This study examined the efficacy of Atractylodes macrocephala Koidz (AMK) protein and polysaccharide extracts as adjuvant or adjuvant booster when given together with porcine pleuropneumonia vaccine. Experimental mice (n = 5/group) were subcutaneously immunized with $25{\mu}g$ ApxIIA #3 antigen, a target protein against A. pleuropneumoniae, together with alum and/or various concentrations ($0-500{\mu}g$) of the AMK extracts, while the control group received PBS only. Immunization with ApxIIA #3 antigen increased the antigen-specific IgG titer and this increase was enhanced in the immunization together with AMK protein, but not polysaccharide extract. Supplementation of AMK protein extract exhibited dose-dependent increases in the antigen-induced protective immunity against A. pleuropneumoniae challenge and in the lymphocyte proliferation specific to the antigen. Glycoproteins present in the AMK extract were the active components responsible for immune response induction. Collectively, the present findings suggest that AMK glycoproteins are useful as immune stimulating adjuvant or adjuvant booster.
Appearance patterns of freshwater fish were investigated in the western Demilitarized Zone (DMZ) adjacent areas near Imjin River in Gyeonggi-do, Korea, of 2014. A total of 61 species belonging to 14 families were found in the river during the survey. The dominant and subdominant species were Zacco platypus (Relative abundance, 20.3%) and Zacco koreanus (12.6%), respectively. Other abundant species include Rhinogobius brunneus (9.2%), Pungtungia herzi (4.4%), Acheilognathus yamatsutae (4.4%), and Pseudogobio esocinus (4.0%). Among the residing species, 24 species (39.3%) were endemic to Korea; and 1 species, Carassius cuvieri, was non-indigenous species. Hemibarbus mylodon is a natural monument, and Lethenteron reissneri, Acheilognathus signifer, Pseudopungtungia tenuicorpa, Gobiobotia brevibarba, G. macrocephala, and G. naktongensis are endangered species in Korea. In addition, Anguilla japonica and Plecoglossus altivelis are catadromous and amphidromous species, respectively. Among the investigated sites, the lower part of Sami Stream (St. 5, 6) and Gunnarm-dam region (St. 11, 14) are particularly valuable for the conservation of species diversity, because many fish species (33~42 species) including a natural monument and several endangered species inhabit in these regions.
Field monitoring was conducted for fish fauna and community assessment at 7 sites from April 2008 to October 2009 in the Chogang Stream. The number of fish samples in this period were 4,669 in 36 species of 9 families. Family Cyprinidae take 66.7 (24 species), Cobitidae, Bagridae, Centropomidae and Odontobutidae occupied 5.6%(each 2 species), respectively. Twenty species (55.6%) including Acheilognathus koreensis and A. yamatsutae were found endemic out of the 36 species. The species of Pseudopungtungia nigra, Gobiobotia macrocephala and Gobiobotia brevibarba were endangered species. The most frequently found one was Zacco koreanus (34.0%, n=1,588) followed by Z. platypus (22.6%, n=1,053) and Coreoleuciscus splendidus (13.3%, n=623). The lower reach of Chogang Stream was more abundance of species, high diversity, evenness and richness, and lower dominance index than those of the upper reach. According to the dendrogram established at 0.5 level of similarity rate, sampling stations were divided into 3 groups. They were divided into upper most stream (St. 1~St. 2), upper stream (St. 3), middle and lower stream (St. 4~St. 7). Overall, it was concluded that the Chogang Stream has been relatively well protected from the anthropogenic disturbance for the legally protected species including the endemic species studied in this study.
Bioassay techniques using young leaves and roots were developed to screen resistance of Atractylis spp. against Phytophthora drechsleri. Among 638 plants collected from various regions of Korea from 1994 to 1996, 67 were pre-screened in fields naturally infested with P. drechsleri, which is the causal pathogen of rhizome rot of Atractylis. Among the pre-screened sources, 18 (ca. 26.8%) were highly resistant to the pathogen in leaf inoculation. In the root inoculation test, abundant sporangia were formed in susceptible plant roots, while only a few or no sporangia were produced on the roots which were found resistant in the leaf inoculation test. Among the selected resistant plants, A. japonica 96066 and 96104 were used to cross with another species, A. macrocephala 96362 that showed high yield with good quality of rhizome but susceptible to the pathogen. The F$_1$hybrids designated as HA03 turned out to be resistant to the pathogen, indicating that resistant gene(s) was inherited. Among intra-species hybrids of A. japonica, HA07 and HA09 were resistant to the pathogen in leaf inoculation and moderate in root inoculation. However, HA08 was susceptible in both inoculation tests. This result suggests that the parent material might be genetically heterogeneous. Further genetic study should be carried out to verify this phenomenon.
This study was carried out to determine the functional works and basic ingredients of Sagoonja-Tang on white pan bread. The -experimental groups consisted of 6% mixtures of five(Ed- confirm the number) kinds powders, : B1 for Sagoonja-Tang powder, B2 for Panax ginseng, B3 for Poria cocos, Koidz, B4 for Atractylodes macrocephala, and B5 for Glycyrrhiza uralensis Fisch. The volume of white pan bread somewhat decreased after adding the ingredients, and the volume differences in order from highest to lowest, were white pan bread control>B2>B3>B4>B1>B5. For white pan bread, pH of control, B3, and B5 decreased on the 2nd day during storage, but increased on the 4th and 7th days. In the texture analyses of white pan bread, B4 and B5 showed a higher degree of hardness than that of control, while B2, and B3 were the lowest. In sensory test, the overall acceptability of regular white pan bread in order from highest to lowest was control>B3>B2>B1 ${\cdot}$ B5>B4.

Atrartylodis Rhizoma(蒼朮)'s origin plants are Atratylodes lancea and A. Chinensis in Chinese, Japanese and Korean pharmacopoeia. A. Japonica is only indigenous in Korea, it is actually used as Atractylodis Rhizoma in Korean market. A. lancea is used in Hunan province, China and A. Chinensis is used in Hubei province, China. It is impossible to distinguish with species differency as macro- and micro-morphology. We tried to distinguish with species differency by HPLC and GC-Mass spectra. Atractylone(mw. 216) which is a marker compound in Atractylodis Rhizoma Alba(白朮) was detected in A. japonica. Atractylodin (mw.182) was detected in A. lancea and two eudesmadien derivatives (mw. 204) were detected in A. chinensis. HPLC chromatogram showed the same patterns. As a result, we propose that A. japonica will be added as Atractylodis Rhizoma (蒼朮)'s origin plant in Korean Pharmacopoeia. Atractylodis Rhizoma Alba(白朮)'s origin plants are A. macrocephala in China, and A. Japonica and A. ovata in Korea and Japan. In GC-Mass analysis, all samples showed same patterns and the main compound was atractylone.
Soybean mosaic virus (SMV) is the predominant viral pathogen that affects the yield and quality of soybean. The natural host range for SMV is very narrow, and generally limited to Leguminosae. However, we found that SMV can naturally infect Pinellia ternata and Atractylodes macrocephala. In order to clarify the molecular mechanisms underlying the cross-family infection of SMV, we used double-stranded RNA extraction, rapid amplification of cDNA ends polymerase chain reaction and Gibson assembly techniques to carry out SMV full-length genome amplification from susceptible soybeans and constructed an infectious cDNA clone for SMV. The genome of the SMV Shanxi isolate (SMV-SX) consists of 9,587 nt and encodes a polyprotein consisting of 3,067 aa. SMV-SX and SMV-XFQ008 had the highest nucleotide and amino acid sequence identities of 97.03% and 98.50%, respectively. A phylogenetic tree indicated that SMV-SX and SMV-XFQ018 were clustered together, sharing the closest relationship. We then constructed a pSMV-SX infectious cDNA clone by Gibson assembly technology and used this clone to inoculate soybean and Ailanthus altissima; the symptoms of these hosts were similar to those caused by the virus isolated from natural infected plant tissue. This method of construction not only makes up for the time-consuming and laborious defect of traditional methods used to construct infectious cDNA clones, but also avoids the toxicity of the Potyvirus special sequence to Escherichia coli, thus providing a useful cloning strategy for the construction of infectious cDNA clones for other viruses and laying down a foundation for the further investigation of SMV cross-family infection mechanisms.

The consumption of herbal medicine and related products (herbal products) have increased in South Korea. At the same time the quality, safety, and efficacy of herbal products is being raised. Currently, the herbal products are standardized and controlled according to the requirements of the Korean Pharmacopoeia, the National Institute of Health and the Ministry of Public Health and Social Affairs. The validation of herbal products and their medicinal component is important, since many of these herbal products are composed of two or more medicinal plants. However, there are no tools to support the validation process. Interest in deep learning has exploded over the past decade, for herbal medicine using algorithms to achieve herb recognition, symptom related target prediction, and drug repositioning have been reported. In this study, individual images of four herbs (Panax ginseng C.A. Meyer, Atractylodes macrocephala Koidz, Poria cocos Wolf, Glycyrrhiza uralensis Fischer), actually sold in the market, were achieved. Certain image preprocessing steps such as noise reduction and resize were formatted. After the features are optimized, we applied GoogLeNet_Inception v4 model for herb image recognition. Experimental results show that our method achieved test accuracy of 95%. However, there are two limitations in the current study. Firstly, due to the relatively small data collection (100 images), the training loss is much lower than validation loss which possess overfitting problem. Secondly, herbal products are mostly in a mixture, the applied method cannot be reliable to detect a single herb from a mixture. Thus, further large data collection and improved object detection is needed for better classification.

Background: Some plants have harmful effects on fungi and bacteria as well as other plants. Incorporating such plant into soil as green manure is effective in reducing population densities of soil pathogens. Methods and Results: Twenty-three species of green manure crops were cultivated after the harvest of 6-year-old ginseng and then incorporated into the soil at the flowering stage. The following year, the root rot ratio of 2-year-old ginseng and soil chemical properties were investigated. In the absence of green manure addition, the $NO_3$ content, electric conductivity (EC), and K content decreased by 95%, 79% and 65%, respectively. In the presence of green manure addition, $P_2O_5$ and $NO_3$ contents reduced by 41% and 25%, respectively. The "survived root ratio" of 2-year-old ginseng significantly increased by 56.2%, 47.5%, and 47.3%, in the Sorghum sudanense, Ricinus communis and Helianthus tuberosus treatment, respectively. In addition, there was a significant increase in the "survived root ratio" in the Secale cereale, Chrysanthemum morifolium, Atractylodes macrocephala, and Smallanthus sonchifolius treatments. The "survived root ratio" of ginseng showed a significant positive correlation with the soil pH and a negative correlation with the $NO_3$ contents, and EC. Conclusions: Cultivation of plant form the Chrysanthemum family as green manure, using mainly the rhizomes was effective for the control of root rot disease of ginseng.
