Molecular Cloning of a cDNA Encoding Novel Tomato ACC Oxidase Using RT-PCR

  • Using RT-PCR, a cDNA encoding tomato (Lycopersicon esculentum) ACC oxidase has partially been cloned, sequenced and identified. The nucleotide suquence of the clone was in the coding region and shared about 80% of homology iwht the other ACC oxidase genes of tomato, and 70∼84% with those of other plants such as Oryza sativa, Nicotiana tabacum and Helianthus annuus. In the wounded tomato leaves, this nucleotide transcripts were accumulated rapidly and declined slowly thereafter. These results suggested that the predicted clone might be another member of tomato ACC oxidase gene family.

운전자 주행 패턴 분석 및 차량의 순항제어 기법 (Human Drivers' Driving Pattern Analysis and An Adaptive Cruise Control Strategy)

  • This paper presents experimental results for human drivers' driving patterns and an Adaptive Cruise Control(ACC) strategy. Analyses have shown that female drivers' driving characteristic values such as time-gap and minimum clearance are larger than those of male drivers'. Human drivers tend to have more clearance margins at high speed than at low speed. At low speed, drivers are much more sensitive to the desired clearance than at high speed. A multi-vehicle detection method is presented to improve ride quality of an ACC. Simulation results have shown that the proposed ACC can provide superior performance compared to the ACC strategy which uses a single-vehicle detection method.

  • Silicone insulators have many advantages over porcelain insulators. Especially silicone insulators have good characteristics of impact hardness, surface insulation, ease of processing, mass productivity and don't have risk of bombardment and vandalism. Recently insulation part made by silicone are becoming widely used. In this paper we introduce the development of Sealing End for 132kV XLPE cable with silicone composite hollow insulator and the adoption of it to a actual transmission line in abroad. This paper contains of design procedure, structure, electrical performance of it.

$Ca^{2+}$ Effect on Conversion of Exogenous 1-Aminocyclopropane-1-Carboxylic Acid to Ethylene in Vigna radiata Protoplasts

  • The possibility that 1-aminocyclopropane-1-carboxylic acid (ACC)-uptake may be dependent on the H+-gradient established across the plsma membrane was tested in protoplasts isolated from 2.5 day old mungbean hypocotyls. The ACC-induced ethylene production was inhibited when the H+-gradient was collapsed by the treatment with carbonycyamide-p-trifluro-methoxy-phenylhydrazone (FCCP). Moreover, the treatment with o-vanadate, a specific inhibitor of plasma membrane H+-ATPase, caused the inhibition of ethylene production. The ACC-induced ethylene production was inhibited by the treatemnt with verapamil (Ca2+-channel blocker), or ethylene glycol-bis($\beta$-aminoethyl ether) N, N, N', N'-tetraacetic acid (EGTA) (Ca2+-chelator). In contrast, the ehtylene production was stimulated by the application of A23187 (Ca2+ ionophore). The inhibitory effect of EGTA in the ethylene producton was magnified in the presence of A23187. From these results, we suggest that the external Ca2+ influx to the cytosol resulted in the stimulatin of ACC oxidase activity after ACC-uptake resulting from a H+-gradient across the plasma membrane.

  • Adaptive Cruise Control (ACC) is one of key features on intelligent Transportation System(ITS). In ACC, the steering is done by a driver, but the engine throttle valve and the brake are controlled electronically. The relative velocity and distance from the preceeding vehicle are measured by radars or image processing units and relevant vehicular spacing is maintained in ACC control systems. In this study, vehicle longitudinal dynamics are modeled to simulate vehicle longitudinal maneuver and to design longtitudinal controllers for ACC vehicles. The control algorithm is designed based on the modeled vehicle longitudinal dynamics using a non-linear sliding mode control method. To verity the performance of the control algorithm, a real time numerical simulation program is developed on a Silicon Graphics workstation using C-language . A real time graphic program is alos develpe and integrated with the numerical simulation program.

흰쥐의 간세포에서 호르몬에 의한 Acetyl-CoA Carboxylase Promoter I Activity 조절에 대한 연구 (Hormonal Regulation of Acetyl-CoA Carboxylase Promoter I Activity in Rat Primary Hepatocytes)

  • Acetyl-CoA carboxylase (ACC) is the enzyme that controls no devo fatty acid biogynthesis, and this enzyme catalyzes the carboxylation pathway of acetyl-CoA to malonyl-CoA. Acetyl-CoA carboxylase gene expression was regulated by nutritional and hormonal status. The present study was performed to identify the regulation mechanism of ACC gene promoter I. The fragments of ACC promoter I -1.2-kb region wert recombined to pGL3-Basic vector with luciferase as a reporter gene. The primary hepatocytes from the rat were used to investigate the hormonal regulation of ACC promoter I activity. ACC PI (-1.2)/Luc plasmid was trtransferred into primary hepatocytes using lipofectin. Activity of luciferase was increased two-fold by 10-9M, three-fold by 10-8M, 10-6M, 3.5-fold by 10-6M, and 4.5-fold by 10-7M insulin treatment, respectively. In the presence of dexamethasone (1 $\mu$M), the effects of insulin increased about 1.5-fold, showing the additional effects of dexamethasone. Moreover, the activity of luciferase increased with insulin+dexamethasone, insulin+T3, dexamethasone+T3, and dexamethasone+insulin+T3 treatment approximately 6-, 4-, 6.5-, and 10-fold, respectively. Therefore it can be postulated that 1) these hormones coordinately regulate acetyl-CoA caroxylase gene expression via regulation of promoter activity, 2) the -1.2-kb region of ACC promoter I may have the response element sequences for insulin, dexamethasone, and T3.

Effects of Light on the Expression of 1-Aminocyclopropane-1-Carboxylic Acid Synthase and Oxidase Genes in Mung Bean Hypocotyls

  • The effects of light on the regulation of ethylene biosynthesis during development of mung bean seedlings were investigated by monitoring the differential expression of seven 1-aminocyclopropane-l-carboxylate (ACC) synthase and two ACC oxidase genes. Among them, only the expression of VR-ACS1, VR-ACS6, VR-ACS7, VR-ACO1 and VR-AC02 was observable in etiolated mung bean hypocotyls. When the seedlings were de-etiolated for 1 d under a light/dark cycle of 16 h/8 h, the expression of VR-ACS6, VR-ACS7 and VR-ACO2 was controlled negatively by light. The expression of VR-ACS1 showed a tendency to increase until 6 h after a dark-to-light transition and then decreased at 12 h. On the other hand, the expression of VR-ACO1 was mostly constitutive up to 12 h after the dark-to-light transition. The opening of hypocotyl hooks during de-etiolation in the light was stimulated by the inhibition of the action of endogenous ethylene in the presence of 1-MCP. These results suggest that the negative regulation of light on the expression of ACC synthase and ACC oxidase genes eventually results in the inhibition of ethylene production with an acceleration of the opening of apical hooks.

  • 최근 인터넷의 급속한 발달과 스마트폰의 부각으로 인해서 디지털 콘텐츠의 활용이 증가되고 있으나 다양한 공격으로 지적 재산권을 침해할 수 있다. 디지털 콘텐츠의 공모공격에 대처하기 위한 대표적인 핑거프린팅 기술인 BIBD AND-ACC 방식은 매트릭스 연산의 복잡성으로 많은 사용자가 사용하기에는 연산문제가 발생한다. 따라서 본 논문에서는 BIBD 이론을 기반으로 한 공모보안 코드를 생성하여 다수의 콘텐츠 구매자에게 분배 가능한 삽입 기법을 제안한다. 제안한 기법은 컴퓨터 시뮬레이션을 통해 증폭계수 ${\alpha}$가 1일때 PSNR이 50.84dB로 실제 사용에 문제가 없는 화질임을 확인하였고 공모공격에 가담한 공모자 추출이 가능함을 실험을 통해 확인하였다.

Characterization of the TAK1 gene in Apis cerana cerana(AccTAK1) and its involvement in the regulation of tissue-specific development

  • TGF-$\beta$ activated kinase-1 (TAK1) plays a pivotal role in developmental processes in many species. Previous research has mainly focused on the function of TAK1 in model organisms, and little is known about the function of TAK1 in hymenoptera insects. Here, we isolated and characterized the TAK1 gene from Apis cerana cerana. Promoter analysis of AccTAK1 revealed the presence of transcription factor binding sites related to early development. Real-time quantitative PCR and immunohistochemistry experiments revealed that AccTAK1 was expressed at high levels in fourth instar larvae, primarily in the abdomen, in the intestinal wall cells of the midgut and in the secretory cells of the salivary glands. In addition, AccTAK1 expression in fourth instar larvae could be dramatically induced by treatment with pesticides and organic solvents. These observations suggest that AccTAK1 may be involved in the regulation of early development in the larval salivary gland and midgut.

Effects of Insulin on Acetyl-CoA Carboxylase in Bovine Mammary Secretory Cells

  • Objectives of this study were to determine effects of insulin on acetyl-CoA carboxylase (ACC) activity and correlate this activity with relative amounts of ACC in MAC-T cells. MAC-T cells were grown in Medium 199 supplemented with fetal bovine serum (5%), cortisol ($1{\mu}g/ml$), and insulin ($1{\mu}g/ml$). At confuluence, the cells were transferred to $100mm^2$ culture dishes coated with the extracelluar matrix. After 10 h of incubation, the media were replaced with media without fetal bovine serum and the concentration of insulin was lowered to 5 ng/ml. After 24 h, the media were changed to contain the varying concentrations of insulin and incubations continued for 48 h. The addition of insulin resulted in increases in the specific activity of ACC. The maximal effects of insulin on the ACC activity occurred at concentrations of insulin, 1,000 ng/ml. In contrast, the relative change in lactate dehydrogenase (LDH) activity in response to increasing insulin concentration was minimal as compared to the effects of insulin on ACC. Transblot and enhanced chemiluminescence (ECL) analysis indicated that the increase in ACC activity in MAC-T cells caused by insulin were due to actual increases in amounts of enzyme.
