새삼, 실새삼 및 갯실새삼 추출물이 Clone M-3 세포주의 Melanin 생합성 및 Tyrosinase 활성에 미치는 영향과 세포독성 및 항산화효과 (Inhibitory Effects on Melanin Biosynthesis and Tyrosinase Activity; Cytotoxicity in Clone M-3 and Antioxidant Activity by Cuscuta japonica, C. australis, and C, chinensis Extracts)

  • Water extracts, ethanol extracts, and juice of Cusuta japonica, C, australis, and C. chinensis were prepared, and their cytotoxicity, antioxidant activity and inhibitory effects on tyrosinase activitiy and melanin biosynthesis were estimated by using melanoma Clone M-3. From this study; the following conclusions were attained. Extracts of Cuscuta japonica, C. australis, and C. chinensis showed noticeable cytotoxicity except ethanol extracts from the stem of C. australis. A maximual cytotoxicity was observed with tile ethanol extract from the seed of C, australis (87.39%). While the ethanol extract from the seed of C. japonica (91.88%) showed the most pronounced inhibitory effect on melanin biosynthesis, the water extract from the stem of C. japonica (1.05%) possessed very little inhibitory effect. The most inhibitory effect on tyrosinase activity was observed with the water extract from the stem of C. australis (76.67%). Howeverr the water extract from the stem of C. japonica showed a very poor effect on the inhibition of tyrosinase activity All the preparations, except extracts from the seed of C. australis were able to remove reactive oxygen species (ROS) in a dose-dependent manner. The juice of C. japonica demonstrated the strongest activity (59.02%).

감귤류 종자 추출물의 향균활성 (Antimicrobial Activity of Extracts from Citrus Seeds)

  • To develope natural food preservatives antimicrobial effect of the natural products against food-related bacteria and yeast The purpose of this study was evaluate antimicrobial effect of the citrus seeds. antimicrobial activities of methanol extracts from the citrus seeds investigate against Escherichia coli O26, Staphylococcus aureus 6358, Saccharomyces cerevisiae IBM 4274, Bacillus licheuiformis 9945a and Alcoligenes faecalis. Citrus seeds is containing to moisture 4∼6.5%, curd protein 11∼15%, curd lipid 32∼46%, curd carbohydrate 22∼45 % and ash 2∼4 %, that is containing to flavornoid 12∼48mg% and phenolic compound 22∼53mg%. Solidity content of the methanol extract from the citrus seeds was 0.8∼1.2%. Almost all of the methanol extracts from citrus seeds exhibited growth inhibiting activities for most of microorganisms tested. The methanol extracts from Citrus grandis, C. sunki, C. sulcata showed the growth inhibitory effects against Escherichia coli O26. The methanol extracts from C. obovoidea, C. sulcata, C. aurantium showed the growth inhibitory effects against Staphylococcus aureus 6358. The methanol extracts from C. obovoidea, C. sulcata, C. tangerina showed the growth inhibitory effects against Saccharomyces cerevisiae IBM 4274. The methanol extracts from C. obovoidea, C sunki, C. sulcata, C. tangerinan, C. natsudaidai, C. iyo, C. aurantium showed the growth inhibitory effects against Bacillus licheuiformis 9945a. The methanol extracts from C. obovoidea, C sunki, C. sulcata, C. aurantium showed the growth inhibitory effects against Alcoligenes faecalis. Among this especially, Showed growth inhibiting activity of the methanol extracts from Citrus sulcata that about microorganisms investigated. If apply searching suitable application method about such the citrus seeds antimicrobial activity, role as good antimicorbial material in storage or cooking of food, processing is expected.

LPS로 유도한 Raw 264.7 세포에서 A.C.C. 추출물의 항염증 효과 (The anti-inflammation effects of A.C.C. extracts on the LPS-induced Raw 264.7 cell)

  • 본 연구의 목적은 황백을 포함한 총 14가지의 한약재를 증류 추출하여 얻은 시료(A.C.C. 추출물)의 항염증 활성을 확인하고 임상적인 효과를 평가하는 것이다. 이를 위해, Lipopolysaccharide(LPS)로 자극한 RAW 264.7세포에서 세포가 방출하는 nitric oxide(NO) 생성량과 염증성 사이토카인인 tumor necrosis factor(TNF)-${\alpha}$, interleukin(IL)-$1{\beta}$ 및 IL-6 생성량의 변화를 확인하였다. 그 결과, A.C.C. 추출물은 세포 독성없이 LPS가 증가시킨 NO와 염증성 사이토카인들의 생성을 강력하게 억제하였다. 또한, A.C.C. 추출물은 Pseudomonas aeruginosa, Staphylococcus aureus, MRSA(Methicillin-resistant Staphylococcus aureus), Candida albicans and Streptococcus mutans 등의 세균들에서 99.9%의 강력한 세균 감소율을 보여 주었다. 이는 A.C.C. 추출물이 항염증 효과와 함께 강력한 항균 효과를 가진 유효한 성분인 것을 의미한다. 또한, 기저귀 발진, 가려움증, 땀띠 증상을 겪고 있는 영 유아에게 A.C.C. 추출물을 도포하였을 때, 발진, 아토피, 짓무름, 가려움증, 땀띠 등의 증상 개선 효과를 나타내었으며, 영 유아 피부의 땀띠가 2주일 경과 후 상당히 완화된 것을 육안으로 확인할 수 있었다. 이는 A.C.C. 추출물이 임상적으로도 항염증 및 항균작용을 통하여 기저귀 발진 및 땀띠 등의 증상을 완화 시키는 물질임을 확실히 한 것이다. 그러므로, A.C.C. 추출물이 염증성 질환에 대한 효과적인 대안이 될 수 있을 것으로 사료된다.

Multidrug-resistance reversing activity of the local Citrus fruits in Jeju Island, Korea

  • We examined whether extracts from 14 local citrus spp. on Jeju Island (Korea) contained chemosensitizing activity that would increase the cytotoxic effect of vincristine(VCR) in drug-resistant cancer cells. We report that methanol extracts from fruits and flowers of some species had a chemosensitizing effect that reversed P-glycoprotein (Pgp)-mediated multidrug resistance (MDR). Using drug-sensitive AML-2/WT and drug-resistant AML-2/D100 in the absence of VCR in human acute myelogenous leukemia cells we found that fruit or flower extracts alone generally had low cytotoxicity $(IC_{50}>200\;{\mu}g/ml)$. In studies examining the effect of extracts on 120 ng/ml VCR cytotoxicity in drug-resistant AML-2/D100 cells, we found that immature fruit extracts had greater chemosensitizing activity than either extracts from mature fruit or flower. Of the 14 species examined, the immature fruit extract from Inchangkyool (Citrus ichangiensis) showed the hishest chemosensitizing index(CI) valus. Immature fruit extracts of Hongkyool(C. tachibana), Byungkyool(C. platymamma), Cheongkyool(C. nippokoreana) and Jinkyool (C. sunki) also strongly potentiated VCR cytotoxicity in AML-2/D100 cells. The chemosensitizing effect of peel extracts was 2-10-fold that of whole fruit extracts from Hongkyool (C. tachibana), Byungkyool (C. platymamma) and Inchangkyool (C. inchangiensis). The CI values for flower extracts were higher than those for mature fruit extracts, but lower than those for immature fruit extracts. These results indicate that immature citrus fruits contain compounds that do not exert their activity solely through cytotoxicity. In particular, Incahngkyool (C. inchangiensis), Byungkyool(C.platymamma), Cheongkyool(C. nippokoreana) and Hongkyool (C. tahibana) may be useful sources of chemosensitizing compounds.

Fungicidal Activities of 51 Fruit-Derived Extracts in vivo against Six Phytopathogenic Fungi

  • Methanol extracts from 51 fruits were tested for their fungicidal activities against six phytopathogenic fungi in a greenhouse. The efficacy varied with both the plant pathogen and fruit species used. At 10 and 5 mg/pot, methanol extracts of Poncirus trifoliata peel and seed gave over 80% control values against Pyricularia grisea, and strong fungicidal activities against Rhizoctonia solani were showed from the extracts of Citrus paradisi peel and Punica granatum leaf. In a test with Botrytis cinerea at 5 mg/pot, the extracts of C. sinensis seed and D. kaki leaf produced potent fungicidal activities, and the extracts of C. crenata peel and leaf, Ch. sinensis seed, P. trifoliata peel, and Z. jujuba leaf had strong fungicidal activities. At 5 mg/pot, strong fungicidal activities were produced in the extracts of P. trifoliata peel and seed against Phytophthora infestans and in the extracts of P. ussriensis var. macrostipes fruit and seed, C. crenata peel, C. crenata leaf, C. paradisi peel, P. trifoliata peel, P. granatum peel, and Z. jujuba leaf against Puccinia recondita. In a test with E. graminis, potent activities at 10 mg/pot were produced from the extracts of Ch. sinensis seed, C. sinensis seed, P. trifoliata leaf, P. ussriensis var. macrostipes fruit and seed, and Vitis vinifera seed. In the control effect of seven extracts against B. cinerea strains resistant to carbendazim, procymidone, and diethofencarb, extracts of C. crenata peel and leaf, Ch. sinensis seed, and P. trifoliata peel were highly effective against all strains of B. cinerea. Furthermore, potent fungicidal activities were produced from the extracts of C. sinensis seed and D. kaki leaf against the SSR, SRR, and RRS, and Z. jujuba leaf against the SSR and RRS strains. As a naturally occurring fungicide, these fruit-derived materials could be useful as new fungicidal products against phytopathogenic fungi.

뽕나무(Morus alba)와 꾸지뽕나무(Cudrania tricuspidata)의 부위에 따른 항산화 활성 및 3T3-L1세포 지방축적 억제 효과 (Antioxidative Activities and Inhibitory Effects on Lipid Accumulation of Extracts from Different Parts of Morus alba and Cudrania tricuspidata)

  • In this study, we examined antioxidative effects and the anti-adipogenesis effect of different parts of Cudrania tricuspidata (C), and Morus alba (M). Total polyphenol contents were highest in M-root ($34.56{\pm}0.045mg\;GAE/g$), and there was no significant difference, between C-root and M-leaf. Total flavonoid contents of C-root were highest ($23.07{\pm}0.004mg\;QE/g$). To examine antioxidant activities of C and M extracts, DPPH and ABTS radical scavenging activity, and FRAP assay, was used. Results show that antioxidant activities of C and M extracts increased, in a dose-dependent manner. Adipocytes are generated by preadipocyte differentiation, during adipogenesis. Matured adipocytes accumulate in abnormal and cause obesity. We investigated effects of leaf and root extracts of C and M, on lipid accumulation, in 3T3-L1 adipocytes. Changes in cell morphology, and degrees of lipid accumulation in adipocytes, were evaluated by Oil Red O staining. Root extracts of C and M, reduced lipid content in a dose-dependent manner. Therefore, root extracts of C and M, may be good candidates for managing obesity.

오자연종환(五子衍宗丸) 추출물의 항산화 효과에 관한 연구 (Study of the Antioxidant Effect of Ojayeonjong-hwan)

  • Objective: In this study, the antioxidant activity of Ojayeonjong-hwan extracts was compared, and the following results were obtained. Methods: For hydrothermal and ethanol extracts, DPPH free radical and ABTS cationic radical erasing activity and reducing power using the FRAP method were compared, and the association between the antioxidant power of each extract and total phenol content was investigated. Significant results were obtained through in vitro apoptosis analysis through FFITC staining, mitochondrial membrane potential analysis, and ROS level measurement using C2C12 myoblastoma. Results: 1. In a comparison of DPPH free radical and ABTS cationic radical scavenging activity, water, and 70% ethanol extracts of Ojayeonjong-hwan (WEO and EEO) showed superior radical scavenging ability. 2. In the results of reducing power using the FRAP method, WEO and EEO showed antioxidant activity, which was shown to be dependent on the total phenol content contained in the extracts. 3. In comparison to the protective effect against H2O2-induced oxidative stress in C2C12 myoblasts, water extracts had no significant effect, but 70% ethanol extracts inhibited H2O2-mediated cytotoxicity in a concentration-dependent manner. 4. The cytotoxic protective effect of EEO against oxidative stress in C2C12 myoblasts was correlated with its inhibitory effects on H2O2-induced apoptosis and cell-cycle arrest. 5. In H2O2-treated C2C12 myoblasts, the apoptosis inhibitory effects of EEO were associated with the suppression of mitochondrial dysfunction and DNA damage. 6. The protective effects of EEO against H2O2-induced oxidative stress in C2C12 myoblasts were directly related to the inhibition of ROS generation. Conclusions: Ojayeonjong-hwan extracts all have protective potential against oxidative stress.

Effects of three local Malaysian Channa spp. fish on chronic inflammation

  • Water and chloroform/methanol extracts of the three local Malaysian snakehead fish, Channa striatus (striped snakehead), Channa micropeltes (giant snakehead) and Channa lucius (blotched snakehead) were evaluated for inhibitory activity in chronic inflammation, using cotton pellet granuloma test. Both water extracts of C. striatus and C. micropeltes showed marked inhibition of the transudative and proliferative components of chronic inflammation (42.9 and 31.2% respectively for C. striatus, 35.6 and 26.2% for C. micropeltes) when compared to those of mefenamic acid (25.1 and 21.3% respectively) and piroxicam (36.1 and 26.2% respectively). The chloroform/methanol extracts did not exhibit any anti-inflammatory effects. These results indicated that C. striatus has more anti-transudative and anti-proliferative activities than the extract of C. micropletes. C. lucius extract in contrast, did not inhibit these two components. This present study indicated the beneficial effects of the water extracts of C. striatus and C. micropeltes, but not C. lucius on chronic inflammation.

Antifungal Activity of Methanolic of Centella asiatica and Andrographis panicuiata

  • The antifungal activity of methanolic extracts of Centella asiatica and Andrographis paniculata leaves was observed against fourteen fungi, viz., Alternaria alternata, A. brassicae, A. brassicicola, A. solani, A. tenuissima, Cercospora blumae, Curvularia lunata, C. penniseti, and Drechslera monoceras, D. oryzae, D. turitica, Fusarium albizziae and F. udum. Different concentrations of the methanolic extract (1000, 2000, 3000, 5000, 7000, 10000ppm) were used. The effect of mixed leaf extract (1500 ppm of C. asiatica + 1500 ppm of A. paniculata) and its 1:2 ad 1:4 dilutions were also studied. The individual extracts of both the plants showed significant inhibitory effect on spore germination of all the fungi tested. F. udum, F. albizzae, D. oryzae, D. turtica, and D. monoceras were particularly sensitive to these extracts. In general, the extract of C. asiatica showed a higher inhibitory effect in all concentrations against all the fungi as compared to A. paniculata, except for A. brassicae A. solani, D. oryzae, D. penniseti and Curvularia sp. The inhibitory effect of extracts increased when they were used in combination with or without dilutions against A. brassicicola, A. solani A. brassicae, A. alternata, A. tenussima, C. blumae, C. lunata, C. penniseti and Curvularia species. Higher efficacy of active ingredient of these extracts under field condition is envisaged against plant pathogens.

항생제(抗生劑) 내성(耐性) 화농균(化膿菌)의 생육(生育)을 억제(抑制)하는 한약재(韓藥材) 탐색(探索)에 관(關)한 硏究(연구) (A Study on Screening of Oriental Medicines Against Antibiotics-resistant Staphylococcus aureus)

  • The various oriental herbal medicines, which have usually been used for treatment of reducing fever, purging intense heat and detoxication, were screened to determine the antibacterial activity and the minimal inhibitory concentration against antibiotics-resistant Staphylococcus aureus. The results obtained were as follows: 1. Among the 23 oriental medicines tested, the water-soluble extract of Coptis japonica showed very excellent antibacterial activity against antibiotics-sensitive and -resistant S. aureus. A little activity was found in the extracts of Scutellaria baicalensis, Eriobotrya japonica and Prunus mume against antibiotics-sensitive S. aureus and in that extracts of P. mume, Schizandra chinensis and S. baicalensis against antibiotics-resistant S. aureus. When C japonica was used, the high antibacterial activity was shown against Bacillus subtilis and other extracts showed a little or no antibacterial activity against B. subtilis and E. coli as a control. 2. The highest antibacterial activities against antibiotics-resistant S. aureus were found in the ethanol-soluble extract of C japonica. Other ethanol-soluble extracts of S, baicalensis, S. chinensis and P. mume showed a little activity against both antibiotics-sensitive and antibiotics-resistant S. aureus. 3. Among the medicinal herbs tested, the water- and ethanol extracts of C. japonica showed very excellent antibacterial activity against the pathogenic S. aureus and both Escherichia coli and Bacillus subtilis used as a control. 4. When the water-soluble extracts of C. japonica and S. baicalensis, minimal inhibitory concentrations (MICs) against antibiotics-sensitive S. aureus were $20mg/m{\ell}\;and\;30mg/m{\ell}$, respectively. The MICs of the ethanol-soluble extracts of C. japonica and P. scabriosaefolia were $7.5mg/m{\ell}\;and\;12mg/m{\ell}$, respectively. 5. For the MICs against antibiotics-resistant S. aureus, the water-soluble extracts of C. japonica and P. mume showed $15mg/m{\ell}\;and\;10mg/m{\ell}$, respectively. The MICs of the ethanol-soluble extracts of C. japonica and P. mume were $13mg/m{\ell}\;and\;20mg/m{\ell}$, respectively. As a result, the highest antibacterial activity was found in both water- and ethanol-soluble extracts of C. japonica against antibiotics-resistant S. aureus. And the water- and ethanol-soluble extracts of S. baicalensis, S. chinensis and P. mume showed comparatively high antibacterial activities against both antibiotics-sensitive and -resistant S. aureus.
