant L1210 (L1210AdR, L1210VcR, or L1210Cis) sublines were measured. Expression and amplification of GCS, GGT, and GST-${\pi}$ genes were also observed in the parent Ll210 and the drug-resistant Ll210 sublines. The concentration of GSH was increased 5.34 fold in L1210Cis, 2.83 fold in L1210VcR, and 1.78 fold in L1210AdR, compared to L1210. The activities of GCS and GGT were increased in drug-resistant L1210 sublines. The GST activity was increased in L1210VcR and L1210Cis but decreased in L1210AdR compared to Ll210. Expression of GCS, GGT, and GST-${\pi}$ genes were increased in the resistant L1210 sublines compare to the parent L1210 in northern blot analyses. Overexpression of GCS, GGT, and GST-${\pi}$ were observed in the resistant sublines, and the increases of the concentration of glutathione and the activities of GCS and GGT in the resistant sublines may be involved in a part of the drug-resistance in the resistant sublines.

FTCARP: A Fault-Tolerant Routing Protocol for Cognitive Radio Ad Hoc Networks

  • Cognitive Radio (CR) has been recently proposed as a promising technology to remedy the problems of spectrum scarcity and spectrum underutilization by enabling unlicensed users to opportunistically utilize temporally unused licensed spectrums in a cautious manner. In Cognitive Radio Ad Hoc Networks (CRAHNs), data routing is one of the most challenging tasks since the channel availability and node mobility are unpredictable. Moreover, the network performance is severely degraded due to large numbers of path failures. In this paper, we propose the Fault-Tolerant Cognitive Ad-hoc Routing Protocol (FTCARP) to provide fast and efficient route recovery in presence of path failures during data delivery in CRAHNs. The protocol exploits the joint path and spectrum diversity to offer reliable communication and efficient spectrum usage over the networks. In the proposed protocol, a backup path is utilized in case a failure occurs over a primary transmission route. Different cause of a path failure will be handled by different route recovery mechanism. The protocol performance is compared with that of the Dual Diversity Cognitive Ad-hoc Routing Protocol (D2CARP). The simulation results obviously prove that FTCARP outperforms D2CARP in terms of throughput, packet loss, end-to-end delay and jitter in the high path-failure rate CRAHNs.

  • Mobile grid can provide the grid service to grid users regardless of space and time. There are many restrictions to use grid service in mobile environment such as battery power of mobile device, movement pattern of mobile device etc. So it is difficult to apply the grid technologies of wired network to wireless network environments. Therefore, to provide the mobile grid service, we suggested the mobile grid architecture using ad-hoc network. Also we showed that hybrid routing protocol is efficient for mobile grid service by considering the battery power of mobile device and simulations of evaluating the delay time of three routing protocols using NS-2. We will study the methods of resource allocations and network reliability to provide the mobile grid service.

Interactive Roles of Local versus Global Primed Identity and Advertisement Framing on Brand Evaluation (브랜드평가에 대한 아이덴티티의 점화와 광고프레임의 상호작용효과)

  • This article aims to explore the interactive roles of types of primed identity (local versus global identity) and types of ad framing on brand evaluations. The authors designed 2 experiments in which each experiment followed a $2{\times}2$ between-subject design. The empirical results showed that a gain-framed ad induced more positive emotional responses than a loss-framed ad, and the positive affective responses lead to more favorable brand evaluation. Furthermore, the results showed that there were interactive effects of primed identity and types of advertisement frame on brand evaluation. In the additional analysis, the results showed that when people with local identity were exposed to the gain-framed ad, they would engage in a higher level of integration processing than those in the control group, which in turn induced more favorable evaluation to the local brand. That is, the integration processing mode played a mediating role between the interaction (local id priming ${\times}$ ad frame) and the local brand evaluation. However, in the case of global brand evaluation, the integration processing mode did not play such a mediating role.

  • This study was performed to optimize the integrated anaerobic digestion (AD) and microbial electrolysis cells (MECs) for the enhanced hydrogen production. The optimum operational conditions of integrated AD and MECs were obtained using response surface methodology. The optimum substrate concentration and operational pH were 10 g/L and 6.8, respectively. In the confirm test, 1.43 mol $H_2/mol$ hexose was achieved, which was 2.5 times higher than only AD. After 40 to 60 hour at seeding, the volatile fatty acids (VFAs) in reactor of AD were not changed. However the VFAs of reactor of AD-MECs were reduced by 61.3% (acetate: 76.4%, butyrate: 50.0%, lactate: 55.0%).

  • KorQuAD 2.0은 총 100,000+ 쌍으로 구성된 한국어 질의응답 데이터셋이다. 기존 질의응답 표준 데이터인 KorQuAD 1.0과의 차이점은 크게 세가지가 있는데 첫 번째는 주어지는 지문이 한두 문단이 아닌 위키백과 한 페이지 전체라는 점이다. 두 번째로 지문에 표와 리스트도 포함되어 있기 때문에 HTML tag로 구조화된 문서에 대한 이해가 필요하다. 마지막으로 답변이 단어 혹은 구의 단위뿐 아니라 문단, 표, 리스트 전체를 포괄하는 긴 영역이 될 수 있다. Baseline 모델로 구글이 오픈소스로 공개한 BERT Multilingual을 활용하여 실험한 결과 F1 스코어 46.0%의 성능을 확인하였다. 이는 사람의 F1 점수 85.7%에 비해 매우 낮은 점수로, 본 데이터가 도전적인 과제임을 알 수 있다. 본 데이터의 공개를 통해 평문에 국한되어 있던 질의응답의 대상을 다양한 길이와 형식을 가진 real world task로 확장하고자 한다.

Galectin-9 Induced by Dietary Prebiotics Regulates Immunomodulation to Reduce Atopic Dermatitis Symptoms in 1-Chloro-2,4-Dinitrobenzene (DNCB)-Treated NC/Nga Mice

  • Atopic dermatitis (AD) is a skin disorder that causes chronic itch. We investigated the inhibitory effects of a mixture of prebiotic short-chain galacto-oligosaccharides and long-chain fructooligosaccharides (scGOS/lcFOS), inulin, or β-glucan on AD development in 1-chloro-2,4-dinitrobenzene (DNCB)-treated NC/Nga mice. Mice were randomly assigned to six groups: untreated mice, AD control, positive control (DNCB-treated NC/Nga mice fed a dietary supplement of Zyrtec), and DNCB-treated NC/Nga mice fed a dietary supplement of prebiotics such as scGOS/lcFOS (T1), inulin (T2), or β-glucan (T3). The prebiotic treatment groups (T1, T2, and T3) showed suppression of AD symptoms, Th2 cell differentiation, and AD-like skin lesions induced by DNCB. In addition, prebiotic treatment also reduced the number of microorganisms such as Firmicutes, which is associated with AD symptoms, and increased the levels of Bacteroidetes and Ruminococcaceae, which are associated with alleviation of AD symptoms. Our findings demonstrate the inhibitory effects of prebiotics on AD development by improving the Th1/Th2 cytokine balance and beneficial symbiotic microorganisms in in vitro and in vivo models.

Aged Doraji (Platycodon grandiflorum) Ameliorates Cyclophosphamide-Induced Immunosuppression in Mice (Cyclophosphamide에 의해 유도된 면역저하 마우스 모델에서 홍도라지 추출물의 면역 기능 조절 효과)

  • This study was done to examine immunomodulative effects of aged doraji (AD) in the immune-suppressed mice induced by cyclophosphamide. The immune-stimulating effects of ethanol AD extract in in vivo at 75 and 150 mg/kg body weight (BW) for AD and 2AD groups were evaluated and compared to the normal doraji group (2ND, 150 mg/kg BW) treated with a doraji without aging process. After the 10 days of oral supplement, body and immune related organ weights, serum immunoglobulin G (IgG) and cytokines levels, splenocytes proliferation rate, and splenic NK cell activity were measured as immune-related biomarkers. Body weight and serum IgG level increased in the 2AD group. But, the serum Th2 cytokine (IL-6, $TNF-{\alpha}$) levels were lower in the AD and 2AD groups, respectively. Splenic T cell and B cell proliferation and NK cell activity increased in the doraji groups and the significant increases were found only in the 2AD group. Thus, the aged doraji extract may affect body weight, serum IgG level, splenocytes proliferation, and splenic NK cell activity, and normalize the Th2 cytokine levels in the immune-suppressed mice. The results suggest that the aged doraji improves effectively immune system rather than the normal one.

Physiological Responses of Starry Flounder Platichthys stellatus during Freshwater Acclimation with Different Speeds in Salinity Change (염분변화 속도를 달리한 담수순화 과정에서 강도다리 Platichthys stellatus의 생리학적 반응)

  • Physiological responses (hematological factors, cortisol, glucose, osmolality, $Na^+$, $K^+$ and $Cl^-$) in starry flounder Platichthys stellatus were investigated during freshwater acclimation in the conditions of different speeds in salinity change with acute-decrease (AD) or stepwise-decrease (SD I and II). In AD of acute-decrease salinity, hematocrit (Ht), red blood cell (RBC) and hemoglobin (Hb) were rapidly increased more than SD I of stepwise-decrease salinity. But in case of SD II, Ht, RBC and Hb were no significant difference from beginning to end of this experiments. In AD, cortisol level significantly increased from $2.1{\pm}1.0{\mu}g/mL$ at the beginning to $13.7{\pm}0.2{\mu}g/mL$ at 6 hours and recovered to the basal levels ($3.1{\mu}g/mL$) at 10 days. In SD I, cortisol level was significantly increased from $2.1{\pm}1.0{\mu}g/mL$ at the beginning to $13.6{\pm}0.6{\mu}g/mL$ at 6 hours and recovered to the basal levels ($3.1{\pm}0.4{\mu}g/mL$) at 10 days. In SD II, cortisol level was a little increased from $2.1{\pm}1.0{\mu}g/mL$ at the beginning to $10.5{\pm}2.5$, $10.8{\pm}5.6{\mu}g/mL$ at 6, 12 hours and recovered to the basal level at 48 hours. Glucose level of AD, SD I, II were no significant difference from beginning to end of this experiments. Osmolality was $286.8{\pm}3.3\;mOsm/kg$ at the beginning. In SD II of stepwise-decrease, osmolality was no significant difference during rearing in freshwater (FW). But AD of stepwise-decrease and SD I of stepwise-decrease, osmolality was a little decreased end of this experiments. In AD of acute-decrease, only $Cl^-$ level was showed no significant difference from beginning to end of experiment and $Na^+$, $K^+$ levels were decreased. In case of SD I, $Cl^-$ level was showed no significant difference from beginning to end of experiment and $Na^+$, $K^+$ levels were decreased.

The Effect of Angelica gigas Nakai Extract and Bacillus Polyfermenticus KJS-2 on Atopic Dermatitis induced by DNCB in mice (DNCB로 유도된 아토피 피부염에 대한 당귀 추출물과 Bacillus polyfermenticus KJS-2의 효과)

  • Objectives: The purpose of this study was to investigate the effects of Angelica gigas Nakai extract(AGNE) and Bacillus polyfermenticus KJS-2 (BP2) on atopic dermatitis (AD) induced by 2, 4-dinitrochlorobenzene (DNCB) in mice. Methods: In the experiment, we divided mice into four groups: a control group, a DNCB group, an AGNE group, and an AGNE+BP2 group. Then we examined the changes in scratching frequency, clinical aspects on dorsum skin, immunoglobulin (IgE), cytokines ($TNF-{\alpha}$, IL-6) and expression of COX-2. Resutls: From the experiment, the scratching frequency was significantly dropped in AGNE group and AGNE+BP2 group. Clinical observations of dorsum skin, there were a severe keratotic lesion and drop of dead skin cell in DNCB group, but symptoms of AD were decreased 39.6% in AGNE group and 49.6% in AGNE+BP2 group during 3 weeks. IgE, $TNF-{\alpha}$ and IL-6 were decreased significantly in both AGNE and AGNE+BP2 group. Expression of COX-2 was also decreased significantly in both groups. Conclusions: In conclusion, these data suggest that AGNE can decrease symptoms of AD and BP2 makes AGNE more effective. So AGNE can be useful herbal therapy for AD.
