Characterization of an antimicrobial Chitinase Purified from the Grapefruit Extract (자몽 추출물로부터 분리된 항균성 Chitinase의 특성)

  • An antimicrobial chitinase was purified from grapefruit extract and its properties were characterized. The chitinase was purified with a single step chromatography on regenerated chitin affinity gel column. The molecular weight of the purified chitinase was 29 kDa. The grapefruit extract contained the chitinase protein more than 50% of its total soluble proteins measured by coomassie stained protein bands. When the purified chitinase was incubated with polymers of N-acetylglucosamine (NAG), such as mycelia of Fusarium oxysproum and swollen chitin, they were degraded to oligosaccharides, and the oligosaccharides were then further hydrolyzed by the same enzyme to monomer and dimer of NAG. This result suggests that the chitinase contained both endo- and exo- chitinase activities. The chitinase was stable to heat and pH treatment; its activity was not diminished by the heat treatment upto 7$0^{\circ}C$ for 1 hr, and it showed a pH stability in the range of pH 4.0 to 12.0.

Needs Analysis of Experiential Learning Program for Eco-friendly School Farm Activation -Target of Teachers in Elementary School- (친환경 스쿨팜 활성화를 위한 체험학습용 프로그램 요구분석 -초등학교 교사를 대상으로-)

  • This research was conducted to analysis the Elementary School Teachers' needs necessary for developing the eco-friendly school farm program. To perform these research problems, 167 people from five Elementary School group (A, B, C, D and E). Of respondents of the survey, 160 questionnaires were significant. The frequency analysis was used with SPSS 17.0 statistics program. Teacher of the needs analysis results are as follows: Teachers want to help school farm program by gardening expert, experienced volunteer. Costs of instructor and material must be supported by the government, local government. Teachers are demanding an integrated program for horticulture. The teachers would have preferred an easy plant to grow and appear in textbooks. Therefore teachers' needs to be a convert-gence in the eco-friendly school farm program manual.

Optimization of Cellulase Production from Paenibacillus jamilae BRC 15-1 (Paenibacillus jamilae BRC15-1의 Cellulase 생산 최적화)

  • In this study was selected the cellulolytic microorganism and investigated optimum condition of cellulase production for the cellulosic bioethanol production. A bacterial strain Paenibacillus jamilae BRC15-1, was isolated from soil of domestic reclaimed land. For optimizing cellulase production from the selected strain, various culture parameters were investigated such as culture medium, pH (pH 4~10), temperature ($25{\sim}50^{\circ}C$) and culture time (2~72 h). As a result, P. jamilae BRC15-1 efficiently produced cellulase from cellulosic biomass under following conditions: 24 h of culture time (pH 7, $40^{\circ}C$) in manufactured media of CMC (carboxymethyl cellulose) with peptone. Optimum saccharifying condition of crude enzyme produced from P. jamilae BRC15-1 was identified on pH 6 and $40^{\circ}C$ of reaction temperature, respectively. This crude enzyme from P. jamilae BRC15-1 was used for saccharification of pretreated sweet sorghum (Sorghum bicolor var. dulciusculum Ohwi) bagasse under the optimal condition. Finally, pretreated sweet sorghum bagasse including 0.1 g of glucan was saccharified by crude enzyme of P. jamilae BRC15-1 into 2.75 mg glucose, 0.79 mg xylose and 1.12 mg arabinose.

Analysis of Water Quality Improvement of Ceratophyllum demersum under Laboratory Condition - by Nutrients Removal Efficiency (실험실 조건에서 붕어마름의 수질개선 효과 분석 - 영양염류 제거 효율을 중심으로)

  • To evaluate the ability of the submerged plant, Ceratophyllum demersum's (C. demersum) to remove nutrients and to inhibit growth of cyanobacteria, a total of 6 mesocosms were conducted in a batch reactor for 9 days. From the 84 hr of the experiment, C. demersum was stabilized and showed daily cycle trends according to changes in pH and DO levels. The concentration of nutrients, $NH_3{^+}$, $NO_3{^-}$ and $PO_4{^3}$ continuously decreased until 9 days of the experiment, with the rapid decrease in nutrient concentration for the first 24 hours. High correlation coefficient ($r^2{\geq}0.96$, p<0.001) between the amount of C. demersum's biomass per unit area and the nutrients removal level were derived, and greater C. demersum's biomass per unit area showed higher removal efficiency of nutrients. However, there were differences in the C. demersum's activity level between batch reactors with higher and similar density of the C. demersum, but nonetheless water purification effect appears to have a significant influence due to attached algae and microorganisms. The growth rate of harmful cyanobacteria, Microcystis aeruginosa (M. aeruginosa) with C. demersum's density of 2,500 g $fw/m^2$ (100% of cover degree) was 0.31 /day, compared to the growth rate of 0.47 /day for the control group (0% of cover degree). In terms of number of cells, the control group had 1.7 times higher number of cells than the experimental group, proving that C. demersum has the ability to inhibit the growth of harmful cyanobacteria.

  • Polymer electrolyte membrane fuel cells (PEMFCs) use the bipolar plate of various materials between electrolyte and contact electrode for the stable hydrogen ion exchange activation. The bipolar plate of various materials has representatively graphite and stainless steel. Specially, stainless steels have advantage for low cost and high product rate. In this study, SUS 316 was effectively coated with 600 nm thick F-doped tin oxide (SnOx:F) by electron cyclotron resonance-metal organic chemical vapor deposition and investigated in simulated fuel cell bipolar plates. The results showed that an F-doped tin oxide (SnOx:F) coating enhanced the corrosion resistance of the alloys in fuel cell bipolar plates, though the substrate steel has a significant influence on the behavior of the coating. Coating SUS 316 for fuel cell bipolar plates steel further improved the already excellent corrosion resistance of this material. After coating, the increased ICR values of the coated steels compared to those of the fresh steels. The SnOx:F coating seems to add an additional resistance to the native air-formed film on these stainless steels.

The Effectiveness of Infrared Thermography in Patients with Whiplash Injury

  • Objective : This study aims to visualize the subjective symptoms before and after the treatment of whiplash injury using infrared (IR) thermography. Methods : IR thermography was performed for 42 patients who were diagnosed with whiplash injury. There were 19 male and 23 female patients. The mean age was 43.12 years. Thermal differences (${\Delta}T$) in the neck and shoulder and changes in the thermal differences (${\Delta}dT$) before and after treatment were analyzed. Pain after injury was evaluated using visual analogue scale (VAS) before and after treatment (${\Delta}VAS$). The correlations between ${\Delta}dT$ and ${\Delta}VAS$ results before and after the treatment were examined. We used Digital Infrared Thermal Imaging equipment of Dorex company for IR thermography. Results : The skin temperature of the neck and shoulder immediately after injury showed $1-2^{\circ}C$ hyperthermia than normal. After two weeks, the skin temperature was normal range. ${\Delta}T$ after immediately injuy was higher than normal value, but it was gradually near the normal value after two weeks. ${\Delta}dT$ before and after treatment were statistically significant (p<0.05). VAS of the neck and shoulder significantly reduced after 2 week (p=0.001). Also, there was significant correlation between ${\Delta}dT$ and reduced ${\Delta}VAS$ (the neck; r=0.412, p<0.007) (the shoulder; r=0.648, p<0.000). Conclusion : The skin temperature of sites with whiplash injury is immediately hyperthermia and gradually decreased after two weeks, finally it got close to normal temperature. These were highly correlated with reduced VAS. IR thermography can be a reliable tool to visualize the symptoms of whiplash injury and the effectiveness of treatment in clinical settings.

Seismic Capacity Evaluation of Rectangular RC Columns Strengthened with Steel Bars (강봉으로 보강된 RC 사각기둥의 내진 성능 평가)

  • With the steady increase in the annual number of earthquakes in South Korea, the need to apply seismic reinforcement on public facilities has recently increased. To reinforce seismic capacity, spaced full-column-height steel bars are attached to column faces. In this study, nonlinear finite element analysis was conducted to analyze the effect of external reinforcement steel bars on the seismic capacity of RC columns with a square or rectangular cross-section. For verification, the analysis results were compared with test results. Results showed that the finite element analysis reasonably predicted the actual structural behavior of RC columns with steel bars. In addition, both the analysis and the test results showed that the failure mode was converted from brittle failure to ductile fracture, owing to the external reinforcement steel bars. Both loading capacity and ductility were increased as well. Therefore, the external reinforcement steel bar can effectively enhance the seismic capacity of existing RC columns. This study is expected to contribute to relevant research areas such as the development of design methods.

Comparison of Antioxidant Activities of Hydrolysates of Domestic and Imported Skim Milk Powders Treated with Papain

  • Milk proteins have many potential sequences within their primary structure, each with a specific biological activity. In this study, we compared and investigated the bioactivities of hydrolysates of the domestic (A, B) and imported (C, D) skim milk powders generated using papain digestion. MALDI-TOF analysis revealed that all milk powder proteins were intact, indicating no autolysis. Electrophoretic analysis of hydrolysates showed papain treatment caused degradation of milk proteins into peptides of various size. The antioxidant activity of the hydrolysates, determined using 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and total phenolic contents (TPC) assays, increased with incubation times. In all skim milk powders, the antioxidant activities of hydrolysates were highest following 24 h papain treatment (TPC: A, 196.48 μM GE/L; B, 194.52 μM GE/L; C, 194.76 μM GE/L; D, 163.75 μM GE/L; ABTS: A, 75%; B, 72%; C, 72%; D, 57%). The number of peptide derived from skim milk powders, as determined by LC-MS/MS, was 308 for A, 283 for B, 208 for C, and 135 for D. Hydrolysate A had the highest antioxidant activity and the most potential antioxidant peptides amongst the four skim milk powder hydrolysates. A total of 4 β-lactoglobulin, 4 αs1-casein, and 56 β-casein peptide fragments were identified as potential antioxidant peptides in hydrolysate A by LC-MS/MS. These results suggest that domestic skim milk could have applications in various industries, i.e., in the development of functional foods.

A Study on Combustion Reaction Mechanism of Korean Anthracites (國産無煙炭의 燃燒反應機構에 關한 硏究)

  • The rate Constants and energies of activation for the Combustion reaction of Korean anthracites have obtained by DTA method using the following rate equation derived by authors. $K=\frac{C_3{\cdot}W_0}{{\Delta}H{\cdot}{\Delta}C{\cdot}M{\cdot}S_A}(\frac{dy}{dt}+A(y-y_3))$ The anthracites of various ranks were treated at the different temperatures in the furnace. The probable combustion reaction mechanisms have discussed with the results obtained by the X-ray diffraction method, IR spectroscophic analysis, and gas chromatography. By the intensity of d(002) Values, it was confirmed that a parts of the amorphous carbon was converted to graphite form by heat treatment. The appreciable amounts of CO gas were expelled in the combustion process and it appeared that a little amount of the gas came from the catalytic decomposition of anthracites, The functional groups such as -OH, -SH, -NH, $-CH_2-CH_3,$ -CO, -COC-. and polycondensed aromatic rings in anthracites have observed by IR spectrophotometric analysis.

Structure direct agent-assisted hydrothermal synthesis and small gases adsorption behavior of pure RHO zeolite (구조유도물질 18-crown-6 ether를 이용한 순수한 RHO 제올라이트 수열합성과 작은 가스 흡착 거동)

  • In the present study, pure RHO zeolite was hydrothermally synthesized by using 18-crown-6 ether as a structure directing agent(SDA), and the small gases adsorption was investigated. Synthesized RHO zeolite was a cube shape particle of which average edge length was around $1.2{\mu}m$ and composed of primary crystallites having a diameter of around 100 to 200 nm. RHO zeolite structure was stable under 3h calcination at $600^{\circ}C$. Water adsorption data announced that RHO zeolite has a specific surface area of 483.32 m2/g and its micropore diameter was about 4 A. Gas adsorption was studied in the pressure range of 50 to 500 kPa for $CO_2$, $N_2$, $O_2$ and $H_2$. It was evident that RHO zeolite showed a strong $CO_2$ adsorption behavior. Especially, RHO zeolite showed a transient $CO_2$ adsorption behavior. The 3h $CO_2$ up-take at 50 kPa and 500 kPa was 1.283 and 3.357 mmol/g, respectively. The $CO_2/H_2$ selectivity was around 16 at 500 kPa. Compared with gas adsorption data for some representative microporous adsorbents, it was certain that RHO zeolite is a beneficial adsorbent for $CO_2/H_2$ separation.
