미더덕 껍질 추출물을 함유한 곤약의 제조 및 품질 분석 (Qualities of Konjac Containing Tunic Extract from Styela clava)

  • 수산물 가공 부산물로 여겨지는 미더덕 껍질을 소재로 한 식품을 개발하기 위하여 건조한 미더덕 껍질에 물을 가하여 끓인 추출물을 얻은 후, 이를 이용하여 미더덕 껍질 추출물 함유 곤약을 제조하였다. 미더덕 껍질 추출물은 농도가 증가할수록 ABTS 라디칼 소거능이 증가한 것으로 보아 항산화능을 보유하고 있었다. 미더덕 껍질 추출물 함유 곤약의 색도는 미더덕 껍질 추출물의 함량이 증가할수록 L값은 감소하지만 a값과 b값은 증가하였고, 갈색도도 증가하는 경향을 보였다. 또한, 미더덕 껍질 추출물의 함량이 증가할수록 곤약의 강도와 경도가 증가하였으며, 절곡검사 결과 모든 곤약에서 유연성과 탄력성이 우수하였다. 관능검사 결과, 맛과 향은 농도가 높아질수록 미더덕 특유의 맛과 향이 높게 나타났으며, 질감과 촉촉한 정도는 대조군을 포함한 모든 곤약에서 유의차를 보이지 않았고, 전체적인 선호도는 25% 농도의 미더덕 껍질 추출물을 함유한 곤약에서 가장 높았다. 이상의 결과는 적절한 농도의 미더덕 껍질 추출물을 이용하면 기존의 곤약과 비교하여 영양적, 물성적, 관능적으로 소비자들에게 긍정적인 반응을 보일 수 있는 제품이 제조될 수 있음을 시사한다.

감귤 콤부차 발효액의 인체 방광암세포에 대한 성장억제와 Apoptosis에 미치는 영향 (Growth Inhibition and Induction of Apoptosis in Human Bladder Cancer Cells Induced by Fermented Citrus Kombucha)

  • 본 연구에서는 홍차버섯이라고 알려진 콤부차(Kombucha, K)에 플라보노이드 성분 및 각종 기능성 물질이 풍부한 감귤액을 첨가하여 감귤의 생리활성 물질들이 콤부차로 이행되는 효과를 기대하여 감귤 콤부차(citrus Kombucha, CK)를 배양한 후 항산화 능력 및 인체 방광암세포(T-24와 5637)를 이용한 항암 효과를 확인하고 더 나아가 암의 증식을 억제시킬 수 있는 천연소재 탐색을 목적으로 연구를 진행하였다. 항산화 및 총페놀 함량 결과는 K보다 CK의 항산화 능력과 페놀 함량이 높게 확인되었으며 방광암세포 T-24와 5637에 K 혹은 CK를 24시간 처리한 후 MTT assay를 통해 세포독성을 확인한 결과 농도 의존적으로 생존율이 감소하였다. 특히 T-24 세포에서는 CK를 처리하였을 때 현저한 세포의 형태적 변화를 확인하였다. Western immunoblot을 통해 apoptosis 관련 단백질들의 발현을 확인하였는데 T-24에 CK 처리하였을 때 Bcl-2의 발현은 크게 감소하였으며, pro-caspase-9, pro-caspase-8, pro-caspase-3는 농도가 높아질수록 감소하였으며, cleaved caspase-9, cleaved caspase-8, cleaved caspase-3는 농도가 높아질수록 증가하는 경향을 보였다. 또한, cleaved PARP가 증가함을 확인할 수 있었다. 이상의 결과에서 일반 콤부차보다 감귤액을 첨가한 감귤 콤부차가 인체 방광암세포 T-24에 caspase에 의한 apoptosis가 유도됐음을 확인할 수 있었다.

레몬그라스 분말을 첨가한 머핀의 품질특성 및 항산화 효과 (Quality Characteristics and Antioxidant Activity of Muffins containing Lemongrass Powder)

  • 레몬그라스 분말을 첨가한 머핀의 품질특성 및 항산화를 측정하기 위하여 레몬그라스 분말 0%, 1%, 3%, 6%, 9%를 첨가하여 머핀을 제조하였다. 머핀의 높이, 부피 및 비체적은 분말의 첨가량이 증가할수록 감소하였다. 레몬그라스 분말 첨가군의 수분 함량과 점도는 대조군과의 유의적 차이가 나타내지 않았으며, 색도 측정 결과, 명도와 황색도는 레몬그라스의 농도가 증가할수록 유의적으로 감소하였으나, 적색도는 증가하였다. Texture를 측정한 결과, 레몬그라스 분말의 씹힘성은 첨가량이 증가할수록 감소하였다. 관능검사는 색, 향, 맛, 조직감, 기호도를 측정했으며, 그 결과 3% 첨가군에서 가장 높은 관능검사 결과를 보여주었다. 레몬그라스 머핀의 DPPH 라디칼 소거능 측정 결과, 레몬그라스의 농도가 증가함에 따라 유의적으로 항산화 활성이 증가하였으므로, 레몬그라스의 첨가가 머핀의 기능성에 좋은 영향을 나타낼 것으로 사료된다.

당 침지액 농도에 따른 건조 둥근 마의 품질 특성 및 항산화 효과 (Quality Characteristics and Antioxidant Activity of Dioscorea bulbifera, under Various Oligosaccharide Pre-Soaking Condition)

  • This study was investigated the quality characteristics and antioxidant of dried Dioscorea bulbifera with various pre-soaking concentrations of oligosaccharide. Dioscorea bulbifera are prepared by additions of 0, 4, 6, 8 and 10% oligosaccharide solution, and dried at $50^{\circ}C$. The effects of pre-soaking percent of Dioscorea bulbifera slices were evaluated by the moisture, soluble solid, pH, titratable acidity, color, browning degree, texture, antioxidant activities and sensory test. According to the percent of pre-soaking oligosaccharide solution was increased, the moisture was increased but soluble solids and titratable acidity were decreased. With respect to the result of colors, Dioscorea bulbifera slices that underwent the 10% pre-soaked process (85.86%) were lighter than control (73.88%). However, the redness and yellowness scores were the lowest than control. The springiness and cohesiveness of texture showed no significant differences among all groups. Gumminess and chewiness of texture results were increased according to per-soaking concentration increase. Also the polyphenol, flavonoid and DPPH (${\alpha},{\alpha}$-diphenyl-${\beta}$-picrylhydrazyl) and ABTS [2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)] radical scavenging activities were significantly increased with increasing immersion concentration. The Dioscorea bulbifera slices supplemented with 6% pre-soaking oligosaccharide solution treatment showed the highest total sensory score. The results of our study indicated that when pre-soaking oligosaccharide solution is used to immerse the Dioscorea bulbifera slice, it has browning inhibition and antioxidant effect.

단일 생약재 및 생약복합제 에탄올추출물의 항산화 및 항염증 활성 (Antioxidant and Anti-inflammatory Activities of Ethanol Extracts from Medicinal Herb Mixtures)

  • 본 연구에서는 문헌을 통하여 항산화 혹은 항염증 활성을 가지는 6종의 생약재와 이들 생약재를 혼합하여 제조한 생약복합제의 항산화 및 항염증 활성을 측정하였다. 총 폴리페놀 함량은 어성초, 홍화, MIX-2 에탄올추출물이 각각 58.98, 60.79, 57.74 mg/g의 함량으로 다른 시료들 보다 높게 측정되었다. 또한 플라보노이드 함량도 어성초(HC), 홍화(CT), MIX-2에탄올추출물이 각각 36.86, 19.42, 14.74 mg/g으로 높게 측정되었다. 단일 생약재와 생약복합제 에탄올추출물의 라디칼 소거능을 측정한 결과 DPPH 라디칼의 RC50 값은 어성초, 홍화, MIX-2 에탄올추출물에서 각각 69.64, 50.70, $63.49{\mu}g/mL$로 추출물 가운데 우수한 DPPH 라디칼 소거능을 보였고, ABTS 라디칼의 $RC_{50}$ 값도 어성초, 홍화, MIX-2가 각각 24.06, 18.91, $41.61{\mu}g/mL$로 DPPH 라디칼 소거능과 유사한 경향을 보였다. 시료들의 항염증 효능을 측정하기 위해 5-LO 및 COX-2 효소 저해활성을 측정한 결과 단일 생약재 에탄올추출물보다 생약복합제 에탄올추출물이 5-LO와 COX-2 효소를 동시에 저해하는 효과가 증가되는 것으로 나타났다. 염증유발 효소 저해활성이 우수했던 생약복합제 에탄올추출물들의 NO 소거활성을 비교한 결과, MIX-2 에탄올추출물이 $50{\mu}g/mL$의 농도에서부터 유의적으로 NO의 생성을 저해하는 것을 확인하였다. 따라서 단일 생약재 보다는 생약복합제가 서로 혼합되면서 시너지 효과가 나타나는 것으로 생각되며 앞으로 명확한 기전에 대한 연구들이 지속된다면 기능성 식품 소재로서의 이용이 가능할 것으로 생각된다.

호박잎 분말을 첨가한 스폰지 케이크의 품질 특성, 항산화 활성 및 저장 기간에 따른 노화 억제 효과 (Effects of Pumpkin (Cucurbita moschata Duch.) Leaf Powder on Quality Characteristics, Antioxidant Activities, and Retarding Retrogradation by Shelf-life of Sponge Cake)

  • This study investigated the quality characteristics, antioxidant activities, and retarding retrogradation of sponge cakes made with 0%, 1%, 3%, 5% and 7% pumpkin (Cucurbita moschata Duch.) leaf powder. Specific capacity significantly increased with the addition (0.33~0.38), but baking loss and dough yield were not significantly different (p<0.05). In color, there was a decrease in the L-value, a-value and b-value, but ${\Delta}E$ increased in proportion to the amount of pumpkin leaf powder. Moisture content increased in the 0~3% additions, from 27.90~31.68%, but decreased in 5% and 7% (22.37% and 28.15%, respectively). pH tended to decrease significantly according to the amount of pumpkin leaf powder (p<0.05). Hardness increased with the addition of pumpkin leaf powder, and pumpkin leaf groups presented higher springiness and cohesiveness than the control. Chewiness was not significantly differ (p<0.05). In retarding retrogradation, Avrami exponent (n) showed that addition of 5% (0.1329) had more retarding retrogradation effect than the control (0.4319), whereas time constant (1/k) showed both 3% (100.00) and 5% (70.42) addition had more effect than control (18.45). Total phenols and flavonoids content increased proportionate to the addition levels. ABTS radical scavenging activity tended to increase according to the level of pumpkin leaf powder. In sensory properties, 5% addition showed maximum color, and 3% addition had the highest scores in flavor, moistness, sweetness, chewiness and overall acceptability. These results suggested that pumpkin leaf was a good addition to improve the quality characteristics, health and sensory preferences. The most appropriate proportion to have the effect of retarding retrogradation in sponge cake is 3%.

Differences in Functional Materials between White and Black Rice Varieties

  • Rice is the world's three major crops that is the staple food of Asian. Japonica type rice is consumed in Korea. Rice cultivation area decreases steadily. That is affected by decrease of the farm income with the inventory of 1.7 million tons, which is 2 to 3 times more than the optimal amount due to the decrease in the consumption of rice per person and cultivate high yield varieties. In recent years, as income level has improved, interest in health has been high and consumption for health food has been steadily increasing. For health food, rice is added by adding grains such as millet, sorghum, oats, beans and colored rice rather than white rice. In 1997, RDA(Rural Development Administration) developed black rice 'Heuknambyeo', and then 20 varieties were bred until 2017. In CBARES(Chungbuk Agricultural Research and Extension Services), we have developed new rice varieties 'Cheongpungheukchal' in 2010, 'Cheongpungheukhyangchal' in 2014, 'Cheongpungheukchal' is high in farming preference because of high yield. Black rice has high content of GABA and water-soluble phenol, DPPH radical scavenging activity, and consumers are interested in the function in the body. Therefore, functional and antioxidant activities(anthocyanin, total polyphenol, ${\alpha}-glucosidase$ inhibition) and antioxidant activities(ABTs, DPPH) were analyzed by comparing white and black rice. Testing varieties are 'Chucheongbyeo', best quality cultivars 'Daebo' and 'Samgwang' as white rice which are cultivated much in Chungbuk area, and black rice are 10 varieties including 'Cheongpungheukchal', 'Cheongpungheukhyangchal', 'Josaengheukchal' and so on. It has transplanted on 25th May, at CBARES research paddy by 100% fertilizer recommendation rate. Harvesting time was 50 days after heading by varieties, and has researched growth properties, yield and yield components, functional and antioxidant activities. Anthocyanin content was not measured because there was no pigmented in white rice, the highest value of anthocyanin content was 'Shintoheukmi' and in the range of 125.6~249.6mg/100g by black rice varieties. Total polyphenol content was high 'Cheongpungheukchal' and 'Shintoheukmi' and in the range of 96.68~244.34 mg/100g in black rice, white rice lower than blackish rice at 19.84~22.51mg/100g. ${\alpha}-glucosidase$ inhibition was high 'Cheongpungheukchal', 'Cheongpungheukhyangchal' and 'Shintoheukmi' in the rage of 75.87~98.85% by black rice varieties, especially 'Samgwang' was 80.75% and the other white rice was higher than 58~68%.

복분자와 오디의 항산화 특성 (Antioxidant Activities of Rubus coreanus Miquel and Morus alba L. Fruits)

  • 다양한 추출용매를 사용하여 얻은 복분자와 오디의 추출용액 중에서 가장 높은 항산화 활성을 보인 60% acetone과 이들의 분획물을 대상으로 측정한 항산화 활성에서 시료별로는 오디, 분획별로는 ethyl acetate 분획의 $EC_{50}$ 값이 낮게 나타났다. 복분자와 오디의 항산화 성분 중 총 페놀성 화합물과 총 플라보노이드 함량이 안토시아닌 함량에 비해 매우 높게 나타났다. 항산화 성분과 항산화 활성의 상관성은 총 페놀성 화합물과 총 플라보노이드에서는 높은 상관성을 나타냈으나 안토시아닌에서는 오디에서만 높은 상관성을 나타내었다. 복분자와 오디의 주된 phenolic acid는 protocatechuic acid, p-hydrobenzoic acid 및 salicylic acid이었으며 주된 안토시아닌은 cyanidin-3-glucoside이었다. 위의 결과들을 종합해보면 복분자와 오디의 항산화 성분은 추출에 사용된 용매의 종류에 따라 함량 및 조성이 변화하며 그 결과로서 항산화 활성에 영향을 주게 됨을 확인할 수 있었다. 또한 오디 ethyl acetate 분획물은 DPPH assay와 reducing power에서 BHT보다 각각 2.2와 1.2배 낮은 $EC_{50}$ 값을 보여 천연 항산화 소재로서의 가능성을 보였다.

먹물버섯(Coprinus comatus )의 항산화 활성 및 Zucker rat에 대한 항비만 효과 (Antioxidant activity and anti-obesity effect of Coprinus comatus in Zucker rat (fa/fa))

  • The aim of the study was to investigate the anti-obesity effect of Coprinus comatus (CC) in high-fat diet-fed Zucker rat (fa/fa). Obesity was induced by feeding on high-fat diet (HFD) containing 60% kcal fat for 10 weeks, in which CC extracts were administrated through the gastrointestinal tract at a concentration of 200 mg/kg BW/day for 10 weeks. The total polyphenol and flavonoid contents of CC extracts were found to be $18.5{\pm}1.1mg$ of catechin equivalent/g, and $5.24{\pm}0.54mg$ of quercetin equivalent/g extract, respectively. The DPPH, ABTS, and hydroxyl radical scavenging activities of CC extracts were 15.34 %, 17.25%, and 16.18%, respectively. In animal study, CC administration significantly reduced the body weight, while there were no significant differences in the daily food intake between the HFD-fed Zucker rats and HFD plus CC-fed rats. CC treatment decreased epididymal and perirenal fat weights in HFD-fed Zucker rats. Significant decreases in the levels of triglyceride and total cholesterol in the serum and liver were observed in the CC-treated group compared with HFD-fed Zucker rats. Serum HDL-cholesterol levels in the CC-treated group were increased compared with the HFD-fed groups. Serum AST and ALT activities in the CC group were significantly lower than those of the HFD-fed group. Taken together, these data demonstrated that CC has potential in preventing high-fat diet induced obesity and is a good candidate for an anti-obesity agent.

가미계작지모탕약침(加味桂芍知母湯藥鍼)이 퇴행성관절염 병리인자 및 동물병태 모델에 미치는 영향 (Anti-inflammation and Anti-oxidation Effects of Gamikyejakjimo-tang Herbal Acupuncture on Pathologic Factor and MIA-induced Osteoarthritis Rat)

  • Objectives This study was intended to clarify the anti-inflammation and anti-oxidation effects of gamikyejakjimo-tang herbal acupuncture (GKHA) for osteoarthritis. Methods Osteoarthritis was induced by injection of MIA into right knee joint cavities of rats. Rats were divided into a total of 4 groups (n=8). The 4 groups were normal group, control group, positive comparison group and expeimental group. Indomethacin and GKHA were medicated for a total of 4 weeks. After that, functions of liver and kidney by AST, ALT, creatinine, BUN, DPPH and ABTS free radical scavenging activity, ROS (reactive oxygen species) production, NO (Total Nitric oxide), IL-$1{\beta}$, IL-6, TNF-${\alpha}$ production, weight changes in the hind legs of MIA-induced osteoarthritis rat, serum PGE2, TIMP-1, MMP-2, MMP-9, LTB4, hs-CRP, and white blood cells, neutrophils, lymphocytes, monocytes were measured. The volume of cartilage was observed by micro CT arthrography. H&E and Safranin-O staining were used to examine the injury of synovial tissue. Results 1. In the hind leg weight bearing measurement, level of weight was increased. 2. AST, ALT, BUN, creatinine were decreased. 3. The production of total white blood cell was decreased, and the production of neutrophils, lymphocytes, monocytes were significantly decreased. 4. The production of NO, PGE2, TIMP-1, MMP-2, MMP-9, LTB4 were significantly decreased, and the production of hs-CRP was also decreased but with no significance. 5. The cartilage volume was significantly increased. 6. In H&E staining and Safranin-O staning, the cartilage cell appeared to be proliferated, and proteoglycans appeared to be increased. Conclusions Based on the results above, Gamikyejakjimo-tang Herbal Acupuncture has anti-oxidation and anti-inflammation effects, which leads to suppressing the underlying causes and the progression of osteoarthritis.
