Purity Improvement of Calcium Lactate and Calcium Citrate Prepared with Shell of Anadarac tegillarca granosa (꼬막 패각으로 제조한 젖산칼슘과 구연산칼슘의 순도 향상에 대한 연구)

  • Ash of Anadarac tegillarca granosa shell was used for preparation of calcium lactate and calcium citrate, and improvement of their purity was carried out by using ammonium chloride process (ACP) and ammonium nitrate process (ACP). Purity of calcium lactate and calcium citrate made by the reaction of ash of Anadarac tegillarca granosa shell with lactic acid solution and with citric acid solution was 94.35-96.72 and $87.58-93.06\%$, respectively. However, purity of calcium lactate and calcium citrate prepared with purified calcium carbonate pre-purified from the ash of Anadarac fegiliarca granosa shell using ACP and ANP method was 99.53-100.34 and $99.32-99.88\%$, respectively. The purity of these calcium products were higher than those of calcium lactate and calcium citrate made with ash of Anadarac tegillarca granosa shell. Whiteness of calcium lactate and calcium citrate prepared with purified calcium carbonate pre-purified using ACP and ANP method was 94.8-98.5 and 99.4-101.5, respectively. Whiteness of these calcium products was higher than that of calcium lactate (91.8) and calcium citrate (92.9) made with the ash of Anadarac tegillarca granosa shell. Therefore, we estimated that calcium lactate and calcium citrate prepared with purified calcium carbonate using ACP and ANP method could be used potentially as a food additive for calcium supplement.

  • In order to modify lipid production of Perilla qualitatively as well as quantitatively by genetic engineering, genes involved in carbon metabolism were isolated and characterized. These include acyl-ACP thioesterases from Perilla frutescens and Iris sp., four different $\beta$-ketoacyl- ACP synthases from Perilla frutescens, and two $\Delta$15 a-cyl-ACP desaturases(Pffad7, pffad3). Δ15 acyl-ACP desa turase (Δ15-DES) is responsible for the conversion of linoleic acid (18:2) to $\alpha$-linolenic acid (ALA, 18:3). pffad 3 encodes Δ15 acyl-desaturase which is localized in ER membrane. On the other hand, Pffad7 encodes a 50 kD plastid protein (438 residues), which showed highest sequence similarity to Sesamum indicum fad7 protein. Northern blot analysis revealed that the Pffad7 is highly expressed in leaves but not in roots and seeds. And Pffad3 is expressed throughout the seed developmental stage except very early and fully mature stage. We constructed Pffad7 gene under 355 promoter and Pffad3 gene under seed specific vicillin promoter. Using Pffad7 construct, Perilla, an oil seed crop in Korea, was transformed by Agrobacterium leaf disc method. $\alpha$-linolenic acid contents increased in leaves but decreased in seeds of transgenic Perilla. Currently, we are transforming Perilla with Pffad3 construct to change Perilla seed oil composition. We isolated three ADP-glucose pyrophosphorylase (AGP) genes from Perilla immature seed specific cDNA library. Nucleotide sequence analysis showed that two of three AGP (Psagpl, Psagp2) genes encode AGP small subunit polypeptides and the remaining (Plagp) encodes an AGP large subunit. PSAGPs, AGP small subunit peptide, form active heterotetramers with potato AGP large subunit in E. coli expressing plant AGP genes.

Effect of Testoeterone and Cyclic AMP-theophylline on Ultrastructure and Several Enzymes' Activities in Rat Epididymis (Testosterone과 Cyclic AMP-theophyllin이 흰쥐 부정소의 미세구조 및 몇 가지 효소활성에 미치는 영향)

  • This study was made to investigate the effect of testosterone and cyclic-AMP (cAMP) on rat epididymis. Peritoneai injections of testosterone and cAMP to rats were earned out The activities of acid phosphatase (ACP), alkaline phosphatase (ALP) and lactate dehydrogenase (LDH) were measured and ultrastructural changes of luminal epithelial cells were observed. As a result, the activity of ACP was significanily decreased on the third day, that of ALP on the fifth day and that of LDH on the seventh day respectively in castrated group. In addition, the activities of ACP and ALP were significanily increased when treated with testosterone for 5 days, that of LDH when treated with testosterone for 7 days. In case of cAMP and cAMP - theophylline injection, the activities of ACP and LDH were increased but the range of increase was of no significance. However a significant increase in the activity of ALP was seen on both cases. On electron microscopic examination, gradually deformed Golgi complex, destructed mitochondria and disrupted stereociha were observed in castrated group. In case of testosterone injection, disrupted Golgi complex, mitochondria and stereocilia showed recovery. When cAMP and cAMP-theophylline were injected as an alternative, various cytoplasmic organelles as well as Golgi com- plex were recovered but stereocilia remained unrecovered.

  • KAERI is developing the Advanced Spent Fuel Conditioning Process (ACP) as a pre-disposal treatment process for spent fuel. In this process, the management process must operate in intense radiation fields as well as in a high temperature. Therefore, remote maintenance has played a significant role in this process. Hence suitable remote handling and maintenance technology needs to be developed along with the design of the process concepts. To do this, we developed the digital mockup for the ACP. The digital mockup provides the capability of verifying the remote operability of the process without fabrication of the process equipment. In other words, by applying virtual reality to the remote maintenance operation, a remote operation task can be simulated in the digital mockup. Through utilizing this graphic simulation in this digital mockup, general guidelines can be established for designing equipment intended for remote handling and maintenance. Also, the designer of the equipment that must be remotely maintained should ensure that there is adequate access to the process equipment. The graphic simulator will substantially reduce the cost of the develo363pment of the remote handling and maintenance procedure as well as the process equipment.

  • The Advanced Spent Fuel Conditioning Process(ACP), which is the process of the reduction of uranium oxide by lithium metal in a high temperature molten salt bath for spent fuel, was developed at Korea Atomic Energy Research Institute (KAERI). Since the ACP equipment is located in an intense radiation field (hot cell) as well as in a high temperature, it must be remotely operated and maintained. The ACP hot cell is very narrow so the workspace of the wall-mounted mechanical Master-Slave Manipulators(MSMs) is restricted. A Bridge Transported Servo Manipulator(BTSM) system has been developed to overcome the limitation of an access that is a drawback of the mechanical MSMs. The BTSM system consists ot a bridge crane with telescoping tubeset, a slave manipulator, a master manipulator, and a control system. We applied a bilateral position-position control scheme with friction compensation as force-reflecting controller. In this paper, the transmission characteristics on the tendon-and-pulley train is numerically formulated and analyzed. Also, we evaluate the performance of the force-reflecting servo manipulator.

  • The remote operation of the Advanced Spent Fuel Conditioning Process (ACP) is analyzed by using the 3D graphic simulation tools. The ACP equipment operates in intense radiation fields as well as in a high temperature. Thus, the equipment should be designed in consideration of the remote handling and maintenance. As well as suitable remote handling and maintenance technology needs to be developed along with the design of the process concepts. To develop such remote operation technology, we developed the graphic simulator which provides the capability of verifying the remote operability of the ACP without fabrication of the process equipment. In other words, by applying virtual reality to the remote maintenance operation, a remote operation task can be simulated in the graphic simulator, not in the real environment. The graphic simulator will substantially reduce the cost of the development of the remote handling and maintenance procedure as well as the process equipment, while at the same time developing a remote maintenance concept that is more reliable, easier to implement, and easier to understand.

Effect of Acylation on the Structure of the Acyl Carrier Protein P

  • Acyl carrier protein is related with fatty acid biosynthesis in which specific enzymes are involved. Especially, acyl carrier protein (ACP) is the key component in the growing of fatty acid chain. ACP is the small, very acidic protein that covalently binds various intermediates of fatty acyl chain. Acylation of ACP is mediated by holo-acyl carrier protein synthase (ACPS), which transfers the 4'PP-moiety of CoA to the 36th residue Ser of apo ACP. Acyl carrier protein P (ACPP) is one of ACPs from Helicobacter plyori. The NMR structure of ACPP consists of four helices, which were reported previously. Here we show how acylation of ACPP can affect the overall structure of ACPP and figured out the contact surface of ACPP to acyl chain attached during expression of ACPP in E. coli. Based on the chemical shift perturbation data, the acylation of ACCP seems to affect the conformation of the long loop connecting helix I and helix II as well as the second short loop connecting helix II and helix III. The significant chemical shift change of Ile 54 upon acylation supports the contact of acyl chain and the second loop.

Methyl-Branched Fatty Acids, Inhibitors of Enoyl-ACP Reductase with Antibacterial Activity from Streptomyces sp. A251

  • Bacterial enoyl-ACP reductase (FabI) has been demonstrated to be a novel antibacterial target. In the course of our screening for FabI inhibitors, we isolated two methyl-branched fatty acids from Streptomyces sp. A251. They were identified as 14-methyl-9(Z)-pentadecenoic acid and 15-methyl-9(Z)-hexadecenoic acid by MS and NMR spectral data. These compounds inhibited Staphylococcus aureus FabI with $IC_{50}$ values of 16.0 and $16.3\;{\mu}M$, respectively, but did not affect FabK, an enoyl-ACP reductase of Streptococcus pneumonia, at $100\;{\mu}M$. Consistent with their selective inhibition for FabI, they blocked intracellular fatty acid synthesis as well as the growth of S. aureus, but did not inhibit the growth of S. pneumonia. Additionally, these compounds showed reduced antibacterial activity against fabI-overexpressing S. aureus, compared with the wild-type strain. These results demonstrate that the methylbranched fatty acids show antibacterial activity by inhibiting FabI in vivo.

Panosialins, Inhibitors of Enoyl-ACP Reductase from Streptomyces sp. AN1761

  • In the continued search for inhibitors of enoyl-acyl carrier protein (ACP) reductase, we found that four acylbenzenediol sulfate metabolites from Streptomyces sp. AN1761 potently inhibited bacterial enoyl-ACP reductases of Staphylococcus aureus, Streptococcus pneumoniae, and Mycobacterium tuberculosis. Their structures were identified as panosialins A, B, wA, and wB by MS and NMR data. They showed stronger inhibition against S. aureus FabI and S. pneumoniae FabK with $IC_{50}$ of 3-5 ${\mu}M$ than M. tuberculosis InhA with $IC_{50}$ of 9-12 ${\mu}M$. They also exhibited a stronger antibacterial spectrum on S. aureus and S. pneumoniae than M. tuberculosis. In addition, the higher inhibitory activity of panosialin wB than panosialin B on fatty acid biosynthesis was consistent with that on bacterial growth, suggesting that they could exert their antibacterial activity by inhibiting fatty acid synthesis.

  • The process of coprecipitation of biocomposite hydroxyapatite/chitosan from aqueous solution at low temperature in alkali environnement was examined. We have shown that initially we have the formation of amorphous octocalcium phosphates $(Ca_8(HPO_4)(PO_4)_5,\;nH_2O:\;OCP)$ and the transferring from OCP to amorphous calcium phosphate $(Ca_9(PO_4)_3,\;nH_2O:\;TCP)$, and then from TCP to calcium-deficient hydroxyapatite $(Ca_{10-X}\;(HPO_4)_X(PO_4)_{6-x}(OH)_{2-X}\;:\;ACP)$ and hydroxyapatite $(Ca_{10}(PO_4)_6(OH)_2\;:\;HAP)$. The transformation of ACP to HAP was inhibited in the presence of chitosan. The result suggests that there is an affinity binding between ACP and chitosan and subsequently blocking the active growth site of ACP.
